Bacteriophages densely pack their long double-stranded DNA genome inside a protein capsid. The conformation of the viral genome inside the capsid is consistent with a hexagonal liquid crystalline structure. Experiments have confirmed that the details of the hexagonal packing depend on the electrochemistry of the capsid and its environment. In this work, we propose a biophysical model that quantifies the relationship between DNA configurations inside bacteriophage capsids and the types and concentrations of ions present in a biological system. We introduce an expression for the free energy that combines the electrostatic energy with contributions from bending of individual segments of …

Characterization Of Application-Driven Sol-Gel Coated Meshes And Composites, Armaghan Naderi
LSU Doctoral Dissertations
Surface engineering is a multi-disciplinary research area that encompasses disciplines such as materials science, chemistry, mechanical engineering, and electrical engineering. Coating is a surface modification technique in surface engineering, applied on the surface of a substrate (i.e., bulk material), to cover/protect it from environmental degradation or to modify/improve surface properties for specific applications. This PhD research presents an in-depth investigation of sol-gel ceramic coatings to modify surface characteristics of bulk materials for two specific applications: 1) antistatic/antidust applications, and 2) biomaterial implant applications.
First, zirconia antistatic thin films on glass substrates, and their application to polymer-based composites, was studied. Zirconia …

The terrestrial gamma-ray flash (TGF) and Energetic Thunderstorm Rooftop Array (TETRA-II) detected 22 X-ray/gamma-ray flash events associated with lightning between October 2015 and March 2019 across three ground-based detector locations in subtropical and tropical climates in Louisiana, Puerto Rico, and Panama. Each detector array consists of a set of bismuth germanate scintillators that record X-ray and gamma-ray bursts over the energy range 50 keV–6 MeV (million electron volts). TETRA-II events have characteristics similar to both X-ray bursts associated with lightning leaders and TGFs: sub-millisecond duration, photons up to MeV energies, and association with nearby lightning (typically within 3 km). Seed Treatment With Jasmonic Acid And Methyl Jasmonate Induces Resistance To Insects But Reduces Plant Growth And Yield In Rice, Oryza Sativa, Santhi Bhavanam, Michael Stout

When applied exogenously to plants, jasmonates [i.e., jasmonic acid (JA) and methyl jasmonate (MeJA)] increase plant resistance against herbivores, and their use in pest management has been suggested. For integration into pest management programs, the benefits of the resistance induced by jasmonates must outweigh the costs of jasmonates on plant growth and yield. A previous field study in rice found that seed treatment with MeJA reduced densities of the rice water weevil, Lissorhoptrus oryzophilus, but also reduced plant growth. Yields from MeJA plots were similar to yields from control plots. Because this study was conducted under field conditions with natural …

Total daily energy expenditure (“total expenditure”) reflects daily energy needs and is a critical variable in human health and physiology, but its trajectory over the life course is poorly studied. We analyzed a large, diverse database of total expenditure measured by the doubly labeled water method for males and females aged 8 days to 95 years. Total expenditure increased with fat-free mass in a power-law manner, with four distinct life stages. Fat-free mass-adjusted expenditure accelerates rapidly in neonates to ~50% above adult values at ~1 year; declines slowly to adult levels by ~20 years; remains stable in adulthood (20 to …

High-entropy alloys (HEAs) have intriguing material properties, but their potential as catalysts has not been widely explored. Based on a concise theoretical model, we predict that the surface of a quaternary HEA of base metals, CoCrFeNi, should go from being nearly fully oxidized except for pure Ni sites when exposed to O2 to being partially oxidized in an acidic solution under cathodic bias, and that such a partially oxidized surface should be more active for the electrochemical hydrogen evolution reaction (HER) in acidic solutions than all the component metals. These predictions are confirmed by electrochemical and surface science experiments: the …

Previous studies of the lakes of the McMurdo Dry Valleys have attempted to constrain lake level history, and results suggest the lakes have undergone hundreds of meters of lake level change within the last 20ĝ€¯000 years. Past studies have utilized the interpretation of geologic deposits, lake chemistry, and ice sheet history to deduce lake level history; however a substantial amount of disagreement remains between the findings, indicating a need for further investigation using new techniques. This study utilizes a regional airborne resistivity survey to provide novel insight into the paleohydrology of the region. Mean resistivity maps revealed an extensive brine …

Mitochondria are essential to the onset and progression of cancer through energy production, reactive oxygen species regulation, and macromolecule synthesis. Genetic and functional adaptations of mitochondria to the tumor environment drive proliferative and metastatic potential. The advent of DNA and RNA sequencing removed critical barriers to the evaluation of genetic mediators of tumorigenesis. However, to date, methodological approaches to evaluate tumor mitochondrial function remain elusive and require technical proficiency limiting the feasibility, ultimately diminishing diagnostic and prognostic value in both experimental and clinical settings. Here, we outline a simple and rapid method to quantify rates of oxidative phosphorylation (OXPHOS) and …

Prioritization of limited funding for pipelines maintenance is a major issue that concerns municipalities nationwide. Conducting a probabilistic assessment can provide a complete characterization of the performance of structural elements and systems along with optimizing the limited resources. The most widely used probabilistic performance indicator is reliability, a measure of the probability of failure corresponding to a particular limit state (e.g., ultimate strength or serviceability). Reliability methods can be used to identify which pipeline sections within a particular system require the most urgent inspection or repair. To this end, an automated data-driven framework for large diameter reinforced concrete pipes (RCPs) …

Plant structural traits can act as barriers for herbivore attachment, feeding, and oviposition. In particular, epicuticular waxes (EWs) on the aerial surfaces of many land plants offer protection from biotic and abiotic stresses. In rice (Oryza sativa L.), mutations that reduce EWs have been previously reported. However, whether such mutations affect rice water weevil (Lissorhoptrus oryzophilus Kuschel) and fall armyworm (Spodoptera frugiperda Smith) performance has not been investigated yet. These pests cause significant economic problems in important rice-producing areas of the United States. The aim of our study was to characterize the EWs of EW mutants and wild-type rice plants …

A seed blend refuge has been implemented in the U.S. Corn Belt for Bt maize resistance management. The fall armyworm, Spodoptera frugiperda (J.E. Smith), is a target pest of Bt maize in the Americas. The larvae of this pest are mobile, which may affect the efficacy of seed blend refuges. In this study, field and greenhouse trials were conducted to determine the performance of Bt-susceptible (aabb) and -heterozygous dual-gene-resistant (AaBb) genotypes of S. frugiperda in seed blends of non-Bt and pyramided Bt maize. Three field trials evaluated larval survival, larval growth, and plant injury with aabb in seed blends of …

Wireworms have become a significant menace to cereals in the Northern Great Plains. Therefore, research toward developing effective control methods such as biological control with entomopathogenic nematodes (EPNs) is warranted. Two strains, each of two EPN species, Steinernema carpocapsae (Weiser) and Steinernema riobrave Cabanillas, Poinar, and Raulston in the form of infected Galleria mellonella (L.) cadavers were evaluated against wireworms in field and greenhouse. In field experiments, none of the four EPN strains were found effective against wireworms. However, in the greenhouse test, three of the strains, S. carpocapsae (All and Cxrd) or S. riobrave (355) applied in cadavers killed …

Ultraviolet (UV) radiation has been widely used to monitor insects and conduct surface sterilizations at 365 nm and 254 nm wavelengths, respectively. The global increase in the use of UV light has raised concerns about environmental impacts and effects on non-target organisms. We, therefore, examined the effects of three UV radiation wavelengths (365 nm, 308 nm and 254 nm) on the survival and antioxidant activities of a tephritid fruit fly, Bactrocera dorsalis. Exposure to UV radiation significantly increased B. dorsalis cohort mortality and the duration of pre-oviposition. Moreover, the application of UV-B and UV-C significantly decreased egg laying. The activities …

Wheat stem sawfly, [Cephus cinctus (Hymenoptera: Cephidae)], females display complex behaviors for host selection and oviposition. Susceptible hollow stem wheat (Triticum aestivum L.) cultivars release a greater amount of attractive compound, (Z)-3-hexenyl acetate and receive a greater number of eggs compared to resistant solid stem wheat cultivars. However, barley (Hordeum vulgare L.) is becoming a more common host for C. cinctus in Montana. Therefore, how do host selection and oviposition behaviors on barley cultivars compare to what happens when encountering wheat cultivars? To answer this question, we carried out greenhouse experiments using two barley cultivars: 'Hockett' and 'Craft'. Between these …

Dietary protein restriction and dietary methionine restriction (MR) produce a comparable series of behavioral, physiological, biochemical, and transcriptional responses. Both dietary regimens produce a similar reduction in intake of sulfur amino acids (e.g., methionine and cystine), and both diets increase expression and release of hepatic FGF21. Given that FGF21 is an essential mediator of the metabolic phenotype produced by both diets, an important unresolved question is whether dietary protein restriction represents de facto methionine restriction. Using diets formulated from either casein or soy protein with matched reductions in sulfur amino acids, we compared the ability of the respective diets to …

As an obligate salt marsh species, Seaside Sparrows (Ammospiza maritima) are vulnerable to numerous threats including climate change, coastal erosion, sea-level rise, and both natural and anthropogenic disasters. Of the 9 recognized subspecies, 2 are extinct and 1 is endangered. Previous genetic analyses of mitochondrial DNA (mtDNA) and microsatellite loci showed that current taxonomy does not accurately reflect underlying genetic diversity, with possible consequences for the distribution of conservation resources. To further inform Seaside Sparrow management, we comprehensively describe genetic structure among 24 range-wide sampling locations that include all extant subspecies. We inferred population structure from several thousand single-nucleotide polymorphisms …

Background & objectives: Examine maternal gestational diabetes mellitus (GDM), macrosomia and DNA methylation in candidate genes IGF1, IGF2, H19, ARHGRF11, MEST, NR3C1, ADIPOQ and RETN. Materials & methods: A total of 1145 children (572 GDM cases and 573 controls) from the Tianjin GDM study, including 177 with macrosomia, had blood DNA collection at median age 5.9 (range: 3.1-10.0). We used logistic regression to screen for associations with GDM and model macrosomia on 37 CpGs, and performed mediation analysis. Results: One CpG was associated with macrosomia at false discovery rate (FDR) < 0.05 (cg14428359 in MEST); two (cg19466922 in MEST and cg26263166 in IGF2) were associated at p < 0.05 but mediated 26 and 13%, respectively. Conclusion: MEST and IGF2 were previously identified for potential involvement in fetal growth and development (Trial Registration number: NCT01554358 [ClinicalTrials.gov]).

Background and aims: A diminution in skeletal muscle mitochondrial function due to ectopic lipid accumulation and excess nutrient intake is thought to contribute to insulin resistance and the development of type 2 diabetes. However, the functional integrity of mitochondria in insulin-resistant skeletal muscle remains highly controversial. Methods: 19 healthy adults (age:28.4 ± 1.7 years; BMI:22.7 ± 0.3 kg/m2) received an overnight intravenous infusion of lipid (20% Intralipid) or saline followed by a hyperinsulinemic-euglycemic clamp to assess insulin sensitivity using a randomized crossover design. Skeletal muscle biopsies were obtained after the overnight lipid infusion to evaluate activation of mitochondrial dynamics proteins, …

Ambient air particulate matter (PM) and PM-associated environmentally persistent free radicals (EPFRs) have been documented to contribute to pollution-related health effects. Studies of ambient air PM potentially bear artifacts stemming from the collection methods. We have investigated the applicability of PM phytosampling (PHS) as a supplementary tool to a classic PM sampler in respect of achieving better PM chemical composition assessment (primarily organic fraction). Phytosampling is a static PM collection method relying on the particle entrapment by the plant’s leaf through electrostatic forces and surface trichomes. We have investigated the differences in the EPFR and polycyclic aromatic hydrocarbon (PAH) speciation …

Choosing among types of genomic markers to be used in a phylogenomic study can have a major influence on the cost, design, and results of a study. Yet few attempts have been made to compare categories of next-generation sequence markers limiting our ability to compare the suitability of these different genomic fragment types. Here, we explore properties of different genomic markers to find if they vary in the accuracy of component phylogenetic trees and to clarify the causes of conflict obtained from different data sets or inference methods. As a test case, we explore the causes of discordance between phylogenetic …

Separate introductions or post-introduction evolution may lead to multiple invader genotypes or cytotypes that differ in growth rates, biomass or chemical profile responses (phenotype) to a range of environments. If the invader has high trait plasticity to a range of resource levels, then sediment N or P enrichment may enhance invasiveness. However, the ways in which ploidy, plasticity, and available N or P interact are unknown for most species despite the potential to explain spread and impacts by invaders with multiple introduced lineages. We conducted a common garden experiment with four triploid and six diploid populations of Butomus umbellatus, collected …

Concrete is a remarkable construction material. However, its low tensile strength makes it prone to cracking, which negatively affects its durability. To address this issue, bacterial concrete has been implemented as a self-healing alternative due to its capability to seal microcracks through microbial-induced calcium carbonate precipitation (MICCP). In this study, a bacterial strain (i.e, Bacillus Pseudiformus) was encapsulated through three different methods: encapsulation through hydrogel beads, vacuum impregnation on lightweight aggregates, and attachment to cellulose nanocrystals. Furthermore, three precursor types were used, magnesium acetate, calcium lactate, and sodium lactate were implemented. Compressive strength tests and flexural strength tests were performed …

Young people aged 10–24 years constitute 24% of the world's population; investing in their health could yield a triple benefit—eg, today, into adulthood, and for the next generation. However, in physical activity research, this life stage is poorly understood, with the evidence dominated by research in younger adolescents (aged 10–14 years), school settings, and high-income countries. Globally, 80% of adolescents are insufficiently active, and many adolescents engage in 2 h or more daily recreational screen time. In this Series paper, we present the most up-to-date global evidence on adolescent physical activity and discuss directions for identifying potential solutions to enhance …
Phragmites Australis Dieback In The Mississippi River Delta: Chemical Profiles Of Soil Types And Restoration Potential, Herie Lee
LSU Master's Theses
Since 2016, there has been widespread dieback of P. australis in the Lower Mississippi River Delta (hereafter referred to as Lower MRD, which is defined as the Birds Foot Delta) with relatively little to no signs of recovery. The cause of the current dieback is not fully understood. This thesis explores P. australis with emphasis on chemical profile characterization of different soil types and its effects on plant growth and the potential for restoration in the Lower MRD.

The distribution and vertical fluxes of particulate organic carbon and other key elements in the Arctic Ocean are primarily governed by the spatial and seasonal changes in primary productivity, areal extent of ice cover, and lateral exchange between the shelves and interior basins. The Arctic Ocean has undergone rapid increase in primary productivity and drastic decrease in the areal extent of seasonal sea ice in the last two decades. These changes can greatly influence the biological pump as well as associated carbon export and key element fluxes. Here, we report the export of particulate organic and inorganic carbon, particulate nitrogen …

ABSTRACT
Arthropod-borne viruses (arboviruses) are the etiological agents of much morbidity and mortality, especially in low- and middle-income countries. Many of these viruses are spread and maintained by mosquitoes, particularly the urban mosquito Aedes aegypti. Zika virus (ZIKV) is responsible for one of the largest vector-borne disease outbreaks in the past decade, affecting millions in Central and South America including a wave of microcephaly among newborns. Mayaro virus (MAYV) is a mosquito-borne virus endemic to South America and is predicted to become an emergent public health threat. Describing the vector-virus transmission systems are critical for understanding the potential spread …

Challenging The Illusion Of Diversity: A New Theoretical Paradigm Towards Newsroom Inclusion., Paromita Saha
LSU Doctoral Dissertations
Over the past forty years, the news industry has attempted to diversify the racial composition of their newsrooms from the perspective of representing their markets better mirroring best business practices in the industry. The statistics show that an affirmative style of recruitment has produced minimal, superficial results with a high turnover of minority journalists who are hardly promoted to editorial positions, as well as skewed representations of minority racial groups. The literature indicates that hegemonic ideological forces and the White male professional ethos behind journalism is not conducive to minority journalists producing meaningful changes to news content. My study indicates …
Degumming Of Hemp Fibers Using Combined Microwave Energy And Deep Eutectic Solvent, Bulbul Ahmed
Degumming Of Hemp Fibers Using Combined Microwave Energy And Deep Eutectic Solvent, Bulbul Ahmed
LSU Master's Theses
Hemp is considered as one of the sustainable agricultural fiber materials. Degumming or surface modification of hemp bast is needed to produce single fibers for ensuing textile and industrial applications. The traditional degumming process necessitates a high amount of alkali, which causes detrimental environmental pollution. This study offers a new method to degum hemp fibers with reduced use of harmful alkali and precious water resources. In this work, hemp bast fibers were degummed by using combined microwave energy and deep eutectic solvent (DES). The properties of hemp fibers manufactured by this method were investigated and compared with the traditional alkali …
