Caloric restriction (CR) is a strategy that attenuates aging in multiple nonhuman species. The Comprehensive Assessment of Long-term Effects of Reducing Intake of Energy (CALERIE) trials are part of a research program aiming to test the effects of CR on aging and longevity biomarkers in humans. Building on CALERIE phase 1, CALERIE phase 2 (CALERIE 2) was the largest study to date to assess sustained CR in healthy humans without obesity. In a 24-month randomized controlled trial comprising 218 participants at baseline, CALERIE 2 showed that moderate CR, 11.9% on average, induced improvements in aging-related biomarkers without adversely affecting psychological …

Aims/hypothesis: In vitro and rodent studies suggest that pioglitazone, a thiazolidinedione, can promote adipogenesis in adipose tissue (AT); however, there is a lack of in vivo studies in humans to support these findings. The objectives of this randomised, placebo-controlled, parallel-arm trial were to test if pioglitazone stimulates in vivo adipogenesis in the subcutaneous adipose tissue depots and if these measures were related to metabolic health outcomes in women with obesity. Office-based activity reduces sedentariness, yet no randomized controlled trials (RCTs) have assessed how such activity influences visceral adipose tissue (VAT). This study examined the effect of an office-based, multicomponent activity intervention on VAT. The WorkACTIVE-P RCT enrolled sedentary office workers (body mass index: 31.4 (standard deviation (SD) 4.4) kg/m2 ) to an intervention (n = 20) or control (n = 20) group. For 3 months, the intervention group received an office-based pedal desk, further to an intervention promoting its use and increased walking. The control group maintained habitual activity. Background:This study evaluated the potential of activating the fuel-sensing enzymes Adenine monophosphate (AMP)-activated protein kinase and the deacetylase sirtuin1, to promote weight loss. This study investigated the amendment of sugarcane bagasse biochar (SCBB) on soil fertility, crop yield, and nutrient loss in two different-textured soils under sugarcane (Saccharum spp.) production. Eleven megagrams per hectare of freshly incorporated biochar increased yield of plant cane by 22% in light-texture soil (LS) and 12% in heavy-textured soil (HS). Although the overall yield of the ratoon crop was lower, the biochar treatment produced 20 and 14% higher yields than the control at the LS and HS sites, respectively. Objective: The purpose of this study was to estimate the associations of genetically determined maternal blood glucose levels with obesity-related outcomes among children from pregnancies with and without gestational diabetes mellitus (GDM). Methods: A total of 1,114 mothers with (N = 560) and without (N = 554) GDM and their children were included in the present study. A maternal genetic risk score (GRS) for blood glucose was constructed on the basis of 17 single-nucleotide polymorphisms identified from a recent genome-wide association study. Regular physical activity improves physical and mental health across all ages, and provides many social, environmental, and economic co-benefits. Physical activity includes all forms of active recreation, sports participation, cycling and walking for transportation, as well as activity undertaken at home or for work. Despite well-established benefits, one quarter of adults and three quarters of adolescents are not sufficiently active. The policy and practice of promoting physical activity integrates multidisciplinary knowledge from across clinical and social medicine, exercise science, health psychology, public health, transport, and environmental studies, to improve global health. The scope of this article is to associate breakfast consumption (BC) with moderate to vi-gorous physical activity (MVPA) intensity and the nutritional status of children aged 9-11. The sample consisted of 432 children from São Caetano do Sul participating in the International Study of Childhood Obesity Lifestyle and the Environment. Data were collected between 2012 and 2013. The weekly BC was obtained using the self-reported method. To measure MVPA, children used ac-celerometers and were classified into two groups (< 60 versus ≥60 min/day). The nutritional status was presented by body mass index (BMI). Logistic regression models were used, adjusted for gender, age, race, parental educational level and eating score. The mean BC was 5.25 (CI95%: 5.1-5.4) days/week and the MVPA was 59.29 (CI95%: 57.3-61.7) min/day. In the total sample, 55.8% of the children did not perform ≥60 min/day of MVPA and 50.2% were overweight or obese. The daily BC was not associated with MVPA, however, it reduced the odds of children being overweight or obese (OR: 0.51; CI95%: 0.34-0.76; p=0.001). Daily BC reduced the odds of excess weight or obesity in children, though it did not increase the chances of children meeting the recommendations of MVPA. Public policies should encourage daily BC to prevent excess weight or obesity.

Objective: To evaluate the association between nutritional status, physical activity, sedentary behavior, diet, and lifestyle in childhood with respiratory diseases during adolescence. Methods: Prospective study conducted in Saõ Caetano do Sul, Saõ Paulo-Brazil, as part of the International Study of Childhood Obesity, Lifestyle and the Environment (ISCOLE). During childhood, indicators of lifestyle (body composition, physical activity, sedentary behavior, diet) and family environment were measured in 2012 and 2013. The inflammatory response is an obstacle to success in both allogeneic and autologous islet transplantation. In autologous islet transplantation (AIT), however, the recipient is also the donor, permitting pretreatment of donor/recipient for a controlled duration prior to transplantation. We sought to exploit this feature of (AIT) by pretreating donor/recipients with chronic pancreatitis undergoing total pancreatectomy and autologous islet transplantation (TPAIT) to test the hypothesis that peri-transplant treatment with the FDA-approved anti-inflammatory hydroxychloroquine (HCQ) improves graft function. Background: Family-based behavioral therapy is an efficacious approach to deliver weight management counseling to children and their parents. However, most families do not have access to in-person, evidence-based treatment. We previously developed and tested DRIVE (Developing Relationships that Include Values of Eating and Exercise), a home-based parent training program to maintain body weight among children at risk for obesity, with the intent to eventually disseminate it nationally alongside SafeCare, a parent support program that focuses on parent-child interactions. Introduction: Changes to mental, physical, and physiological functions drive the complex processes underlying the variable progression of human aging. Nutritional interventions are one of the most promising non-pharmacological therapeutics to attenuate aging in humans. This narrative review aims to describe the implications of moderate and prolonged calorie restriction (CR) in healthy adults without obesity that occur beyond weight loss. Dictionary learning and sparse coding techniques overcome limitations of traditional voxel-level analyses of task-based functional magnetic resonance imaging (fMRI) data by identifying broader temporal and spatial patterns of brain activity. However, prior applications of these methods to task-related fMRI data are not simultaneously optimized to find temporal patterns of activity that change in concert with changes in task conditions and spatial patterns that leverage existing neuroscience knowledge. As the population of elderly individuals in South Africa (SA) grows, there is a need to promote the continued health of these persons as they progress through the life cycle. Food Based Dietary Guidelines (FBDGs) for the SA elderly were developed to address this need. These thirteen guidelines for the elderly collectively offer a basis of health practices that the elderly can follow to ensure that they are taking the right steps toward maintaining their health. Studies have shown that the elderly are at a higher risk of developing malnutrition due to physiological and pathological changes. Several studies have confirmed that older South Africans have insufficient dietary diversity, resulting in nutritional deficiencies. Furthermore, poor and uninformed dietary choices are associated with the development of several diseases and increased mortality. The study was performed to investigate some changes in physicochemical characteristics and ginsenoside profiles of Korean ginseng after being subjected to immersion in fermented tea liquid (FTL) [at a ratio of ginseng powder: FTL; 1:4, 1:6, and 1:8, w/w] and steam treatments (with and/or without steam). The ratio at 1:4 w/w showed the highest yield of bioactive components and was, therefore, selected to evaluate the effects of steam conditions [temperatures (60, 80, and 100°C) and times (1, 2, and 3 hr)]. Excess abdominal visceral adipose tissue (VAT) is associated with cardiometabolic risk factors in adolescents. VAT is mainly measured using Magnetic Resonance Imaging (MRI), yet dual-energy x-ray absorptiometry (DXA) is more affordable and available. The purpose was to compare adolescent VAT values obtained by MRI and DXA. A sample of 330 adolescents 10–16 years of age were recruited (52.3% female, 58.5% White). Abdominal VAT was measured using a General Electric (GE) Discovery MRI scanner with imaging software. Background: Few studies have examined both parent and child preferences regarding family-based weight management programs (WMPs) delivered in primary care settings, especially among racial minority populations. The purpose of this study was to determine the perceptions that parents and their children/adolescents have about the components that should be included in a family-based WMP and to identify perceived preferences, benefits, and/or barriers to participation. Nutrient stoichiometry and trade-offs focusing on the mass balance of multiple nutrient elements play important roles in the response mechanism of plants to environmental stresses in wetlands. Nonetheless, little is known about the response dynamics and trade-offs of nutrient elements of wetland plants, especially potassium (K), under hydrological fluctuation conditions. Aims: To examine the effect of lifestyle (diet and physical activity) interventions on the prevalence of GDM, considering the method of GDM ascertainment and its association with early pregnancy characteristics and maternal and neonatal outcomes in the LIFE-Moms consortium. Methods: LIFE-Moms evaluated the effects of lifestyle interventions to optimize gestational weight gain in 1148 pregnant women with BMI ≥ 25 kg/m2 and without known diabetes at enrollment, compared with standard care. Background: Intentional weight loss is associated with lower risk of heart failure (HF) and atherosclerotic cardiovascular disease among patients with type 2 diabetes. However, the contribution of baseline measures and longitudinal changes in fat mass (FM), lean mass (LM), and waist circumference (WC) to the risk of HF and myocardial infarction (MI) in type 2 diabetes is not well established. Background: Individual perceptions of personal and national threats posed by COVID-19 shaped initial response to the pandemic. The aim of this study was to investigate the changes in residents' awareness about COVID-19 and to characterize those who were more aware and responsive during the early stages of the pandemic in Louisiana.

Calorie restriction (CR) is a potent modulator of longevity in multiple species. A growing body of evidence shows that sustained periods of CR without malnutrition improves risk factors involved in the pathophysiology of type 2 diabetes, cardiovascular diseases, cancer, and neurological disorders in humans. Innovative dietary strategies such as intermittent fasting and protein restriction have recently emerged as alternative approaches that improve markers of aging. Iron homeostasis offers a significant bacterial vulnerability because pathogens obtain essential iron from their mammalian hosts, but host-defenses maintain vanishingly low levels of free iron. Although pathogens have evolved mechanisms to procure host-iron, these depend on well-regulated iron homeostasis. To disrupt iron homeostasis, our work has targeted iron mobilization from the iron storage protein bacterioferritin (BfrB) by blocking a required interaction with its cognate ferredoxin partner (Bfd). Aedes albopictus, the Asian tiger mosquito, is an important nuisance mosquito species and known vector of arboviruses such as dengue, chikungunya, and Zika. Despite their cosmopolitan distribution around the world, there is a paucity of accurate predictive models based on rates of development at different temperatures (degree-day models). Objective: Estrogen protects animals from obesity through estrogen receptor α (ERα), partially by inhibiting overeating in animals fed ad libitum. However, the effects of estrogen on feeding behavior in hungry animals remain unclear. In this study, we examined the roles of 17β-estradiol (E2) and ERα in the regulation of feeding in hungry female animals and explored the underlying mechanisms. Methods: Wild-type female mice with surgical depletion of endogenous estrogens were used to examine the effects of E2 supplementation on acute refeeding behavior after starvation. Brain glucose-sensing neurons detect glucose fluctuations and prevent severe hypoglycemia, but mechanisms mediating functions of these glucose-sensing neurons are unclear. Here we report that estrogen receptor-α (ERα)-expressing neurons in the ventrolateral subdivision of the ventromedial hypothalamic nucleus (vlVMH) can sense glucose fluctuations, being glucose-inhibited neurons (GI-ERαvlVMH) or glucose-excited neurons (GE-ERαvlVMH). Hypoglycemia activates GI-ERαvlVMH neurons via the anoctamin 4 channel, and inhibits GE-ERαvlVMH neurons through opening the ATP-sensitive potassium channel. Further, we show that GI-ERαvlVMH neurons preferentially project to the medioposterior arcuate nucleus of the hypothalamus (mpARH) and GE-ERαvlVMH neurons preferentially project to the dorsal Raphe nuclei (DRN). Homeotherms maintain a stable internal body temperature despite changing environments. During energy deficiency, some species can cease to defend their body temperature and enter a hypothermic and hypometabolic state known as torpor. Recent advances have revealed the medial preoptic area (MPA) as a key site for the regulation of torpor in mice. The MPA is estrogen-sensitive and estrogens also have potent effects on both temperature and metabolism. Here, we demonstrate that estrogen-sensitive neurons in the MPA can coordinate hypothermia and hypometabolism in mice.
