Monkeypox virus (MPXV) is the causative agent of monkeypox (mpox), a rare viral disease that affects humans [1]. It is primarily found in Africa and is transmitted to humans through contact with sick animals, particularly rodents and monkeys, or through human-to-human transmission [2]. From the beginning of May 2022, cases of mpox have been recorded from non-endemic nations, and the illness has continued to be reported in other endemic nations. Underexplored environments such as soil samples continue to be an untapped source of bacterial strains with great potential to produce secondary metabolites for medicinal applications. As a result, these microorganisms represent a broad and yet unknown reservoir of new strains capable of producing these novel compounds. The current research primarily seeks to perform the isolation, purification, and characterization of secondary metabolites from a soil bacterium (Janthinobacterium lividum TAJX1901). The isolated soil bacterium was successfully cultured on rich media agar plates, followed by extraction using methanol and chloroform. Microsporidia are a distant fungal pathogen that have severe clinical consequences for the immunocompromised. Previous work identified a microsporidian pathogen protein termed Microsporidian ADAM or MADAM. This protein has close sequence homology to other ADAM proteins (A Disintegrin and Metalloproteinase) in two microsporidian species, Encephalitozoon intestinalis and E. cuniculi. ADAM proteins have a wide range of functions, including binding to integrins and host signaling. It is known that many pathogens manipulate integrins to invade host cells, and it is predicted that microsporidia are also exploiting this host target. Introduction: The entire United States has felt the effects of the overdose epidemic, but rural Appalachia has experienced disproportionate overdose deaths. This variation in overdose mortality can be attributed to the risk environment for overdose in rural Appalachia. Overdose prevention behaviors are individual level behaviors that people who inject drugs can implement that may have the potential to reduce their risk of overdose, however limited research exists regarding the utilization of those behaviors. Glioblastoma is the most commonly diagnosed central nervous system primary malignancy and it is considered a terminal diagnosis with few treatment options available. Glioblastoma tumors frequently develop treatment resistance due in part to their highly heterogenic nature. The heterogeneity of glioblastoma is partially attributed to the presence of glioma stem-like cells (GSC), which are highly invasive and resistant to chemotherapy and irradiation treatments. Signaling of the receptor tyrosine kinase MET is a known regulator of GSC. Glioblastoma patients have an increasingly poor prognosis that corresponds with increasing MET expression. (July 12, 2023) East Tennessee State University has introduced a revamped infectious disease fellowship that now incorporates critical care, which will enhance physicians’ capabilities during pandemics.
Through the newly launched Infectious Disease Critical Care Fellowship, a comprehensive three-year program, fellows will receive specialized training in diagnosing and treating severe infections while also gaining increased exposure to critical care medicine. In collaboration with Ballad Health, this initiative aims to elevate the standard of subspecialty care, benefiting the local community and beyond.

The purpose of this study was to explore the resiliency of Head Start teachers during the COVID-19 pandemic. The study explored the role social support (as an external factor of resilience) and self-efficacy (as an internal factor of resilience) played in the resilience of Head Start teachers during the COVID-19 pandemic. This study employed a sequential explanatory mixed methods design involving two phases: a quantitative and a qualitative phase. Participants in this study were 99 teachers in rural eastern Tennessee and some rural parts of Virginia, North Carolina, and New Mexico, who were Head Start teachers during the COVID-19 pandemic. …

Determining the activity among species-specific bat populations within specific habitat selections can help contribute to the conservation of Appalachian bat species. This study examines the differences in species-specific bat activity patterns between three ponds of variable sizes and a southeast-facing open field with a wooded edge. Four Song Meter SM4BAT FS bioacoustics detectors were used on a 15-acre property in Fall Branch, TN, with a wildlife acoustics detector being placed at three ponds and one field. The Central Appalachian region of the United States disproportionately experiences higher rates of cardiovascular diseases (CVD) and associated risk factors. Primary risk factors for CVD include hypertension, hypercholesterolemia, and tobacco use, all of which have the potential to be mitigated through lifestyle behavior changes. Dietary and physical activity practices are the two main driving forces for the development of hypertension and hypercholesterolemia, while tobacco use is a behavioral choice on its own. Subsequentially, shifting lifestyle choices to emphasize healthier living has the potential to reduce CVDs within the Central Appalachian region. The study examined the frequent frames adopted by the CDC in the communication of the Covid-19 vaccine and how it impacts vaccine uptake through quantitative content analysis. The study builds on the framing theory and elaboration likelihood model. This study’s content collected 327 posts by the CDC on covid-19 vaccinations from December 2020- December 2022. A research randomizer was used to choose 200 posts as the final sample from the data that had been gathered. 122 posts out of the 200 total posts that the randomizer tallied were pertinent to the research questions for the study. Tick populations have been immigrating into northeast Tennessee from east Tennessee, Kentucky, Virginia, and North Carolina. Counties in states bordering northeast Tennessee harbor tick species associated with human illness. Human diseases transmitted by ticks include ehrlichiosis, spotted fever rickettsial group diseases, tularemia, anaplasmosis, babesiosis, Lyme disease, alpha-gal syndrome, Heartland virus, Powassan virus, and southern tick-associated rash illness (STARI). These diseases cause morbidity and mortality in human populations and may pose a high risk to individuals, wildlife, and livestock. The Cherokee National Forest covering the east Tennessee border provides a permissible environment for ticks to immigrate and thrive. Residents of northeast …

2-ω-carboxyethylpyrrole (CEP) is a product of docosahexaenoic acid (DHA) oxidation, which forms covalent adducts with different proteins. CEP-modified proteins can interact with macrophage receptor, integrin αDβ2. This study aims to compare αDβ2 binding to its physiological ligand, fibrinogen, and CEP-modified fibrinogen, which is formed during inflammation. We hypothesize that modification of fibrinogen changes its ligand-binding properties to integrin αDβ2 which can affect macrophage migration and retention. Recombinant αD I-domain and αDβ2-transfected HEK293 cells were used for the experiments. Regulation of the bacterial second messenger cyclic-di-GMP in Salmonella Typhimurium allows it to delicately alter phenotypes to optimize invasion and survive intracellularly in epithelial cells and macrophages to become virulent and cause infection. Cyclic-di-GMP concentration is regulated by the presence of external stimuli, sensory diguanylate cyclases (DGCs) and phosphodiesterases (PDEs), and cyclic-di-GMP binding effectors. Previous studies established that maintenance of low cyclic-di-GMP concentrations is required for survival in macrophages, and that deletion of 3 active PDEs reduces this survival. The severity of the COVID-19 pandemic and the high-profile nature of the public health response make it a natural context for exploring the current state of public health ethics. This paper explores this topic from two perspectives: justification and compliance. Libertarianism and utilitarianism are two frameworks that dominate the question of how public health interventions are justified. Consequently, this paper analyzes the events of the pandemic to determine how these frameworks fared in terms of offering reliable means of justifying the interventions needed to curb the spread of COVID-19. It shares a mostly commensal relationship with its hosts but can develop into an opportunistic pathogenic infection under conditions of immune suppression. Background
The use of vaccines is among the most cost-effective tools for preventing infectious diseases and their complications. However, poor uptake and increasing exemption to routine childhood vaccination have been linked with outbreaks of infectious diseases such as measles, pertussis, and more recently poliomyelitis in the US. The objective of the study is to determine the prevalence, pattern, and predisposing factors of vaccine exemptions to childhood immunization amongst parents of children in the Northeast Region from 2017 to 2021.

Human immunodeficiency virus (HIV) and hepatitis C virus (HCV) infections induce a myriad of disturbances to CD4 T cell functions, including mitochondrial compromise, excessive inflammation, increased telomeric DNA damage and attrition, cellular exhaustion and senescence, and accelerated aging. In this dissertation, the mechanisms underlying metabolic failure, accelerated aging, and cellular dysfunctions were evaluated in CD4 T cells from healthy subjects (HS) treated with a telomere-targeting drug (KML001) or HCV-infected individuals or people living with HIV (PLHIV) compared to HS. We observed that KML001-induced telomere injury resulted in mitochondrial swelling and decreased mitochondrial membrane potential, cellular respiration, mitochondrial DNA (mtDNA) copy …

Anti-retroviral therapy (ART) is the current approach to suppress active HIV replication to a level of undetected viral load, but it is not a curative approach. Newer and sophisticated gene editing technologies could indeed be a potent antiviral therapy to achieve a clinical sterilization/cure of HIV infection. For the past several decades, the United States led the world in incarceration rates. With nearly 2.3 million people being held in state or federal prisons or local jails in 2019, incarceration rates in the United States are over four times higher than in other developed countries. Disparities exist by gender, race, ethnicity, and other special populations. Males are 13 times more likely to be incarcerated than females. Additionally, black males are 5.7 times and Hispanic males are 2.8 times more likely to be incarcerated than white males. A Comparison Of The Mental Health Of Police Officers And Correctional Officers In Rural Appalachia, Sierra Thomas

The purpose of this study was to explore perceptions of mental health among police officers and correctional officers within rural Appalachia. The main goal of this research was to better understand how the occupational demands of working in the criminal justice field can impact one’s mental health over time. Several research questions were explored, including the prevalence of various mental health problems, associated stressors, the structure of support among officers, and the perceptions of mental health treatment services. Data were gathered through semi-structured interviews with 21 police and correctional officers located in rural Appalachia. Results provided a better understanding of …

JOHNSON CITY – East Tennessee State University will host the American Society for Microbiology (ASM) Fall Regional In-Person Conference on Friday and Saturday, Oct. 21-22.
The conference will be hosted by the university’s ASM student chapter and the Department of Health Sciences in the College of Public Health. JOHNSON CITY (Sept. 30, 2022) – Noted scientist, physician and administrator Dr. James E.K. Hildreth will give a free public lecture at East Tennessee State University on Thursday, Oct. 13.
Hildreth, president and CEO of Meharry Medical College, will speak at 6:30 p.m. in the Grand Soldiers Ballroom of the Carnegie Hotel, following a 5:30 p.m. reception.
