This dissertation focuses on the study of water-soluble calixarene derivatives as receptors for fluorescence probes and as chiral selectors. The synthesis of sulfonated calixarenes was first pursued and a method for the separation of sulfonated calixarenes (4), (6), (8) and the 4-hydroxylbenzene sulfonate monomer by capillary zone electrophoresis with direct UV absorbance detection is described. Fluorescence and absorption studies of water-soluble sulfonated calix(6)arene (SCX6) are reported. A red shift in the absorption spectrum and a change in the fluorescence spectrum of the calixarenes are observed upon an increase in pH from 2.0 to 13.0. The spectroscopic changes are attributed to …

This dissertation puts forth an original theory of cortical neural processing that is unique in its view of the interplay of chaotic and stable oscillatory neurodynamics and is meant to stimulate new ideas in artificial neural network modeling. Our theory is the first to suggest two new purposes for chaotic neurodynamics: (i) as a natural means of representing the uncertainty in the outcome of performed tasks, such as memory retrieval or classification, and (ii) as an automatic way of producing an economic representation of distributed information. Since before Spanish Contact, the Pech Indians have occupied a large portion of northeastern Honduras. Like other native American populations, they have suffered significant territorial reductions and cultural alterations at the hands of European colonists and modern ladino immigrants. Utilizing the methodologies of cultural geography, ethnohistory, and ethnogeography, the Pech, formerly known as the Paya, are scrutinized to illustrate the process by which indigenous peoples are reduced and incorporated into a developing national setting. Hepatitis A virus (HAV), the causative agent of food and water borne acute hepatitis worldwide, is very difficult and expensive to propagate in tissue culture. Large scale production of HAV proteins in E. coli by recombinant DNA techniques would be invaluable for antisera production, biochemical analyses and in developing sensitive diagnostic tests. Therefore, PCR amplified DNA fragments encoding the HAV capsid proteins were cloned individually into prokaryotic expression vectors for production of either recombinant fusion or non-fusion proteins. Cloning and expression of HAV capsid genes, which induced toxic effects in certain host E. coli, was tolerated better by protease-deficient host …

The tetraphosphine ligand, Et$\sb2$P(CH$\sb2)\sb2$P(Ph)CH$\sb2$P(Ph)(CH$\sb2)\sb2$PEt$\sb2 ,$ et,ph-P4, was designed to be a binucleating ligand for use in bimetallic hydroformylation catalysis. This ligand can be tailored for both electronic and steric factors at the internal and external phosphorus atoms. It is hoped that through careful tuning of this ligand, the rate and selectivity of our bimetallic catalyst can be optimized for various substrates. Modification of the original ligand may also lead us to a better understanding of the mechanism involved in the catalytic cycle. We apply stochastic techniques towards the solution of the two-dimensional Laplace's equation in boundary value problems encountered in the calculation of electrostatic potentials in electron lenses and deflectors. The justification of these techniques arises from an astonishingly simple but far-reaching principle, which has been known for a long time but has been rarely used: the potential at any point in the interior of a charge-free region can be calculated by performing random walks starting at this point and terminating at the boundary of the region--the potential is then the average of the potential boundary values (assumed known) over the random …

This research was conducted to obtain a better understanding of the amino acid needs of finishing pigs. Twelve experiments were conducted using the urea nitrogen method to determine the lysine requirement of gilts and barrows. Two experiments were conducted to determine the optimum ratio of total sulfur amino acids to lysine for finishing pigs (74 to 111 kg); and three experiments were conducted to evaluate the net energy content of low crude protein, crystalline amino acid supplemented diets for finishing pigs (70 to 117 kg). Three experiments are reported that attempt to further our knowledge of the preparation and production of rapid sequential aiming movements. Specifically, these experiments examined possible reasons why in sequential aiming responses, the second movement segment (MT2) is consistently performed more slowly than first and third movement segments (MT1 and MT3). In the first experiment, participants struck 1, 2, or 3 targets in sequence without the benefit of visual feedback and without time stress to determine if MT2 is slowed due to a visually based on-line trajectory-correction process. The results showed that MT2 was not slower than MT1 under these condition. …

This dissertation examines the relationship between technological change and spatial industrial restructuring through a case study of the 1840-1880 British ocean-going iron and steam shipbuilding industry. The study tests the hypothesis that a shipbuilding center's share of the national British shipbuilding market was associated with its ability to generate or rapidly adopt technological change. The study begins by establishing iron steamship technological changes introduced by British shipbuilders and the industry's attendant spatial restructuring It then develops two site-specific variables: industrial viability and innovative ability. This study utilized cross-sectional data obtained from the 1991 National Survey of Fishing, Hunting, and Wildlife-Associated Recreation to analyze the expenditures and consumption of nonconsumptive wildlife related recreation in the Lower Mississippi Valley Ecosystem area. In the process of selecting an appropriate model most consistent with individuals' consumption behavior associated with nonconsumptive wildlife related recreation, the tobit model and the double-hurdle model for both the primary nonresidential and residential expenditure models were evaluated. Based on the Lagrange multiplier test and the likelihood ratio test results, the double-hurdle model fit the data much better than the tobit model. The hypothesis that …

Alligator mississippiensis contains all of the components of the hepatic microsomal mixed function oxidase system. In general, basal and induced levels of P450 specific content, immunochemically detected abundance and examined activities are less than in mammals. In alligator at least two classes of P450s are inducible by xenobiotics: those induced by 3-methylcholanthrene (3MC) and those induced by phenobarbital (PB). PB and 3MC induce multiple P450 isoforms as detected by western blot and enzymatic analyses. The induced level of alkoxyphenoxazone O-dealkylation (AROD) was 10- to 100-fold lower in alligator than in rat, with the exception of phenylbenzyloxyphenoxazone O-dealkylation. In contrast to …

An investigation of the plasma nitriding mechanism under intensified conditions (average particle energy up to 1200 eV) was carried out using a triode configuration. The significance of the different physical processes taking place during intensified plasma nitriding, such as sputtering, implantation, defect generation, redeposition and diffusion were investigated experimentally, while theoretical predictions were obtained by using the TRIM (transport of ions in matter) code. A titanium-base alloy was selected as the substrate material. Sputtering yield in an inert atmosphere was found to exhibit three regimes as a function of incoming particle energy. Magnetization and de Haas - van Alphen (dHvA) measurements have been made on the high temperature superconductor Ba$\sb{0.6}$K$\sb{0.4}$BiO$\sb3$ and the heavy fermion CeB$\sb6$. First ever observations of the dHvA effect in Ba$\sb{0.6}$K$\sb{0.4}$BiO$\sb3$ are reported. Two dHvA spectrometers (pulsed field and field modulation) were used to measure two samples. Four frequencies are found in the field modulation data with the 11.6 kT orbit in good agreement with band structure calculations. The three lowest frequencies were measured with the pulsed field spectrometer. All dHvA measurements were made in the superconducting mixed state. These measurements indicate that Ba$\sb{0.6}$K$\sb{0.4}$BiO$\sb3$ has a Fermi surface. Magnetization …

There are numerous techniques available for fitting a surface to any supplied data set. The feature-based modeling technique takes advantage of the known, geometric shape of the data by deforming a model having this generic shape to approximate the data. The model is constructed as a rational B-spline surface with characteristic features superimposed on its definition. The first step in the fitting process is to align the model with a data set using the center of mass, principal axes and/or landmarks. Regulation of transcription most often occurs at the stage of initiation. RNA polymerase binding to the transcription start site, the promoter, is influenced by many nucleotide sequence elements. The predominant recognition sequences are those bound by the $\sigma$ subunit of RNA polymerase located at $-$10 and $-$35 relative to initiation site of most promoters. Another element involved in this regulation is intrinsic DNA curvature. In this study, the contribution of intrinsically curved DNA immediately upstream of the promoter to the interaction between Escherichia coli RNA polymerase and this DNA was examined. The main goal of the study was to provide the basic technical frame work for the use of native fish species as environmental sentinels for potential human exposure to genotoxic agents in general and to 1,2-dichloroethane (ethylene dichloride) in particular. First, LC/MS/ESI/ID and LC/MS/MS/ESI/ID methods for detection and quantification of S- (2-(N$\sp7$-guanyl)ethyl) glutathione DNA adduct from biological samples was developed. The methods were tested by exposing groups of rats with three dose levels of 1,2-dibromoethane. They allowed detection of $\sim$1 adduct in 10$\sp7$ bases. This study provides a description and analysis of the effects of a mathematics reform initiative on classrooms in two rural communities. It describes the sites, details interactions that take place in the classroom, and enumerates teachers' perceptions of the change. A mixed methodology was used to support the findings. Qualitative data were collected through classroom observations and interviews and quantitative data were gathered through questionnaires. Quantitative results indicated that teachers who participate in the training have the opportunity to learn more about new teaching strategies in mathematics and are likely to change their teaching practices in mathematics. In this doctoral research, we propose several novel protocols and topologies for the interconnection of massively parallel processors. These new technologies achieve considerable improvements in system performance and structure simplicity. Currently, synchronous protocols are used in optical TDM buses. The major disadvantage of a synchronous protocol is the waste of packet slots. To offset this inherent drawback of synchronous TDM, a pipelined asynchronous TDM optical bus is proposed. Field cage studies were conducted to measure the effects of late season beet armyworm, Spodoptera exigua (Hubner), infestations (0, 1, 3, and 6 egg masses per 5.1 row m) on defoliation, fruit damage, and yield of cotton. Light penetration through the canopy was significantly higher in infested plots. Although a trend for increased numbers of damaged fruiting forms with increases in egg mass density was observed, there were no significant differences between infested and control plots. There were no significant differences in the cumulative number of shed fruiting forms or yield between infested and control plots. Field tests were conducted …

The endemic goby Sicyopterus stimpsoni is unique among five species of Hawaiian freshwater fishes because it undergoes rapid metamorphosis of cranial structures during postlarval development. These fishes are amphidromous and return after a prolonged stay in the ocean to streams where they are confined to estuaries until completion of cranial restructuring. Within 48 hours after entering fresh water, head width increases significantly, while total length and head length remains unchanged. Weight decreases by approximately 15% during this period. Aerenchyma tissue consists of gas channels that provide a pathway for O$\sb2$ transport to waterlogged roots. Aerenchyma gas spaces are formed by cell separation or cell lysis during development. Light microscopy was used to determine how aerenchyma develops in Sagittaria lancifolia, a freshwater marsh species, when plants were grown in hydroponic solution. Petiole aerenchyma was formed by cell separation caused by differential cell expansion and division. In the root cortex, certain radial files of cells separated slightly from adjacent files and then lysed to form gas spaces. Exploration into the nature of ultrathin, surface-confined films capable of electronic conduction has been the primary purpose of this dissertation. Specifically, the confinement of pyrrole monomer to Au surfaces through organosulfur linkages has been investigated. The synthesis of these new $\omega$-(N-pyrrolyl)alkanethiols is described herein. Cyclic voltammetry was utilized in this work to polymerize the monolayers and characterize the resulting films. The Production Of Glucooligosaccharides By Leuconostoc Mesenteroides Atcc 13146 And Lipomyces Starkeyi Atcc 74054., Sun Kyun Yoo

Glucooligosaccharides of less than DP 10 were produced either by mixed cultures of Leuconostoc mesenteroides ATCC 13146 and L. starkeyi ATCC 74054, or by an acceptor reaction produced by Leuconostoc mesenteroides ATCC 13146. The yields of glucooligosaccharides produced by mixed culture and acceptor reaction reached up to 49 and 90% of theoretical yield. The composition of carbohydrates at the end of fermentation by mixed culture was 31.4% oligosaccharides, 17.6% dextran, 31.4% mannitol, 2.1% fructose, and 2.4% of sucrose. Encouraged by the Enlarged Homestead Act, higher than average rainfall, and various boosters, thousands of Americans homesteaded in Montana and the northern Great Plains during the 1910s. The agricultural economy boomed during World War I, but the post-war contraction during the 1920s, coupled with drought, caused serious economic problems for farmers. In response to the problems in the agricultural economy, a Land Utilization movement emerged, led by agricultural economists such as Lewis C. Gray. The research presented in this dissertation involves the construction of a near-infrared time-correlated single photon counting system which utilizes a mode-locked Ti:sapphire or pulsed diode laser as the excitation source and single photon avalanche diode as the photodetector and the subsequent study of two simple algorithms, the Maximum Likelihood Estimator (MLE) and the Rapid Lifetime Determination (RLD) methods, for fluorescence lifetime determination.
