The Impact Of The Nonsignaling Domain On The Effect Of Tim3 Car T Cell In Acute Myeloid Leukemia, Kristine Cate Singayan Pe

Chulalongkorn University Theses and Dissertations (Chula ETD)

Translation of CAR T cell therapy for AML has been challenging due to the lack of target specificity. One promising target is TIM3, which is overexpressed on AML cells and absent in majority of healthy hematopoietic cells. Previous reports have shown that each of CAR’s structural designs impacts CAR functionality. Here, we evaluated the influence of non-signaling domains on a preclinical TIM3-targeting CAR construct. To investigate the impact of the spacer and transmembrane non-signaling domain, we designed anti-TIM3 CARs containing either IgG2-CH3 spacer and CD28 transmembrane (TM) domain or CD8α spacer-TM, linked to intracellular 4-1BB and CD3ζ signaling domain. Anti-TIM3 …

Looped Homology Theory Of Weighted Reflexive Graphs, Sahawat Wongkhunen Jan 2023

Looped Homology Theory Of Weighted Reflexive Graphs, Sahawat Wongkhunen

Chulalongkorn University Theses and Dissertations (Chula ETD)

In recent years, the notions of discrete homotopy for graphs, such as A-homotopy, x-homotopy and fundamental groupoid for graphs, have been introduced. Next, these theories are developed and extended for further studies, motivating us to study the discrete homology theory of graphs. This dissertation develops the notion of a looped fundamental groupoid for graphs to the isotropy group of the vertex v in the fundamental groupoid. We show that it is similar to the classical fundamental group of a topological space and the A-theory of graphs in some sense. Then, we apply this theory's idea to the looped fundamental group …

Theoretical Study Of Nickel-Complex Electrocatalyzed Carbon Dioxide Conversion, Wilasinee Santiwarodom

Chulalongkorn University Theses and Dissertations (Chula ETD)

The electrochemical conversion of CO2 to CO using nickel-N4-Schiff base complexes as catalysts was studied by Density Functional Theory (DFT). Three nickel complexes from Juthathan et. al. were analyzed: 1-Ni, 2-Ni, and [2-Ni]Me. Two CO2 conversion pathways, external and internal proton transfer, were proposed, and their reaction energy profiles were computed. The external proton transfer pathway, which involves three steps, does not contain a transition state. All steps are exothermic, with the overall reaction energies being lowest for 1-Ni (-5.72 eV), followed by 2-Ni (-5.56 eV) and [2-Ni]Me (-5.54 eV). The internal proton transfer pathway consists of four steps, including …

Cisplatin - Loaded Calcium Citrate Nanoparticlesfor Inhibition Of Lung Cancer Cell Growthand Quantum Mechanical Study Of Cisplatin And Citrate, Lipika Oopkaew

Chulalongkorn University Theses and Dissertations (Chula ETD)

The International Organization for Research on Cancer (International Agency for Research on Cancer (IARC) issued a statement saying lung cancer is a cancer with high incidence rates of new cases and deaths. Although there are current therapeutic approaches such as surgery, radiation therapy, and chemotherapy. However, these treatments exhibit ineffectiveness; the main limitation is severe side effects to normal cells or non-target organs. To address this challenge, the development of targeted therapy by nanoparticle-based drug delivery system is applied to increase drug stability and control the specificity to cancer cells with target-specific proteins that play an important role in intracellular …

Bi-Directional Evolutionary Structural Optimization For 3d-Printed Prefabricated Concrete Panels, Pochara Amornsittipipat Jan 2023

Bi-Directional Evolutionary Structural Optimization For 3d-Printed Prefabricated Concrete Panels, Pochara Amornsittipipat

Chulalongkorn University Theses and Dissertations (Chula ETD)

This thesis seeks to develop an optimal design methodology for 3D-printed prefabricated concrete panels. Although 3D-printed precast panels are advantageous for producing flexible forms, reducing workforce demand, and minimizing environmental impacts, the product must not be too heavy to transport and install. Therefore, the consideration of weight emphasizes the need for structural optimization. This research presents truss-like structural modeling based on the properties of 3D-printed concrete, suggests an optimal design methodology using the Bi-directional Evolutionary Structural Optimization (BESO) algorithm to achieve minimum weight, conducts parametric studies to understand the relationship between significant parameters and the optimal weight achieved, and demonstrates …

Image-Base Adaptive Proportional Topology Optimization Of Infill Structure Using Polygon Sbfe Approach, Sompong Rungsawang Jan 2023

Image-Base Adaptive Proportional Topology Optimization Of Infill Structure Using Polygon Sbfe Approach, Sompong Rungsawang

Chulalongkorn University Theses and Dissertations (Chula ETD)

The use of infill structures is widespread in additive manufacturing applications, offering porous domains that are well-suited for withstanding various physical loadings. This thesis introduces a novel approach, namely Proportional Topology Optimization (PTO), leveraging an automatic image-based quadtree Scaled Boundary Finite Element (SBFE) framework. The SBFE, where sub-domains are treated as polygonal elements in 2D, is employed to address the hanging node issue. The methodology is extended to design infill structures by minimizing compliance concerning force variations, subject to local volume fraction constraints rather than a global volume constraint for each element in the design domain. Material density for each …
