Four phthalocyanine (Pc)-peptide conjugates designed to target the epidermal growth factor receptor (EGFR) were synthesized and evaluated in vitro using four cell lines: human carcinoma A431 and HEp2, human colorectal HT-29, and kidney Vero (negative control) cells. Two peptide ligands for EGFR were investigated: EGFR-L1 and -L2, bearing 6 and 13 amino acid residues, respectively. The peptides and Pc-conjugates were shown to bind to EGFR using both theoretical (Autodock) and experimental (SPR) investigations. The Pc-EGFR-L1 conjugates 5a and 5b efficiently targeted EGFR and were internalized, in part due to their cationic charge, whereas the uncharged Pc-EGFR-L2 conjugates 4b and 6a …

A sustainable system is not necessarily a high-quality one, but it could be. We could, for example, “survive” on the desperate edge, as the remnants in a self-fouled and deteriorating environment. Why won’t a future sustainable system be just another industrial model of mass efficiency and throughput? Perhaps the incompatible outcomes are a choice between the sometimes nearly invisible civilizing aspects of culture nurturing respect, equality, and cooperation on one hand, and the greed and self-indulgences undermining social tolerance, empathy, and cooperation that ends up promoting violence and dehumanization. The human heritage is subtle, indestructible, and worth nurturing if we …

High Dispersal Ability Inhibits Speciation In A Continental Radiation Of Passerine Birds, Santiago Claramunt, Elizabeth P. Derryberry, Jr V. Remsen, Robb T. Brumfield

Dispersal can stimulate speciation byfacilitating geographical expansion across barriers or inhibit speciation by maintaining gene flow among populations. Therefore, the relationship between dispersal ability and speciation rates can be positive or negative. Furthermore, an 'intermediate dispersal' model that combines positive and negative effects predicts a unimodal relationship between dispersal and diversification. Because both dispersal ability and speciation rates are difficult to quantify, empirical evidence for the relationship between dispersal and diversification remains scarce. Using a surrogate for flight performance and a species-level DNA-based phylogeny of a large South American bird radiation (the Furnariidae), we found that lineages with higher dispersal …

The killifish Fundulus heteroclitus is abundant in osmotically dynamic estuaries and it can quickly adjust to extremes in environmental salinity. We performed a comparative osmotic challenge experiment to track the transcriptomic and physiological responses to two salinities throughout a time course of acclimation, and to explore the genome regulatory mechanisms that enable extreme osmotic acclimation. One southern and one northern coastal population, known to differ in their tolerance to hypo-osmotic exposure, were used as our comparative model. Both populations could maintain osmotic homeostasis when transferred from 32 to 0.4 p.p.t., but diverged in their compensatory abilities when challenged down to …

Herpes simplex virus 1 (HSV-1) viral glycoproteins gD (carboxyl terminus), gE, gK, and gM, the membrane protein UL20, and membrane-associated protein UL11 play important roles in cytoplasmic virion envelopment and egress from infected cells. We showed previously that a recombinant virus carrying a deletion of the carboxyl-terminal 29 amino acids of gD (gDΔct) and the entire gE gene (ΔgE) did not exhibit substantial defects in cytoplasmic virion envelopment and egress (H. C. Lee et al., J. Virol. 83:6115-6124, 2009). The recombinant virus ΔgM2, engineered not to express gM, produced a 3-to 4-fold decrease in viral titers and a 50% reduction …

The histone deacetylase sirtuin 1 (SIRT1) inhibits adipocyte differentiation and suppresses inflammation by targeting the transcription factors peroxisome proliferator-activated receptor γ and nuclear factor κB. Although this suggests that adiposity and inflammation should be enhanced when SIRT1 activity is inactivated in the body, this hypothesis has not been tested in SIRT1 null (SIRT1 -/-) mice. In this study, we addressed this issue by investigating the adipose tissue in SIRT1 -/-mice. Compared with their wild-type littermates, SIRT1 null mice exhibited a significant reduction in body weight. In adipose tissue, the average size of adipocytes was smaller, the content of extracellular matrix …

Phylogenomics offers the potential to fully resolve the Tree of Life, but increasing genomic coverage also reveals conflicting evolutionary histories among genes, demanding new analytical strategies for elucidating a single history of life. Here, we outline a phylogenomic approach using a novel class of phylogenetic markers derived from ultraconserved elements and flanking DNA. Using species-tree analysis that accounts for discord among hundreds of independent loci, we show that this class of marker is useful for recovering deep-level phylogeny in placental mammals. In broad outline, our phylogeny agrees with recent phylogenomic studies of mammals, including several formerly controversial relationships. Our results …

Genomic Insights Into Adaptation To High-Altitude Environments, Z. A. Cheviron, R. T. Brumfield

Elucidating the molecular genetic basis of adaptive traits is a central goal of evolutionary genetics. The cold, hypoxic conditions of high-altitude habitats impose severe metabolic demands on endothermic vertebrates, and understanding how high-altitude endotherms cope with the combined effects of hypoxia and cold can provide important insights into the process of adaptive evolution. The physiological responses to high-altitude stress have been the subject of over a century of research, and recent advances in genomic technologies have opened up exciting opportunities to explore the molecular genetic basis of adaptive physiological traits. Here, we review recent literature on the use of genomic …

SPECT is primarily used in the clinic for cardiac myocardial perfusion imaging. However, for SPECT, sensitivity is impaired due to the need for collimation. System resolution FWHM is poor as well (∼ 1 cm). In this work the resolution of a curved detector was theoretically derived. The advantage of a curved detector over a flat detector with pinhole collimation was demonstrated for cardiac applications using theoretical derivations as well as a ray-tracing voxel-based forward projector. For the flat detector using parameters close to what was expected for the new multi-pinhole GE Discovery system, it is shown that using a paraboloid …

Coral reefs are experiencing unprecedented degradation due to human activities, and protecting specific reef habitats may not stop this decline, because the most serious threats are global (i.e., climate change), not local. However, ex situ preservation practices can provide safeguards for coral reef conservation. Specifically, modern advances in cryobiology and genome banking could secure existing species and genetic diversity until genotypes can be introduced into rehabilitated habitats. We assessed the feasibility of recovering viable sperm and embryonic cells post-thaw from two coral species, Acropora palmata and Fungia scutaria that have diffferent evolutionary histories, ecological niches and reproductive strategies. In vitro …

Message from President: With the advent of the political season and the opening of the Louisiana Legislature, universities up, down, and across our marshy state go public. Lacking competition from football and situated in a media market that generates fewer distractions than do major metropolitan areas, universities and their troubles routinely grab frontpage or top-of-the-hour placement in journalistic productions. Loud and colorful battles, whether against budget woes or recalcitrant lawmakers or consultants with reorganization plans, draw public attention to the big issues surrounding education but also distract public attention from the more subtle causes of university distress, many of which …

A 2-yr study of the impacts of the red imported fire ant, Solenopsis invicta Buren, on the success of Cyrtobagous salviniae Calder & Sands as a biological control agent of common salvinia, Salvinia minima Baker, was undertaken in southern Louisiana. Floating moats were constructed around 8 release sites where C. salviniae was introduced as a biological control agent. These moats were used to restrict access to the release site by red imported fire ants in half the quadrats. Moating was successful in limiting the number of red imported fire ants in the release sites over the course of the study …

Classical epidemic theory focuses on directly transmitted pathogens, but many pathogens are instead transmitted when hosts encounter infectious particles. Theory has shown that for such diseases pathogen persistence time in the environment can strongly affect disease dynamics, but estimates of persistence time, and consequently tests of the theory, are extremely rare. We consider the consequences of persistence time for the dynamics of the gypsy moth baculovirus, a pathogen transmitted when larvae consume foliage contaminated with particles released from infectious cadavers. Using field-transmission experiments, we are able to estimate persistence time under natural conditions, and inserting our estimates into a standard …

Health benefits of resistant starch (RS), a dietary fermentable fiber, have been well documented in young, but not in old populations. As the essential step of more comprehensive evaluations of RS on healthy aging, we examined the effects of dietary RS on tolerance, colonic fermentation, and cytokine expression in aged mice. Healthy older (18-20 months) C57BL/6J male mice were fed control, 18% RS, or 36% RS diets for 10weeks. Body weight gain, body composition, and fat pad weights did not differ among the three groups after 10weeks, indicating good tolerance of the RS diet. Fermentation indicators (cecum weights, and cecal …

Androgen signaling through androgen receptor (AR) is critical for prostate tumorigenesis. Given that AR-mediated gene regulation is enhanced by AR coregulators, inactivation of those coregulators is emerging as a promising therapy for prostate cancer (PCa). Here, weshowthat the N-acetyltransferase arrest-defect 1 protein (ARD1) functions as a unique AR regulator in PCa cells. ARD1 is up-regulated in human PCa cell lines and primary tumor biopsies. The expression of ARD1 was augmented by treatment with synthetic androgen (R1881) unless AR is deficient or is inhibited by AR-specific siRNA or androgen inhibitor bicalutamide (Casodex). Depletion of ARD1 by shRNA suppressed PCa cell proliferation, …

The epithelial Na + channels (ENaCs) are present in kidney and contribute to Na + and water homeostasis. All three ENaC subunits (α, β, and γ) were demonstrated in the cardiovascular regulatory centers of the rat brain, including the magnocellular neurons (MNCs) in the supraoptic nucleus (SON) and the paraventricular nucleus (PVN). However, the functional significance of ENaCs in vasopressin (VP) and oxytocin (OT) synthesizing MNCs is completely unknown. In this study, we show with immunocytochemical double-labeling that the α-ENaC is colocalized with either VP or OT in MNCs in the SON and PVN. In addition, parvocellular neurons in the …

Message from President: Nearly two months have slipped away since LSU completed a football season that produced an astonishing record of victories: a record that, even without a national championship as garnish, must surely count as a super-size main course in the meatiest of sports. Remarkably, what lingers from the past season is not the sweet aroma of victory but rather something of a bad taste. Football, like most sports, allegorizes the institutions that sponsor it. Identifying the reason that one of the greatest seasons in sports history is creating only a paucity of post-season joy is a good way …
