It has been known that the transverse orientation of cortical microtubules (MTs) along the elongation axis is essential for normal cell morphogenesis, but whether cortical MTs are essential for normal cell wall synthesis is still not clear. In the present study, we have investigated whether cortical MTs affect cell wall synthesis by direct alteration of the cortical MT organization in Arabidopsis thaliana. Disruption of the cortical MT organization by expression of an excess amount of green fluorescent protein-tagged α-tubulin 6 (GFP-TUA6) in transgenic Arabidopsis plants was found to cause a marked reduction in cell wall thickness and a decrease in …

Perceptions of interpersonal dominance and affiliation have been extensively examined throughout past research. In the current study, the purpose was to fill in the some of the gaps of existing research well, specifically the gap created by current confusion in the literature regarding the effects of sex, status, and emotional display on ratings of dominance and affiliation. Also, interactions between the primary variables of interest (sex, status, and emotional display) were observed. Stock assessment methods that quantify the status of fishery resources are critical to effective fisheries management. There is a need for stock assessment methods applicable for management of tropical species based on limited, un-aged catch data. I applied four stock assessment approaches to situation with limited life-history information and with short-term, un-aged catch data. The four approaches are: life-history invariants, length-based catch analysis, individual-based modeling, and surplus production modeling. This study reports a characterization of the iron-sulfur clusters of the MoFe protein expressed by the nifBnifZ deletion strain of A. vinelandii. VT-MCD spectroscopic and magnetization analysis of as-isolated ΔnifBΔnifZ MoFe protein were undertaken. VT-MCD spectra of as-isolated ΔnifBΔnifZ MoFe protein contains bands remarkably similar to those seen in as-isolated ΔnifH MoFe protein, assigned to arise from an [4Fe-4S]+ cluster. The low-temperature magnetization curve of as-isolated ΔnifBΔnifZ MoFe protein reveals the MCD spectra bands stem from an S = ½ spin system with diamagnetic contributions of some other species. Coastal forests in Louisiana are in decline due to natural and human caused changes in the hydrology of the region. Baldcypress and water tupelo trees have been further stressed by caterpillar herbivory in recent decades. Regeneration of water tupelo is crucial for cavity creation for nesting habitat for secondary cavity nesting bird populations. Insectivorous birds have been shown to decrease insect-caused leaf damage on trees in other ecosystems. Two experiments examining effects of insectivorous birds on tree growth were conducted in degraded areas of the Maurepas Swamp in southeastern Louisiana. Polyelectrolyte multilayers (PEMs) have been used as stationary phases for various chiral and achiral separations in open-tubular electrochromatagraphy (OT-CEC). However, the physical characteristics of PEMs are not well understood. The production of PEM coatings involves flowing alternating rinses of positive and negative charged polyelectrolytes onto a surface. Although PEMs are typically deposited on fused silica capillaries, there is growing interest to use this approach in conjunction with microchip devices to enhance separation characteristics. Impacts red imported fire ants (RIFA) exert on native faunal communities were monitored in two pine-dominated ecosystems in Louisiana. After suppression of established RIFA populations with Amdro®, cotton mice (Peromyscus gossypinus), herpetofaunal, ground-dwelling invertebrate, Lycosidae, and non-target ant communities were compared between untreated-control and treated plots with respect to possible ecological impacts of RIFA on these communities. Efficacy of Amdro® (A. I. 0.7% hydramethylnon) was tested at Alexander State Forest and Sandy Hollow WMA, and was found to be effective at both sites for 99-42.3% and 97-48%, respectively, suppression of RIFA on treated plots, for three to seven months, with …

The ultimate goal of this research is to characterize data from the laboratory, pilot, and industrial scale rice mills. Pilot and laboratory scale data are presented in this research. Two long grain rice cultivars were milled with two different scale mills. Cheniere and Cypress were milled with a McGill No. 2 mill and a pilot scale mill (Satake). Both material streams, rice kernels and bran, were collected and weighed. Measurements of Degree of milling, transparency, and whiteness were made with a milling meter (Satake). The purpose of this study was to investigate the effects of Typha latifolia and Phragmites communis root matter in degradation of sediments contaminated with aged 1,2,3,4- Tetrachlorobenzene (1,2,3,4-TeCB). In the present study, complete reductive dechlorination of 1,2,3,4-TeCB was observed in root matter systems, contrasting sediment with no roots present. Microcosms amended with Phragmites communis roots achieved the greatest first-order reaction rate constant over a 4-week period (0.113 d-1, half-life time of 6.13 days). Recently, much evidence has been brought forth into the scientific community supporting the idea that RNA Polymerase III transcribed regions of DNA may serve as chromosomal landmarks for silencing. Transfer RNA genes are known to involve themselves in several extra-transcriptional functions within the chromosome, including the pausing of replication forks, Ty element integration, tRNA position effects (repression of neighboring genes), acting as a barrier to the spread of heterochromatin, and over-riding nuclosome positioning sequences. The coastal wetlands play a central role in maintaining productivity of the Gulf of Mexico while protecting coastal regions during severe storms. To help offset the loss of coastal marsh, there is growing interest in the beneficial use of dredge spoil for reconstruction of eroded areas. Hydraulic dredges are routinely employed to maintain navigation and the unconsolidated sediments can be pumped considerable distances to create gently sloping wetland areas. Previous studies have found the spaced-retrieval memory intervention technique to be successful in training people with probable Alzheimer’s disease (AD) to learn new, simple associations. In the present study, we expanded on these previous findings by training eight participants with probable AD to learn the names and pictures of countries via spaced retrieval. We demonstrated the flexibility of the technique, gave insight as to the effects of distracters on the spaced-retrieval training performance, and demonstrated the memorial effects of adding pictorial support to the text.

This research examines the contemporary practice of midwifery in Louisiana, a state that very early on had progressive legislation, yet remains a tough place for a midwife to practice. What, then, are the social forces that affect the ability to practice midwifery in Louisiana? I try to answer that question by examining the narratives of midwives and their clients, and evaluating the options these women have access to in this state. The narratives provide opportunities to observe the authoritarian knowledge of biomedicine in our society, and apply Foucault’s theory of power/knowledge. There are numerous ways to estimate postmortem interval (PMI), or time since death, including body temperature, rigor mortis, insect activity, and decomposition. Individually, many of these indicators are prone to inaccuracy due to the influence of the external environment upon them. This study proposed that in addition to or in conjunction with these and other indicators, certain enzymes could be used to accurately determine PMI, namely aspartate aminotransferase (AST), alcohol dehydrogenase (ADH), creatine kinase (CK), and lactate dehydrogenase (LDH). Static Low Density Media filters have been used over the past two decades for nitrification and solids removal in high-density recirculating aquaculture systems. The SLDM filters are submerged biofilters which contain a plastic media with a density less than that of water. The media beds remain packed, or static, during operation except for periodic expansion of the bead bed during a backwash. To begin to understand how nektonic species and communities respond to environmental variables before habitat modification, I examined patterns of habitat use by fishes and decapod crustaceans in a seemingly pristine mesohaline system that drains into Bay Batiste, southeastern Louisiana. Extracts of adult Haemonchus contortus were purified and used as a vaccine against the blood feeding parasite in goats as previously studied in sheep. The proteins used were H11 and H-gal-GP, hidden gut antigens from the microvillar membrane of the gut of the worm and combined with Quil A as adjuvant, then administered to a group of goats kept on concrete then artificially infected with H. contortus. Agrobacterium tumefaciens-mediated transformation is generally used for genetic transformation of higher plants. Several experimental factors important for the increase of beta-glucuronidase (GUS) reporter gene activity were evaluated in this study using leaf disks of tobacco (Nicotiana tabacum L. cv. Xanthi). We found that co-cultivation temperature at 20C is the most critical factor to obtain the reproducible enhancement of GUS activity. pCAMBIA 1305.01 resulted in higher GUS activity than the other two pCAMBIA vectors 1301 and 1305.02. Anoxia Tolerance, Anaerobic Metabolism, And The Lack Of A Mitochondrial Permeability Transition In The Ghost Shrimp, Lepidophthalmus Louisianensis, Schmitt, 1935, Jeremy Dale Holman

The ghost shrimp, Lepidophthalmus louisianensis, burrows up to meters deep in oxygen-limited marine sediments along the Gulf coast. During low tides these animals are subjected to extended periods of anoxia. The main objective of this study was to assess survival under anoxia and evaluate the physiological mechanisms that underlie the anoxia tolerance of this species. I observed large specimens of L. louisianensis (>2g) having an LT50 of 64 h under anoxia at 25º C. Smaller specimens (<1g) have a significantly higher LT50 of 113 h under identical conditions (P<0.0001). I measured whole body lactate levels in shrimp exposed to anoxia for up to 72 h, and recorded significant accumulation of this anaerobic end product (ANOVA, P<0.001). I also measured adenylates and arginine phosphate in shrimp exposed to anoxia for up to 48 h, and after a 24-h recovery period. Adenylates were not significantly altered during the anoxia regime, and reductions in arginine phosphate occurred after 12 and 24 h, but returned to normoxic values during recovery (ANOVA, P<0.001). While reserves of arginine phosphate are used to some extent to buffer losses in ATP, substantial contribution to the maintenance of energetic status comes from the high rate of anaerobic glycolysis. Rapid growth of warm-season grasses such as bermudagrass (Cynodon dactylon) and bahiagrass (Paspalum notatum) is associated with a decline in their nutritional value. This study was initiated to provide production and composition data with different cultivars of bermudagrass (common, Russell, Jiggs) and bahiagrass (Tifton-9, Pensacola, Argentina). The Edwards Aquifer in central Texas is one of the largest carbonate aquifers in the United States, supplying nearly two million people with water. The role of microorganisms in the development of deep carbonate aquifers has not been thoroughly investigated. The Edwards Aquifer is composed of a freshwater zone and a sulfidic, saline water zone. Down-well video surveys show an abundance of white filamentous and planktonic biomass floating in the water column, indicating that the saline water may serve as a habitat for microbes and provide a unique opportunity to investigate microbially enhanced deep subsurface karstification. Characterizing Salmonella Fecal Shedding Among Racehorses In Louisiana, Anna Marie Chapman

Salmonella is an important intestinal pathogen in horses capable of infecting populations without demonstrating clinical illness. This study was performed to determine the prevalence of Salmonella fecal shedding among racehorses in Louisiana. Three serial fecal samples were collected from 429 Thoroughbred horses housed at four racetracks. Feces were tested for Salmonella by microbiologic culture with selective primary enrichment and delayed secondary enrichment (DSE). Samples were also evaluated for the presence of Salmonella by polymerase chain reaction (PCR) using genus-specific oligonucleotide primers. Neural network architectures such as backpropagation networks, perceptrons or generalized Hopfield networks can handle complex inputs but they require a large amount of time and resources for the training process. This thesis investigates instantaneously trained feedforward neural networks that can handle complex and quaternion inputs. The performance of the basic algorithm has been analyzed and shown how it provides a plausible model of human perception and understanding of images. The motivation for studying quaternion inputs is their use in representing spatial rotations that find applications in computer graphics, robotics, global navigation, computer vision and the spatial orientation of instruments.
