Real-Time Observation Of Dynamic Sarcomeric Addition In An In Vivo-Like Cardiomyocyte Culture Model, Zhonghai Wang
Real-Time Observation Of Dynamic Sarcomeric Addition In An In Vivo-Like Cardiomyocyte Culture Model, Zhonghai Wang
All Dissertations
Cardiac hypertrophy is the enlargement of individual cardiac muscle cell (cardiomyocyte) in both size and mass, which is achieved by addition of sarcomeres, the basic contractile unit. Cardiomyocytes elongate by adding sarcomeres in series and thicken by adding sarcomeres in parallel. Though it is generally accepted that sarcomeric addition can be initiated by increased mechanical loading, the sarcomeric addition process under various mechanical overloads on molecular level remains largely unknown. Previous research on sarcomeric addition largely rely on animal models of induced cardiac hypertrophy; those experiments provide little direct evidence for sarcomeric addition process as a response to increased mechanical …

Survival, Recovery, And Inactivation Of Human Norovirus Surrogates, Feline Calicivirus And Murine Norovirus, On Carpets, David Buckley
All Dissertations
Worldwide, enteric viruses are the main cause of acute gastroenteritis (AGE). Among these viruses, human noroviruses (HuNoV) are leading cause of AGE and account for ca. 20% of all diarrheal cases, a top-five cause of death worldwide. In humans, these viruses spread via person-to-person contact, food, water, and/or the environment. Person-to-person contact is the most common mode of HuNoV transmission. Yet, environmental transmission has been linked to several outbreaks and prolonged others. HuNoV survival and inactivation on hard environmental surfaces have been extensively studied. However, nonlaunderable soft surfaces, such as carpet, have received little attention despite epidemiological evidence suggesting their …
Proteinaceous Resin And Hydrophilic Encapsulation: A Self-Healing-Related Study, Ting Zheng
Proteinaceous Resin And Hydrophilic Encapsulation: A Self-Healing-Related Study, Ting Zheng
All Dissertations
Inspired by living organisms, self-healing materials have been designed as smart materials. Their automatic healing nature is achieved through the use of capsule in which the healing agent is encapsulated. The occurrence of cracks leads to ripping of the capsule, along with crack propagation and release of the healing agent that wets the crack surface to eventually heal (bond) the crack. Such automatic repair of the crack significantly extends the service life of the material. A vast majority of existing self-healing systems have been designed for the epoxy matrix - the most common commercially used thermoset - that possesses low …

Observations in a previous study on biomarker responses in fish collected from urban creeks in Greenville, SC, indicated that there might be considerable differences in the expression of biotransformation enzymes in chub and sunfish species. To further investigate these species differences a dosing experiment was performed in which bluehead and creek chub (Nocomis leptocephalus and Semotilus atromaculatus), and redbreast sunfish, pumpkinseed, and bluegill (Lepomis auritus, L. gibbosus, and L. macrochirus) were injected with benzo[a]pyrene (BaP) as a model compound for common pollutants in urban creeks. Fish were injected with BaP doses of 0, 25 and 50 mg/kg, …

The southern Appalachian Mountains have experienced rapid human population growth rates since the 1980s. Land used practices are shifting from rural to residential. The majority of development has been low density, and is often near biologically diverse areas such as National Forests and National Parks. The long-term effects of urbanization in the southeastern Appalachian Mountains are not clearly understood and even less is known with respect to stream salamander response to urbanization. In order to determine the temporal influence of exurban housing on southern Appalachian streams we sampled 27 first- and second-order streams in watersheds containing exurban developments ranging in …

Assessment Of Anemia Levels In Infants And Children In High Altitude Peru, Roxanne Amerson, Lisa Miller, Michelle Glatt, Jenna Baker
Publications
When prevalence rates of anemia exceed 40%, the World Health Organization recognizes this as a severe public health problem. In Peru, approximately 43.5% (urban) and 51.1% (rural) of children between the ages of 6 and 36 months have anemia. Currently, limited data exists regarding prevalence rates in many of the high altitude regions of Peru. The main purpose of this pilot study was to establish evidence of anemia in infants and children (7 months through 5 years of age) living in the rural, mountainous region of Ollantaytambo District. This pilot study utilized a quantitative, cross-sectional design to assess the presence …

The Influence Of Exurban Landscapes And Local Site Characteristics On Riparian Vegetation, Nathan Weaver, Kyle Barrett, Don L. Hagan
The Influence Of Exurban Landscapes And Local Site Characteristics On Riparian Vegetation, Nathan Weaver, Kyle Barrett, Don L. Hagan
Publications
The southern Appalachian Mountains have experienced large population growth and a change in land use in the past 30 years. The majority of development has been low density, suburban land, known as exurban development. The long-term effects of exurbanization on riparian vegetative communities in the southeastern Appalachian Mountains are not well known. We sought to determine if vegetative community composition and structure change as a function of watershed–level variables such as time since neighborhood development or percent impervious surface within the watershed. We also assessed local–scale measures of disturbance such as canopy cover and basal area. Over two years we …

Introducing tumor-derived cells into normal mammary stem cell niches at a sufficiently high ratio of normal to tumorous cells causes those tumor cells to undergo a change to normal mammary phenotype and yield normal mammary progeny. This phenomenon has been termed cancer cell redirection. We have developed an in vitro model that mimics in vivo redirection of cancer cells by the normal mammary microenvironment. Using the RNA profiling data from this cellular model, we examined high-level characteristics of the normal, redirected, and tumor transcriptomes and found the global expression profiles clearly distinguish the three expression states. To identify potential redirection …

Because xenosensing nuclear receptors are also lipid sensors that regulate lipid allocation, we hypothesized that toxicant-induced modulation of HR96 activity would alter lipid profiles and the balance between adult survival and neonate production following exposure in Daphnia magna. Adult daphnids were exposed to unsaturated fatty acid- and toxicant- activators or inhibitors of HR96 and later starved to test whether chemical exposure altered allocation toward survival or reproduction. The HR96 activators, linoleic acid and atrazine, decreased reproduction as expected with concomitant changes in the expression of HR96 regulated genes such as magro. The HR96 inhibitors, docosahexaenoic acid (DHA) and triclosan, …

The city of Greenville, SC is a rapidly expanding urban area located on the Reedy River in the upstate of South Carolina. Historical and current point-source pollutants and runoff from impermeable surfaces have resulted in a contaminated river environment, which through previous studies was demonstrated to be reflected in biological effects in fish species in the river. Because it was not known how much smaller tributaries in the urbanized area were contributing to the pollution of the main stem of the Reedy River, we collected fish (bluehead chub (Nocomis leptocephalus), creek chub (Semotilus atromaculatus), bluegill ( …

Connector Or Disconnector: Analyzing Impacts Of The Interstate 75/85 Connector On Historic Neighborhoods In Atlanta, Georgia, Caroline Darnell
All Theses
As a response to growing traffic in Atlanta, almost constant road work has been occurring in the city for the past 75 years. The growth of the city and the ever-changing road system has had dramatic impact on the physical deterioration of the historic fabric of the city. One of the most intense impacts came with the construction and reconstruction of the Interstate 75/85 Connector running north to south through the center of downtown. This research examines impacts of the construction of the interstate in the 1950s and the reconstruction in the 1980s to determine the level of physical deterioration …
Novel Device For Improved Diagnosis And Monitoring Of Rotator Cuff Injury, Brittany Nicole Hall
Novel Device For Improved Diagnosis And Monitoring Of Rotator Cuff Injury, Brittany Nicole Hall
All Theses
Rotator cuff disease impacts approximately over 50% of the population above the age of 60, causing pain and ultimately possible loss of shoulder function. The rotator cuff is composed of muscles and tendons that work in tandem to support the shoulder and aid in the movement of the arm. History of trauma and increased age can lead to a rotator cuff tear, which can range in severity from a partial-thickness tear to a full-thickness, total rupture. Currently, diagnostic techniques for rotator cuff disease are based on physical assessment, detailed patient history, and medical imaging, primarily X-ray, MRI and ultrasonography. However, …
Framework For Embedding Physical Systems Into Virtual Experiences, Saurabh Hindlekar
Framework For Embedding Physical Systems Into Virtual Experiences, Saurabh Hindlekar
All Theses
We present an immersive Virtual Reality (VR) experience through a combination of technologies including a physical rig; a gamelike experience; and a reï¬ned physics model with control. At its heart, the core technology introduces the concept of a physics-based communication that allows force-driven interaction to be shared between the player and game entities in the virtual world. Because the framework is generic and extendable, the application supports a myriad of interaction modes, constrained only by the limitations of the physical rig (see Figure 1). To showcase the technology, we demonstrate a locomoting robot placed in an immersive gamelike setting.
The Influence Of Sociodemographic, Location, And Health Characteristics On Environmental Risk Perception, Alicia Michel Scalia
The Influence Of Sociodemographic, Location, And Health Characteristics On Environmental Risk Perception, Alicia Michel Scalia
All Theses
Environmental risk perception is how much risk people perceive as existing due to environmental hazards. This is a topic relevant to our time, a time of environmental policy shifts and concern about pollution, and is an important concept for policy makers to understand. This study explored the following questions: Do sociodemographic characteristics, such as gender, race, age and education, influence environmental risk perception? Do location characteristics, such as if the area surrounding the home is polluted or not, and length of residence, influence environmental risk perception? And finally, do health characteristics, both self-rated health and diagnosed conditions, influence environmental risk …
Effects Of Phosphorus Concentration On Algal Composition And Bioprocessing Potential, Cassidy Laird
Effects Of Phosphorus Concentration On Algal Composition And Bioprocessing Potential, Cassidy Laird
All Theses
As an increasing amount of carbon dioxide (CO2) and other greenhouse gases pollute the lower atmosphere, and as various chemicals and elements pollute the water and soil, many search for potential solutions. One possibility is bio-mitigation using algae. Algae use CO2 and common polluting nutrients like nitrogen and phosphorus to grow. The growth, in turn, produces biomass and other byproducts that are converted to commodities such as biofuels, pharmaceuticals, cosmetics, and food supplements. Cosmetics and gelatin products are commonly sold, while other chemical products have bottlenecks in the process that require further research to make it marketable. The potential bioproducts …

Target-Specific Capture Of Relevant Gaseous Compounds Using Natural And Synthetic Nano And Micro Materials, Fernanda Delbuque Guerra
All Dissertations
By definition, volatile organic compounds (VOCs) are organic compounds that easily become vapor or gases. Many VOCs are hazardous air pollutants and often known for their low, often unpleasant, threshold odor, which can be below parts per billion in some cases. Nanotechnology-based materials for the remediation of gaseous pollutants has been the focus of many researchers due to their unique chemical and physical properties and enhanced reactivity when compared to bulkier materials. Among the many challenges preventing the global use of nanomaterials are the high costs of fabrication, on-site recovery, limited scale-up, potential toxicity since many of them rely on …
Acetate Transport Is Essential For Survival And Virulence Of Cryptococcus Neoformans, Grace Naanyu Kisirkoi
Acetate Transport Is Essential For Survival And Virulence Of Cryptococcus Neoformans, Grace Naanyu Kisirkoi
All Dissertations
Cryptococcus neoformans is a basidiomycetous fungal pathogen that claims 625,000 lives annually worldwide. Particularly among the immunocompromised, it is the leading cause of fungal meningitis following pulmonary colonization and dissemination to the central nervous system via the blood-brain barrier. The success of C. neoformans as a pathogen is largely attributable to metabolic scavenging during starvation conditions within the host's environment. Lung alveolar macrophages, which are among the first immune effectors to combat an initial pulmonary infection, present a glucose- and amino acid-poor environment that likely necessitates metabolism of nonpreferred carbon sources such as lactate and acetate for establishment of a …

Toxicity Of Microplastics To Aquatic Organisms, Sarah Au
All Dissertations
Plastic is used in products that span from cutlery to the main components in automobiles, and airplanes. Both municipal solid waste streams and marine debris are comprised of largely plastic products, and as annual global plastic production and use is unlikely to decrease, the presence of plastic in the environment will likely increase. Microplastics (MPs) are plastic fragments that have at least one dimension that is less than 5 mm, and have been found throughout water bodies around the world. MPs are in the same size fraction as most sediment particles and microorganisms, allowing them to be easily ingested by …
Development Of A Tissue Engineered Mitral Valve, Christopher Patrick Deborde
Development Of A Tissue Engineered Mitral Valve, Christopher Patrick Deborde
All Dissertations
Heart valve diseases affect nearly 8 million people every year in the United States. Of these patients, 72% are affected by mitral valve diseases. Stenosis, regurgitation, and prolapse of the mitral valve are the primary pathologies affecting valve function resulting in atrial fibrillation, arterial thromboembolism, pulmonary edema, pulmonary hypertension, cardiac hypertrophy and heart failure. Surgical options to repair or replace the mitral valve are only palliative, especially for children with congenital defects, and do not exclude the need for reoperation. A tissue-engineered option is feasible and holds great potential through the combination of decellularized scaffolds, patient stem cells, and heart …

Abducted By Reading Machines: Pragmatism And The Discourse Of Distant Reading, Matthew Scott Duncan
Abducted By Reading Machines: Pragmatism And The Discourse Of Distant Reading, Matthew Scott Duncan
All Theses
This project begins with an investigation of a case study in literary analysis that attempts to bring together close and distant reading techniques to enhance the output of both methods. The investigation reveals not only a breakdown in the attempted corroboration of truth claims and evidence across methods, but a tension felt more broadly in the discourse surrounding distant reading as a methodological and disciplinary position. In response to this problematic, I access C.S. Peirce and John Dewey to construct an argument for a pragmatic model for mediating between the more traditional methods employed in literary studies and the computational …

A Shared Resilience In Contemporary Southern Literature: Dorothy Allison's Bastard Out Of Carolina And Robin Boylorn's Sweetwater: Black Women And Tales Of Resilience, Laurie Pfister Epps
All Theses
As literary scholars, Southern Literature as we know it today needs to be all-inclusive of its people - all genders and all races. The South offers a most unique blend of colorful people that share a deep scar of a past full of injustices; however, it is through the richness in women writers' descriptions of the South and phenomenal characters with very realistic lives and circumstances that we can recognize many similarities of resilience and struggles with that identity and its daily attributes. In exploring the idea of a intercultural or intersection of Southern women and their lives, I want …
