Two new approaches for reducing the order of large scale continuous systems are presented. The first approach is a modified version of the aggregation technique. It uses the matching properties of the steady state output covariance and the Markov parameters of the high order system to those of the r('th) order model. This approach also uses a new algorithm derived for the computation of the controllability and observability Grammians, to produce controllable, observable and stable low order models. An Analysis Of Mercury Accumulation Within The Upper Estuarine Food Chain Of Lake Charles, Louisiana, Jeffrey W. Tingle

Analysis of the level of mercury contamination within the sediments and fish associated food chain of Lake Charles, Louisiana, an area located within the Calcasieu River system, were assessed. Sediment levels were found to be slightly elevated (0.33µg Hg/g) from natural background levels (0.20 µg Hg/g). Levels found within the fish of the region were also found to be slightly elevated (0.287 µg Hg/g) from natural levels. This is below the hazardous level designated by the Food and Drug Administration (0.50 µg Hg/g). Locus-Specific Detection Of Hla-Dq And -Dr Antigens By Antibodies Against Synthetic N-Terminal Octapeptides Of The Β Chain, Thomas Deufel, Anne Grove, Hans Kofod, Åke Lernmark

Antibodies against synthetic peptides representing the class-II antigen HLA-DR and -DQ β chain N-terminal sequences were prepared in rabbits. The two octapeptides only share two amino acids and enzyme-linked immuno-assays showed the antisera only to bind to its own antigen. Both peptide antisera detected a 29 kDa component in immunoblots of Raji and AL-34 cell plasma membrane proteins separated by SDS gel electrophoresis. The binding of either N-terminal peptide antiserum was selectively inhibited only by the peptide used as antigen. Indirect immunofluorescence.analysis by flow cytofluorometry showed specific surface immunofluorescence in 1:100-1:1000 dilutions in lymphoblastoid and blood mononucleated cells. In the …

The Azido[14C]Atrazine Photoaffinity Technique Labels A 34-Kda Protein In Scenedesmus Which Functions On The Oxidizing Side Of Photosystem Ii, James G. Metz, Terry M. Bricker, Michael Seibert

We have used azido[14C]atrazine to photoaffinity label thylakoids from wild-type (WT) Scenedesmus and a mutant, LF-1, which is blocked on the oxidizing side of photosystem II (PS II). One protein is labeled in each case, at 34 kDa in the WT and 36 kDa in LF-1. Previous comparison of the WT with LF-1 had been used to assign a PS II donor side function to the 34-kDa protein. These results suggest that this photoaffinity technique does not label the herbicide-binding protein involved in electron transfer on the reducing side of PS II. © 1985.

Studies On The Molecular Cloning Of A Gene For Aryletherase From A Ligninolytic Erwinia Sp. (Lignin, Biodegradation, Bacterial)., Kenneth Edwin Narva

Recombinant DNA techniques were utilized to clone a gene from the ligninolytic system of a saprophytic Erwinia sp. into Escherichia coli using pBR322 as a vector. The cloned gene encodes aryletherase activity and was selected from amp('r) tet('S) transformants of E. coli Cs412 using salicin as the selective carbon source. This compound has structural characteristics similar to some lignin model compounds. The cloned gene enclodes activity which oxidizes aromatic lignin model compounds resulting ultimately in the breakage of arylether bonds while leaving the aromatic nucleus intact. Extracts of foliage from the insect resistant soybean genotype PI227687 were bioassayed for activity against the soybean looper, Pseudoplusia includens (Walker), in feeding deterrency and growth inhibition tests. Dichloromethane extract had a pronounced deterrent effect on feeding, while petroleum ether and methanol extracts stimulated feeding. The water extract exhibited neutral activity. Flash chromatography of the dichloromethane extract yielded four fractions with moderate deterrent properties, one which was strongly deterrent, and one which was stimulative. Similar fractionation of the methanol extract yielded four fractions; one stimulated feeding, two deterred feeding, and one was neutral. Feeding P. includens larvae on artificial diet …

Recombinant DNA technology was used to produce a probe for detection of Clavibacter xyli subsp. xyli, the sugarcane ratoon stunting disease agent. C. xyli, subsp. xyli chromosomal DNA was digested with the restriction enzyme Pst 1, inserted into the plasmid pUC 19, and transformed into Escherichia coli JM 83. Recombinant clones containing six different sizes of C. xyli subsp. xyli DNA sequences were obtained. Plasmid K3-5, with an insert of 4.9Kb, and C. xyli subsp. xyli chromosomal DNA, were radiolabelled by nick translation using dCTP or dATP (alpha-('32)P). The purpose of experiment one was to develop and validate an open circuit technique (OCT) employing a mixing-chamber for measurement of gas exchange during non-steady-state exercise in humans and ponies. Serial measurements of oxygen uptake ((')VO(,2)) using human subjects and ponies were made during rest to exercise transitions using the OCT incorporating a time delay to match mixed expired gas concentrations with the appropriate ventilatory volume. Oxygen uptake calculated by the OCT did not differ significantly (P > 0.05) from (')VO(,2) obtained by Douglas bag procedures. As part of a biochemical systematic investigation of the tribe Helianteae (Compositae) for their secondary metabolites, Calea deltophylla Cowan from the subtribe Galinsoginae, Squamopappus skutchii (S. F. Blake) Jansen, Harriman and Urbatsch, from the Ecliptinae, and Podachaenium eminens (Lagasca) Shultz-Bip. from the Verbesininae were chemically investigated. Calea deltophylla provided the known compounds ageratochromene and eupatarone and three new compounds, 3-methoxy-4-hydroxyseneciophenone, caldeltophyllide and deltophyllin. Squamopappus skutchii yielded the known sesquiterpenoid (beta)-eudesmol, two new guaianolides, skutchiolides A and B, and the new eudesmanolide chapinolin. P. eminens provided three known guaianolides. The structures of the compounds were established by chemical and spectroscopic methods …

Rice flour was modified with either 0.1, 1.0 and 5.0% acetic or succinic anhydride for 1, 6 and 18 hr under basic conditions. The pH was maintained above 7.0 using either sodium hydroxide or sodium carbonate. The results showed that modified rice flours had lower fat content as compared with the unmodified rice flour (control). The type of anhydride significantly influenced the fat content of rice flour. Ash content of rice flour increased significantly due to the anhydride concentrations and alkali type. A study was conducted to investigate the use of bacterial numbers and metabolites as estimators of the potential shelf-life of perishable dairy products, i.e., whole milk, skim milk and cottage cheese. The study was conducted in three phases. Karyotypes of nine species (Megalops atlanticus, Elops saurus, Brevoortia patronus, B. smithi, B. tyrannus, Dorosoma cepedianum, D. petenense, Harengula clupeola, and Anchoa mitchilli) and one hybrid (B. smithi x B. tyrannus) of teleost fishes revealed diploid complements ranging from 28 to 50 chromosomes and comprised of uniarmed elements or combinations of uniarmed and small and large biarmed elements. The occurance of large metacentric-submetacentric chromosomes was usually associated with a reduction in total chromosome number suggesting fusion events in the evolution of karyotypes from ancestral complements with 48 to 52 acrocentrics. An ecological study of the young-of-the-year of three year-classes of gulf menhaden (Brevoortia patronus) was conducted from July 1981 to December 1983 in Fourleague Bay, Louisiana. This study documented gulf menhaden population ecology and the importance of gulf menhaden in the nutrient and energy balance of estuaries. Young-of-the-year gulf menhaden immigrated in winter and moved to tidal creeks and ponds, where they remained until midsummer. They then moved into open bays, where they remained until the fall. This migration pattern coincides with sequentially available, but spatially separate, productivity peaks in marsh and open bays. In the fall, they emigrated to …

Insight into the mineral, protein, phytate relationships in rice bran was gained by examining the solubility behavior of these components. Solubility profiles of the phosphorus and nitrogen components as a function of pH did not correspond, except in the pH 1.0-2.0 range. This is evidence that association between protein and phytate only occurred in this low pH range. Potassium, magnesium, and calcium solubilities as a function of pH corresponded to those of the phytic acid salts of these minerals. Soluble magnesium and potassium phytates were isolated on Sephadex G-25-50 and identified by ICP, confirming their presence in rice bran. The …

Variation in alleles at gene loci encoding proteins was used to determine zoogeographic and evolutionary differentiation in thamnophiine snakes. Differentiation among populations within a species, among species within a genus, and among genera was investigated. In the southern banded water snake, Nerodia fasciata, analysis by F-statistics and genetic distances of allele frequencies indicated that extensive differentiation has occurred between those forms adapted to a saltwater environment (N. f. clarkii and N. f. compressicauda) and those adapted to a freshwater environment (N. f. fasciata, N. f. confluens, and N. f. pictiventris). Oogenesis commenced on day 3 (day 0 = adult ecdysis) in Acheta domesticus L., which, based on degeneration of the flight muscles, was approximately when juvenile hormone was released. Starvation did not prevent the onset of oogenesis, although egg production was greatly reduced. Virgin females did not oviposit and the oviducts filled with eggs by day 15. Compaction appeared to be a factor limiting oogenesis. The digestive tract was compressed and feeding reduced, which might be the primary factors terminating oogenesis. The concentration of juvenile hormone esterase (JHE) did not change as virgins filled with eggs, which indicated that JHE …

The inheritance of resistance to the sheath blight disease of rice (Oryza sativa L.), caused by Rhizoctonia solani Kuhn, was studied using as parent lines four susceptible commercial varieties--Lebonnet, Labelle, Starbonnet and Leah, and three resistant lines: L201, RU7902185 and RU7902191. The parental lines, F(,2) and F(,3) progeny of each cross were inoculated when the seedlings were 35 days old using the isolate LR172, and then kept in a humidity chamber for two weeks. The infection level of each plant was evaluated using a 0-9 rating system where 0 indicates no symptoms and 9 indicates the most severe infection. Infection …

The superfamily Tyrannoidea (Aves) has been the subject of numerous systematic investigations, yet the relationships among and within the five component families remain ambiguous. Relationships within the superfamily were investigated using starch gel electrophoresis of 20 enzyme loci in 14 cotinga genera, 12 manakin genera, the flycatcher genus Myiarchus, and each of the genera from the monogeneric families Phytotomidae and Dxyruncidae. Electrophoretic characters were found to contain considerable information concerning familial and subfamilial relationships within this New World assemblage. Although there is considerable evidence to support the contributions of life event stress to subsequent psychological and physical symptoms, the correlations reported between obtained stress scores and illness measurements have been low. This study was undertaken to improve prediction of physical and psychological complaints subsequent to life event stress by more comprehensive measurement of life event characteristics and by assessment of locus of control, social supports, and somatization as moderating variables in the life stress-illness process. Antibody responses to the apparently species specific phenolic-glycolipid-1 (Phen-Gl-1) antigen of Mycobacterium leprae were examined in humans and armadillos using enzyme-linked immunosorbent assays (ELISA). Statistical definitions for the interpretation of positive and negative reactions were derived. A retrospective serological survey of armadillos indicated that leprosy in the wild armadillo is a naturally acquired zoonosis. Presently 12.5% of the armadillos in 2 parishes in south central Louisiana have detectable IgM antibodies to Phen-Gl-1. Approximately 2.7% of these histologically exhibit clinical disease. Antibodies were not detected in Florida armadillo sera. Variations in prevalence rates were noted, and may be due to environmental …

Two groups of domestic cats with established social structures were tested for their response to novel stimuli in a variety of test situations. Clearly observable individual differences in responses were seen which were consistent over a series of test sessions and remained stable despite regular environmental disruptions. Individual differences in behavioral response were found not to be related to object dominance (food competition) or social dominance (freedom of movement in social encounters) unlike similar studies with social animals. No relationship was found in either group between rank in object dominance and rank in social dominance. Significant correlations were demonstrated between …

The hypoxia tolerance (LC-50) of Thais haemastoma was inversely related to acclimation temperature but was not related to acclimation salinity between 10 and 30(DEGREES)C and 10 and 30('o)/ooS. T. haemastoma is moderately tolerant to anoxia, the LT-50 (time to 50% mortality) being 12-22 days. Oxyregulatory ability varies from poor to good, and varies inversely with temperature. The oxygen consumption rate was lower after 28 days exposure to hypoxic conditions than at full oxygen saturation at all salinities at 20 and 30(DEGREES)C.
