The availability of large amounts of genetic data from the mitochondrial DNA of species has created an unprecedented opportunity for the study of evolutionary processes. Being our closest relatives on the evolutionary tree the primates are a prime candidate for the study of evolutionary processes. The ability of removing toxic organic compounds in natural systems is important due to the capacity of these toxicants to increase risk of diseases when they are ingested by humans. This study developed a mechanistic model to estimate the removal efficiency of toxic organics in wetlands using the tanks-in-series model. Sensitivity analyses were performed for different values of the hydraulic loading rate and two kinds of wetlands: bottomland hardwood forest and freshwater marsh. This thesis addresses optimization techniques for cryopreservation of biological systems. A part of the thesis (Chapter 2) reports a simplified procedure to predict the optimal rate of freezing biological systems once the cell level parameters are known a priori. The key cell level parameters investigated were, reference permeability of the membrane to water (Lpg), apparent activation energy (ELp), inactive cell volume (Vb), diameter (D), and the ratio of the available surface area for water transport to the initial volume of intracellular water (SA/WV). The functional analysis methodology developed by Iwata, Dorsey, Slifer, Bauman, and Richman (1982/1994) has been successfully used to identify the variables that maintain the problem behavior of individuals with developmental disabilities. However, in some cases, the results of functional analysis may be inconclusive. Altering parameters of reinforcement, such as the schedule, the quality, or magnitude of the reinforcer, may increase the likelihood of obtaining clear functional analysis results. The island of Puerto Rico is confronting a crisis in waste management due to inadequate management from the local government, the decreasing number of landfills available, high population density, and paucity of places for waste disposal. This research develops a least-cost model for the disposal and transportation of non-hazardous solid waste. Location-allocation (LA) and Geographic Information Systems (GIS) software are used to analyze the efficiency of the present pattern of waste allocation and to identify a near-to-optimal assignment of waste for the landfills in operation today and the landfills that will be open by 2008. The objective of Experiment 1 was to determine the effect of bovine follicular fluid (bFF) on nuclear maturation. Treatment A (Control) oocytes were stained with Hoechst-33342 immediately after aspiration from follicles, whereas, oocytes in Treatment B were held in bFF for 12 hours at 38¢ªC and then stained to determine nuclear status. No significant difference was detected between treatment groups. Results indicate that bFF inhibits resumption of meiosis. A continuing challenge for wood and wood-based composite building materials has been durability--the power of resisting agents or influences which tend to cause changes, decay, or dissolution; lastingness. In the case of wood-based products, this often requires a resistance to biological degradation and moisture related dimensional instability (particularly in particle and flake or strand boards). The effect of cotton aphids, Aphis gossypii (Glover), on cotton plant development and yield was studied during 2000 and 2001. Tests were conducted at the Macon Ridge Station (Winnsboro, Louisiana) in 2000 and 2001, and at the Northeast Research Station (St. Joseph, Louisiana) in 2001. Tests were performed in irrigated and non-irrigated regimes using individual plant comparisons, micro-plots, and whole plots. In both years and at both locations, cotton aphid densities induced chlorosis, and a downward cupping of leaves during the infestation period. The insecticides, pymetrozine and thiamethoxam, were applied to control natural infestations of cotton aphids. Although pymetrozine and …

The usefulness of detecting genomic instability in the form of microsatellite instability (MSI) has been examined in a number of cancers. Squamous cell carcinoma (SCC) and adjacent normal and lichen sclerosis (LS) samples were taken from patients with radical vulvectomies. Vulvar SCC samples were compared with adjacent normal samples using four oligonucleotide markers (BAT-25, BAT-26, D10S208 and D10S587) found to be informative with other carcinomas. Using polymerase chain reaction techniques, 30 vulvar SCC DNA samples were examined for MSI. BAT-26 displayed the highest level of MSI and was considered to be the most sensitive marker in studying vulvar SCC. There …

Molecular dynamics (MD) simulations of the wheat antimicrobial peptide â-purothionin were carried out in explicit water (~14,000 atoms) using an all-atom model. The structural properties of the peptide as a function of MD simulation, temperature, and presence of the mono- and divalent metal ions were investigated. The accuracy and scalability tests of the code Peach 3.8 for our system were performed on the Intel Xeon-based Linux cluster SuperHelix at Louisiana State University (LSU). Cryopreservation of animal oocytes will permit germplasm of valuable or unique females to be preserved for extended times. The objective of this research was to derive a procedure to cryopreserve bovine oocytes by vitrification to be used as recipients for somatic cell nuclear transfer (SCNT). Caprine oocytes vitrified by the same procedure were assayed by cytological examination of microtubules. In the first two of three experiments, bovine oocytes matured in vitro were vitrified in a mixture of ethylene glycol (EG), dimethylsulfoxide (Me2SO) and trehalose, and then subjected to in vitro fertilization (IVF) or SCNT. Concerns over the adverse effects of pesticides on human health and the environment have led to the development of stricter pesticide regulations and the outright banning of many pesticides in the U.S. Bifenthrin and fipronil are important pesticides used in the nursery industry for the control of imported fire ants (Solenopsis invicta Buren, S. richteri Forel, and their hybrids) to meet the requirements of the Imported Fire Ant Quarantine. Microfabrication technology has proven to be a valuable tool for creating polymer-based devices utilized in chemical and biochemical assays. Although, reducing the size of the device allows for short analysis times and reduces the reagent demand to ultrasmall volumes (< 1 nanoliter), a resulting consequence is the constraint placed on the limits of detection associated with the detector hardware required for readout. To overcome such constraints, laser-induced fluorescence (LIF) is often employed as a detection method as it provides low detection limits, which approach the single molecule level. Unfortunately, most LIF systems do not offer the benefits of miniaturization, with the detector components (i.e. laser, optics, filters) often times requiring a much larger footprint compared to the device. Another readout strategy that has shown promise for these devices is conductivity detection. Detection can be accomplished using either conventional-size or microfabricated electrodes, which can be integrated on the device. Although conductivity has been commonly used to detect inorganic or small organic species, the potential for detection of biological species has received little attention. In this work, an integrated conductivity detector was developed for the analysis of amino acids, peptides, proteins, and oligonucleotides (double-stranded DNA). Using the detector, mass detection sensitivities in the range of 10-18 - 10-21 moles were achieved. To increase the throughput of the system a state-of-the-art, multichannel device with a conductivity array detector was devised. This device, which consists of a 16-channel fluidic network and a printed circuit board, is geared toward automating three-processing steps onto a single fluidic platform including purification, preconcentration and detection for downstream parallel processing.

A quality assurance testing of the ECLOX-M™, a rapid water quality indicator used in potential terrorist attacks, was evaluated for its adequacy in nine regional Louisiana water systems. The ECLOX-M™ system uses enhanced chemiluminescence to measure toxicity in water by integration of a mixture of luminol (C8H7O3N3), and an oxidant in the presence of a catalyst enzyme - horseradish peroxidase (HRP). Although great improvements have been achieved through traditional breeding methods, cotton breeders are facing many problems, i.e., narrow genetic base, inability to use alien genes and difficulty in breaking gene linkages. Genetic transformations and quantitative trait loci (QTL) analyses are main tools used by breeders to overcome these problems. In this dissertation, an optimized cotton regeneration system from shoot apices was developed. The Herpes Simplex Virus life cycle contains a number of membrane fusion events that must function properly to ensure a productive infection, including: virus attachment and entry into susceptible cells, de-envelopment at the outer nuclear lamellae, and virus-induced cell-to-cell fusion. I evaluated hypotheses regarding the nature of habitat specialization by comparing the microhabitat distribution and demography of L. cernua and P. basiramia, two Florida rosemary scrub habitat specialist species, with their habitat generalist congeners, L. deckertii and P. robusta. Specifically, I addressed the following two hypotheses: (1) that habitat specialist species may occur in a narrower range of microhabitat conditions than habitat generalist species, and (2) that demographic parameters of habitat specialist species may be more variable than those of their habitat generalist congeners. The research presented in this dissertation involves the synthesis, modification, characterization, and the application of polymeric surfactants in micellar electrokinetic chromatography (MEKC). Sodium undecenoyl-L-Leucinate (L-SUL) was synthesized and modified by use of alkali metal counterions, alcohols, and Triton X-102. Characterization of the modified poly-L-SUL was performed in order to elucidate the structural changes on the surfactants by use of four techniques. This family is well known for its cultivated edible fruits and seeds (mangos, pistachios, and cashews), dermatitis causing taxa (e.g., Comocladia, Metopium, Semecarpus, Toxicodendron, etc.), and lacquer plants (Toxicodendron and Gluta spp.). Generalized mixed model methodology and MCMC simulations were used to estimate genetic parameters for calving rate and calf survival with the normal, probit, and logistic models. Calving rate and calf survival were defined as 0 each time a cow failed to calf or a calf failed to survive to weaning age, otherwise they were set to 1. Data were available on 1,458 cows and on 5,015 calves. Chlamydomonas reinhardtii Dangeard does not have two major phospholipids, PS and PC. This fact renders C. reinhardtii a desirable system for investigations of the PE biosynthetic pathway and its regulation independent of PC and PS biosynthesis. The cDNA coding for ECT protein in C. reinhardtii was cloned from a cDNA library. The ECT cDNA encodes a protein of 443 amino acid residues. The ECT protein in C. reinhardtii has a repetitive internal sequence in its N- and C-terminal halves. The tempo of evolution and the causes of rate variation among lineages are central foci of evolutionary biology. I evaluated two hypothesized sources of variation in molecular evolutionary rate, and I applied a variable molecular clock to estimate the timescale of diversification in three families of Neotropical birds. First, I examined the phylogenetic evidence for molecular punctuated equilibrium, the hypothesis that speciation drives accelerated molecular evolution. Recent findings that rates of DNA evolution and speciation are linked implicate molecular punctuated equilibrium as an important cause of rate variation among lineages. Two new methods for the efficient parallel computation of the unsteady incompressible Navier-Stokes equations are presented. Such efficient methods are desired for large scale parallel computations of unsteady turbulent flows such as Direct Numerical Simulations (DNS). The performance of the new methods has a distinct advantage over the artificial compressibility method, in that the methods exhibit robust convergence for a variety of flow problems without extensive need for tuning computational parameters. A phage collection (M1114), which is a mixture of two collections, M710-3R and Silica 426 each derived from a human synthetic phage antibody library (Fab 2lox), was used further enriched for dextran binding using enzyme-linked immunosorbent assays (ELISA) screening. The effects of dextran concentration on phage binding affinity were tested using indirect sandwich ELISA on phage collections, M1114-m74 and AE-M1114-m74-2R. Most of the phage bound to dextran (T2000) coated on a sandwich ELISA.
