Recently, it has been shown that the high species richness in western Amazonia is related to habitat heterogeneity. Some habitats are rapidly disappearing or deteriorating as a result of anthropogenic activities, resulting in loss of species. One such unique habitat is white-sand forest (hereafter WSF), which is patchily distributed and restricted to a few areas in the northern Peruvian Amazonia. The kinetics of fecal coliforms (FC) decay and regrowth were analyzed under laboratory conditions using filtered dairy wastewater under anaerobic conditions. The mean specific growth rates during the regrowth phase, µr , in the batch study were 1.79, 1.46, and 1.27d-1 for initial organic carbon concentrations of 478, 235 and 127 mg/L COD, respectively. The substrate concentrations had a significant impact on the FC regrowth. A maximum specific growth rate (mmax) of 1.92 d-1, and half-saturated coefficient (ks) of 60.92mg/L were determined from these data. A Tn7-based transposition system carrying a kanamycin resistance gene has been used to disrupt a known virulence gene of Brucella melitensis 16M and Brucella abortus 2308 in-vitro. The suicide plasmid pBAvirB1::kan-29 was created, in part, by cloning the B. abortus virB1 gene into a vector that is not stably maintained in Brucella spp. The cloned virB1 gene was then mutagenized by in-vitro transposition of a Tn7-based transposon. This construct was used in-vivo to insert a disrupted virB1 gene in place of the wild type virB1 found in B. abortus 2308 and B. melitensis 16M by homologous recombination. The resulting virB1 …

The present work aimed at recovering, purifying, and testing rice cystatin or oryzacystatin I (OCI) bioavailability, to provide scientific evidence that rice cystatin can be used as a functional food ingredient. OCI was extracted from rice bran using 25 mM sodium phosphate buffer containing 0.15 M NaCl at pH 7.0. The resulting homogenate was heat treated, cooled and precipitated with ammonium sulfate. The recovered protein was dialyzed against 50 mM sodium acetate buffer pH 4.8 and sequentially purified by cation exchange, size exclusion, and anion exchange chromatography. Age ratio estimates obtained annually by the Cooperative Waterfowl Parts Collection Survey (PCS) serve as important estimates of annual waterfowl recruitment. To determine if age and sex ratios are biased due to repeat sampling of hunters across years, I examined PCS data collected from 1991-2000. Mean seasonal harvest increased with number of consecutive years hunters responded to the PCS. Proportions of juveniles in the mallard (Anas platyrhynchos) harvest and harvest of all species combined decreased with increasing seasonal harvest level. Efficacy of Bt corn hybrids expressing an insecticidal delta-endotoxin through different transformation events was evaluated in Louisiana. Efficacy levels were determined for southwestern corn borer (SWCB), Diatraea grandiosella Dyar; sugarcane borer (SCB), Diatraea saccharalis (F.); corn earworm (CEW), Helicoverpa zea (Boddie); and fall armyworm (FAW), Spodoptera frugiperda (J. E. Smith). Insect (neonates and third instars) susceptibility was evaluated on Bt and non-Bt corn plants (V6 and R1 stages) in field plots and plant tissue in laboratory studies. Bt corn hybrids with MON810 and CBH351 transformation events sustained significantly (P < 0.05) less injury caused by SWCB, SCB, and FAW during infestations at mid-whorl stage. SWCB, SCB, and CEW infestations during silking stage were significantly reduced in Bt hybrids with the MON810 event. However, CEW infestations …

This study focuses on radii excavated from the Poole-Rose ossuary and analyzes the occurrence and patterning of degenerative joint disease (DJD) on the proximal and distal joint surfaces. The Poole-Rose ossuary, located in eastern Ontario, is dated to A.D. 1550 +/- 50. The Poole-Rose population, dating to the Late Woodland period, were agricultural in their subsistence activities. The disarticulated patterning of the skeletal remains suggests this site was associated with the “Feast of the Dead,” a mass interment burial ceremony. This ceremony took place about every eight to twelve years. Frequencies of lipping, porosity, and eburnation were reported in degree …

Sex determination of adult remains has become a well-established practice in forensic anthropology. However, the assessment of sex in fetal remains is a highly contested subject. The purpose of this research is to ascertain whether fetal and early infant specimens can be sexed accurately using five bones of the cranium and the mandible. Little is known regarding the feeding behavior of many shark species. Even less is known about shark feeding habits in Louisiana coastal waters. The stomach contents of gillnet-captured blacktip sharks (n=356), Carcharhinus limbatus, and Atlantic sharpnose sharks (n=55), Rhizoprionodon terraenovae, were examined in this study. Roughly half of the blacktip stomachs (52%) and sharpnose stomachs (45%) contained prey items. The primary prey item in terms of percent number, occurrence, and weight for blacktips and sharpnose was the gulf menhaden, Brevoortia patronus. Both blacktips and sharpnose appear to mainly be piscivores; however, no members of Sciaenidae, the most common family of …

Achondroplasia and hypochondroplasia are two forms of skeletal dysplasias caused predominantly by single base mutations in the fibroblast growth factor receptor 3 gene (FGFR-3). The mutation for achondroplasia is a G1138A/C substitution and the mutation for hypochondroplasia (occurring about 50% of the time) is a C1620A/G substitution. Recent genetic studies have shown that spontaneous mutations for achondroplasia and hypochondroplasia occur exclusively on the paternally derived chromosome, suggesting that these mutations occur preferentially during spermatogenesis. For unknown reasons, the mutation rates at these FGFR-3 nucleotides appear to occur at a much higher frequency than nucleotide specific mutation rates observed in other …

Roadside death memorials are a response to the sudden, tragic death of a loved one; and are appearing with increasing regularity in developed and developing countries across the globe. In Mexico, however, wayside memorials and shrines of religiosity are a centuries-old tradition. This work, an effort to understand why the exact location of a person’s death is so important that a sacred place must be created where no place is intended, is basic and exploratory research. The discussion is positioned at the cross-section of three on-going conversations: 1) current discourses on Conjure in African American women’s literature, 2) analyses of Africanisms in black culture, and 3) previous scholarship on recurring topics in African American women’s writing. Understanding the influence of different perturbations on the fate of spilled oil in marine ecosystems is useful in assessing the environmental impact and remedial investigation. The effect of flooding and spill recurrence on the fate of an experimental crude oil spill (2 L/m2) was investigated using salt marsh intact cores, incubated for about 3 months, by monitoring residual petroleum hydrocarbons, heterotrophic microbial activity (fluorescein diacetate assay) and soluble organic carbon (SOC). The effects of summer soil solarization on the production and economics of four lettuce cultivars were evaluated in two plantings conducted during the fall growing season of 2001, to determine the feasibility of integrating strip-solarization in plasticulture cropping systems. Soil was solarized for 53 and 34 days for the first and second plantings, respectively, using transparent (T4, T5) and black films (T6) that were kept in place as plastic mulches in raised beds through the fall season. This research investigated the capability of wetland soils to function as a medium for the degradation of 1,1,2,2-TeCA and its daughter products, and the influence of different hydrogen donors and nutrients on this process. The rate constants calculated for this study, show that removal of 1,1,2,2-TeCA is occurring in the microcosms receiving hydrogen donors as well as the biotic controls. The parent compound was undergoing complete degradation to ethene, which was confirmed by ethene analysis, as well as monitoring the intermediate compounds. The goals of this thesis were to: 1) induce tetraploidy in zebrafish; 2) characterize single-female spawning in Nile tilapia for egg collection; 3) design and construct hatching systems for tilapia eggs; 4) induce tetraploidy in Nile tilapia. Tetraploidy was induced in zebrafish Danio rerio, by applying a 41 C heat shock to eggs for 2 min at 11, 13, 15, 17, or 20 min after fertilization (AF). A trend of increasing percent tetraploid yield occurred the later the shocks were applied. Shocks applied at 11 min AF yielded 1% tetraploidy and shocks applied at 20 min AF yielded 10%. There …

This thesis describes the development of a particle tracer technique for making short-term, in situ measurements of aquatic bed sediment biodiffusion coefficients. The bioturbation process in the upper sediment layers of streams, lakes, estuaries, and the marine environment moves particles and porewater. When present, organic chemicals, metals, colloids, etc., are transported across these layers and exchanges may occur at the sediment-water interface. Fickian biodiffusion coefficients that characterize such particle movements, Db (cm2/yr), are used for assessing chemical diagenesis rates and contaminant fluxes and are specific to each site. City Park Lake is a shallow, subtropical, urban hypereutrophic lake located in Baton Rouge, Louisiana. By the late 1970’s, this artificial lake had experienced extreme eutrophication and suffered from frequent algal blooms and fish kills. Restoration of City Park Lake occurred in 1983 and consisted of dredging nutrient-laden sediments and the rehabilitation of sewage pipelines. Pre- and post-restoration water quality monitoring programs have provided valuable information by which to gage the long-term success of the restoration work. The current study evaluated the effect of frozen storage and hydration under vacuum on density and breaking strength of the rat femoral neck. Femurs were frozen in saline for periods of 15, 34, 122, or 831 days. No significant effect on bone density was detected for freezing periods of 15, 34, and 122 days, indicating that frozen storage of specimens in saline is moisture-preserving for periods up to four months. Freezing periods of 34 and 122 days were used to examine the effect of frozen storage on bone biomechanical bahavior. Plastic strain increased for the 34-day storage period (p=0.0453) and …

Governmental agencies have set regulatory values on the concentration of dye-related color that can be released in textile mill effluents. A proprietary biotic and abiotic technology treatment train was built at a candidate facility in northwestern Georgia to reduce the organic content and dye-related color content in the textile mill’s effluent. Laboratory experimentation began with bench scale 4 L immobilized microbe (IMBR) bioreactor tests to biologically treat primary wastewater streams, namely skein dye and space dye, from this candidate facility. Three pig (Sus scrofa L.) carcasses were exposed during the summer in three different biogeoclimatic zones ranging in elevations from 6,700 to 11,100 feet in order to determine the rates of carrion decomposition and arthropod succession patterns on carrion in southwest Colorado. The carcasses were exposed in three scenarios: sun-exposed, shaded, and sun-exposed/shaded. Of the total 63 taxa collected, thirty species overlapped between two biogeoclimatic zones. A strong elevational preference is indicated for the Sarcophagidae taxon. A previously undescribed Boettcheria species was collected at 11,100 feet. Smooth cordgrass (S. alterniflora) is a perennial grass that dominates the salt marsh in tidal wetlands along the Atlantic and Gulf Coast of North America and has been used for preventing soil erosion and restoring wetlands. Accessions collected from south Louisiana were studied to investigate flowering phenology, pollen viability, crossability, and seed production. S. alterniflora exhibited protogynous flowering where stigmas exserted 2 to 5 days from the floret prior to anthesis. Pollen shed primarily between 8:00 and 10:00 AM.
