Propidium monoazide (PMA) is a molecular tool used to assess viability of microorganisms. Currently, PMA is thought to discern viability through membrane permeability; PMA enters only membrane compromised cells, irreversibly crosslinks to theirDNAand precipitates theDNAout of solution, preventing it from being amplified during polymerase chain reaction (PCR). Using PMA on a sample of live and dead microorganisms results in only theDNAof living organisms being amplified and identified. Therefore, a comparison ofPCRresults with and without PMA allows one to determine the live fraction and total population, respectively.

: Plant biomass is a rich source of sugars that can be converted to biofuels by engineered microbes. However, because the lignocellulose in biomass is insoluble in aqueous conditions and recalcitrant to enzymatic degradation, thermochemical treatment is required to break apart the lignin and cellulose polymers before sugars can be released. The most effective chemicals for doing this are known as ionic liquids, which are salts that are molten at temperatures below 100° C. Although these solvents have many unique properties that are ideal for solubilizing lignocellulose, they have been found to inhibit the growth of bacterial strains used to …

Zooplankton represent a vital link between phytoplankton and fish, like the endangered Delta Smelt. Human interferences (nitrates from waste water, flow alteration, invasive species introduction…) have altered the structure of the San Francisco Estuary (SFE) ecosystem. We use stable isotope analysis to improve our knowledge of the planktonic food web in the SFE and gain insights into its evolution over the past decades. We use the ratios of certain isotopes (Nitrogen, Carbon, Sulfur, etc.) in different species of zooplankton to tell us what it is feeding on as well as the trophic level it feeds in. As a Wildlife Biology major at Cal Poly, I was able to express my passion for the ocean and beaches in planning and co-designing a beach and dive clean up Earth Day celebration as my senior project. This event took place at Avila Beach on April 21st and was hosted by the Central Coast Aquarium. It was open to the public in attempts to promote outreach and awareness of coastal conservation, allowing volunteers to participate in the clean-ups and visit the Aquarium. The event was a success, involving over 100 volunteers, collecting and recycling 150 pounds of trash and also …

The purpose of this project was to develop a promotional video about Supervised Agricultural Experience (SAE) programs in a high school agriculture program. It was created to show the importance of SAE’s and the benefits students can gain from having an SAE. In order to capture a wide array of opinions the project includes interviews both from agriculture teachers and students to diversify the importance of having SAE’s in an agriculture program. The video shows how both students and teachers benefit from having SAE’s and why they are so crucial to a successful high school agriculture program.

The Multiphasic And Dynamic Nature Of Flow In Adventure Experiences, Susan Houge Mackenzie

This study evaluated phases of adventure experiences by identifying flow and reversal theory states over a 3-day white-water river surfing course. Data were collected with novice river surfers (n = 5) via in-depth qualitative interviews using head-mounted video cameras. Findings suggested that "opposing" experiential phases (i.e., telic and paratelic) may be symbiotic in adventure experiences and may facilitate flow experiences. These results may account for the dynamic nature of enjoyment, flow, and motivational states within adventure experiences. Future research should seek to validate the phasic models presented herein and evaluate their potential applicability to other adventurous activities.

Background: Survival and colony formation by transplanted tissue derived connective tissue progenitor cells (CTPs) are thought to be important factors in the success of clinical tissue engineering strategies for bone regeneration. Transplantation of cells into defects larger than a few millimeters expose cells to a profoundly hypoxic environment. This study tested the hypothesis that delaying the onset of hypoxia will improve the survival and performance of CTPs in vitro.

Microbial fuel cells (MFC) utilize bacteria to generate an electrical current that can be used in the decomposition of sludge and human urine. In a MFC there is an anode (for oxidation of organic compounds), cathode (reduction of oxygen or carbon dioxide), and a proton exchange membrane (PEM, allows protons to migrate); reduction-oxidation reactions between the anode and cathode produce a measurable current. Bacteria that are found in sludge can be used to produce electrons in a voltaic cell, but optimizing conditions for harnessing the energy is crucial to making a MFC efficient. Situated at the northwest end of Santa Cruz County and occupying circa 30 square miles of sharply contrasted terrain, the Scott Creek Watershed concentrates within its geomorphological boundaries, at least 10-12% of California's flora, both native and introduced. Paradoxically, the use/abuse that the watershed has sustained over the past 140+ years, has not necessarily diminished the biodiversity and perhaps parallels the naturally disruptive but biologically energizing processes (fire, flooding, landslides and erosion), which have also been historically documented for the area for +60 years. Research to increase the efficiency of conventional solar cells is constantly underway. The goal of this work is to increase the efficiency of conventional solar cells by incorporating quantum dot (QD) nanoparticles in the absorption mechanism. The strategy is to have the QDs absorb UV and fluoresce photons in the visible region that are more readily absorbed by the cells. The outcome is that the cells have more visible photons to absorb and have increased power output. The QDs, having a CdSe core and a ZnS shell, were applied to the solar cells as follows. Nanotechnology is the manipulation of matter and devices on the nanometer scale. Below the critical dimension length of 100nm, materials begin to display vastly different properties than their macro- or micro- scale counterparts. The exotic properties of nanomaterials may trigger the start of a new technological revolution, similar to the electronics revolution of the late 20th century. Current applications of nanotechnology primarily make use of nanoparticles in bulk, often being made into composites or mixtures. The multiple-objective exploratory study investigates effects of various silvicultural management regimes commonly applied to coast redwood (Sequoia sempervirens [D. Don] Endl.) forests in Santa Cruz and San Mateo Counties, California, USA. A temporary forest inventory was installed in 24 harvest origin stands and 4 natural origin stands throughout the study area (sample area = 1189 acres). Data from the systematic sample of 233 one-quarter acre nested cluster plots (sample intensity = 4.9%) rendered overall forest descriptions in terms of species composition, density, and structure. Today complimentary medicine is being increasingly sought out. Ayurveda and TCM, are among the oldest systems of medicine and have been developed for over thousands of years in India and China respectively. This paper details the philosophies, medical theories, anatomy, diagnosis, and treatments of both of these systems, including a comparison. Both of these modalities of healing operate with a microcosm – macrocosm paradigm. This makes them fundamentally similar, and compatible with each other. Ayurveda uses Tridoshic theory to apply treatments ranging from diet, massage, meditation, yoga among other therapies to bring Vata, Pitta, and Kapha into balance. TCM is …
