Four Ni(II) complexes have been obtained with the pyridyltriazole ligand, bpt, or the bis(pyridyltriazole) ligand, m-xpt, in chapter 2. This work was carried out as a comparison with the previously established coordination chemistry of copper(II) with bpt and m-xpt. Initially, the reaction between Ni(II) and bpt only produced one mononuclear compound, [Ni(bpt)2(H2O)2](NO3)2 (1). Second harmonic generation (SHG) is used to investigate the factors that impact nanoparticle-based drug-delivery applications. In the first study, molecular adsorption and transport kinetics of a positively-charged dye, malachite green isothiocyanate (MGITC), is characterized at the surface of different colloidal liposomes in water using SHG spectroscopy. The molecular interactions of MGITC is compared to our previous investigations with malachite green (MG). In comparison to MG, MGITC demonstrates stronger adsorption and faster transport through lipid membranes. Correspondingly, the SHG experimental results are in excellent agreement with the molecular dynamics (MD) simulations results. A key finding illustrates the importance of functional groups, …

Isolation And Characterization Of Bacteria In A Toluene-Producing Enrichment Culture Derived From Contaminated Groundwater At A Louisiana Superfund Site, Madison Mikes

In an effort to better understand the role that various microbes may play in toluene production, bacteria from a toluene-producing enrichment culture derived from contaminated groundwater at a Superfund site were cultivated and isolated on low nutrient solid media. A total of 14 solid medium formulations containing varying pH ranges, carbon sources, solidifying agents, and incubation gas headspaces were used to obtain 278 isolates in pure culture. Isolated bacteria, identified using partial 16S rRNA gene sequences, were most closely related with the genera Anoxybacillus, Azospira, Bacillus, Bradyrhizobium, Cellulosimicrobium, Micrococcus, and Propionicimonas.

Cellulose nanofibers (CNFs) with nanoscale dimension, high aspect ratio, and easily modified surface chemistry show great potential as novel rheological and filtration modifiers in bentonite water-based drilling fluids (BT-WDFs). However, CNFs would suffer from poor redispersibility in an aqueous suspension if they were fully dried for transportation, storage, and field application. Herein, we report a simple, versatile, and scalable strategy to prepare water-redispersible CNFs through compounding them with a water-soluble, commercially available drilling fluid additive, polyanionic cellulose (PAC), and subsequent drying. Salt marshes are dynamic systems able to laterally expand, contract, and vertically accrete in response to sea level rise. Here, we present the grand challenges that need to be addressed to fully characterize marsh morphodynamics. The review focuses on physical processes and quantitative models. Without predictive models, it is impossible to determine the future marsh evolution under accelerated sea level rise. In these models, one of the challenges is to resolve both horizontal and vertical dynamics within the same framework. Chlamydia trachomatis Scc4 (formerly CT663) engages the transcription machinery and the pathogenic type III secretion system (T3SS). Both machines are required for Chlamydia infection. These requirements and the limited ability for genetic manipulation in Chlamydia have hampered dissection of Scc4's contributions. Here, by developing bacterial systems that permit the controlled expression and stable maintenance of Scc4, we assess Scc4's effects on chlamydial growth phenotype, secretion, and the patterns of T3SS gene expression. The indiscriminate use of nitrogenous fertilizers continues unabated for commercial crop production, resulting in air and water pollution. The development of rice varieties with enhanced nitrogen use efficiency (NUE) will require a thorough understanding of the molecular basis of a plant's response to low nitrogen (N) availability. The global expression profiles of root tissues collected from low and high N treatments at different time points in two rice genotypes, Pokkali and Bengal, with contrasting responses to N stress and contrasting root architectures were examined. In Drosophila, TADs are often separated by inter-TAD regions containing active housekeeping genes and associated insulator binding proteins. This raises the question of whether insulator binding proteins are involved primarily in chromosomal TAD architecture or gene activation, or if these two activities are linked. The pathogenesis of the rice pathogenic bacterium Burkholderia glumae is under the tight regulation of the tofI/tofR quorum-sensing (QS) system. tepR, encoding a group I bacterial enhancer-binding protein, negatively regulates the production of toxoflavin, the phytotoxin acting as a major virulence factor in B. glumae. In this study, through a transcriptomic analysis, we identified the genes that were modulated by tepR and/or the tofI/tofR QS system. Leaves, as the most important photosynthetic organ of plants, are intimately associated with plant function and adaptation to environmental changes. The scaling relationship of the leaf dry mass (or the fresh mass) vs. leaf surface area has been referred to as “diminishing returns”, suggesting that the leaf area fails to increase in proportion to leaf dry mass (or fresh mass). Host plant traits strongly affect survivorship of insect herbivores, and host suitability is especially important for the wheat stem sawfly, Cephus cinctus Norton, which spends its entire egg, larval, and pupal periods in a single stem. Measuring larval survival inside stems from egg hatch through diapause-mediated dormancy is a potential measure of population size for the next year but is also useful in assessing effects of growing season precipitation and temperature. Energy cane bagasse (ECB) is the major residue after the cane is milled to extract juice for sugar, biochemical, and/or biofuel production. The sustainable conversion of ECB into high value-added products can help reduce agricultural waste, and enhance utilization of bioresources. In the present work, we demonstrate that ECB can be converted into cellulose nanofibers (CNFs) that can serve as high value-added additives in bentonite water-based fluids (BT-WDFs). Particularly, cellulose fibers (CFs) were rapidly isolated from ECB by microwave-assisted NaOH / NaClO2 treatments. The salinization of freshwater lakes by agricultural activities poses a threat to many lake ecosystems around the world. Quantitative, medium-to long-term studies are needed to understand how some common agricultural practices, such as the discharge of crop irrigation in the vicinities of large lakes, may affect lake salinization. Sperm vitrification as an alternative approach to conventional cryopreservation (equilibrium freezing) allows quick and low-cost sample preservation and is suitable for small-bodied aquatic species with miniscule testis, fieldwork at remote locations, and small-scale freezing for research purposes. Western diets high in fat and sucrose are associated with metabolic syndrome (MetS). Although the prevalence of MetS in women is comparable to that in men, metabolic adaptations in females to Western diet have not been reported in preclinical studies. This study investigates the effects of Western diet on risk factors for MetS in female mice. Based on our earlier studies in male mice, we hypothesized that dietary supplementation with extracts of Artemisia dracunculus L. For decades, ecologists have studied fundamental questions of how Amazonian biodiversity is maintained, and whether that diversity can persist following deforestation. The long history of avian research at the Biological Dynamics of Forest Fragments Project, near Manaus, Brazil, has helped advance this understanding in the context of a broader research program focused on rainforest fragments embedded in a dynamic matrix. We evaluated the cryoprotective effects of methanol on zebrafish sperm at different concentrations, exposure times, and stages during cryopreservation. Samples were collected by crushing of dissected testes or abdominal stripping. After exposure to 0%, 2%, 5%, 8%, and 10% methanol for 0-11 min, fresh sperm (1 × 106 cells/mL) did not show changes in plasma membrane integrity (measured by flow cytometer), but cell size changes (light scatter) were observed after exposure to 8% or 10%. Bark water vapour conductance (g bark) is a rarely considered functional trait. However, for the few tree species measured to date, it appears high enough to create stem water deficits associated with mortality during droughts, when access to water is limited. I tested whether g bark correlates with stem water deficit during drought conditions in two datasets of tropical trees: One of saplings in forest understories during an annual dry season and one of potted saplings in a shadehouse during extreme drought conditions. Exercise seems to enhance the beneficial effect of bariatric (Roux-en-Y gastric bypass [RYGB]) surgery on insulin resistance. We hypothesized that skeletal muscle extracellular matrix (ECM) remodeling may underlie these benefits. Women were randomized to either a combined aerobic and resistance exercise training program following RYGB (RYGB + ET) or standard of care (RYGB). Insulin sensitivity was assessed by oral glucose tolerance test. Muscle biopsy specimens were obtained at baseline and 3 and 9 months after surgery and subjected to compre-hensive phenotyping, transcriptome profiling, molecular pathway identification, and validation in vitro. Alcohol (i.e., ethanol) is consumed regularly by much of the adult population; yet, the health effects associated with its use are not well-characterized. Clinical interventions to investigate the effects of moderate alcohol consumption on metabolic outcomes, including adiposity and cardiovascular risk factors, are limited and have yielded conflicting data. In addition, no study has reported the effects of routine alcohol intake during weight loss in a controlled feeding trial. Background/objectives: The Comprehensive Assessment of Long-term Effects of Reducing Intake of Energy Phase 2 (CALERIE) study showed that individuals who are nonobese were able to undergo significant calorie restriction (CR), yet the time course changes in adherence, weight, and appetite are unknown. This analysis aimed to investigate the time course changes in adherence, body weight, and appetite during the CALERIE study. Subjects/methods: Overall, 143 participants (body mass index: 21.9–28.0 kg/m2) were randomized to a CR group that aimed to achieve 25% CR for 2 years. In the Look AHEAD trial, randomization to Intensive Lifestyle Intervention (ILI) or Diabetes Support and Education (DSE) did not result in differences in cognitive outcomes. However, menopause and APOE genotype are factors that affect the response to this intervention. The effect of this intervention on a single cognitive assessment was examined in 3 groups of women: premenopausal or < 5 years postmenopausal (N = 594), within 5–10 years (n = 388), and ≥10 years postmenopausal (n = 963), and as a function of continuous years since menopause. The late postmenopausal group in the ILI had worse composite z-scores compared to those in the DSE, whereas the younger premenopausal or early postmenopausal women in the ILI had better composite z-scores than the DSE. A significant interaction between years since menopause and intervention arm, but not baseline age, was observed on executive function domains. ILI appeared only to benefit cognitive function among non-APOE4 carriers during premenopause or early postmenopause. These findings emphasize the importance of assessing menopause and APOE status to understand how weight loss impacts cognition.

Background: Elevated cardiovascular disease risk factor burden is a recognized contributor to poorer cognitive function; however, the physiological mechanisms underlying this association are not well understood. We sought to assess the potential mediation effect of left ventricular (LV) remodeling on the association between lifetime systolic blood pressure and cognitive function in a community-based cohort of middle-aged adults. The novel SARS-CoV-2 coronavirus that emerged in the city of Wuhan, China, last year has since become the COVID-19 pandemic across all continents. To restrict the spread of the virus pandemic, the Government of India imposed a lockdown from 25 March 2020.
