Experimental And Computational Tools For Single Cell Analysis In Cancer Diagnostics, Manibarathi Vaithiyanathan Jan 2020

Experimental And Computational Tools For Single Cell Analysis In Cancer Diagnostics, Manibarathi Vaithiyanathan

LSU Doctoral Dissertations

Substantial evidence shows that cellular heterogeneity commonly exists within an isogenic or clonal population. Whether in isolation or caused through a combination of the above events, cellular heterogeneity can dramatically influence cellular decision making and cell fate, however, this can be masked by the average response from a population. One approach to solve this issue is to analyze a population at the individual cell level. The goal of this work is to develop high-throughput experimental and computational platforms to screen and quantify single cancer cells for specific intracellular enzyme activities. An interdisciplinary approach was taken to 1) better understand the …

Impacts Of The Deepwater Horizon Oil Spill On Reproduction And Ontogeny Of Gulf Killifish (Fundulus Grandis), Charles Alexander Brown

LSU Doctoral Dissertations

On April 20, 2010, the Deepwater Horizon oil spill released an estimated 779 million liters of Macondo-252 crude oil into the Gulf of Mexico, making it the largest marine oil spill in history. Over a thousand kilometers of marshland that many species of fish use as a spawning grounds and nurseries was oiled, exposing breeding adult fish and their offspring to oil. My dissertation investigates the impacts of the Deepwater Horizon oil spill and associated remediation efforts on the reproduction and ontogeny of a sentinel ecotoxicological species, the Gulf killifish (Fundulus grandis). Concerns were raised regarding use of …

Influence Of Neighboring Clonal-Colonies On Aflatoxin Production By Aspergillus Flavus, Rebecca R. Sweany, Kenneth E. Damann Jan 2020

Influence Of Neighboring Clonal-Colonies On Aflatoxin Production By Aspergillus Flavus, Rebecca R. Sweany, Kenneth E. Damann

Faculty Publications

Aspergillus flavus is an ascomycete fungus that infects and contaminates corn, peanuts, cottonseed, and treenuts with acutely toxic and carcinogenic aflatoxins. The ecological function of aflatoxin production is not well understood; though not phytotoxic, aflatoxin may be involved in resisting oxidative stress responses from infection or drought stress in plants. Observation of aflatoxin stimulation in 48-well plates in response to increasing inoculated wells sparked an investigation to determine if A. flavus volatiles influence aflatoxin production in neighboring colonies. Experiments controlling several culture conditions demonstrated a stimulation of aflatoxin production with increased well occupancy independent of pH buffer, moisture, or isolate. …

A Study Of Particle-Laden Flows From Meso And Micro-Scale Perspectives, Daniel Guedes De Oliveira Jan 2020

A Study Of Particle-Laden Flows From Meso And Micro-Scale Perspectives, Daniel Guedes De Oliveira

LSU Doctoral Dissertations

Particle-laden flows are investigated numerically from a meso-scale perspective using Computational Fluid Dynamics coupled with Discrete Element Method (CFD-DEM) and from a micro-scale perspective using Particle Resolved Direct Numerical Simulation (PR-DNS). For the former, the dynamics of a pseudo-2D pulsed fluidized bed (PFB) consisting of 400,000 to 800,000 particles was investigated (Chapters 2 and 3). The focus is on the capabilities of CFD-DEM to (1) reproduce pattern formation in these systems and (2) further the understanding of the dynamics of PFB's as a function of pulsation parameters. In Chapter 4, a two-spheres system is investigated with a recently implemented PR-DNS …

Concentrations, Sources And Health Risks Of Particulate Matter In Southeast Us: Current Status And Future Changes, Fenglin Han

LSU Doctoral Dissertations

Evaluation of long-term air quality trends and prediction of future air quality may help to design and demonstrate effectiveness of control strategies and guide future air quality management. As one of the six criteria air pollutants (CAPs) regulated by the US Environmental Protection Agency (EPA), particulate matter with aerodynamic diameter of 2.5 µm or less (PM2.5) is of great interests since it has adverse effects on human health, visibility and climate. This study aims to understand the current status and future changes in concentrations, sources, and health risks of PM2.5 in Southeast US. Firstly, observations were analyzed …

Ageism And Embodied Stereotypes: A Study Of Adult Learners In Community College At Midlife, Marla Jane Erwin Jan 2020

Ageism And Embodied Stereotypes: A Study Of Adult Learners In Community College At Midlife, Marla Jane Erwin

LSU Doctoral Dissertations

Adult students are generally classified as a single group for study, yet developmental psychologists recognize separate developmental periods during adulthood that suggest adult students at midlife may experience development within higher education differently that younger adult students, in part due to ageism expressed at individual, institutional and internalized levels. This project applies the concept of lifespan developmental periods to distinguish students at midlife as a focus of inquiry using a mixed method design. Twenty-nine faculty and 205 students responded to the Relating to Older People Evalution (ROPE; Cherry & Palmore, 2008) to assess self-reports of both positive and negative ageist …

Coastal wetlands, such as saltmarshes and mangroves that fringe transitional waters, deliver important ecosystem services that support human development. Coastal wetlands are complex social-ecological systems that occur at all latitudes, from polar regions to the tropics. This overview covers wetlands in five continents. The wetlands are of varying size, catchment size, human population and stages of economic development. Economic sectors and activities in and around the coastal wetlands and their catchments exert multiple, direct pressures. These pressures affect the state of the wetland environment, ecology and valuable ecosystem services. All the coastal wetlands were found to be affected in some …

Impaired Purine Homeostasis Plays A Primary Role In Trimethoprim-Mediated Induction Of Virulence Genes In Burkholderia Thailandensis, Sudarshan S. Thapa, Anne Grove

Faculty Publications

© 2020 John Wiley & Sons Ltd One of the most commonly prescribed antibiotics against Burkholderia infections is co-trimoxazole, a cocktail of trimethoprim and sulfamethoxazole. Trimethoprim elicits an upregulation of the mal gene cluster, which encodes proteins involved in synthesis of the cytotoxic polyketide malleilactone; trimethoprim does so by increasing expression of the malR gene, which encodes the activator MalR. We report that B. thailandensis grown on trimethoprim exhibited increased virulence against Caenorhabditis elegans. This enhanced virulence correlated with an increase in expression of the mal gene cluster. Notably, inhibition of xanthine dehydrogenase by addition of allopurinol led to similar …

The Worst Passions Of Human Nature: White Supremacy In The Civil War North, Andrew Turner Jan 2020

The Worst Passions Of Human Nature: White Supremacy In The Civil War North, Andrew Turner

Civil War Book Review

The New York City riots of July 1863 exhibited the potential of the violent tension that ran in a steady stream throughout the Northern states during the middle of the Civil War. As white New Yorkers rioted to oppose the Enrollment Act of 1863 imposed by the United States government, they quickly turned their ire toward African Americans, with deeply seated white supremacy and racially motivated fear prompting their deadly reprisals against innocent black New Yorkers. Sadly, as illustrated by Paul Escott’s needed new book, The Worst Passions of Human Nature: White Supremacy in the Civil War North, …

Purpose Studies suggest ketogenic diets (KD) produce favorable outcomes (health and exercise performance); however, most rodent studies have used a low-protein KD, which does not reflect the normal- to high-protein KD used by humans. Liver has an important role in ketoadaptation due to its involvement in gluconeogenesis and ketogenesis. This study was designed to test the hypothesis that exercise training (ExTr) while consuming a normal-protein KD (NPKD) would induce additive/synergistic responses in liver metabolic pathways. Methods Lean, healthy male C57BL/6J mice were fed a low-fat control diet (15.9% kcal protein, 11.9% kcal fat, 72.2% kcal …

The Gulf of Mexico (GoM) region is prone to disasters, including recurrent oil spills, hurricanes, floods, industrial accidents, harmful algal blooms, and the current COVID-19 pandemic. The GoM and other regions of the U.S. lack sufficient baseline health information to identify, attribute, mitigate, and facilitate prevention of major health effects of disasters. Developing capacity to assess adverse human health consequences of future disasters requires establishment of a comprehensive, sustained community health observing system, similar to the extensive and well-established environmental observing systems. We propose a system that combines six levels of health data domains, beginning with three existing, national surveys …

Epidemics have traditionally been viewed as the widespread occurrence of infectious disease within a community, or a sudden increase above what is typical. But modern epidemics are both more and less than the diffusion of viral entities. We argue that epidemics are 'fire objects', using a term coined by Law and Singleton: They generate locative fears through encounters that focus attention on entities that are unknown or imprecisely known, transforming spaces and humans into indeterminate dangers, alternating appearance and absence. The Ebola epidemic of 2014 had more complex impacts than the number of infections would suggest. We employ multi-sited qualitative …

The mobilization of iron from BfrB, which requires binding by a cognate ferredoxin (Bfd), is essential to the regulation of cytosolic iron levels in P. aeruginosa. This paper describes the structure-guided development of small molecule inhibitors of the BfrB-Bfd protein-protein interaction. The process was initiated by screening a fragment library and followed by obtaining the structure of a fragment hit bound to BfrB. In fishes, arginine vasotocin (AVT) expression is related to social and/or reproductive status in most male fishes studied to date, and is linked to territorial defense, paternal care, and courtship. Despite a plethora of studies examining AVT in male fishes, relatively little is known about how AVT expression varies with female reproductive state or its role in female social behaviors. We used multiple methods for examining the AVT system in female African cichlid fish Astatotilapia burtoni, including immunohistochemistry for AVT, in …

One of the most devastating fungal diseases of soybean in the southern USA is Cercospora leaf blight (CLB), which is caused mainly by Cercospora cf. flagellaris. Recent studies found that the fungal effector AVR4, originally identified in Cladosporium fulvum as a chitin-binding protein, is highly conserved among other Cercospora species. We wanted to determine whether it is present in C. cf. flagellaris and, if so, whether it plays a role in the pathogen infection of soybean. We cloned the Avr4 gene and created C. cf. flagellaris ∆avr4 mutants, which produced little cercosporin and significantly reduced expression of cercosporin biosynthesis genes. …
