© 2019 American Society for Clinical Investigation. MAPK signaling consists of an array of successively acting kinases. ERK1 and -2 (ERK1/2) are major components of the greater MAPK cascade that transduce growth factor signaling at the cell membrane. Here, we investigated ERK1/2 signaling in skeletal muscle homeostasis and disease. Using mouse genetics, we observed that the muscle-specifc expression of a constitutively active MEK1 mutant promotes greater ERK1/2 signaling that mediates fber-type switching to a slow, oxidative phenotype with type I myosin heavy chain expression. Herein, we report the synthesis and characterization of a nanosensor developed from a group of uniform materials based on organic salts (GUMBOS). This sensor is composed of three ionic species, namely, fluorescein, rhodamine B, and tetradecyltrihexyl phosphonium (P-66614) ions. Nanoparticles prepared from this three-component GUMBOS, termed nanoGUMBOS, exhibited increased fluorescence at two wavelengths with increases in pH values. The ratio of fluorescence emission corresponding to FL and RhB components of these nanoGUMBOS also changed with pH, allowing ratiometric analysis of pH through fluorescence measurements. An isothiocyanato-functionalized phthalocyanine (Pc) was synthesized in good yield from the corresponding amine-substituted Pc. This Pc reacted with ethanolamine, biotin hydrazine, and biotin diethylamine under mild conditions (room temperature in DMF or DMSO in the presence of TEA) to produce the corresponding thiourea products in 60-75% yields. All Pcs showed intense Q absorptions in DMF around 677 nm, emissions centered at 683 nm, and fluorescence quantum yields in the range 0.18-0.27. The Pcs were phototoxic to human carcinoma HEp2 cells (IC50 similar to 7 at 1.5 J/cm(2)) and localized in multiple organelles, including the lysosomes, Golgi and ER. One biotin-Pc …

© Zhang et al. Mitochondria change their shape through fusion and fission in order to adapt to their metabolic milieu. Mitofusin-2 (MFN2) is a key regulatory protein in this process, mediating mitochondrial fusion and interaction with endoplasmic reticulum. Targeted deletion of Mfn2 in oocytes resulted in mitochondrial dysfunction and female subfertility associated with impaired oocyte maturation and follicle development. Oocytes lacking MFN2 showed shortened telomeres and increased apoptosis, resulting in compromised oocyte quality and accelerated follicular depletion, consistent with a reproductive aging phenotype.

Dosage compensation of the mammalian X chromosome (X) was proposed by Susumu Ohno as a mechanism wherein the inactivation of one X in females would lead to doubling the expression of the other. Ohno's hypothesis predicts that the expression of the single X chromosome in males needs compensatory upregulation to balance its dosage with that of the diploid autosomes. Additionally, X chromosome inactivation ensures that quadruple expression of the two X chromosomes is avoided in females. Copyright © 2019 Salter et al. Northern bobwhites (Colinus virginianus) are small quails in the New World Quail family (Odontophoridae) and are one of the most phenotypically diverse avian species. Despite extensive research on bobwhite ecology, genomic studies investigating the evolution of phenotypic diversity in this species are lacking. Here, we present a new, highly contiguous assembly for bobwhites using tissue samples from a vouchered, wild, female bird collected in Louisiana. Sir Oliver Scott, a philanthropist and radiation biologist and, therefore, the epitome of a gentleman and a scholar, was an early Director of the BECC Radiobiology Research Unit at Mount Vernon. The impacts of extended flooding on microbial communities and their activities in natural and agricultural wetlands have been well-documented, but there is little basis for predicting the responses of urban soil microbial communities to infrequent, short-term flooding. © 2019, the authors. Benthic assemblages of the Antarctic continental shelf are dominated by sessile and slow-moving, epifaunal invertebrates. This community structure persists because shell-crushing (durophagous) predators are absent or ecologically insignificant in shelf habitats. Durophagous teleosts, elasmobranchs, and crustaceans have been excluded by cold waters over the Antarctic shelf for millions of years. Now, as shallow waters warm rapidly, predatory king crabs (Lithodidae) living in the upper bathyal zone could emerge onto the shelf and into nearshore habitats. Filamentous fungi rapidly evolve in response to environmental selection pressures in part due to their genomic plasticity. Parastagonospora nodorum, a fungal pathogen of wheat and causal agent of septoria nodorum blotch, responds to selection pressure exerted by its host, influencing the gain, loss, or functional diversification of virulence determinants, known as effector genes. Whole genome resequencing of 197 P. nodorum isolates collected from spring, durum, and winter wheat production regions of the United States enabled the examination of effector diversity and genomic regions under selection specific to geographically discrete populations. 1,026,859 SNPs/InDels were used to identify novel loci, as well …

Neonicotinoid insecticides have come under scrutiny for their potential role in honey bee declines. Additionally, reduced access to forage in agricultural areas creates the potential for risk interactions with these pesticides in regions critical for honey production. In this study, we sought to determine whether sufficient access to pollen during larval development could mitigate stress associated with oral clothianidin exposure in honey bee adults. An apiary was established where pollen traps deprived half of the colonies of pollen, which was then supplemented to the others. Newly emerged wheat blast disease is a serious threat to global wheat production. Wheat blast is caused by a distinct, exceptionally diverse lineage of the fungus causing rice blast disease. Through sequencing a recent field isolate, we report a reference genome that includes seven core chromosomes and mini-chromosome sequences that harbor effector genes normally found on ends of core chromosomes in other strains. No mini-chromosomes were observed in an early field strain, and at least two from another isolate each contain different effector genes and core chromosome end sequences. Grain of various species of millet is a staple food of rural communities in sub-Saharan Africa, the Indian subcontinent, and China. In addition, millet is used as poultry and cattle feed in the United States, and the foliage is fodder for cattle in India. The crop is damaged by at least 150 insect pests during its growth and development. Although the current status of all of these pests is not known, shoot flies, stem borers, leaf-sucking, and the panicle-attacking insects are considered economically important. Dosage compensation of the mammalian X chromosome (X) was proposed by Susumu Ohno as a mechanism wherein the inactivation of one X in females would lead to doubling the expression of the other. This would resolve the dosage imbalance between eutherian females (XX) versus male (XY) and between a single active X versus autosome pairs (A). Expression ratio of X- and A-linked genes has been relatively well studied in humans and mice, despite controversial results over the existence of upregulation of X-linked genes. © the authors. An often overlooked component of research on factors that drive amphibian geographic distributions is description of species range shape. Broad-scale range disjunction has implications for phylogeography, ecology, and conservation, but descriptions of fragmentation are usually based on subjective visual assessment of range maps. Here, we describe a method for objectively quantifying range fragmentation and use this method to describe the patterns of amphibian species range shapes in the southeastern United States, home to the highest amphibian species richness in North America.
