Conceptus-Derived Immunosuppressive Factors And Their Role In Fetal Allograft Protection., Terri Lynn Roth

The role of conceptus-derived immunosuppressive factors in protecting the fetal allograft was investigated by first examining the immunosuppressive activity derived from ovine, caprine and hybrid conceptus trophoblast tissue. Medium conditioned by 20 day-old conceptus trophoblast tissue from each species suppressed sheep and goat lymphocyte proliferation (P $$.05) between the immunosuppressive effects of the three media on either sheep or goat lymphocytes. In subsequent studies, horse conceptus-conditioned medium (HCCM) derived from embryos at days 9, 15 and 26 of gestation, suppressed mitogen-stimulated horse lymphocytes (P $<$.001). HCCM maintained its suppressive activity following heating and repeated freeze-thawing. Using microconcentrator centrifugation, it was determined that the suppressor factor(s) was $>$100,000 MW. We have studied the interaction of cigarette smoke with two proteins: alpha-1-proteinase inhibitor ($\alpha$1PI) and ferritin. The inactivation of $\alpha$1PI, the major plasma inhibitor of serine proteases, by cigarette smoke has been implicated in the eitology of pulmonary emphysema. Release of iron from ferritin, the principal source of physiological iron, by cigarette smoke could explain increased amounts of iron observed in the pulmonary alveolar macrophages of smokers. These studies show that cigarette smoke reacts with proteins to cause amino acid modifications by two mechanisms: oxidation of methionine and nonoxidative modification of the basic amino acids. Ozonation of methyl oleate has been shown for the first time to form ozonides in aqueous micelles of sodium dodecyl sulfate (SDS) and in L-$\alpha$-distearoyl-phosphatidylcholine (DSPC) liposomes. The total yield of ozonides in 0.02 M SDS micelles exposed to 26 ppm ozone at pH 7.4 and 22$\sp\circ$C is 11.1%. This 11.1% consists of 7.5% of the normal ozonide (MOO2), ca. 4.2% of trans-isomer and ca. 3.3% of cis-isomer, and the rest 3.6% is accounted for by the cross ozonides (MOO1 and MOO3). The chromatographic properties of polymer-coated Unisphere aluminas for reversed-phase liquid chromatography were evaluated and compared to those of Si-C$\sb{18}$ phases. A greater difference in solvent strength between acetonitrile and methanol was observed for the Unisphere columns. Approximately equal methylene selectivity was also obtained for both stationary phase types. Evaluation of polar group selectivity showed the absence of hydroxyl group participation in solute retention for the aluminas. In terms of kinetic properties, acceptable reduced plate heights were obtained for the Unisphere columns at optimum velocities. However, the van Deemter curves for the aluminas showed a more rapid loss in efficiency with …

The effect, in vitro, of water soluble components of the particulate phase of cigarette smoke on the elastase inhibitory capacity (EIC) of human alpha-1-proteinase inhibitor ($\alpha\sb1$PI) is studied. Basic features of aqueous cigarette tar extract-mediated damage to $\alpha\sb1$PI are presented, as are mechanistic studies addressing the identities of damaging species and the nature of damage to $\alpha\sb1$PI. Aqueous cigarette tar extract (ACT) causes a slow loss of EIC, and retains this $\alpha\sb1$ PI damaging ability for hours. Most of the effort to elucidate the mechanism of oxygen-mediated regulation of tetrapyrrole biosynthesis in Rhodobacter capsulatus has focused on the first step in the biosynthetic pathway, the condensation of glycine with succinyl-CoA to form $\delta$-aminolevulinic acid. This reaction, in R. capsulatus, is catalyzed by $\delta$-aminolevulinate synthase, the product of the hemA gene. In order to facilitate further study of the hemA gene and its product, its nucleotide sequence was determined. The open reading frame encoding $\delta$-aminolevulinate synthase was determined to be 1,206 base pairs, encoding a 50,491 Da polypeptide of 401 amino acids. The DNA sequence was compared with …

The major chemical constituents of Magnolia acuminata, M. ashei, M. cordata, M. dealbata, M. fraseri, M. freemani, M. little gem, M. macrophylla, M. tripetala and M. virginiana were studied as part of a continuing search for biologically active secondary metabolites from regional plant sources and the investigation of their biosynthesis and large scale production. All these Magnolia species are rich in fatty acids and several also contain monoterpenes, sesquiterpenes, sesquiterpene lactones and lignans. The organic extract of the leaves of Magnolia virginiana provided a new sesquiterpene lactone, 4,14-dehydromagnograndiolide, and the new sesquiterpene lactol costunolactol. Anaerobic formation of dimethylsulfide (DMS) and methylmercaptan (MSH) in anoxic sulfide-containing slurries from marine and fresh water sediments was stimulated by addition of syringate (4-hydroxy,3,5,-dimethoxybenzoate) and 3,4,5,-trimethoxybenzoate. The release of DMS and MSH occurred during the consumption of the aromatic monomers and ceased after their depletion. DMS was the dominant methylated sulfur compound in fresh water sediments, in contrast to marine sediments where MSH was predominant. No production of volatile organic sulfur compounds was observed in slurries containing gallate (3,4,5,-trihydroxybenzoate) or in autoclaved controled. About 50-65% of the methoxy carbon could be accounted for by peak accumulation of DMS and …

The HS subfamily of Alu sequences is comprised of a group of nearly identical members. Individual subfamily members share 97.7% nucleotide identity with each other and 98.9% nucleotide identity with the HS consensus sequence. Individual subfamily members are on the average 2.8 million years old, and were probably derived from a single source 'master' gene sometime after the human/great ape divergence. The recent Alu family member insertions provide a better image of the structure of Alu retroposons before they have had the opportunity to change significantly. Patterns in coastal wetland loss in the northern Gulf of Mexico were examined using aerial imagery from 1955–56 and 1978. Five qualitative types of wetland changes are evident: (1) spoil bank-parallel pond formation, (2) pond formation with apparent random distribution for the smallest ponds, but very clumped distribution for larger ponds, (3) semi- or complete impoundment resulting in open water formation, (4) cutting off of stream channels upstream of where a spoil bank crosses a natural channel, and (5) erosion at the land-water interface. In this experiment we have identified and partially characterized the immunosuppressive activity of preimplantation horse conceptus-conditioned medium (HCCM). Horse conceptuses were nonsurgically flushed from mares at Days 9-10 (n = 6), 15-16 (n = 3), and 25-26 (n = 3). After incubating the conceptuses for 24 h in RPMI-1640 supplemented with 15% fetal calf serum (FCS) and 1% penicillin/streptomycin, HCCM was obtained from cultures and tested for immunosuppressive activity in lymphocyte proliferation assays. The causes of the extensive (0.86%/yr; 288,414 ha/yr) and well documented dramatic and accelerating rate of coastal wetland loss in the northern Gulf of Mexico were investigated by an interdisciplinary university research team to discern the role of outer continental shelf development. The landscape changes and potential causal agents are emphasized herein. Natural driving factors include sea level rise and geological com- paction, which appear to remain constant this century, and sediment supply from the Mississippi River which has declined by 50%,since the 1950s. Several parameters affecting the behavior of Chromium(+VI) in deep well injection zones were studied. Temperature, pH, concentration of inorganic salt in wastewater, and organic chemicals co-injected with wastewater were considered as they affect the adsorption and reduction rates of Cr(VI). Water content, bulk density of shale and sand, and temperature were examined for their effect on the diffusion rate of Cr(VI) in porous media. Adsorption and reduction of Cr(VI) were investigated using three clays, Na-Montmorillonite, Illite, and Kaolinite, and sand from an injection well formation in southern Louisiana, and a confining layer shale obtained from a second Louisiana injection well. …

I examined how wave action and predators affect the feeding of the intertidal snail, Stramonita haemastoma. In laboratory wave simulators, the numbers of oysters eaten and tissue mass consumed were reduced compared to controls, but prey size selection was not affected. Handling times and profits (g dry tissue/handling time) of snails feeding on oysters did not differ between snails in wave simulators and controls. Over both treatments, small oysters reduced snail handling times but provided profits similar to that of large oysters. In contrast, field experiments showed that wave exposure was correlated with an increase in the number of oysters …

Bovine Leukemia Virus (BLV) is an exogenous type-C retrovirus which can induce an aleukemic (AL) stage, a persistent lymphocytotic (PL) stage, or a lymphosarcomatous (LS) stage in infected animals. After experimental infection with BLV, whole blood and serum samples were obtained from 12 holstein heifers for 103 weeks and used to correlate clinical signs (visual observation and white blood cell counts) with changes in humoral immune response (Agar gel immunodiffusion using envelope gp50 as primary antigen, and ELISA and Western immunoblots using whole BLV as antigen). Juvenile penaeid shrimp were studied by a variety of methods in a southwest Louisiana, brackish marsh. Most of the work was conducted in two study ponds, one with a fixed-crest weir and the other without. In mark and recapture studies, both brown shrimp Penaeus aztecus and white shrimp P. setiferus were found to grow faster in marsh behind a fixed-crest weir. There was no apparent weir effect on mortality of either species. Brown shrimp emigrated an average of 12 d later from marsh behind the weir; white shrimp were sometimes delayed by the weir. The biosynthesis of a 36 kilodalton polypeptide of Chlamydomonas reinhardtii was induced by photoautotrophic growth on low CO2. Fractionation studies using the cell-wall-deficient strain of C. reinhardtii, CC-400, showed that this polypeptide was different from the low CO2-induced periplasmic carbonic anhydrase. In addition, the 36 kilodalton polypeptide was found to be localized in intact chloroplasts isolated from low CO2-adapting cultures. This protein may, in part, account for the different inorganic carbon uptake characteristics observed in chloroplasts isolated from high and low CO2-grown C. reinhardtii cells.

Freshly precipitated iron or manganese oxides were added to surface sediments from a salt marsh and from the intertidal region of Lowes Cove, Maine. In the presence of added manganese, sulfate was formed under anoxic conditions, suggesting a manganese dependent sulfide oxidation. Sulfate formation was not observed with iron additions. Sulfate reduction was substantially but not completely inhibited by either metal oxide, even though both were added at levels well in excess though both were added at levels well in excess of natural concentrations. Rates of methane emission from intact cores were measured during anoxic dark and oxic light and dark incubations. Rates of methane oxidation were calculated on the basis of oxic incubations by using the anoxic emissions as an estimate of the maximum potential flux. This technique indicated that methane oxidation consumed up to 91% of the maximum potential flux in peat sediments but that oxidation was negligible in marl sediments. Oxygen microprofiles determined for intact cores were comparable to profiles measured in situ. Thus, the laboratory incubations appeared to provide a reasonable approximation of in situ activities. Glucose transport in skeletal muscle is mediated by two distinct transporter isoforms, designated muscle/adipose glucose transporter (Glut4) and erythrocyte/HepG2/brain glucose transporter (Glut1), which differ in both abundance and membrane distribution. The present study was designed to investigate whether differences in insulin responsiveness of red and white muscle might be due to differential expression of the glucose transporter isoforms. Glucose transport, as well as Glut1 and Glut4 protein and mRNA levels, were determined in red and white portions of the quadriceps and gastrocnemius muscles of male Sprague-Dawley rats (body wt. approx. 250 g). Glycosylation can affect the physical and biochemical properties of the polypeptide chain in glycoproteins. Asparagine‐N‐linked polylactosaminyl glycosylation of the chymotryptic 44‐kDa gelatin‐binding domain from human placental fibronectin confers protease resistance [Zhu, B. C. R., Fisher, S. F., Panda, H., Calaycay, J., Shively, J. E. & Laine, R. A. (1984) J. Biol. Chem. 259, 3962–3970] and weakens the binding to gelatin [Zhu, B. C. R. & Laine, R. A. (1985) J. Biol. Abstract The patterns and controls of methane oxidation in a Danish wetland sediment have been determined using a combination of slurry and intact core techniques. Results from slurries indicated that methane oxidation was effectively inhibited by low concentrations of nitrapyrin (9 μM) and acetylene (0.5 μM) but that oxidation was relatively insensitive to pH between 6 and 8; in addition, high concentrations of ammonia (1 mM) decreased oxidation, especially at alkaline pH. Kinetic analyses of methane oxidation in slurries indicated that Vmax was high relative to values reported for other sediments, that Vmax changed seasonally, that Km was consistently low …

Abstract Anaerobic formation of dimethylsulfide (DMS) and methylmercaptan (MSH) in anoxic sulfide‐containing slurries from marine and fresh water sediments was stimulated by addition of syringate (4‐hydroxy,3,5,‐dimethoxybenzoate) and 3,4,5,‐trimethoxybenzoate. The release of DMS and MSH occurred during the consumption of the aromatic monomers and ceased after their depletion. DMS was the dominant methylated sulfur compound in fresh water sediments, in contrast to marine sediments where MSH was predominant. No production of volatile organic sulfur compounds was observed in slurries containing gallate (3,4,5,‐trihydroxybenzoate) or in autoclaved controled. Chromosomal areas from metaphase spreads of male bovine leukocytes were digitized and sex chromosomes identified using videomicroscopy. Autosomal areas were ranked in descending order within a cell and assigned to two categories based on alternating rank. X and Y chromosome areas were assigned to respective categories. Areas were divided by 4 to make their sum equivalent to sperm DNA content. Data were analyzed before and after inclusion of sex chromosomal areas. The Smackover Formation in the southeastern Mississippi salt basin has been buried to maximum depths of 3 to over 6 km and subjected to maximum temperatures of 100 to over 200 $\sp\circ$C. This formation contains a lower laminated mudstone member, a middle mudstone to wackestone member, and an upper grainstone member. In Clarke County, the upper Smackover member exhibits a north-south gradient in dolomite content, original mineralogy of ooids, and lithology. These regional gradients resulted from an increase in Mg/Ca concentrations of Smackover sea water northward across the carbonate platform. Shallow burial diagenesis ($<$4.5 km) was dominated by calcite cementation. Calcite cements in Clarke County are luminescently zoned and exhibit a consistent sequence of luminescent zonation. However, the intensity of luminescence and number of zones in the calcite cements vary across the county. This is due to variation in the chemical composition of the host carbonate rock and local hydrology. Deep burial diagenesis ($>$4.5 km and $<$7 km) is dominated by calcitization of anhydrite, precipitation of post-bitumen calcite cement, and formation of elemental sulfur. The chemical composition of post-bitumen calcite cement suggests precipitation from a pore water similar to the present Smackover brine. The high temperature replacement of anhydrite by calcite, the presence of post-bitumen calcite cement, the sulfur isotopic composition of sulfates and elemental sulfur, and the high concentration of hydrogen sulfide indicate that rapid thermochemical sulfate reduction of solid sulfate is taking place in the deep part of the Mississippi salt basin at temperatures of $>$150 $\sp\circ$C. …$7>$4.5>

Over the past decade there has been growing concern about the syndrome of bulimia nervosa. Current theoretical models of the etiology and symptomatology of bulimia predict that the psychopathology of the eating disorder worsens over time. The specific purposes of this study was to examine the effect of onset age and duration of the disorder on the primary and secondary psychopathology of bulimia nervosa at the time of presentation for treatment. The results suggest that binging and purging worsen with increasing duration and older age of onset, accompanied by increased weight and greater distortion of body size estimation. Functional activities of monocyte-derived macrophages and serum killing ability in a population of Brucella abortus-naive cows were evaluated. The ability of these macrophages to phagocytize and control intracellular survival of B.abortus and to produce oxidant was measured. Fresh normal bovine sera killed significantly more B.abortus than reactor serum or fetal bovine serum. When heated to destroy complement, none of the sera were capable of significant killing. The ability of sera from B.abortus-naive cows to kill the bacteria was normally distributed, with all animals within $\pm$ 2 sd of the mean.
