DNA methylation is an important epigenetic modification that undergoes dynamic changes in mammalian embryogenesis, during which both parental genomes are reprogrammed. Despite the many immunostaining studies that have assessed global methylation, the gene-specific DNA methylation patterns in bovine preimplantation embryos are unknown. Using reduced representation bisulfite sequencing, we determined genome-scale DNA methylation of bovine sperm and individual in vivo developed oocytes and preimplantation embryos. An ethanolic extract of Artemisia scoparia (SCO) has metabolically favorable effects on adipocyte development and function in vitro and in vivo. In diet-induced obese mice, SCO supplementation significantly reduced fasting glucose and insulin levels. Given the importance of adipocyte lipolysis in metabolic health, we hypothesized that SCO modulates lipolysis in vitro and in vivo. Free fatty acids and glycerol were measured in the sera of mice fed a high-fat diet with or without SCO supplementation. In the current study, we characterized the expression and histone modification profiles of the alternative promoters found within imprinted Igf2r, Mest, Zac1, Peg3, Snrpn and non-imprinted Myc loci. This work was done in order to deepen the understanding of interactions occurring during sorption of anthropogenic organic compounds and dissolved organic matter (DOM) onto soil. As well as the transformation DOM undergoes during the sorption process and the overall transformation OM undergoes in the environment. This was achieved through the use of a “bottom-up” synthetic investigation, “top-down” natural system study, and a combination of these approaches.
The “bottom-up” approach, discussed in Chapter 2, involved the synthesis of a modular multi-tiered engineered soil surrogate (ESS) designed to mimic OM and general trends identified in the literature that impacts sorption behavior. …

The work presented in this dissertation employs task-specific materials for sensing and protein separation applications. These materials were derived from a group of uniform materials based on organic salts (GUMBOS). GUMBOS are organic salts similar to ionic liquids, but have melting points ranging from 25 to 250 °C. As with ionic liquids, the properties of GUMBOS can be easily tuned by changing the counter-ion. Thus, task-specific GUMBOS can be designed and prepared with properties that are beneficial for applications in sensing or protein separation. Louisiana is home to 40% of the coastal wetlands in the lower 48 states, yet accounts for 80% of the coastal wetland loss in this region. This loss is attributed to decreased sediment supply, levee alteration, sea level rise, channelization, and subsidence. The levee system in Louisiana disconnected coastal wetlands from the main stem of the Mississippi River (MSR), reducing the amount of land-building sediment that reaches coastal wetlands. This disconnection also allows a greater percentage of river discharge, including agricultural contaminants and runoff, to flow into the northern Gulf of Mexico (NGOM).

Chapter 1 contains a brief overview of the history, synthesis, and properties of BODIPY dyes as well as that of the biomedical applications of bio-imaging and photodynamic therapy. Additionally, an overview of the theoretical framework of density functional theory and its time-dependent variant are provided. In Chapter 2, the effects of structural modification on the electronic structure and electron dynamics of cationic meso-(4-pyridyl)-BODIPYs were investigated. Climate is one of the most important limiting factors in animal production. Cattle under the effect of heat stress have reduced fertility. Negative effects during gamete maturation and embryo development have been observed at the morphological, biochemical, transcriptional and developmental levels. A child’s play is facilitative of language development, creating a situation where words are needed to speak to and for toy characters, socially engage with peers, and develop play narratives. Children with language disorders, in particular autism spectrum disorder, often fail to engage in play activities at levels of their typically developing peers. However, under highly structured conditions, or scaffolded conditions, children with autism have demonstrated the ability to engage in play at similar levels as their typically developing peers. The goal of this longitudinal study was to measure the attitudes and behaviors of participants of the Marsh Maneuvers program over the course of the last 25 years. The goal of the Marsh Maneuvers program is to educate students into becoming environmentally literate adults. Environmental literacy is considered herein to be people possessing knowledge about and positive attitudes and behaviors towards the environment. The current study sought to test the relationship between course design, as described by the rubric produced by Quality Matters, and online university student performance. Due to the link between student motivation and active learning behaviors, and thus performance, it was predicted that the better-designed courses would facilitate student motivation. It was also predicted that goal orientation would moderate this relationship. While a significant relationship was observed between student motivation and course performance, no relationship was observed between course quality, as measured by the QM Rubric, and motivation, or performance. Utilizing winter annuals in livestock pastures reduces feed costs of hay, and provides additional vegetative cover that is beneficial to the soil. However, how this practice affects the soil microbial communities has not been well studied. Two sites were included in this study. Both were commercial cattle grazing operations that over-seeded a mixture of grasses, legumes, and brassicas into pastures dominated by bahiagrass (Paspalum notatum). Soil health was assessed using a suite of physical, chemical, and biological properties. Soil physical assessments included bulk density and aggregate stability. Crassostrea virginica, the eastern oyster, forms reefs that provide critical services and benefits to the resiliency of the surrounding ecosystem. Changes in environmental conditions, including salinity and temperature, can dramatically alter the services oysters provide by affecting their population dynamics. Climate warming may further exacerbate the effects of salinity changes as precipitation events increase in frequency, intensity, and duration. Temperature and salinity independently and synergistically influence gene expression and physiology in marine organisms. A three-dimensional, high-resolution, Finite-Volume Coastal Ocean Model (FVCOM) was used to study the dynamics of Barataria Estuary located in the Southeastern Louisiana. Three numerical experiments with different discharge scenarios, including the actual discharge (average ~ 160 m3 s-1) from the Davis Pond Diversion (DPD) over three months from April to June 2010, no discharge (NO), and the proposed Mid-Barataria Diversion (MBD) with a constant discharge of 850 m3 s-1, were conducted to investigate the impacts of river diversions on salinity gradients and residence times in the estuary. Ceria is an earth-abundant material that has been widely used in heterogeneous catalysis, environmental catalysis, and energy applications thanks for its ability to readily convert between different oxidation states. The objective of this study is to theoretically elucidate the reaction mechanisms for the conversion of model organic compounds on ceria, in order to gain insights for the design of cost-effective and selective ceria-based catalysts. In research conducted using indeterminate soybean [Glycine max (L.) Merr.], fourteen injury criteria observed following dicamba at 0.6 to 280 g ae ha-1 (1/1000 to ½ of 560 g ha-1 use rate) were rated using a scale of 0= no injury, 1= slight, 2= slight to moderate, 3= moderate, 4= moderate to severe, and 5= severe. Greatest crop injury 15 d after treatment (DAT) was observed following dicamba applied at 0.6 to 4.4 g ha-1 at V3/V4 for upper canopy pale leaf margins (3.8 to 4.2) and at R1/R2 for terminal leaf cupping (4.1 to 5.0) …

The ability to harvest this energy can be useful for supplying low-consumption wearable and implantable devices. Thermoelectric materials are usually used to harvest human body heat for wearable devices; however, thermoelectric generators require temperature gradient across the device to perform appropriately. Since they need to attach to the heat source to absorb the heat, temperature equalization decreases their efficiencies. Growth models estimate life history parameters that are used in the management of fisheries stocks. The most commonly used growth model in fisheries is the von Bertalanffy growth model (VBGM), yet it has been shown to provide a poor fit for length-at-age data of some species and other models exist. I reviewed 196 peer-reviewed age and growth studies and 50 NOAA stock assessments to examine temporal trends in the use of growth models and model selection in fisheries. First, the identification of possible sources of distraction and their associated crash/near-crash risk. © 2018 IBRO Current evidence suggests that the epithelial Na + channel (ENaC) in the brain plays a significant role in the development of hypertension. ENaC is present in vasopressin (VP) neurons in the hypothalamus, suggesting that ENaC in VP neurons is involved in the regulation of blood pressure. Our recent study demonstrated that high dietary salt intake caused an increase in the expression and activity of ENaC that were responsible for the more depolarized basal membrane potential in VP neurons. Anti-proliferative responses such as senescence and apoptosis are often used by normal cells to combat oncogenic insults and to prevent tumorigenesis. However, oncogenic mutations are frequently found in cancers, suggesting that additional mutations may occur to facilitate the bypass of these anti-proliferative responses. It is believed that some of these additional mutations may also contribute to the chemoresistance of cancers. This dissertation focused on identifying novel genes and biological processes implicated in chemoresistance and tumorigenesis.

Deep neural networks trained over large datasets learn features that are both generic to the whole dataset, and specific to individual classes in the dataset. Learned features tend towards generic in the lower layers and specific in the higher layers of a network. Methods like fine-tuning are made possible because of the ability for one filter to apply to multiple target classes. The Cordilleran Flycatcher (Empidonax occidentalis) is one of many North American birds whose distribution crosses the US–Mexican border and for which little is presently known about summer distribution and breeding biology south of this border. In addition, the Cordilleran Flycatcher presents ornithologists with a number of challenging questions surrounding its taxonomy, migration, distribution, population structure, and species limits. A contact-type endoscope surface enhanced Raman scattering (SERS) probe includes a gradient-index (GRIN) lens, a transparent substrate adhered to the GRIN lens, and a rough metallic layer adhered to an opposite side of the transparent substrate from the GRIN lens. The GRIN lens focuses light from a Raman spectrometer onto the rough metallic layer, and the rough metallic layer is positioned at the distal end of the contact-type endoscope SERS probe.

Among these, MRI exhibits good soft tissue contrast, but is currently less commonly used for nonclinical research than X-ray CT, even though the latter requires contrast-staining that shrinks and distorts soft tissues. When the objective is the creation of a realistic and complete 3D model of soft tissue structures, MRI data are more demanding to acquire and visualize and require extensive post-processing because they comprise noncubic voxels with dimensions that represent a trade-off between tissue contrast and image resolution.
