Here, we present a protocol to isolate progenitor cells from mouse epididymal visceral adipose tissue and construct bulk RNA and assay for transposase-accessible chromatin with sequencing (ATAC-seq) libraries. We describe steps for adipose tissue collection, cell isolation, and cell staining and sorting. We then detail procedures for both ATAC-seq and RNA sequencing library construction. This protocol can also be applied to other tissues and cell types directly or with minor modifications. For complete details on the use and execution of this protocol, please refer to Liu et al. (2023).1

We continue our work on the study of spherically symmetric loop quantum gravity coupled to two spherically symmetric scalar fields, one which acts as a clock. As a consequence of the presence of the latter, we can define a true Hamiltonian for the theory. The spherically symmetric context allows to carry out precise detailed calculations. Here we study the theory for regions of large values of the radial coordinate. Community colleges have teaching faculty whose primary focus is educating their students and providing service to their institution. Community college teaching research has focused on innovation, students’ expectations, and the use of active learning techniques to foster student success. Research on blended courses has focused on instructional practices, learner engagement, and the benefits of blended learning for the community college student population. Translation is critical for development as transcription in the oocyte and early embryo is silenced. To illustrate the translational changes during meiosis and consecutive two mitoses of the oocyte and early embryo, we performed a genome-wide translatome analysis. Acquired data showed significant and uniform activation of key translational initiation and elongation axes specific to M-phases. Although global protein synthesis decreases in M-phases, translation initiation and elongation activity increases in a uniformly fluctuating manner, leading to qualitative changes in translation regulation via the mTOR1/4F/eEF2 axis. The use of environmental DNA (eDNA) as a sampling tool offers insights into the detection of invasive and/or rare aquatic species and enables biodiversity assessment without traditional sampling approaches, which are often labor-intensive. However, our understanding of the environmental factors that impact eDNA removal (i.e., how rapidly eDNA is removed from the water column by the combination of decay and physical removal) in flowing waters is limited. Quantitative polymerase chain reaction (qPCR) measurement of antibiotic resistance genes (ARGs) in untreated municipal wastewater may prove useful in combating the antimicrobial resistance crisis. However, harmonizing and optimizing qPCR-based workflows is essential to facilitate comparisons across studies, and includes achieving highly-effective ARG capture through efficient concentration and extraction procedures. Louisiana has high coastal wetland loss rates due to natural processes such as subsidence and anthropogenic activities such as construction of river levees and dams, pervasive alteration of surface hydrology by local industries such as oil and gas, and navigation. With the exception of the Atchafalaya River discharge area, most of Louisiana's marsh coastline is retreating and coastal marshes are degrading. In the inactive degrading delta regions, there exists a previously uncharacterized landform referred to colloquially as coastal ‘land bridge’ marshes. Increasing atmospheric carbon dioxide (CO2) concentrations have been linked to effects in a wide range of ecosystems and organisms, with negative effects of elevated CO2 documented for marine organisms. Less is known about the dynamics of CO2 in freshwaters, but the potential exists for freshwater organisms to be challenged by elevated CO2. In flowing freshwaters CO2 exhibits more variability than in lakes or the ocean, yet spatiotemporally extensive direct measures of CO2 in freshwater are rare. The United States Inland Creel and Angler Survey Catalog (CreelCat) contains a national compilation of angler and creel survey data collected by natural resource management agencies across the United States (including Washington, D.C. and Puerto Rico). These surveys are used to help inform the management of recreational fisheries, by collecting information about anglers including what they are catching and harvesting, the amount of effort they expend, their angling preferences, and demographic information. As of May 1, 2023, CreelCat houses over 14,729 surveys from 33 states, Puerto Rico, and Washington, D.C., comprising 235 data fields across 8 tables. These tables contain …

The global COVID-19 pandemic resulted in many jurisdictions implementing orders restricting the movements of people to inhibit virus transmission, with recreational angling often either not permitted or access to fisheries and/or related infrastructure being prevented. Following the lifting of restrictions, initial angler surveys and licence sales suggested increased participation and effort, and altered angler demographics, but with evidence remaining limited. Here, we overcome this evidence gap by identifying temporal changes in angling interest, licence sales, and angling effort in world regions by comparing data in the ‘pre-pandemic’ (up to and including 2019); ‘acute pandemic’ (2020) and ‘COVID-acclimated’ (2021) periods. Despite the intensifying interest in flash drought both within the United States and globally, moist tropical landscapes have largely escaped the attention of the flash drought community. Because these ecozones are acclimatized to receiving regular, near-daily precipitation, they are especially vulnerable to rapid-drying events. This is particularly true within the Caribbean Sea basin where numerous small islands lack the surface and groundwater resources to cope with swiftly developing drought conditions. Natural geochemical markers in the otolith of yellowfin tuna (Thunnus albacares) were used to establish nursery-specific signatures for investigating the origin of fish captured in the western Atlantic Ocean (WAO). Two classes of chemical markers (trace elements, stable isotopes) were used to first establish nursery-specific signatures of age-0 yellowfin tuna from four primary production zones in the Atlantic Ocean: Gulf of Mexico, Caribbean Sea, Cape Verde, and Gulf of Guinea. Human-induced nitrogen–phosphorus (N, P) imbalance in terrestrial ecosystems can lead to disproportionate N and P loading to aquatic ecosystems, subsequently shifting the elemental ratio in estuaries and coastal oceans and impacting both the structure and functioning of aquatic ecosystems. The N:P ratio of nutrient loading to the Gulf of Mexico from the Mississippi River Basin increased before the late 1980s driven by the enhanced usage of N fertilizer over P fertilizer, whereafter the N:P loading ratio started to decrease although the N:P ratio of fertilizer application did not exhibit a similar trend. Spotting people with dementia early is challenging, but important to identify people for trials of treatment and prevention. We used brain scans of people with Alzheimer’s disease, the commonest type of dementia, and applied an artificial intelligence method to spot people with Alzheimer’s disease. We used this to find people in the Healthy UK Biobank study who might have early Alzheimer’s disease. The people we found had subtle changes in their memory and thinking to suggest they may have early disease, and we also found they had high blood pressure and smoked for longer. Background: Perinatal outcomes vary for women with gestational diabetes mellitus (GDM). The precise factors beyond glycemic status that may refine GDM diagnosis remain unclear. We conducted a systematic review and meta-analysis of potential precision markers for GDM. Methods: Systematic literature searches were performed in PubMed and EMBASE from inception to March 2022 for studies comparing perinatal outcomes among women with GDM. We searched for precision markers in the following categories: maternal anthropometrics, clinical/sociocultural factors, non-glycemic biochemical markers, genetics/genomics or other -omics, and fetal biometry. Background: The greatest change in the treatment of people living with type 1 diabetes in the last decade has been the explosion of technology assisting in all aspects of diabetes therapy, from glucose monitoring to insulin delivery and decision making. Background: The objective was to test the efficacy of a scalable, virtually delivered, diabetes-tailored weight management program on glycemic control in adults with type 2 diabetes (T2D). Methods: This was a single arm, three-site clinical trial. Participants had baseline HbA1c between 7–11% and BMI between 27–50 kg/m2. Primary outcome was change in HbA1c at 24 weeks. Secondary outcomes were changes in body weight, waist circumference, the Diabetes Distress Scale (DDS), quality of life (IWQOL-L), and hunger (VAS). Phyllachora maydis is a fungal pathogen causing tar spot of corn (Zea mays L.), a new and emerging, yield-limiting disease in the United States. Since being first reported in Illinois and Indiana in 2015, P. maydis can now be found across much of the corn growing regions of the United States. Knowledge of the epidemiology of P. maydis is limited but could be useful in developing tar spot prediction tools. Tidal marsh insect communities are influenced by both salinity and aspects of vegetation texture (vegetation diversity, plant density, and architectural structure). These factors affect the abundances and intra- and interguild interactions of these insects and should result in broad-scale variation in the distribution and ecological functioning of tidal marsh insect communities along these gradients. Mosquitoes are a known public nuisance and can vector various diseases. Historically, New Orleans, LA, has long been acquainted with the burden of mosquito-borne diseases, such as malaria and yellow fever in the 20th century and West Nile virus in the 21st century. In the era of targeted therapy, there has been increasing concern about the development of oncology drugs based on the “more is better” paradigm, developed decades ago for chemotherapy. Recently, the US Food and Drug Administration (FDA) initiated Project Optimus to reform the dose optimization and dose selection paradigm in oncology drug development. To accommodate this paradigm shifting, we propose a dose-ranging approach to optimizing dose (DROID) for oncology trials with targeted drugs. Insect sepsis is a severe consequence that arises from the invasion of the hemocoel by symbionts of entomopathogenic nematodes and bacteria. In the present study, we unveiled the heightened virulence of the entomopathogenic nematode Steinernema feltiae and the entomopathogenic bacteria Xenorhabdus bovienii, which operate symbiotically, against the wax moth Galleria mellonella. Maximum mortality was observed at 25°C while the optimal infestation efficiency was 20 nematodes per host. After infestation, G. mellonella displayed rapid darkening and softening, accompanied by an escalated esterase activity at 9 h. The X. bovienii, released by S. feltiae, underwent substantial proliferation and discharged toxins that attacked …

Objective: The tarnished plant bug (TPB), Lygus lineolaris (Palisot de Beauvois) (Hemiptera: Miridae), is a pest damaging many cultivated crops in North America. Although partial transcriptome data are available for this pest, a genome assembly was not available for this species. This assembly of a high-quality chromosome-length genome of TPB is aimed to develop the genetic resources that can provide the foundation required for advancing research on this species. The ironwood tree (Casuarina equisetifolia, family Casuarinaceae), an indigenous agroforestry species in Guam, has been threatened by ironwood tree decline (IWTD) since 2002. Formation of bacterial ooze by the wilt pathogen from the Ralstonia solanacearum species complex and wetwood bacteria (primarily Klebsiella species) has been linked to IWTD. In addition, termite infestation of trees was statistically associated with IWTD. Termites are known carriers of a diverse microbiome. Therefore, we hypothesized that termites could be vectors of bacteria linked to IWTD. Evidence suggests that perceptual and action related features of concepts are grounded in the corresponding sensory-motor networks in the human brain. However, less is known about temporal features of event concepts (e.g., a lecture) and whether they are grounded in time perception networks. We examined this question by stimulating the right dorsolateral prefrontal cortex (rDLPFC)—a part of time perception network—using HD-tDCS and subsequently recording EEG while participants performed semantic and time perception tasks. Drawing upon relational turbulence theory and the experiencing life transitions model, this study examined communication and relationship qualities as married couples experienced work-family changes during the COVID-19 pandemic. Sixty-four American married, different-sex partners affected by job-related changes due to COVID-19 reported their relationship qualities and their own and their spouse’s engagement in transition processing communication weekly for 10 weeks. Hypotheses addressed associations between relationship qualities and communication experiences both averaged across the 10 weeks and indexed by within-week deviations from those averages, controlling for the previous week’s reports. The assumption that stress negatively impacts marital relationships is widely accepted; however, the majority of research has focused on marital satisfaction as the outcome of interest. Relational turbulence is a quality of romantic associations on par with—but distinct from—satisfaction, in which partners conceptualize their relationship as chaotic or tumultuous. This paper draws on relational turbulence theory (RTT) and stress spillover research to propose that day-to-day stress corresponds with perceptions of relational uncertainty and interdependence, which contributes to increases in relational turbulence. Extreme weather and climate events pose significant risks to rural water and wastewater systems. We examine the vulnerability of the water sector to weather and climate extremes in rural, predominantly Indigenous and underserved coastal areas and analyze how networks support resilience. Drawing on the analysis of 39 web-based questionnaire responses and 19 interviews with rural water and wastewater managers and service providers in southern Louisiana and western Alaska, this article reports a range of interrelated historical, environmental, and social factors that influence vulnerability to extreme weather events. Mud dauber wasps construct soil nests to protect their offspring from predators, extreme temperatures, and rainwater. The mechanical and thermal properties of these nests are important for the reproductive success of mud daubers. The previous study showed that the high densities and strengths of mud dauber nests were due to the repetitive tapping and atmospheric drying used by mud daubers during nest construction.
