© 2017 The Author(s). Using nationwide survey data, we investigate U.S. meat goat producer preferences and willingness to pay for meat goat breeding stock attributes. Discrete choice experiments were employed, and mixed logit and latent class models were used for analysis. Results showed that producers preferred animals that were highly masculine/feminine, had good structure and soundness, and were of the Boer breed, whereas they preferred fewer animals that were older, of Kiko and Spanish breeds, and priced higher. Significant preference heterogeneity was found among the respondents. Larger-scale producers had greater preference for high masculinity/femininity, good structure and soundness, and Boer …

© The Author (2017). Retinoic acid (RA), a bioactive metabolite of Vitamin A, is a critical mediator of cell differentiation. RA blocks adipogenesis, but mechanisms remain to be established. ZFP423 is a key transcription factor maintaining white adipose identity. We found that RA inhibits Zfp423 expression and adipogenesis via blocking DNA demethylation in the promoter of Zfp423, a process mediated by growth arrest and DNA-damage-inducible protein alpha (GADD45A). RA induces the partnering between retinoic acid receptor (RAR) and tumor suppressor inhibitor of growth protein 1 (ING1), which prevents the formation of GADD45A and ING1 complex necessary for locus-specific Zfp423 DNA …

Heme oxygenase from Leptospira interrogans is an important virulence factor. During catalysis, redox equivalents are provided to this enzyme by the plastidic-type ferredoxin-NADP+ reductase also found in L. interrogans. This process may have evolved to aid this bacterial pathogen to obtain heme-iron from their host and enable successful colonization. Herein we report the crystal structure of the heme oxygenase-heme complex at …

Economic Growth And Mortality: Do Social Protection Policies Matter?, Usama Bilal, Richard Cooper, Francis Abreu, Claudia Nau, Manuel Franco, Thomas A. Glass

Faculty Publications

Background: In the 20th century, periods of macroeconomic growth have been associated with increases in population mortality. Factors that cause or mitigate this association are not well understood. Evidence suggests that social policy may buffer the deleterious impact of economic growth. We sought to explore associations between changing unemployment (as a proxy for economic change) and trends in mortality over 30 years in the context of varying social protection expenditures. Methods: We model change in all-cause mortality in 21 OECD (Organization for Economic Cooperation and Development) countries from 1980 to 2010. Data from the Comparative Welfare States Data Set and …

Genistein Crosses The Bioartificial Oviduct And Alters Secretion Composition, Constantine A. Simintiras, Roger G. Sturmey Aug 2017

Genistein Crosses The Bioartificial Oviduct And Alters Secretion Composition, Constantine A. Simintiras, Roger G. Sturmey

Faculty Publications

The dietary derived isoflavone and oestrogen analogue, genistein, is known to perturb fundamental reproductive events such as implantation and embryo cleavage. However the question of whether genistein is able to traverse the oviduct epithelial monolayer and impact oviduct fluid secretion remains unclear. This study tests these research questions using a bioartificial oviduct to show that genistein permeates the oviduct lumen in vitro with a biphasic (burst and plateau) kinetic profile, faster than spontaneous diffusion, and alters the amino acid composition of in vitro derived oviduct fluid (ivDOF) but not as an oestrogen analogue. In addition to offering insights into the …

© Society for Leukocyte Biology. Respiratory syncytial virus (RSV) is one of the leading causes of bronchiolitis in children, and severe RSV infection early in life has been associated with asthma development. Using a neonatal mouse model, we have shown that down-regulation of IL-4 receptor α (IL-4Rα) with antisense oligonucleotides in the lung during neonatal infection protected from RSV immunopathophysiology. Significant down-regulation of IL-4Rα was observed on pulmonary CD11b+ myeloid dendritic cells (mDCs) suggesting a role for IL-4Rα on mDCs in the immunopathogenesis of neonatal RSV infection. Here, we demonstrated that neonatal CD11b+ mDCs expressed higher levels of IL-4Rα than …

Levels of adaptive genetic variation influence how species deal with environmental and ecological change, but these levels are frequently inferred using neutral genetic markers. Major histocompatibility complex (MHC) genes play a key role in the adaptive branch of the immune system and have been used extensively to estimate levels of adaptive genetic variation. Parts of the peptide binding region, sites where MHC molecules directly interact with pathogen and self-proteins, were sequenced from a MHC class I (95/441 tortoises) and class II (245/441 tortoises) gene in threatened and nonthreatened populations of gopher tortoises (Gopherus polyphemus), and adaptive genetic variation at MHC …

In order to reconstruct environments associated with Plio-Pleistocene hominins in southern Africa, researchers frequently rely upon the animals associated with the hominins, in particular, animals in the Family Bovidae. Bovids in southern Africa are typically identified by their teeth. However, identifying the taxon of a bovid tooth is challenging due to various biasing factors. Furthermore, inaccurate identification of fossil bovids can have significant consequences on the reconstructed paleoenvironment. Recent research on the classification of bovid fossil teeth has relied on using elliptical Fourier analysis to summarize the shape of the outline of the occlusal surface of the tooth and the …

© 2017 the authors. The lateral hypothalamus (LHA) integrates reward and appetitive behavior and is composed of many overlapping neuronal populations. Recent studies associated LHA GABAergic neurons (LHAGABA), which densely innervate the ventral tegmental area (VTA), with modulation of food reward and consumption; yet, LHAGABA projections to the VTA exclusively modulated food consumption, not reward. We identified a subpopulation of LHA GABA neurons that coexpress the neuropeptide galanin (LHAGal). TheseLHAGal neurons also modulate food reward, but lack direct VTA innervation. We hypothesized that LHAGal neurons may represent a subpopulation of LHAGABA neurons that mediates food reward independent of direct VTA …

© 2017. Published by The Company of Biologists Ltd. The phosphatidylinositol 3-kinase (PI3K)/protein kinase B (PKB/Akt) pathway plays an important role in regulating cell proliferation, metabolism, and survival. However, the distinct roles of Akt isoforms (Akt1, Akt2, and Akt3) in pluripotent stem cell maintenance are not fully defined. Using mouse embryonic stem cells (ESCs), we show that direct inhibition of Akt activity leads to ESC apoptosis. The Akt3, but not Akt1 or Akt2, activity specifically regulates this effect. Inhibiting Akt3 also leads to a cell cycle arrest at G1 phase. These regulatory roles of Akt3 are dependent on its kinase …

Timing Of Cherry Tree Blooming: Contrasting Effects Of Rising Winter Low Temperatures And Early Spring Temperatures, Peijian Shi, Zhenghong Chen, Gadi V.P. Reddy, Cang Hui, Jianguo Huang, Mei Xiao

Faculty Publications

Phenology reflects the interplay of climate and biological development. Early spring phenological phenomena are particularly important because the end of diapause or dormancy is related not only to heat accumulation in the early spring but also probably to winter low temperatures. Although a warmer winter can reduce overwintering mortality in many insects and plants, it also reduces the accumulation of chilling time that often triggers the end of diapause or dormancy. We examined a continuous 67-year time series of the first flowering date of cherry trees and compared three phenological models based on the temperature-dependent developmental rate: (i) the accumulated …

Shallow Ponds Are Heterogeneous Habitats Within A Temperate Salt Marsh Ecosystem, Amanda C. Spivak, Kelsey Gosselin, Evan Howard, Giulio Mariotti, Inke Forbrich, Rachel Stanley, Sean P. Sylva

Faculty Publications

Integrating spatial heterogeneity into assessments of salt marsh biogeochemistry is becoming increasingly important because disturbances that reduce plant productivity and soil drainage may contribute to an expansion of shallow ponds. These permanently inundated and sometimes prominent landscape features can exist for decades, yet little is known about pond biogeochemistry or their role in marsh ecosystem functioning. We characterized three ponds in a temperate salt marsh (MA, USA) over alternating periods of tidal isolation and flushing, during summer and fall, by evaluating the composition of plant communities and organic matter pools and measuring surface water oxygen, temperature, and conductivity. The ponds …

© 2017 Poultry Science Association Inc. Anise oil (AO) is a phytochemical with antibiotic properties and it reduces the growth of Clostridium perfringens in vitro, which is the main causal agent of necrotic enteritis. A series of battery experiments was conducted to determine an AO dosage to feed to broilers and its effect during 2 C. perfringens challenges, and a floor pen experiment to determine its effect in performance. Anise oil linearly reduced (P < 0.05) ADFI and ADG, but feeding broilers 500 to 2,000 mg/kg of AO resulted in similar performance with positive control (PC) fed broilers. Broilers were challenged on d 10 (Experiment 2) or on d 10 and 13 (Experiment 3). Broilers fed AO had lower (P < 0.05) ADFI and ADG compared with broilers fed PC from zero to 18 d (Experiment 2). Growth performance of broilers fed AO was not affected (P > 0.05) during Experiment 3. In both challenges, the jejunum of broilers fed AO had fewer and milder (P < 0.05) lesions compared with broilers fed PC or negative control (NC) diets. In a floor pen trial, 1,000 mg/kg of AO was fed to broilers from zero to 42 days. Broilers fed AO or PC had similar (P > 0.10) growth performance for the entire trial. Broilers …

The supply of inorganic carbon (Ci) at the site of fixation by Rubisco is a key parameter for efficient CO2 fixation in aquatic organisms including the green alga, Chlamydomonas reinhardtii. Chlamydomonas reinhardtii cells, when grown on limiting CO2, have a CO2-concentrating mechanism (CCM) that functions to concentrate CO2 at the site of Rubisco. Proteins thought to be involved in inorganic carbon uptake have been identified and localized to the plasma membrane or chloroplast envelope. However, current CCM models suggest that additional molecular components …

Pseudomonas fluorescens strain 3JW1, which has a broad-spectrum antimicrobial activity, was studied to investigate whether it affects the amounts of aflatoxin B1 (AFB1) produced by Aspergillus flavus. It was found that the bacterium reduced the amounts of AFB1 in potato dextrose broth (PDB) and peanut medium by 97.8% and 99.4%, respectively. It also reduced AFB1 by ~183 μg/kg (55.8%) when applied onto peanut kernels. This strain reduced AFB1 via three mechanisms. First, it significantly inhibited A. flavus growth; second, our data showed that strain 3JW1 inhibits aflatoxin biosynthesis by A. flavus; and third, P. fluorescens strain 3JW1 is capable of …

Oncology imaging is performed using various modalities including computed tomography (CT), magnetic resonance imaging (MRI), single-photon emission computed tomography (SPECT) and fluorodeoxyglucose-positron emission tomography (FDG-PET). Our goal is to extract useful, otherwise hidden, quantitative biophysical parameters (such as growth-rate, tumor-necrotic-factor, etc.) from these serial images of tumors by fitting mathematical models to images. These biophysical features are intrinsic to the tumor types and specific to the study-subject, and expected to add valuable information on the tumor containment or spread and help treatment plans. Thus, fitting …

Cryopreservation has become an important and accepted tool for long-term germplasm conservation of animals and plants. To protect genetic resources, repositories have been developed with national and international cooperation. For a repository to be effective, the genetic material submitted must be of good quality and comparable to other submissions. However, due to a variety of reasons, including constraints in knowledge and available resources, cryopreservation methods for aquatic species vary widely across user groups which reduces reproducibility and weakens quality control. Herein we describe a standardizable freezing device produced using 3-dimensional (3-D) printing and introduce the concept of network sharing to …
