Non-alcoholic steatohepatitis (NASH) is a clinically important spectrum of non-alcoholic fatty liver disease (NAFLD) in humans. NASH is a stage of NAFLD progression wherein liver steatosis accompanies inflammation and pro-fibrotic events. Presently, there are no approved drugs for NASH, which has become a leading cause of liver transplant worldwide. To discover novel drug targets for NASH, we analyzed a human transcriptomic NASH dataset and found Aldo-keto reductase family 1 member B10 (AKR1B10) as a significantly upregulated gene in livers of human NASH patients. New appetite-regulating antiobesity treatments such as semaglutide and agents under investigation such as tirzepatide show promise in achieving weight loss of 15% or more. Energy expenditure, fat oxidation, and lean mass preservation are important determinants of weight loss and weight-loss maintenance beyond appetite regulation. This review discusses prior failures in clinical development of weight-loss drugs targeting energy expenditure and explores novel strategies for targeting energy expenditure: mitochondrial proton leak, uncoupling, dynamics, and biogenesis; futile calcium and substrate cycling; leptin for weight maintenance; increased sympathetic nervous system activity; and browning of white fat. Precipitated by chronic psychological stress, immune system dysregulation, and a hyperinflammatory state, the sequelae of postacute COVID-19 (long COVID) include depression and new-onset diabetes. We hypothesize that exercise counters the neuropsychiatric and endocrine sequelae of long COVID by inducing the release of circulating factors that mediate the anti-inflammatory response, support brain homeostasis, and increase insulin sensitivity.

In mammalian females, oocytes are stored in the ovary and meiosis is arrested at the diplotene stage of prophase I. When females reach puberty oocytes are selectively recruited in cycles to grow, overcome the meiotic arrest, complete the first meiotic division and become mature (ready for fertilization). At a molecular level, the master regulator of prophase I arrest and meiotic resumption is the maturation-promoting factor (MPF) complex, formed by the active form of cyclin dependent kinase 1 (CDK1) and Cyclin B1. Human cytomegalovirus (HCMV) is a widespread, highly prevalent pathogen with severe consequences in vulnerable populations. HCMV and the related murine cytomegalovirus (MCMV) encode multiple G protein-coupled receptor (GPCR) homologs which are involved in latency, immune evasion, and tissue tropism and dissemination. In this work, we investigate the mechanisms of two CMV-encoded GPCR homologs, HCMV US28 and MCMV M33, in latency and T cell immune evasion. Chapter 2 describes the isolation and immortalization of a salivary gland-derived mesenchymal cell line, termed mSGM. These cells are permissive to MCMV infection and express all three BALB/c mouse MHC class I isotypes, making them …

Brucella spp. are a bacterium that cause brucellosis, a zoonotic disease, which is commonly seen in cattle, sheep, goats, swine, and canines. Brucellosis is a problem worldwide, although it is eradicated in some countries (Garin-Bastuji et al. 1998). The reason for designing recombinant DNA (rDNA) vaccines opposed to utilizing the live-attenuated vaccines on the market is that they cannot be given to pregnant animals without potentially causing abortion, while an rDNA vaccine should be safe for pregnant animals since it does not contain viable bacteria. Also, there are no serological tests that can accurately distinguish between an animal vaccinated …

Aims: The study aimed to explore additive interactions of CDKAL1 rs7747752 and GUDCA/DCA for GDM risk and whether the interactive effects on the risk of GDM was mediated via increasing lysophosphatidylcholines (LPC) 18:0 and/or saturated fatty acid (SFA) 16:0. Methods: A 1:1 age-matched study nested in a prospective cohort of pregnant women (207 pairs) was organized in Tianjin, China. Additive interactions were used to test interaction effects while mediation analyses and Sobel tests were used to test mediation effects of LPC18:0 and SFA16:0 between copresence of rs7747752 and low GUDCA/DCA, and GDM risk. Field studies were conducted in 2018-2021 at the LSU Agricultural Center’s H.R. Caffey Rice Research Station (RRS) near Crowley, LA, and the Dean Lee Research and Extension Center (DLREC) near Alexandria, LA to evaluate off-target florpyrauxifen movement on soybean. Soybean was treated with florpyrauxifen at the V4-V5 or R1-R2 growth stages. Soybean injury was evaluated at 1, 7, 14, and 28 days after treatment (DAT) and soybean plant height (cm) was recorded at 7, 14, and 28 DAT. An electro-acupuncture (EA) system and method for performing EA on a patient are provided. The EA system comprises a wearable neurostimulator device, at least a first pair of electrically-conductive acupuncture needles and a system controller. The wearable neurostimulator device comprises a casing, an EA circuit mechanically coupled to the casing, and an attachment device mechanically coupled to the casing and adapted to removably secure the wearable neurostimulator device to the patient. The first pair of electrically-conductive acupuncture needles is mechanically coupled to the casing and electrically coupled to the EA circuit. The system controller is in communication with the EA …
