Synthesis, Cytotoxicity And Anti-Inflammatory Activity Of Piperine Derivatives, Dina Nur Shinta
Synthesis, Cytotoxicity And Anti-Inflammatory Activity Of Piperine Derivatives, Dina Nur Shinta
Chulalongkorn University Theses and Dissertations (Chula ETD)
Cancer is a significant leading cause of mortality worldwide which has been struggled in the clinical over the years. Parallelly, chronic inflammation becomes one of the risk factors to cause cancer. In searching for new potential anti-cancer and anti-inflammatory agents, an amide, ester, and dimer series of piperine derivatives were synthesized and screened for their anti-cancer and anti-inflammatory activities. Piperine (1) was isolated from fruits of Piper longum L. and hydrolyzed under reflux conditions to obtain piperic acid (2) as a starting material for the synthesis. The conversion of piperic acid to an amide series (3a-j) was involving an azabenzotriazole …

Effects Of Small Molecules In Wnt Signaling On Human Dental Pulp Cells, Chatvadee Kornsuthisopon
Effects Of Small Molecules In Wnt Signaling On Human Dental Pulp Cells, Chatvadee Kornsuthisopon
Chulalongkorn University Theses and Dissertations (Chula ETD)
Osteoblast differentiation requires interaction of several cell signaling to coordinately modulate cell responses. Understanding the integration between signaling pathways provides an insight into molecular mechanism that governs stem cell differentiation. Notch, Wnt, and Interleukin15 (IL15) signaling are among the crucial pathways that control biological processes including osteogenic differentiation. Therefore, this dissertation was focused on modulating these signaling pathways in human dental pulp stem cells (hDPSCs). First, the roles of Wnt signaling in dental pulp homeostasis and dentin regeneration were reviewed. Previous evidence suggested that Wnt might be a promising target cell signaling for dentin regeneration. Thus, effect of Tideglusib, a …

Predicting Treatment Response In Nasopharyngeal Cancer Using Radiomics Features Based On Diffusion-Weighting Imaging (Dwi), Sasawan Wongpisarn
Chulalongkorn University Theses and Dissertations (Chula ETD)
Nasopharyngeal carcinoma (NPC) is common in Southeast Asia, includes Thailand. The patients could treat with concurrent chemoradiation therapy (CCRT). Nowadays, treatment response is assessed from the changed size after treatment. Due to this assessment, it occurs a limitation in considering a changing of heterogeneity within tumor. To overcome this problem, Radiomic is an emerging technique which provides information on tumor characteristics non-invasively with lower cost as compared with biopsy. Several studies used radiomic feature from a single timepoint, especially from pre-treatment to predict the treatment response of various cancer types. On the other hand, the relative change of radiomic feature …

Ocular Surface Microbiome In Diabetes Mellitus, Orathai Suwajanakorn
Chulalongkorn University Theses and Dissertations (Chula ETD)
This cross-sectional, age and gender matched study included 20 eyes of non-diabetic subjects (non-DM group) and 60 eyes of type 2 diabetes mellitus (DM group). Subgroups of DM were classified by diabetic retinopathy (DR) staging into no DR (DM-no DR), non-proliferative DR (DM-NPDR), proliferative DR (DM-PDR), and by glycemic control (well-controlled DM; HbA1c<7%, poorly-controlled DM; HbA1c≥7%). Conjunctival swabs were performed for ocular surface microbiome analysis using conventional culture and next-generation sequencing analysis (NGS). A higher culture-positive rate was found in DM (15%) than in non-DM group (5%) (pvalue=0.437). Antibiotic-resistant organisms were only detected in the DR groups (DM-NPDR and DM-PDR). The NGS analysis showed that potentially pathogenic bacteria predominated in DM, especially in DR. There was dissimilarity in the ocular surface microbiome between DM and non-DM groups. The subgroup analysis showed that the DR group had significantly different microbial community from DM-no DR and non-DM groups (p-value<0.05). The microbial community in the poorly-controlled DM was also significantly different from well-controlled DM and non-DM groups (p<0.001). Using the NGS method, this study is the first to signify the importance of DR and glycemic control status, which affect the changes in the ocular surface microbiome.

Development Of Tetrahydrocurcumin-Loaded Chitosan-Coated-Nanostructured Lipid Carriers For Topical Treatment Of Psoriasis, Hoang Thien Truong
Chulalongkorn University Theses and Dissertations (Chula ETD)
Chitosan (Ch)-coated nanostructured lipid carriers (NLCs) have great potential for topical delivery with high localization of chemotherapeutics. In this study, tetrahydrocurcumin (THC), a primary metabolite of curcumin with enhanced antioxidant and anticancer properties, was used as a model drug to prepare NLCs. Response surface methodology was employed to optimize THC-loaded Ch-coated NLCs (THC-Ch-NLCs) fabricated by high-shear homogenization. The optimized THC-Ch-NLCs had particle size of 244 ± 18 nm, zeta potential of -17.5 ± 0.5 mV, entrapment efficiency of 76.6 ± 0.2% and drug loading of 0.28 ± 0.01 mg/mL. In vitro release study of THC-Ch-NLCs showed sustained release following the …

Development Of Hek-293t Cells Stably Expressing The C-X-C Chemokine Receptor Type 4 (Cxcr4) For Binding Assay Of Potential Anti-Breast Cancer Agents, Thi Thai Ha Dinh
Chulalongkorn University Theses and Dissertations (Chula ETD)
The C-X-C chemokine receptor type 4 (CXCR4) has been recognized as a potential target in cancer drug discovery based on the high expression in cancer cells, particularly invasive breast cancer. This study aims to construct CXCR4-overexpressed cells to screen new anti-breast cancer agents targeting CXCR4 and identify the mediated target for anticancer effect. Firstly, five genetic isotypes of CXCR4, namely A, B, C, D and E, were amplified from Jurkat T cells. Amplicons of each CXCR4 isotype were ligated into cloning vectors and subcloned into expression vectors. They were transformed into competent Escherichia coli strain DH5-alpha cells. The presence of …
Neuroprotective Effect Of Passiflora Edulissims Against Amyloid Beta-Induced Neurotoxicity, Shuqin Cao
Neuroprotective Effect Of Passiflora Edulissims Against Amyloid Beta-Induced Neurotoxicity, Shuqin Cao
Chulalongkorn University Theses and Dissertations (Chula ETD)
Alzheimer’s disease (AD) is a common and devastating disease characterized by pathological aggregations of beta-amyloid (Aβ) plaques extracellularly, and Tau tangles intracellularly. While our understandings of the aetiologies of AD have greatly expanded over the decades, there is no drug available to stop disease progression. This study involves two parts. First, we demonstrate the potential of P. edulis pericarp (PP) extract in protecting against Aβ-mediated neurotoxicity in mammalian cells and Caenorhabditis elegans models of AD. We show PP protects against memory deficit, neuronal loss, and promotes longevity in the Aβ model of AD via stimulation of mitophagy, the selective cellular …

The Effect Of Walking Training With Textured Insole Socks On Standing Balance And Gait In Older Adults With A Risk Of Falls, Chanon Pinvanichkul
Chulalongkorn University Theses and Dissertations (Chula ETD)
Previous research revealed that textured surface treatments improved standing balance performance or had a negative effect on gait performance in older adults. The new design textured insole socks were introduced in this study. This study aimed to evaluate the effects of textured insole socks on standing balance, gait parameters, and a risk of falls in older adults with a risk of falls. Thirty older adults with a risk of falls participated and randomly allocated equally to the intervention (texture insole socks) or control (unmodified socks) groups. Both groups were asked to walk on a 10-meter walkway for 2,000 steps/day, 3 …
