The Microphysiological Systems Laboratory aims to develop colorectal cancer tumor models under a hypoxic environment to assess model response to pharmaceutical compounds in vitro. To perform relevant studies, researchers have attempted to use different hypoxic inducing strategies such as a nitrogen pod and hypoxic incubator to recreate in vivo physiological responses to hypoxia. However, studies would be interrupted due to incubator functionality failure. To ensure successful and physiologically relevant studies, I improved and verified the robustness and reliability of a hypoxic incubator previously designed and manufactured in the lab. The fabrication of microfluidic devices often requires specialized methods. The development of these methods requires careful characterization and understanding of the processes involved. Using primarily PolyJet 3D printing technology, microfluidics offers a wide scope of applications such as microfluidic benthic microbial fuel cells (MBMFCs) and electrostatic artificial muscles. MBMFCs benefit from the confinement of the microbes resulting in close proximity between the electrode and the organisms. Using a modular design called the Sponge, assembly and upscaling is possible. Electrostatic artificial muscles benefit from a microfluidic approach due to the non-linearity of electrostatic attraction creating disproportionate benefits when miniaturized. When designed …

In order to avoid damaging live cells, optical microscope imaging must be conducted under low-excitation light intensity and/or short exposure times, resulting in low signal-to-noise ratios (SNR). Deep learning methods offer an effective solution for removing microscope noise, utilizing algorithms that are able to reconstruct finer features in low SNR images. This research explores the denoising capability of several deep learning methods based on PSNR and SSIM. Tested methods include traditional approaches (BMED), supervised learning (CARE and Restormer), and unsupervised methods (Noise2Fast, N2V, SSD-Unsupervised, and SASSID). Terrestrial microplastics are increasing in abundance and are an emerging contaminant of concern, particularly in agriculture ecosystems where plastic use and subsequent soil contamination are prevalent. Plastic film mulches make up a significant portion of the plasticulture industry and are directly linked to soil microplastic pollution where negative effects to soil biotic and abiotic functions have been observed. In response to this issue, biodegradable film mulches have been promoted as a sustainable alternative to conventional polyethylene-based mulches. Virtual or hybrid options provide a way for marginalized faculty and staff to fully participate in their fields and on their campuses. As such, an equitable and pandemic-informed academic workplace should include fully accessible and well-resourced hybrid participation options for department meetings and events, office work, and classroom duties. In spring 2023, we created a work group on our campus to address the challenges of hybrid work and co-create campus-wide recommendations for hybrid workspaces. This ethnography draws on key informant interviews and participant observation to produce a case study about plastic mulching as an important industry standard in specialty crop production. It documents the historic adoption and present role of plastic mulching technology in California strawberry production and addresses the characteristics of commercial standard management that inhibit the adoption of alternative mulching practices. The Nasogastric Tube Insertion Aid project, developed under the sponsorship of the BiCEP Program at California Polytechnic State University, aims to enhance the accuracy and efficiency of nasogastric (NG) tube placement. This initiative was inspired by challenges observed during the BiCEP summer program, where difficulties in NG tube insertions led to multiple attempts, increased patient discomfort, and serious medical risks, including lung punctures and brain trauma. Survey to quantify the frequency of the five main pathogens (Macrophomina phaseolina, Fusarium oxysporum f. sp. fragariae races 1 and 2, Verticillium dahliae, and Phytophthora spp.) infecting strawberry in the Oxnard growing district of California. M. phaseolina was the most prevalent pathogen detected in diseased plants in the Oxnard growing district, present in 67.6% of samples positive for at least one pathogen. F. oxysporum f. sp. fragariae race 1 (35.3%), V. dahlia (8.8%), F. oxysporum f. sp. fragariae race 2 (5.9%) and Phytophthora spp. (2.9%) followed at lower prevalence. Achieving a blue economy will require reconciling the value of emerging ocean uses with their impacts on the seascape and sectors with historical access to marine resources and areas. To meet this challenge, we developed an analytical framework for conducting marine spatial planning through tradeoff analysis, and applied it to prospective offshore wind energy development in the ∼974 km2 Morro Bay, California, USA Wind Energy Area (WEA). Grazing pressure is important in structuring plant communities, particularly in aquatic environments. Eelgrass (Zostera marina), a keystone species in estuaries across North America, is experiencing widespread population declines. As a keystone species, eelgrass provides numerous important functions, including: 1) water filtration, 2) sediment stabilization, 3) providing refuge and nursery habitat for numerous species, and 4) carbon sequestration. Benefits which could be lost if eelgrass beds continue to diminish. The Vertebrate Integrative Physiology (VIP) lab monitors the population of northern elephant seals at the largest mainland breeding colony, located at Piedras Blancas (San Simeon, CA). As the population expands, more human-seal interactions and conflicts over land use occur. The VIP lab's work informs California State Parks and helps with the management of the rookery. Currently, members of the VIP lab fly a drone over the beaches, capture multiple images, and manually count the seals, which takes around 14 to 21 hours of analysis per survey. Ambush-hunting snakes have long been of great interest in the biology of fasting and starvation, as they are adapted to withstand well over a year without taking a meal. Well-justified ethical constraints have limited our understanding of their biology at the limits of these abilities. Unfortunate circumstances, in which the California Department of Fish and Wildlife confiscated over 50 Western Rattlesnakes (Crotalus oreganus) from an animal hoarder that had long neglected them, culminated in the euthanized animals ending up in the hands of our lab. In 2023, approximately 2.36 million metric tons of olive oil were produced globally. Olive pomace, a byproduct of the flesh and pits left after olive oil extraction, presents environmental challenges when used as landfill due to its high polyphenol and organic contents, or when combusted due to greenhouse gas emissions. Its potential as animal feed is limited, yet it holds promise for methane production via anaerobic digestion (AD), providing a source of renewable energy. Total knee arthroplasty (TKA) is one of the most commonly performed orthopedic surgeries, with nearly one million performed in 2020 in the United States alone. This research developed methods for culturing self-assembling capillaries in an injectable gel as a potential method for vascularizing tissue-on-a-chip models to mimic physiological drug delivery. Additionally, a mathematical model was developed as a tool for understanding nutrient delivery and comparison of potential delivery systems. Organs-on-a-chip provide novel platforms for studying biology and physiology in 3D, allow exploration of tissue engineering on a manageable scale, and serve as models for drug screening and drug-delivery testing.

This concept plan proposes preliminary design and implementation measures to redevelop an area which holds potential to serve as an activated southern entrance to Cannery Row. This project provides an analysis of the existing conditions and subsequently offers a strategy to transform this site into the Coastal Collective, a community-oriented mixed-use development that showcases the area’s history, natural environment, and local residents and businesses. This strategy consists of preliminary design ideas supported by implementation goals, policies, and programs that establish a framework for future redevelopment of this significant, historic waterfront area.

Young voters are not voting. According to the United States Census Bureau, young voters, aged 18-29, have not turned out to vote in Presidential Elections at a rate higher than 50% since 1968. Millions of ballots – with the potential to swing elections, policies, and political momentum – are left blank. It is in the interest of both parties to identify why young voters are not going to the polls. The goal of service life prediction is to accurately predict the useful lifetime of a coating system in a typical service environment. Field testing is the most true-to-life form of service life testing but requires long exposure times, often not quick enough to readily aid redesign in the product development cycle. This paper investigates the persistence of agricultural practices funded by the California Department of Food and Agriculture's Office of Environmental Farming and Innovation (CDFA OEFI). The central inquiry revolves around determining the most effective behavior model for analyzing persistence, comparing the Reasoned Action Approach (RAA), Comprehensive Action Determination Model (CADM), and CADM augmented with structural variables. The study's methodology integrates literature review, analysis of OEFI-funded practices, and statistical modeling to assess persistence levels.
