Because of the importance of potassium efflux in inflammasome activation, we investigated the role of the two-pore potassium (K2P) channel TREK-1 in macrophage inflammasome activity. Using primary alveolar macrophages (AMs) and bone marrow-derived macrophages (BMDMs) from wild-type (wt) and TREK-1–/– mice, we measured responses to inflammasome priming [using lipopolysaccharide (LPS)] and activation (LPS þ ATP). We measured IL-1b, caspase-1, and NLRP3 via ELISA and Western blot. A membrane-permeable potassium indicator was used to measure potassium efflux during ATP exposure, and a fluorescence-based assay was used to assess changes in membrane potential. Inflammasome activation induced by LPS þ ATP increased IL-1b …

As the horticulture consumer base transitions to younger generations, educational outreach collaborations between Extension and industry could be a mutually beneficial way to serve an increasingly diverse customer audience. A recent study investigated drivers of, and opportunities for, increased participation in plant-related activities using an online survey of 2,134 respondents. Both participants and non-participants in plant-related activities were surveyed. Some of the commonly cited reasons for non-participation were a lack of knowledge and overall success, illustrating an opportunity for education to be beneficial. While many plant retailers may not have the personnel or resources to engage widely in education, collaborations …

Humans have profoundly impacted the distribution of plant and animal species over thousands of years. The most direct example of these effects is human-mediated movement of individuals, either through translocation of individuals within their range or through the introduction of species to new habitats. While human involvement may be suspected in species with obvious range disjunctions, it can be difficult to detect natural versus human-mediated dispersal events for populations at the edge of a species' range, and this uncertainty muddles how we understand the evolutionary history of populations and broad biogeographical patterns. Studies combining genetic data with archaeological, linguistic and …

Identification Of A Chondrocyte-Specific Enhancer In The Hoxc8 Gene, Stephania A. Cormier, Claudia Kappen

Division of Basic Science Faculty Publications

Hox genes encode transcription factors whose roles in patterning animal body plans during embryonic development are well-documented. Multiple studies demonstrate that Hox genes continue to act in adult cells, in normal differentiation, in regenerative processes, and, with abnormal expression, in diverse types of cancers. However, surprisingly little is known about the regulatory mechanisms that govern Hox gene expression in specific cell types, as they differentiate during late embryonic development, and in the adult organism. The murine Hoxc8 gene determines the identity of multiple skeletal elements in the lower thoracic and lumbar region and continues to play a role in the …

Advancing ecological restoration assessments requires a more detailed consideration of species interactions and ecosystem processes. Most restoration projects rely on a few metrics not always directly linked with ecological theory. Here, we used Odum's theory of ecosystem development to assess and compare the ecosystem structure and services of created marshes (4–6 years old) with preexisting, reference marshes in a brackish water region of the Mississippi River Delta. We built ecosystem models for created and reference marshes that integrated large datasets of stomach contents, stable isotopes, and taxa abundances. Despite strong resemblance in community structure, created marshes were at an earlier …

Blue supergiants are the brightest stars in their host galaxies, and yet their evolutionary status has been a long-standing problem in stellar astrophysics. In this pioneering work, we present a large sample of 59 early B-type supergiants in the Large Magellanic Cloud with newly derived stellar parameters and identify the signatures of stars born from binary mergers among them. We simulate novel 1D merger models of binaries consisting of post main-sequence giants with helium-rich cores (primaries) and main-sequence companions (secondaries), and consider the effects of interaction of the secondary with the core of the primary along with the mixing induced …

Baryon number conservation is not guaranteed by any fundamental symmetry within the standard model, and therefore has been a subject of experimental and theoretical scrutiny for decades. So far, no evidence for baryon number violation has been observed. Large underground detectors have long been used for both neutrino detection and searches for baryon number violating processes. The next generation of large neutrino detectors will seek to improve upon the limits set by past and current experiments and will cover a range of lifetimes predicted by several Grand Unified Theories. In this White Paper, we summarize theoretical motivations and experimental aspects …

Observations of linear polarization in the 2-8 keV energy range with the Imaging X-ray Polarimetry Explorer (IXPE) explore the magnetic field geometry and dynamics of the regions generating nonthermal radiation in relativistic jets of blazars. These jets, particularly in blazars whose spectral energy distribution peaks at X-ray energies, emit X-rays via synchrotron radiation from high-energy particles within the jet. IXPE observations of the X-ray-selected BL Lac-type blazar 1ES 1959+650 on 2022 May 3-4 showed a significant linear polarization degree of Πx = 8.0% ± 2.3% at an electric-vector position angle ψ x = 123° ± 8°. However, on 2022 June …

We complete the analysis of planetary candidates found by the KMT AnomalyFinder for the 2017 prime fields that cover ∼13 deg2. We report three unambiguous planets: OGLE-2017-BLG-0640, OGLE-2017-BLG-1275, and OGLE-2017-BLG-1237. The first two of these were not previously identified, while the last was not previously published due to technical complications induced by a nearby variable. We further report that a fourth anomalous event, the previously recognized OGLE-2017-BLG-1777, is very likely to be planetary, although its light curve requires unusually complex modeling because the lens and source both have orbiting companions. One of the three unambiguous planets, OGLE-2017-BLG-1275, is …

Oxygenic photosynthetic organisms use Photosystem II (PSII) to oxidize water and reduce plastoquinone. Here, we review the mechanisms by which PSII is assembled and turned over in the model green alga Chlamydomonas reinhardtii. This species has been used to make key discoveries in PSII research due to its metabolic flexibility and amenability to genetic approaches. PSII subunits originate from both nuclear and chloroplastic gene products in Chlamydomonas. Nuclear-encoded PSII subunits are transported into the chloroplast and chloroplast-encoded PSII subunits are translated by a coordinated mechanism. Active PSII dimers are built from discrete reaction center complexes in a process facilitated by …

Genomic selection (GS) is revolutionizing plant breeding. However, its practical implementation is still challenging, since there are many factors that affect its accuracy. For this reason, this research explores data augmentation with the goal of improving its accuracy. Deep neural networks with data augmentation (DA) generate synthetic data from the original training set to increase the training set and to improve the prediction performance of any statistical or machine learning algorithm. There is much empirical evidence of their success in many computer vision applications. Due to this, DA was explored in the context of GS using 14 real datasets. We …

We introduce multilayer structures with the phase-change material Ge2Sb2Te5 (GST) for use as broadband switchable absorbers in the infrared wavelength range. We optimize both the material composition and the layer thicknesses of the multilayer structures, in order to maximize the difference between the absorption for GST in its crystalline phase and the absorption for GST in its amorphous phase in the wavelength range of interest. We show that in the optimized structures near perfect absorption can be switched to very low absorption in a broad wavelength range by switching GST from its crystalline to its amorphous phase. In this work, we explore a simplified model based on both analytical and computational methods for the study of film-boiling droplet motion on microscale ratchets. We consider a specific ratchet design with the length periods and depth of ratchets much smaller than the size of the droplet. We conclude based on our modeling that for the ratchet configuration considered in this paper, the conduction within the vapor film is the dominant means of heat transfer in comparison with convection and radiation. Furthermore, we demonstrate a more manageable two-dimensional model in which analytical approaches coupled with computational approaches yield reasonably accurate …

Reclaimed asphalt pavement (RAP) has been utilized as a potential partial substitute for virgin asphalt binder in asphalt mixtures. However, a primary concern with increasing RAP content in asphalt mixtures is the cracking potential, attributed to the aged RAP asphalt binder (RAP-binder). To address this, the use of petroleum-based and bio-derived recycling agents (RAs) in enhancing the cracking resistance of high-RAP asphalt mixtures has been explored. The objective of this study is to ascertain the effectiveness of six RAs in mitigating cracking in high-RAP asphalt mixtures. The RAs considered include petroleum-crude-oil-derived aromatic oil, soy oil, and four types of tall-oil-derived …

The effectiveness of regional storm surge reduction strategies and tank-level structural mitigation measures in reducing the failure probability of aboveground storage tanks (ASTs) were studied. Given past failures during flood and hurricane events, several studies have developed fragility models for ASTs. However, the suitability of these fragility models for different hurricane hazard scenarios is unknown. Furthermore, to combat climate change and sea level rise, several regional storm surge reduction strategies are being proposed. However, the effectiveness of these strategies in improving the safety of ASTs is also unknown. So, herein, a framework was proposed that facilitates assessing the suitability of …

The present research is primarily focused on investigating the characteristics of environmentally persistent free radicals (EPFRs) generated from commonly recognized aromatic precursors, namely, 1,2-dichlorobenzene (DCB) and 2-monochlorophenol (MCP), within controlled laboratory conditions at a temperature of 230 °C, termed as DCB230 and MCP230 EPFRs, respectively. An intriguing observation has emerged during the creation of EPFRs from MCP and DCB utilizing a catalyst 5% CuO/SiO2, which was prepared through various methods. A previously proposed mechanism, advanced by Dellinger and colleagues (a conventional model), postulated a positive correlation between the degree of hydroxylation on the catalyst’s surface (higher hydroxylated, HH and less …
