The 5-Ht1a-R Knockout Mouse As A Model Of Later Life Anxiety Disorders: Implications For Sex Differences, Tatyana Budylin May 2019

The 5-Ht1a-R Knockout Mouse As A Model Of Later Life Anxiety Disorders: Implications For Sex Differences, Tatyana Budylin

Dissertations, Theses, and Capstone Projects

Anxiety affects nearly twice as many women as it affects men across all cultures and economic groups. Importantly, girls have a higher chance of inheriting anxiety disorders than boys, and many anxiety disorders appear at a very young age. However, little is known about sex differences in brain and behavioral development and how they relate to anxiety in adulthood. Serotonin 1A receptor (5-HT1A-R) mediated signaling has been implicated in depression and anxiety, however most studies that focus on the involvement of the 5-HT1A-R have been conducted in adults. Little is known about how the 5-HT1A …

Mechanical Kingdoms: Sound Technologies And The Avant-Garde, 1928–1933, Lauren Rosati May 2019

Mechanical Kingdoms: Sound Technologies And The Avant-Garde, 1928–1933, Lauren Rosati

Dissertations, Theses, and Capstone Projects

Against accepted histories of the historical avant-garde, which have elevated artistic production in traditional media while suppressing sonic practices, this dissertation argues that artist-engineers working across Europe and the United States independently, if simultaneously, turned their attention to emerging sound technologies as new media for creative experimentation by the early 1930s. This spectrum of activity demonstrates the significance of sound in avant-garde practice, and indicates a wide-ranging artistic engagement with technological devices intended for mass audiences. While the common understanding of the relation between art and technology in this period amounts to one of mere enthusiasm for the novel formal …

Refusing White Privacy, Olivia Dunbar May 2019

Refusing White Privacy, Olivia Dunbar

Dissertations, Theses, and Capstone Projects

In “Refusing White Privacy” I look at theories in White Data and Surveillance Studies around what data is, how it is made to exist, and for whom, in order to intervene in the conceptualization of data as an inevitable residue of human life and relationship. Through this intervention, I show that the alleged crises of privacy ushered in by allegedly non-racial smart technologies (a preoccupation in WDSS) is underwritten by racializing technologies from the Antebellum era to the present.

The Subject Of Jouissance: The Late Lacan And Gender And Queer Theories, Frederic C. Baitinger May 2019

The Subject Of Jouissance: The Late Lacan And Gender And Queer Theories, Frederic C. Baitinger

Dissertations, Theses, and Capstone Projects

The Subject of Jouissance argues that Lacan’s approach to psychoanalysis, far from being heteronormative, offers a notion of identity that deconstructs gender as a social norm, and opens onto a non-normative theory of the subject (of jouissance) that still remains to be fully explored by feminist, gender, and queer scholars. Drawing mostly on the later Lacan, The Subject of Jouissance shows that by locating the identity of the subject in the singularity of its bodily mode of enjoyment (that Lacan calls “jouissance”), and not in the Imaginary illusions of the ego, nor in the Symbolic social structures, Lacan fosters thinking …

A Pretargeted Spect Imaging Strategy Employing Technetium-99m Based On Bioorthogonal Diels-Alder Click Chemistry, Justin H. Walsh May 2019

A Pretargeted Spect Imaging Strategy Employing Technetium-99m Based On Bioorthogonal Diels-Alder Click Chemistry, Justin H. Walsh

Dissertations, Theses, and Capstone Projects

A series of related N3S 99mTc-peptide complexes were synthesized and tested for use in pretargeting SPECT imaging that utilizes the bioorthogonal Diels-Alder click reaction between tetrazine (Tz) and transcyclooctene (TCO). The objective was to optimize the excretory pathways of the 99mTc-peptide complexes for maximum tumor targeting with the in vivo “click” and minimum non-target uptake. The 99mTc–tetrazine constructs were prepared by reaction of 99mTc-peptide complexes (99mTc-FKC, 99mTc-FKCR, 99mTc-DKC, and 99mTc-SKC) with Tz-NHS or Tz-PEG5-NHS to form 99mTc FK(Tz)C, 99mTc-FK(PEG5-Tz)CR, 99mTc-DK(PEG5-Tz)C, …

Stabilizing Forces In Acoustic Cultural Evolution: Comparing Humans And Birds, Daniel C. Mann May 2019

Stabilizing Forces In Acoustic Cultural Evolution: Comparing Humans And Birds, Daniel C. Mann

Dissertations, Theses, and Capstone Projects

Learned acoustic communication systems, like birdsong and spoken human language, can be described from two seemingly contradictory perspectives. On one hand, learned acoustic communication systems can be remarkably consistent. Substantive and descriptive generalizations can be made which hold for a majority of populations within a species. On the other hand, learned acoustic communication systems are often highly variable. The degree of variation is often so great that few, if any, substantive generalizations hold for all populations in a species.

Within my dissertation, I explore the interplay of variation and uniformity in three vocal learning species: budgerigars (Melopsittacus undulatus), …

Between Politics And Morality: Hans Kelsen's Contributions To The Changing Notion Of International Criminal Responsibility, Jason Kropsky May 2019

Between Politics And Morality: Hans Kelsen's Contributions To The Changing Notion Of International Criminal Responsibility, Jason Kropsky

Dissertations, Theses, and Capstone Projects

The pure theory of law analyzes the legal normative basis of jurisprudence. According to its author, Hans Kelsen (1881-1973), the study of law as a science can only arise once “alien elements” associated with sociology, politics, ethics and psychology are extracted from strict legal cognition. But what happens when the international sphere of law that possesses the special quality of holding state officials accountable for core international crimes requires intrusion by extra-legal sources? Does Kelsen’s structural edifice collapse? Or is it reconstituted? In examining how international criminal responsibility, a test case for Kelsen’s positive law claims derives its legitimacy, this …

Working Lives: Artistic Solidarity In Revolutionary Peru (1960–1980), Jose R. Chavarry May 2019

Working Lives: Artistic Solidarity In Revolutionary Peru (1960–1980), Jose R. Chavarry

Dissertations, Theses, and Capstone Projects

This dissertation examines the discourses and experiences of cultural work as a form of intellectual and artistic solidarity in Peru during the 1960s and 1970s. Amid the broader Latin American and global spirit of revolution, anti-imperialism and Third World liberation, in Peru these decades saw a radical transformation in society where rural and urban masses rose against a traditional political and socioeconomic system that maintained colonial structures of domination and oppression of marginalized populations. In an attempt to rein in this desborde popular, as it became known, the nationalist and populist Revolutionary Government of the Armed Forces and a …

Getting At The Surface: A Promoter And Coding Sequence Characterization Of An Odorant Receptor, Irena G. Parvanova May 2019

Getting At The Surface: A Promoter And Coding Sequence Characterization Of An Odorant Receptor, Irena G. Parvanova

Dissertations, Theses, and Capstone Projects

Odorant receptors (ORs) are expressed by mature olfactory sensory neurons (OSNs), in a monogenic and monoallelic fashion (singular gene choice). It has been repeatedly published that the singular expression of ORs is a consequence of epigenetic regulation. The model invokes the depositing of epigenetic marks (off marks) that reflect transcriptional inactivity on all OR genes in immature neurons followed by the activation of one OR allele in maturing neurons. OR expression has been exclusively studied via transgenic animal models, due to the lack of cell line system that can undergo singular gene choice. Here, we used chromatin immunoprecipitation-quantitative polymerase chain …

The Use Of Trazodone Hydrochloride To Reduce Stress In Dogs During The Transition Period Into A Municipal Animal Shelter: Impact On Incidence Of Illness, Length Of Stay, And Outcome, Jennifer L. Abrams

Theses and Dissertations

A large municipal animal shelter in NYC gave dogs low doses of trazodone hydrochloride at the time of intake to evaluate its efficacy in reducing transitional stress. Statistically significant decreases in illness rate and length of stay and increase in adoption rate were observed in the dogs given trazodone.

Synchronicity: The Role Of Midbrain Dopamine In Whole-Brain Coordination, Jeff A. Beeler, Jakob Kiskye Dreyer Apr 2019

Synchronicity: The Role Of Midbrain Dopamine In Whole-Brain Coordination, Jeff A. Beeler, Jakob Kiskye Dreyer

Publications and Research

Midbrain dopamine seems to play an outsized role in motivated behavior and learning. Widely associated with mediating reward-related behavior, decision making, and learning, dopamine continues to generate controversies in the field. While many studies and theories focus on what dopamine cells encode, the question of how the midbrain derives the information it encodes is poorly understood and comparatively less addressed. Recent anatomical studies suggest greater diversity and complexity of afferent inputs than previously appreciated, requiring rethinking of prior models. Here, we elaborate a hypothesis that construes midbrain dopamine as implementing a Bayesian selector in which individual dopamine cells sample afferent …

Stephen, Alex G. Chalco, Usman M. Arshad, Gary W. Thickbroom, Josh Silverstein, K. Zoe Tsagaris, Amy Kuceyeski, Kathleen Friel, Taiza E. G. Santos, Dylan J. Edwards

Publications and Research

The recent enhanced sophistication of non-invasive mapping of the human motor cortex using MRI-guided Transcranial Magnetic Stimulation (TMS) techniques, has not been matched by refinement of methods for generating maps from motor evoked potential (MEP) data, or in quantifying map features. This is despite continued interest in understanding cortical reorganization for natural adaptive processes such as skill learning, or in the case of motor recovery, such as after lesion affecting the corticospinal system. With the observation that TMS-MEP map calculation and quantification methods vary, and that no readily available commercial or free software exists, we sought to establish and make …

The Serial Blocking Effect: A Testbed For The Neural Mechanisms Of Temporal-Difference Learning, Ashraf Mahmud, Petio Petrov, Guillem R. Esber, Mihaela D. Iordanova

Publications and Research

Temporal-difference (TD) learning models afford the neuroscientist a theory-driven roadmap in the quest for the neural mechanisms of reinforcement learning. The application of these models to understanding the role of phasic midbrain dopaminergic responses in reward prediction learning constitutes one of the greatest success stories in behavioural and cognitive neuroscience. Critically, the classic learning paradigms associated with TD are poorly suited to cast light on its neural implementation, thus hampering progress. Here, we present a serial blocking paradigm in rodents that overcomes these limitations and allows for the simultaneous investigation of two cardinal TD tenets; namely, that learning depends on …

Effects Of Vitamin D3 And Its Chemical Analogs On The Growth Of Hodgkin’S Lymphoma, In Vitro, Rajendra Ghardbaran, Bo Zhang, Luis Valerio, Onyekwere Onwumere, Madeline Wong, Jason Mighty, Stephen Redenti

Publications and Research

Objective: Vitamin D receptor (VDR) activities have been noted for a number of B cell malignancies which showed varying sensitivities to vitamin D3 (1,25-dihydroxyvitamin D3, VD3, calcitriol) and its synthetic analogs. The objective of this study was to address the potential effects of VD3 and vitamin D3 analogs (VDAs) on the growth of Hodgkin’s lymphoma (HL), a malignant pathology of B cell origin, in vitro.

Results: Immunofluorescence staining showed the expression of VDR by primary Hodgkin’s (H) and Reed–Sternberg (RS)—HRS-tumor cells in HL histological sections. Western blot analyses revealed expression of VDR in the HL cell lines Hs445, HDLM2, KMH2, …

Visual morphology assessment is routinely used for evaluating of embryo quality and selecting human blastocysts for transfer after in vitro fertilization (IVF). However, the assessment produces different results between embryologists and as a result, the success rate of IVF remains low. To overcome uncertainties in embryo quality, multiple embryos are often implanted resulting in undesired multiple pregnancies and complications. Unlike in other imaging fields, human embryology and IVF have not yet leveraged artificial intelligence (AI) for unbiased, automated embryo assessment. We postulated that an AI approach trained on thousands of embryos can reliably predict embryo quality without human intervention. We …

On Using Ai Bots For Voice Controlled Augmented Reality Applications, Juan Estrella

Publications and Research

Artificial Intelligence (AI) has the potential to benefit society in the realms of medicine, security, manufacturing, entertainment, marketing, and many others. One of the advances of AI is in the field of Natural Language Processing and Speech Recognition; making computers understand what humans say and mean. On the other hand, the term Augmented Reality (AR) refers to the technologies that superimpose digital content generated by computers over the user’s view of the real world. AR technologies enhance the version of the physical environment with computergenerated sensory input such as sound, video, or graphics overlaid on top of the real-world view. …

Studying The Gang Through Critical Ethnography, David C. Brotherton Apr 2019

Studying The Gang Through Critical Ethnography, David C. Brotherton

Publications and Research

Added to the paucity of critical lenses through which the gang has been viewed criminologically is the increasing influence of the US criminal justice system on the global gang discourse. Such a lens has increased in importance as many nation states have followed the example of US repressive gang policies in thinking about crime and deviance, essentially mirroring its adoption of neo-liberalism in thinking about the political economy. In such an approach it is assumed that a coercive social control system is required to discipline and warehouse those “problem populations” excluded by the concentration of wealth and power. Across the …

Spinal Matrix Metalloproteinase 8 Regulates Painafter Peripheral Trauma, Maral Tajerian, J. David Clark Apr 2019

Spinal Matrix Metalloproteinase 8 Regulates Painafter Peripheral Trauma, Maral Tajerian, J. David Clark

Publications and Research

It is well documented that pain chronification requires a host of plastic mechanisms at the spinal cord (SC) level, including alterations in neuronal and glial structure and function. Such cellular plasticity necessitates the existence of a plastic extracellular matrix(ECM). Here, we describe a key role for ECM remodeling in the regulation of chronic pain following peripheral injury. Three weeks following tibia fracture in mice, we show increased levels of MMP8 in the SC. Furthermore, we show that the pharmacological or genetic down regulation of MMP8 ameliorates the pain phenotype observed after injury. These results delineate an extracellular mechanism for pain …

What Is Critical About Critical Librarianship?, Emily Drabinski

Publications and Research

Library work structures intellectual worlds as library workers collect, organize, make accessible, and preserve materials for use. This work is not neutral. Libraries, like all institutions, are produced in and through systems marked by racism, patriarchy, and capitalist modes of production. Critical librarianship offers a framework for thinking about our work that asks how library structures came to be and what ideologies underpin them. Viewing librarianship through this frame allows us to imagine new and better worlds on our way to making them.

Diets low in methionine extend lifespan of rodents, though through unknown mechanisms. Glycine can mitigate methionine toxicity, and a small prior study has suggested that supplemental glycine could extend lifespan of Fischer 344 rats. We therefore evaluated the effects of an 8% glycine diet on lifespan and pathology of genetically heterogeneous mice in the context of the Interventions Testing Program. Elevated glycine led to a small (4%–6%) but statistically significant lifespan increase, as well as an increase in maximum lifespan, in both males (p = 0.002) and females (p < 0.001). Pooling across sex, glycine increased lifespan at each of the three independent sites, with significance at p = 0.01, 0.053, and 0.03, respectively. Glycinesupplemented females were lighter than controls, but there was no effect on weight in males. End‐of‐life necropsies suggested that glycine‐treated mice were less likely than controls to die of pulmonary adenocarcinoma (p = 0.03). Of the 40 varieties of incidental pathology evaluated in these mice, none were increased to a significant degree by the glycine‐supplemented diet. In parallel analyses of the same cohort, we found no benefits from TM5441 (an inhibitor of PAI‐1, the primary inhibitor of tissue and urokinase plasminogen activators), inulin (a source of soluble fiber), or aspirin at either of two doses. Our glycine results strengthen the idea that modulation of dietary amino acid levels can increase healthy lifespan in mice, and provide a foundation for further investigation of dietary effects on aging and late‐life diseases.

Crowd Wisdom Enhanced By Costly Signaling In A Virtual Rating System, Ofer Tchernichovski, Lucas C. Parra, Daniel Fimiarz, Arnon Lotem, Dalton Conley

Publications and Research

Costly signaling theory was developed in both economics and biology and has been used to explain a wide range of phenomena. However, the theory’s prediction that signal cost can enforce information quality in the design of new communication systems has never been put to an empirical test. Here we show that imposing time costs on reporting extreme scores can improve crowd wisdom in a previously cost-free rating system. We developed an online game where individuals interacted repeatedly with simulated services and rated them for satisfaction. We associated ratings with differential time costs by endowing the graphical user interface that solicited …

Carotenoids are lipophilic pigments found in plants and algae, as well as some bacteria, archaea, and fungi that serve two functions—(1) as light harvesting molecules—primary carotenoids, and (2) as antioxidants, acting against reactive oxygen species–secondary carotenoids. Because of their strong antioxidant properties, they are also valuable for the development of anti-aging and photo-protective cosmetic applications. Of particular interest is the carotenoid phytoene, for its colorless and UV absorption characteristics. In this study, we targeted a reduction of phytoene desaturase (PDS) activity with the pigment-inhibiting herbicide 1-methyl-3-phenyl-5-[3- (trifluoromethyl)phenyl]pyridin-4-one (fluridone), which leads to the over-accumulation of phytoene in the recently characterized microalgal …

Dual Complementary Liposomes Inhibit Triple-Negative Breast Tumor Progression And Metastasis, Peng Guo, Jiang Yang, Daxing Liu, Lan Huang, Gillian Fell, Jin Huang, Marsha A. Moses, Debra T. Auguste

Publications and Research

Distinguishing malignant cells from non-neoplastic ones is a major challenge in triple-negative breast cancer (TNBC) treatment. Here, we developed a complementary targeting strategy that uses precisely matched, multivalent ligand-receptor interactions to recognize and target TNBC tumors at the primary site and metastatic lesions. We screened a panel of cancer cell surface markers and identified intercellular adhesion molecule–1 (ICAM1) and epithelial growth factor receptor (EGFR) as optimal candidates for TNBC complementary targeting. We engineered a dual complementary liposome (DCL) that precisely complements the molecular ratio and organization of ICAM1 and EGFR specific to TNBC cell surfaces. Our in vitro mechanistic studies …

Background: Although myelin is composed of mostly lipids, the pathological role of myelin lipids in demyelinating diseases remains elusive. The principal lipid of the myelin sheath is b-galactosylceramide (b-Galcer). Its a-anomer (a-Galcer) has been demonstrated to be antigenically presented by macrophages via CD1d, a MHC class I-like molecule. Myelin, which is mostly composed of b-Galcer, has been long considered as an immunologically-inert neuron insulator, because the antigen-binding cleft of CD1d is highly a-form-restricted.

Results: Here, we report that CD1d-mediated antigenic presentation of myelin-derived galactosylceramide (Mye-GalCer) by macrophages contributed significantly to the progression of experimental autoimmune encephalomyelitis (EAE). Surprisingly, this presentation …
