Limited biological information about Phyllophaga ephilida, a major sweet potato pest in Louisiana, is available. In 2001 and 2002, a study was conducted in the laboratory to investigate the feeding preference of adult Phyllophaga ephilida (Say) (Coleoptera: Scarabaeidae) for the foliage of eight woody plant species: water oak (Quercus nigra L.), live oak (Quercus virginiana Mill.), red maple (Acer rubrum L.), slash pine (Pinus caribaea Morelet), pecan (Carya illinoensis (Wangenh) K. Objective: To assess the effects of energy dilution with non-fermentable and fermentable fibers on abdominal fat and gut peptide YY (PYY) and glucagon-like peptide (GLP)-1 expressions, three rat studies were conducted to: determine the effects of energy dilution with a non-ferment-able fiber, compare similar fiber levels of fermentable and non-fermentable fibers, and compare similar metabolizable energy dilutions with fermentable and non-fermentable fibers. The poorer countries of the world continue to struggle with an enormous health burden from diseases that we have long had the capacity to eliminate. Similarly, the health systems of some countries, rich and poor alike, are fragmented and inefficient, leaving many population groups underserved and often without health care access entirely. Cuba represents an important alternative example where modest infrastructure investments combined with a well-developed public health strategy have generated health status measures comparable with those of industrialized countries. Background: Weight gain in adulthood is common, from modest gains in developing countries to substantial increases in Western societies. Evidence of the importance of energy expenditure in adult weight change has been limited to studies conducted in Pima Indians, in whom resting energy expenditure (REE) was found to be inversely associated with weight gain. Objective: The aim was to determine whether REE was predictive of weight change in lean Nigerian adults. Design: Weight was measured in 744 adults on 2-4 occasions over 5.5 y. The components that define cereal-grain yield potential have not been well defined. The objective of this study was to collect many differing biological measurements from a long-term winter wheat (Triticum aestivum L.) study in an attempt to better define yield potential. Four treatments were sampled that annually received 0, 45, 90, and 135 kg N ha-1 at fixed rates of phosphorus (P) (30 kg ha-1) and potassium (K) (37 kg ha -1). Feedback from community interactions involving mutualisms are a rarely explored mechanism for generating complex population dynamics. We examined the effects of two linked mutualisms on the population dynamics of a beetle that exhibits outbreak dynamics. One mutualism involves an obligate association between the bark beetle, Dendroctonus frontalis and two mycangial fungi. The second mutualism involves Tarsonemus mites that are phoretic on D. frontalis ("commensal"), and a blue-staining fungus, Ophiostoma minus. The presence of O. minus reduces beetle larval survival ("antagonistic") by outcompeting beetle-mutualistic fungi within trees yet supports mite populations by acting as a nutritional mutualist. Objective: Peptide YY (PYY) and glucagon-like peptide-1 are important in the control of energy homeostasis and are both secreted from the gut in response to ingested nutrients. However, more studies are needed on nutrient regulation of their gene expression patterns in specific areas of the gut. This study detailed PYY and proglucagon (the gene that encodes glucagon-like peptide-1) gene expression patterns and regulation in the gut. We further examined the regulation of PYY and proglucagon mRNA by a diet containing fermentation- resistant starch (in vivo) and butyrate (in vitro). The T594M allele of the epithelial sodium channel β-subunit has been proposed as a gain-of-function mutation leading to salt-sensitive hypertension in blacks that is particularly responsive to the specific sodium channel antagonist amiloride. However, the positive associations derive from small convenience samples, and the amiloride challenge study lacked a control group. We determined whether the T594M allele was associated with hypertension and blood pressure (BP) response to amiloride in 2 well-characterized random population samples including 3137 Dallas County subjects and 1666 Jamaican blacks. We report on the fabrication and performance of a gel microfluidic chip interfaced to laser desorption/ionization (LDI) mass spectrometry with a time-of-flight mass analyzer. The chip was fabricated from poly(methylmethacrylate) with a poly(dimethyl siloxane) cover. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis was performed in the channel of the microfluidic chip. After electrophoresis, the cover was removed and either the PDMS chip or the PMMA cover was mounted in a modified MALDI ion source for analysis. During a two year population ecology study in a cave environment, 15 Eurycea (=Typhlotriton) spelaea were observed ingesting bat guano. Furthermore, E. spelaea capture numbers increased significantly during the time that grey bats (Myotis grisescens) deposited fresh guano. We investigated the hypothesis that this behaviour was not incidental to the capture of invertebrate prey, but a diet switch to an energy-rich detritus in an oligotrophic environment. Stable isotope assays determined that guano may be assimilated into salamander muscle tissue, and nutritional analyses revealed that guano is a comparable food source to potential invertebrate prey items. Prp19 is an essential splicing factor and a member of the U-box family of E3 ubiquitin ligases. Prp19 forms a tetramer via a central coiled-coil domain. Here, we show the U-box domain of Prp19 exists as a dimer within the context of the Prp19 tetramer. A high-resolution structure of the homodimeric state of the Prp19 U-box was determined by X-ray crystallography. Mutation of the U-box dimer interface abrogates U-box dimer formation and is lethal in vivo. The objective of this investigation was to evaluate the effect of scraping frequency (2× vs. 6× daily) on N volatilization from manure on the floor of a dairy free-stall barn. Three trials (crossover design) were conducted in the summers of 2001 and 2002, and in the winter of 2003. Nitrogen volatilization was estimated from the change in the N:P ratio in excreta at the time of excretion to the time when manure was scraped from the barn. Total N loss was considered a maximum estimate of NH3-N loss, because small amounts of nonammonia N may be volatilized. Nitrogen was determined …

Three experiments were conducted to investigate the feasibility of using crystalline methionine and lysine as protein supplements for lactating Holstein cows. In the first experiment, Met (DL-methionine) and Lys (L-lysine-HCl) were added to diets used in continuous culture bioreactors to estimate optimal concentrations for use in subsequent in vivo experiments. The second experiment measured ruminal fermentation and nutrient flow to the small intestine when Met and Lys were top-dressed on diets fed to nonlactating cows. Five experiments (Exp.) were conducted to determine the effects of phytase on growth performance and intestinal transit time in chicks fed nutritionally adequate diets and diets deficient in Ca and nonphytate P (nPP). In Exp. 1 and 2, chicks were fed a nutritionally adequate diet from 0 to 6 d or from 0 to 4 d posthatching; assay periods were 8 or 10 d; average initial BW were 98 or 79 g; and average final BW were 371 or 369 g, respectively. An increase in obesity prevalence among patients who initiate dialysis may influence the growth of the total ESRD population as a result of improved survival and decreased likelihood for transplantation. Temporal trends in mean body mass index (BMI) and obesity prevalence were examined among incident patients with ESRD by year of dialysis initiation between 1995 and 2002, and these trends were compared with those in the US population during this same period. Selected missense mutations in the proprotein convertase subtilisin/kexin type 9 serine protease gene (PCSK9) cause autosomal dominant hypercholesterolemia, whereas nonsense mutations in the same gene are associated with low plasma levels of low-density lipoprotein cholesterol (LDL-C). Here, DNA sequencing and chip-based oligonucleotide hybridization were used to determine whether other sequence variations in PCSK9 contribute to differences in LDL-C levels. The coding regions of PCSK9 were sequenced in the blacks and whites from the Dallas Heart Study (n = 3,543) who had the lowest (percentile) and highest (>95 th percentile) plasma levels of LDL-C. Objectives. An adequate description of the trends in cardiovascular disease (CVD) is not available for most of the developing world. Cuba provides an important exception, and we sought to use available data to offer insights into the changing patterns of CVD there. Methods. We reviewed Cuban public health statistics, surveys, and reports of health services. Results. CVD has been the leading cause of death since 1970. A 45% reduction in heart disease deaths was observed from 1970 to 2002; the decline in stroke was more limited. There are moderate prevalences of all major risk factors. Conclusions. The objective of this study was to use fluorescent in situ hybridization (FISH) to determine if a skew in the sex ratio was present at the level of the bovine round spermatid. Paraffin embedded bovine testicular tissue obtained from biopsies were used to perform FISH, and the seminiferous tubules were examined. Our findings show that the skew does exist within the round spermatids. Analysis of variance on X- and Y-chromosome bearing round spermatids as affected by sex, slide, picture set, and tubule was performed. Sex showed that the variation in round spermatid sex ratio differed between slides (P = 0.0001). …

The coastal wetlands play a central role in maintaining productivity of the Gulf of Mexico while protecting coastal regions during severe storms. To help offset the loss of coastal marsh, there is growing interest in the beneficial use of dredge spoil for reconstruction of eroded areas. Hydraulic dredges are routinely employed to maintain navigation and the unconsolidated sediments can be pumped considerable distances to create gently sloping wetland areas. Some areas of these reconstructed wetlands are rapidly colonized by Spartina alterniflora, the dominant vegetative species in saline areas under tidal influence. Previous studies have found the spaced-retrieval memory intervention technique to be successful in training people with probable Alzheimer’s disease (AD) to learn new, simple associations. In the present study, we expanded on these previous findings by training eight participants with probable AD to learn the names and pictures of countries via spaced retrieval. We demonstrated the flexibility of the technique, gave insight as to the effects of distracters on the spaced-retrieval training performance, and demonstrated the memorial effects of adding pictorial support to the text.

This research examines the contemporary practice of midwifery in Louisiana, a state that very early on had progressive legislation, yet remains a tough place for a midwife to practice. What, then, are the social forces that affect the ability to practice midwifery in Louisiana? I try to answer that question by examining the narratives of midwives and their clients, and evaluating the options these women have access to in this state. The narratives provide opportunities to observe the authoritarian knowledge of biomedicine in our society, and apply Foucault’s theory of power/knowledge. Vibrio vulnificus and Vibrio parahaemolyticus are natural inhabitants of estuarine environments of the Gulf of Mexico. V. vulnificus is the leading cause of death, while V. parahaemolyticus is a leading cause of foodborne gastroenteritis from the consumption of seafood in the United States. Refrigeration is commonly used as a preservation method to control the growth of microorganisms in food. The first study was conducted to determine if aminophylline cream application to the waist will reduce waist circumference compared to a control. Topical fat reduction from the thigh in women has been demonstrated, but local fat reduction in other areas or in men has not. Fifty men and women were randomized to 0.5% aminophylline cream to the waist twice a day or to no treatment to the waist. At week 12 there was a significant reduction in BMI from baseline that was not different between the groups. The reduction in waist circumference was 11 ± 1.0 cm in the aminophylline …

Prices of a market basket of 149 foods that could be combined to meet the 2005 Dietary Guidelines for Americans (DGA) were surveyed in 29 stores in an eight parish area of Southeast (SE) Louisiana. Two-week cycle menus and recipes were planned to meet the 2005 DGA in a culturally acceptable way. Linear Programming (LP) analysis was also performed on the market basket to determine the cost-minimizing combination of foods that will sufficiently meet the 2005 DGA. Ozone is an effective sanitizing agent against a broad spectrum of pathogenic and spoilage organisms. Optimization of treatment applications of ozonated water is needed for increased use in the food industry. An experimental apparatus and process has been developed to digitally measure ozone concentrations in processing water at the point of product application. Two application methods were evaluated. Ozone concentrations were measured rapidly at the point of product application. Traditional mayonnaise is manufactured with soybean oil (SBO) and egg-yolk containing ingredients. About 1/4 of American consumers have some forms of cardiovascular disease, accounting for >40% of all deaths in USA. Rice bran oil (RBO), a healthy lipid source, has cholesterol-lowering effects, and could be used to replace SBO in mayonnaise preparation. To take advantage of the health benefits associated with RBO, food products containing RBO need to be developed and characterized. This study was designed to understand the response of the National School Lunch Program (NSLP) and the Food Stamp Program (FSP) in southern Louisiana during Hurricanes Katrina and Rita. This study used portions of the PRECEDE/PROCEED model to develop the questions for the interviews and interpret the barriers and enabling and reinforcing factors with regard to changes in policy, budget, reporting, and program administration after the hurricanes.
