Epidemiological studies have shown that arsenic exposure during early embryogenesis can cause reduced weight gain and neurological deficits later on in life. In addition, in vitro and in vivo studies have indicated that arsenic suppresses neurogenesis and myogenesis. The exact mechanism of how arsenic causes these undesired developmental outcomes is poorly understood, however both skeletal muscle and sensory neuron development require the Wnt/β-catenin signaling pathway to initiate the specific differentiation of precursor cells. Peanut Butter Jelly, directed by Alexander Beaty, is a 51 second computer-animated short film produced by Digital Production Arts. The plot focuses on a fight sequence between a pirate jelly fish and a flyboy jelly fish over a peanut butter jar. The production demanded a photo-realistic computer generated underwater environment, which lead to the need for a custom built volume renderer to render high quality god rays. This thesis illustrates the requirement for a customized volume renderer for the production, the algorithm, and the implementation of the renderer. Osteoarthritis is one of the leading causes of lameness in the athletic horse. Cases of equine osteoarthritis typically present with synovitis, or inflammation of the synovium. This inflammation in the synovium leads to the production of inflammatory cytokines and eicosanoids. Although these downstream products can be beneficial, overproduction can lead to degradation of cartilage and joint disease. Opportunities to help mitigate or decrease inflammation within the joint are of current interest to the equine industry. One method being investigated is nutritional supplementation. My practice takes form around embodied experience. I affect signifiers of the human body within the ordered grid, the scientific text, and the logic of the machine, to highlight the interdependencies of physical bodies and those social constructs that produce and influence identity. Shiga toxin-producing Escherichia coli (STEC), a member of Enterobacteriaceae family, has been recognized as emerging pathogens. Dairy compost is commonly applied to farmland as a soil amendment. Despite the agricultural benefit of manure-based soil amendment, the inadequately treated compost can contribute to fresh produce contamination on the farm. Moreover, the epidemiological results showed that the non-O157 STEC cases have surpassed those of E. coli O157. Therefore, it is critical to evaluate the behavior of non-O157 STEC strains in the dairy manure-based compost. Tissue engineered constructs have proven to possess comparable mechanical and physical characteristics to autologous tissues making them suitable replacements fordiseased anatomy when autologous tissue transplants are unavailable. However, difficulty in reseeding the constructs with cells prevents the tissue-engineered scaffoldsfrom becoming an adequate, equivalent replacement for diseased tissues when compared to autologous transplants. Cells adhere to the outer surfaces of scaffolds but are incapable of penetrating the constructs to provide complete cellularization. My art seeks to question the social value of labor. Throughout history, labor hierarchies influenced by social class and economic stigmas have informed how laborers are viewed in the United States. Physical jobs such as menial and domestic work are a common form of invisible labor that experience debasement and stereotyping. In my art, I use labor-based and ordinary objects as a metaphor for the worker, linking the value or disposability of the object to the societal value of labor. Myocardial infarction (MI) affects nearly 600,000 individuals each year, and the resulting damage initiates a pathophysiological progression towards congestive heart failure (CHF). A shortage of donor organs precludes heart transplantation as a practical solution, and neither surgical intervention nor stem cell therapy have yielded consistent and sufficiently positive results in clinical investigation. To prevent CHF, prospective therapies must target the cause of the maladaptive cardiac remodeling which precedes it--the nonfunctional, dyskinetic infarct scar--and aim to replace it with functional cardiac muscle. Tissue engineering holds promise for the development of novel therapies to either halt or reverse post-MI cardiac remodeling. However, …

Chronic obstructive pulmonary disease (COPD) was found to be the fourth leading cause of death in 2004, but current treatments can only relieve the symptoms and improve the quality of life for the patients. Glucocorticoids have been broadly applied in other inflammatory conditions with promising therapeutic effects, but performed poorly in COPD. The phenomenon called glucocorticoid resistance is responsible for glucocorticoid insensitivity in COPD. The high demand for more efficient purification processes with increased automation and throughput pushes the development of more advanced preparative, pilot, and process scale HPLC instrumentation that is capable of achieving higher purities in a shorter amount of time than are currently achieved using one dimensional separations. Each of the three chapters of this dissertation makes a unique contribution to the fields of public finance or corporate finance. In chapter one I show that tax-price is increasing in workplace risk due to a positive wage-risk response that is observed in the labor supply price for hazardous industries. This result implies that, holding human capital constant, workers in more dangerous industries will demand a relatively smaller public sector. I test this with county-level data on fatality rates and support for the two major party candidates in the 2004 US Presidential election. Taking Republicans to represent the party of …

DNA damage by reactive oxygen species (ROS) is a cause of many chronic diseases. This work examines the ability of sulfur and selenium antioxidants to prevent oxidative DNA damage and the mechanisms for this activity. Although iron- and copper-generated hydroxyl radical are primary causes of damage under oxidative stress conditions, studies typically focus on ROS scavenging rather than antioxidant-metal binding as a mechanism for sulfur and selenium antioxidant behavior. Mass spectrometry studies of sulfur and selenoamino acids (Chapter 2) show that most form CuI and FeII complexes, regardless of their metal-mediated DNA damage prevention abilities. More than 200,000 women are diagnosed with breast cancer each year in the United States, where the majority will undergo a lumpectomy procedure and then radiotherapy as preventative treatment. Both artificial and tissue transplants may obscure the detection of cancer recurrence or limit the effectiveness of radiation treatment. Previous research in our laboratory has been performed to develop injectable lumpectomy void fillers. Although, several breakthroughs have propelled PV technology, it accounts for only less than 1% of the energy produced worldwide. Atlantic killifish, Fundulus heteroclitus, have adapted to, or at least developed a level of resistance to creosote-based polycyclic aromatic hydrocarbons (PAHs) found at the Atlantic Wood (AW) superfund site in the southern branch of the Elizabeth River, VA USA. Historically, 90+% of the mummichogs at this site have hepatic lesions of varying severity, and 30+% of these fish have tumors of various origin. Acetate is excreted as a metabolic end product in many microbes. Acetate production has primarily been studied in bacteria and archaea but is known to occur in eukaryotic organisms as well. For example, acetate is one of the most abundant metabolites excreted by the fungal pathogen Cryptococcus neoformans during cryptococcosis suggesting that acetate production may be important during pathogenesis. One possible pathway for acetate production in C. neoformans involves the enzymes xylulose 5-phosphate/ fructose 6-phosphate phosphoketolase (Xfp), which can generate acetyl phosphate from either fructose 6-phosphate (F6P) or xylulose 5-phosphate (X5P), and acetate kinase (Ack), which can then convert acetyl …

After vanishing from the public eye for more than 50 years, bed bugs have resurged to become one of the most widely discussed and heavily researched insect pests in the westernized world. Our inability to prevent and successfully treat infestations has been the driving force behind this wave of research. I addressed gaps in our understanding of bed bugs by examining behavioral and life history characteristics, as well as insecticide application responses. This dissertation explores the systematics, biogeography, and genomics of the gooseneck barnacle Pollicipes elegans, a marine crustacean of the tropical Eastern Pacific. In Chapter 1, I provide a broad framework for my research by introducing and focusing on the long-­‐standing debate of the mechanisms behind the latitudinal gradient in species diversity, which provided the initial motivation for using Pollicipes elegans as a model system to study the mechanisms leading to genetic differentiation and speciation in tropical regions.
