The impact (i.e., defoliation and mortality) associated with large-scale natural disturbance events is capable of significantly altering ecosystem structure (e.g., biomass) and function (e.g., productivity) in mangrove wetlands. These pulsing events provide an opportunity to assess ecosystem resilience (i.e., recovery to pre-disturbance conditions) by evaluating the immediate and long-term ecosystem response trajectory (i.e., disturbance magnitude, recovery time). While silicon (Si) fertilization is widely practiced in paddy rice production, the establishment of critical soil Si levels has remained understudied. This study was undertaken to: 1) determine the critical soil Si level for rice production in Louisiana using different extraction procedures, and 2) document the relationship between plant-available Si and select essential plant nutrients in soil and their uptake by rice. Field trials were established at 12 sites across Louisiana from 2013 to 2015. Si was applied as silicate slag (SiO3, 14% Si) at the rates of 0, 1, 2, 4, 6, and 8 Mg ha-1. Agricultural lime was …

Strobilurin fungicide resistance threatens rice production in southwestern Louisiana. Strobilurins are the often relied upon method of managing sheath blight development due to the lack of strong genetic resistance to this disease in the currently available rice germplasm. However, isolates of Rhizoctonia solani AG 1-IA causing sheath blight were reported resistant to azoxystrobin near Mowata, LA in 2011. To determine the geographic distribution and severity of azoxystrobin resistance, 40 commercial rice fields in southwestern Louisiana were sampled for isolates of R. solani. Tocopherol conjugates are disclosed that are useful as antioxidants or as pro-drug delivery vehicles. The conjugates are amphiphilic, and tend to localize at the interface between water domains and lipid domains. A prototype embodiment disclosed is an alpha tocopherol-linker-carnosine conjugate that has been designated VECAR.

Objective: We have conducted a study to assess the role of environment on the burden of maternal morbidities and mortalities among women using an external exposome approach for the purpose of developing targeted public health interventions to decrease disparities. Methods: We identified counties in the 48 contiguous USA where observed low birthweight (LBW) rates were higher than expected during a five-year study period. © 2015 Elsevier Inc. Colorimetric protein assays, such as the Coomassie blue G-250 dye-binding (Bradford) and bicinchoninic acid (BCA) assays, are commonly used to quantify protein concentration. The accuracy of these assays depends on the amino acid composition. Because of the extensive use of reductive methylation in the study of proteins and the importance of biological methylation, it is necessary to evaluate the impact of lysyl methylation on the Bradford and BCA assays. Future shifts in phytoplankton composition and productivity are anticipated given that continuing changes are expected in environmental conditions such as temperature, the partial pressure of CO2 (pCO2) and light climate, all of which regulate phytoplankton communities and their physiology through bottom-up control. Culture experiments revealed that future (elevated) pCO2 had no effect on Thalassiosira weissflogii in the absence of environmental stressors, whereas growth rates drastically decreased under future pCO2 when cells were grown under light and temperature stress. Reduction in growth rates and a smaller decline in cellular photosynthesis under high pCO2 were associated with 2- to 3-fold increases in …

The present invention describes a method for identifying one or more of a plurality of sequences differing by one or more single base changes, insertions, deletions, or translocations in a plurality of target nucleotide sequences. The method includes a ligation phase, a capture phase, and a detection phase. The ligation phase utilizes a ligation detection reaction between one oligonucleotide probe, which has a target sequence-specific portion and an addressable array-specific portion, and a second oligonucleotide probe, having a target sequence-specific portion and a detectable label. Twenty-six imprinted genes were quantified in bovine in vivo produced oocytes and embryos using RNA-seq. Eighteen were detectable and their transcriptional patterns were: largely decreased (MEST and PLAGL1); first decreased and then increased (CDKN1C and IGF2R); peaked at a specific stage (PHLDA2, SGCE, PEG10, PEG3, GNAS, MEG3, DGAT1, ASCL2, NNAT, and NAP1L5); or constantly low (DIRAS3, IGF2, H19 and RTL1). These patterns reflect mRNAs that are primarily degraded, important at a specific stage, or only required at low quantities. The mRNAs for several genes were surprisingly abundant. Although aberrant DNA methylation within imprinted domains has been reported in a variety of neoplastic diseases, it remains largely uncharacterized in the context of carcinogenesis. In this study, we induced T-cell lymphoma in mice by employing a breeding scheme involving mouse strains, LSL-KrasG12D and MMTV-Cre. We then systematically surveyed imprinted domains for DNA methylation changes during tumor progression using combined bisulfite restriction analysis and NGS-based bisulfite sequencing. We detected hyper- or hypo-methylation at the imprinting control regions (ICRs) of the Dlk1, Peg10, Peg3, Grb10, and Gnas domains. These DNA methylation changes at ICRs …

Peg3 is an imprinted gene that is predicted to encode a DNA-binding zinc finger protein. This was previously demonstrated through Chromatin ImmunoPrecipitation-based Sequencing experiments. One explanation for why invasive species are successful is that they escape natural enemies from their native range or experience lower attack from natural enemies in the introduced range relative to native species (i.e., the enemy-release hypothesis). However, little is known about how invasive plants interact with co-introduced herbivores or natural enemies of the introduced herbivores. Knowing how climate change affects the population dynamics of insect pests is critical for the future of integrated pest management. Rising winter temperatures from global warming can drive increases in outbreaks of some agricultural pests. In contrast, here we propose an alternative hypothesis that both extremely cold and warm winters can mismatch the timing between the eclosion of overwintering pests and the flowering of key host plants. As host plants normally need higher effective cumulative temperatures for flowering than insects need for eclosion, changes in flowering time will be less dramatic than changes in eclosion time, leading to a mismatch …

Spotted fever group (SFG) rickettsial species are obligate intracellular tick-borne pathogens that are responsible for important human diseases. Previous reports have demonstrated the feasibility of using recombinant surface cell antigen Sca5/OmpB to elicit protective immunity against homologous challenges using murine models of Mediterranean spotted fever and Rocky Mountain spotted fever. In addition, the feasibility of generating cross-protective immunity against related rickettsial species has also been established, but the molecular basis for these phenomena was not explored. Avian diversity in fragmented Amazonian landscapes depends on a balance between extinction and colonization in cleared and disturbed areas. Regenerating forest facilitates bird dispersal within degraded Amazonian landscapes and may tip the balance in favor of persistence in habitat patches. Determining the response of Amazonian birds to fragmentation may be hindered because many species use adjacent second growth matrices thereby limiting the applicability of island biogeography to predict species loss; alternatively, a countryside biogeographic framework to evaluate the value of regenerating forest may be more appropriate. Combustion processes generate different types of particulate matter (PM) that can have deleterious effects on the pulmonary and cardiovascular systems. Environmentally persistent free radicals (EPFRs) represent a type of particulate matter that is generated after combustion of environmental wastes in the presence of redox-active metals and aromatic hydrocarbons. Cytochromes P450 (P450/CYP) are membrane-bound enzymes that are essential for the phase I metabolism of most lipophilic xenobiotics. The EPFR formed by chemisorption of 2-monochlorophenol to silica containing 5% copper oxide (MCP230) has been shown to generally inhibit the activities of different forms of P450s without affecting those of cytochrome P450 reductase …

Colorimetric and fluorometric methods are disclosed for the rapid, accurate, selective, and inexpensive detection of homocysteine, or of homocysteine and cysteine, or of cysteine. The methods may be employed with materials that are readily available commercially. The novel methods are selective for homocysteine, for cysteine, or for total homocysteine and cysteine, and do not cross-react substantially with chemically-related species such as glutathione. The homocysteine-selective method does not have substantial cross-reactivity to the very closely related species cysteine. The Military Macaw (Ara militaris) and the Great Green Macaw (A. ambiguus) are species whose close relationship is reflected in their morphological similarity as well as their geographic ranges. Military Macaws have a disjunct distribution, found in Mexico as well as several areas in South America, while Great Green Macaws have two or more disjunct populations from Honduras to eastern Ecuador. We used mitochondrial sequence data to examine the phylogenetic relationships between these two species, and also among representative samples across their ranges. Background: Protein-protein interactions (PPIs) mediate the vast majority of biological processes, therefore, significant efforts have been directed to investigate PPIs to fully comprehend cellular functions. Predicting complex structures is critical to reveal molecular mechanisms by which proteins operate. Despite recent advances in the development of new methods to model macromolecular assemblies, most current methodologies are designed to work with experimentally determined protein structures. However, because only computer-generated models are available for a large number of proteins in a given genome, computational tools should tolerate structural inaccuracies in order to perform the genome-wide modeling of PPIs. …

Failure to close the neural tube results in birth defects, with severity ranging from spina bifida to lethal anencephaly. Few genetic risk factors for neural tube defects are known in humans, highlighting the critical role of environmental risk factors, such as maternal diabetes. Yet, it is not well understood how altered maternal metabolism interferes with embryonic development, and with neurulation in particular. We present evidence from two independent mouse models of diabetic pregnancy that identifies impaired migration of nascent mesodermal cells in the primitive streak as the morphogenetic basis underlying the pathogenesis of neural tube defects. © 2015 by The American Society for Biochemistry and Molecular Biology, Inc. Secretion of effector proteins into the eukaryotic host cell is required for Chlamydia trachomatis virulence. In the infection process, Scc1 and Scc4, two chaperones of the type III secretion (T3S) system, facilitate secretion of the important effector and plug protein, CopN, but little is known about the details of this event. Here we use biochemistry, mass spectrometry, nuclear magnetic resonance spectroscopy, and genetic analyses to characterize this trimolecular event. We find that Scc4 complexes with Scc1 and CopN in situ at the late developmental cycle of C. trachomatis. …
