Localization And Characterization Of Phenamil-Sensitive Na+ Influx In Isolated Rainbow Trout Gill Epithelial Cells, Scott D. Reid, G. S. Hawkings, F. Galvez, G. G. Goss

Percoll density-gradient separation, combined with peanut lectin agglutinin (PNA) binding and magnetic bead separation, was used to separate dispersed fish gill cells into sub-populations. Functional characterization of each of the sub-populations was performed to determine which displayed acid-activated phenamil- and bafilomycin-sensitive Na+ uptake. Analysis of the mechanism(s) of 22Na+ influx was performed in control and acid-activated (addition of 10 mmoll-1 proprionic acid) cells using a variety of Na+ transport inhibitors (ouabain, phenamil, HOE-694 and bumetanide) and a V-type ATPase inhibitor (bafilomycin). Background: Four multicenter Networks (GenNet, GENOA, HyperGEN, SAPPHIRe) form the National Heart, Lung and Blood Institute Family Blood Pressure Program (FBPP), to search for hypertension/blood pressure (BP) genes. The networks used different family designs and targeted multiple ethnic groups, using standardized protocols and definitions. Linkage genome scans were done on samples within each network (N = 6245 relatives). Methods: The evidence was synthesized using meta-analysis. Results: Combining ethnic groups, no region reached LOD >2, but several small peaks were identified, including chromosome 2p where two other recent reports find hypertension linkage. Ciliary neurotrophic factor (CNTF) is primarily known for its roles as a lesion factor released by the ruptured glial cells that prevent neuronal degeneration. However, CNTF has also been shown to cause weight loss in a variety of rodent models of obesity/type II diabetes, whereas a modified form also causes weight loss in humans. CNTF administration can correct or improve hyperinsulinemia, hyperphagia, and hyperlipidemia associated with these models of obesity. Lake Vida, one of the largest lakes in the McMurdo Dry Valleys of Antarctica, was previously believed to be shallow (<10 >m) and frozen to its bed year-round. New ice-core analysis and temperature data show that beneath 19 m of ice is a water column composed of a NaCl brine with a salinity seven times that of seawater that remains liquid below - 10°C. The ice cover thickens at both its base and surface, sealing concentrated brine beneath. The ice cover is stabilized by a negative feedback between ice growth and the freezing-point depression of the brine. The ice cover …

The crystal structures of the human liver 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase in three different liganding states were determined and compared with those of the rat testis isozyme. A set of amino acid sequence heterogeneity from the two distinct genes encoding the two different tissue isozymes leads to both global and local conformational differences that may cause the differences in catalytic properties of the two isozymes. The sequence differences in a β-hairpin loop in the kinase domain causes a translational shift of several hydrophobic interactions in the dimeric contact region, and its propagation to the domains interface results in a 5° twist of the …

Two centuries of land use in the Mississippi River watershed are reflected in the water quality of its streams and in the continental shelf ecosystem receiving its discharge. The most recent influence on nutrient loading-intense and widespread farming and especially fertilizer use-has had a more significant effect on water quality than has land drainage or the conversion of native vegetation to cropland and grazing pastures. The 200-year record of nutrient loading to offshore water is reflected in the paleoreconstructed record of plankton in dated sediments. The effects of nutrient loading from the Mississippi River basin on the areal extent of hypoxia in the northern Gulf of Mexico were examined using a novel application of a dissolved oxygen model for a river. The model, driven by river nitrogen load and a simple parameterization of ocean dynamics, reproduced 17 yr of observed hypoxia location and extent, subpycnocline oxygen consumption, and cross-pycnocline oxygen flux. With Monte Carlo analysis, we illustrate through hindcasts back to 1968 that extensive regions of low oxygen were not common before the mid-1970s. Studies of incineration of surrogates for hazardous wastes are conducted in the pilot-scale rotary kiln incinerator (RKI) at Louisiana State University (LSU) in Baton Rouge, Louisiana. The purpose of the research is to investigate methods of treating and destroying hazardous wastes in a cost-effective and environmentally sound way. The objective is to provide process data that will contribute to increased knowledge for RKI design and operation. In this study, I experimentally examined how the landscape matrix influenced the movement, oviposition behavior, and spatial distribution of Anagrus columbi, a common egg parasitoid of the planthopper Prokelisia crocea. Both species exist among discrete patches of prairie cordgrass (Spartina pectinata), the sole host plant of P. crocea. Based on out-planted cordgrass pots bearing host eggs (to assess parasitism) or sticky leaves (traps for adult A. columbi), I found that the distribution of adult female A. columbi and pattern of ovipositions within a cordgrass patch were strongly matrix dependent. To date, there is a lack of well-controlled field experiments that disentangle the effects of the intervening matrix from other landscape variables (e.g., patch geography or quality) that might influence animal dispersal among patches. We performed a field experiment to investigate how the movement of a delphacid planthopper (Prokelisia crocea) among discrete patches of prairie cordgrass (Spartina pectinata) is affected by the composition of the matrix (mudflat, native nonhost grasses, and the introduced grass smooth brome [Bromus inermis]). Disturbances that are strongly linked to global climatic cycles may occur in a regular, predictable manner that affects composition and distribution of ecological communities. The El Niño Southern Oscillation (ENSO) influences worldwide precipitation patterns and has occurred with regular periodicity over the last 130 000 years. We hypothesized that ENSO, through effects on local weather conditions, has influenced frequency and extent of fires within Everglades National Park (Florida, USA). The rice water weevil, Lissorhoptrus oryzophilus Kuschel, is the most important insect pest in rice, Oryza sativa L., in the United States, Prior research indicates that rice water weevils feed primarily on monocotyledonous plants. Many monocot weeds occur in rice fields, but little is known about rice water weevil-weed interactions in rice. Host utilization of the rice water weevil was evaluated on rice, cultivar 'Cocodrie', and seven weeds commonly found in rice fields in preference and life cycle compatibility tests in the greenhouse. Clinical, serological, and pathological abnormalities observed in Holstein cows naturally infected with bovine lentivirus 1 bovine immunodeficiency virus (BIV) and other infections were progressive and most commonly associated with weight loss, lymphoid system deficiency, and behavioral changes. Clinical evidence of meningoencephalitis was dullness, stupor, and occasional head or nose pressing postures. The polymerase chain reactions associated the BIV provirus with the lesions in the central nervous system and lymphoid tissues. Multiple concurrent infections developed in retrovirally infected cows undergoing normal stresses associated with parturition and lactation. De novo LINE-1 (long interspersed element-1, or L1) retrotransposition events are responsible for ∼1/1,000 disease-causing mutations in humans. Previously, L1.2 was identified as the likely progenitor of a mutagenic insertion in the factor VIII gene in a patient with hemophilia A. It subsequently was shown to be one of a small number of active L1s in the human genome. Here, we demonstrate that L1.2 is present at an intermediate insertion allele frequency in worldwide human populations and that common alleles (L1.2A and L1.2B) exhibit an ∼16-fold difference in their ability to retrotranspose in cultured human HeLa cells. Although Africa has played a central role in human evolutionary history, certain studies have suggested that not all contemporary human genetic diversity is of recent African origin. We investigated 35 simple polymorphic sites and one Tn microsatellite in an 8-kb segment of the dystrophin gene. We found 86 haplotypes in 1,343 chromosomes from around the world. Although a classical out-of-Africa topology was observed in trees based on the variant frequencies, the tree of haplotype sequences reveals three lineages accounting for present-day diversity. Electrospray ionization and collision induced dissociation on a triple quadrupole mass spectrometer were used to determine the effect of spatial crowding of incremented alkyl groups of two anomeric pairs of peralkylated (methyl to pentyl) disaccharides (maltose/cellobiose and isomaltose/gentiobiose). Protonated molecules were generated which underwent extensive fragmentation under low energy conditions. For both the 1 → 4 and 1 → 6 α and β isomers, at comparable collision energies the methyl derivative exhibited the least fragmentation followed by ethyl, propyl, butyl, and pentyl. Background: Epidemiologic studies show that sudden surges in ambient particulate matter (PM) levels can trigger acute asthma exacerbations. Although diesel exhaust particles (DEPs) act as an adjuvant for allergic sensitization, this is a delayed response and does not explain acute PM effects on airway hyperreactivity (AHR). Objective: Our aim was to determine the acute effects of DEPs on AHR using a mouse model. Methods: Three protocols were developed, 2 of which require OVA sensitization, whereas the third was OVA independent. The transfer of organisms among patches is a key process influencing the spatial structure and regional dynamics of a population; yet, detailed experimental studies of animal movement among patches are uncommon. I performed a series of mark-recapture studies to quantify the movement of a planthopper, Prokelisia crocea (Hemiptera: Delphacidae), among discrete patches of its host plant, prairie cordgrass (Spartina pectinata). Results from these dispersal studies were used to predict the natural distributions and to characterize the spatial population structure of P. crocea. Objective: Interest in mapping genetic variants that are associated with obesity remains high because of the increasing prevalence of obesity and its complications worldwide. Data on genetic determinants of obesity in African populations are rare. Research Methods and Procedures: We have undertaken a genome-wide scan for body mass index (BMI) in 182 Nigerian families that included 769 individuals. Results: The prevalence of obesity was only 5%, yet polygenic heritability for BMI was in the expected range (0.46 ± 0.07). Field and laboratory studies evaluated the influence of selected crop hosts on Helicoverpa zea (Boddie) population dynamics in relation to genetically engineered Bt (Bollgard) and non-Bt cottons. Host specific H. zea colonies were initiated with a colony originally collected from sweet corn. The colony was allowed to complete one generation on meridic diet then split into cohorts and allowed to complete one generation on field corn, grain sorghum, soybean, cotton, or meridic diet in individual 29.5 ml plastic cups. Terpene-Induced Morphological Changes To Exoskeleton Of Formosan Subterranean Termites (Isoptera: Rhinotermitidae): Toxic Effects Of Cis-Nerol, Betty C.R. Zhu, Gregg Henderson, Huixin Fei, Xiao Ying, Roger A. Laine

The terpenoid, cis-nerol was found to be highly toxic to the Formosan subterranean termite, Coptotermes formosanus Shiraki. Morphological abnormalities were observed in the exoskeleton including the spiracles and trichoid sensilla using scanning electron microscopy after exposure of termites to cis-nerol for 2 h. Proteins were found in a water-soluble fraction collected from the whole termite body exposed to cis-nerol for 60 min. It appears that terpenoids cause damage to cell membranes resulting in a significant loss of proteins.

Diel Vertical Migration Of Cladocerans And Copepods In The Atchafalaya River Basin Floodplain, William E. Kelso, D. Allen Rutherford, Norman L. Davidson

We used bal and nocturnal zooplankton collections at 30 sites in the Atchafalaya River Basin (ARB) floodplain (Louisiana) to assess inter-habitat differences in diel vertical movements of numerically abundant cladocerans and copepods during the flood pulse. The magnitude of diel movement at each site was quantified with an index of vertical migration (VM)=log (nocturnal abundance+1)—log (diurnal abundance+1), and sites were grouped into lake (n=12), channel (n=9), and swamp (n=9) habitats based on principal components analysis of physicochemical characteristics. The cladocerans Diaphanosoma birgei and Moina micrura and the copepods Mesocyclops edax, Eurytemora affinis, and Acanthocyclops vernalis all exhibited siecantly greater VM …
