The use of time-of-flight secondary ion mass spectrometry (ToF-SIMS) in plant biology is a relatively unexplored and quickly developing field. The majority of research in plant SIMS involves the application of ToF-SIMS to study dried wood tissues, and only a handful of studies apply SIMS on plant stems, roots, and/or seeds. Our project provides a brief description and review of previous work using SIMS on plant stems, roots, and/or seeds, along with an emphasis on the sample preparation in each study. Bacteria live in environments containing complex ecologies of other microbes that communicate and survive through the action of a variety of small metabolic compounds. One common yet relatively unstudied metabolite is guanidine. Although it can be toxic to cells, recent studies have revealed that guanidine may function as a cellular metabolite through a specialized RNA sequence known as a riboswitch. This paper argues that ethical views that place primary importance on the reduction of extreme suffering imply that, at least in theory, it can be better to allow enhanced non-human animals to come into existence rather than unenhanced non-human animals. Furthermore, they imply that it would be even better if no non-human animals came into existence at all. However, it is unclear, from the perspective of these ethical views, whether enhancement or reduction of future populations is the more effective strategy in practice, and whether it might even be better to instead pursue a seemingly more robust and less controversial …

As of January 2017, the U.S. poultry industry banned the use of antibiotics and now relies on alternatives such as probiotics to help protect animal health. Although probiotic use is not a new concept in the poultry industry, identifying the best combination of bacterial strains to generate an effective probiotic formula requires further investigation. This study aimed to detect a probiotic product of four bacterial strains (Pedioccoccus acidilactici, Pediococcus pentosaceus, Lactobacillus plantarum, and Bacillus subtilis) in a feeding trial with broiler chickens. Meiosis is a specialized form of cell division in sexually reproducing eukaryotes. Crossovers are physical connections formed between homologous chromosomes during meiosis; these connections help ensure normal segregation of homologous chromosomes at meiosis I. However, the yeast Saccharomyces cerevisiae and other eukaryotes can still segregate homologs properly even in the absence of some crossovers. This is due to a backup mechanism known as distributive segregation, which correctly segregates non-crossover chromosomes at a higher rate than if segregation were completely random. Bovine in vitro produced embryos suffer from poor developmental competence and altered metabolism which hinders their cryotolerance. Overall, the goals of this thesis were to improve oocyte and embryo culture with the antioxidant polydatin and to optimize slow freezing procedures. This thesis was designed as three experiments, and in each experiment, oocytes were aspirated from abattoir ovaries, matured for 23h, fertilized with semen from 1 of 3 bulls, and cultured in synthetic oviductal (SOF) based medium (SCF1) in 38.5 °C in 5% O2, 5% CO2 and 90% N2. Developing a tissue-engineered Blood Vessel Mimic (BVM) to represent diabetic macrovascular disease could expedite design of new vascular devices specifically tailored to diabetic patients. In contribution toward this model, this thesis assessed Human Umbilical Vein Endothelial Cell (HUVEC) responses to high glucose conditions. Interleukin 6 (IL-6) and Cluster of Differentiation 36 (CD36) were selected to signify oxidative stress activity, a hallmark of diabetic macrovascular disease. Next, activity of potential reference genes B2M, HPRT1, and ACTB was assessed. All genes were found to exceed acceptable variability, so the E-ΔC T method of data analysis was selected. Roads pose two central problems for wildlife: wildlife-vehicle collisions (WVCs) and habitat fragmentation. Wildlife exclusion fencing can reduce WVCs but can exacerbate fragmentation. In Chapter 1, I summarize the relevant studies addressing these two problems, with a focus on large mammals in North America. Chapters 2 and 3 summarize field assessments of technologies to reduce WVCs and maintain connectivity, specifically jumpout ramps and underpasses, along Highway 101 near San Luis Obispo, CA. In a fenced highway, some animals inevitably breach the fence and become trapped, which increases the risk of a wildlife-vehicle collision. Earthen escape ramps, or “jumpouts”, can allow …

I will discuss how animal laboratory research can be ethically analyzed using Donaldson and Kymlicka’s political theory of animal rights. To accomplish this, I will not presuppose their strong animal rights framework. Donaldson and Kymlicka’s approach revolves around some basic human-animal relationships, reflecting the relational turn in applied ethics writ large. However, they do not discuss laboratory animal research in any detail, and so an extension to that domain of animal use is in order. Donaldson and Kymlicka’s emphasis on human-animal relationships is useful for reminding ourselves that in laboratories various staff or personnel can develop bonds with captive animals …

The importance of calcium in nickel tolerance was studied in the nickel hyperaccumulator plant Alyssum inflatum by gene transformation of CAX1, a vacuolar membrane transporter that reduces cytosolic calcium. CAX1 from Arabidopsis thaliana with a CaMV35S promoter accompanying a kanamycin resistance gene was transferred into A. inflatum using Agrobacterium tumefaciens. Transformed calli were subcultured three times on kanamycin-rich media and transformation was confirmed by PCR using a specific primer for CAX1. More than 16,000 tons of olive cake was produced in the United States in 2017. Olive cake is a by-product of olive oil extraction, which has limited animal feed potential, and poses an environmental threat when landfilled due to its high organic load and polyphenol content. This residue has potential for biofuel (bioethanol and biomethane) production because it is rich in polysaccharides such as pectin, hemicellulose, and cellulose. ABET has revised the criteria 3 (Student Outcomes) and 5 (Curriculum) of the general criteria. These changes have made it through the rigorous approval process and will be effective for the 2019-20 academic year. This paper analyzes the differences between the existing criteria and the proposed changes. Through a specific detailed example, the paper illustrates how an existing assessment system can seamlessly be adjusted for the new criteria. Most of the embedded indicators and direct measures of attainment can still be used. The results simply need to be organized differently.

The amount of structural damage image data produced in the aftermath of an earthquake can be staggering. It is challenging for a few human volunteers to efficiently filter and tag these images with meaningful damage information. The proposed solution is to automate post-earthquake reconnaissance image tagging activities by training a computer algorithm to classify each occurrence of damage per building material and structural member type. Scanning electron microscopy (SEM) video recording is used to characterize the solidification of small volumes of 99.999% pure gallium (Ga) and eutectic gallium-indium (eGaIn) under a high vacuum environment. Specimen are superheated to 55℃ using a hot plate, cast into spherical droplets, and cooled in situ by means of a Peltier cooling stage. Special attention is given to the preparation of the specimen prior to viewing because of gallium and its alloys’ nature to form an oxide layer when melted and air cooled. The oxide acts as a skin that inhibits the observation of microstructural features during solidification. Heated samples …

Summary of Functional Requirements
This project integrates LTSpice with a python script that runs a genetic algorithm to bias a differential amplifier. The system biases the amplifier with 2 different voltages, the base voltage for the PNP BJTs of the active loads and a voltage controlling the current of the current sink. The project runs via a python script, gets data from LTSpice’s command line call, and iteratively runs until the system is biased to achieve the greatest gain on an arbitrary input voltage.

Vermicompost transforms agricultural waste into a high quality soil amendment, though market acceptance remains in its infancy. This study examines how growers’ willingness to pay for vermicompost is affected by grower’s crop, region, income per acre, knowledge level of vermicompost and compost, previous use of compost, and the willingness to pay for compost. The survey results pulled together 223 responses from California growers. It was discovered growers’ had less knowledge of vermicompost than compost but were willing to pay more for vermicompost. Many of the various properties of neutrinos are still a mystery. One unknown is whether neutrinos are Majorana fermions or Dirac fermions. Cuoricino and CUORE are experiments that aim to solve this mystery. Noise reduction in these experiments hinges on the ability to discern among alpha, beta and gamma particle detections using the thermal pulses they create. In this paper, we look at Cuoricino data and attempt to classify pulses, not as alpha, beta or gamma particles, but rather as signal, noise or calibration data. Autonomous vehicle navigation is a complex and challenging task. Land and aerial vehicles often use highly accurate GPS sensors to localize themselves in their environments. These sensors are ineffective in underwater environments due to signal attenuation. Autonomous underwater vehicles utilize one or more of the following approaches for successful localization and navigation: inertial/dead-reckoning, acoustic signals, and geophysical data. This study is a detailed investigation of the dynamic interaction between a stream and an alluvial aquifer at Swanton Pacific Ranch in the Scotts Creek watershed in Santa Cruz County, California. The aquifer is an important source of groundwater for cropland irrigation and for aquatic ecosystem support. This report documents all the research, ideation, and mockups used to determine right ventricle motion and develop a system capable of reproducing that motion on a tissue sample. The model is intended for evaluating anchoring systems being developed by Edwards Lifesciences for use with tricuspid valve therapies. Several design solutions were considered for the primary functions of recreating motion of the right ventricle and attaching tissue to the device. One of the detrimental aspects of the space en- vironment is Atomic Oxygen, AO. Subclinical atherosclerosis is an important area of research to evaluate stroke risk and predict localization of plaque. The current methods for detecting atherosclerosis risk are insufficient because it is based on The Framingham Risk Score and carotid intima media thickness, therefore an engineering detection model based on quantifiable data is needed. Laminar and turbulent flow, dictated by Reynolds number and relative roughness, was modeled through the carotid artery bifurcation to compare shear stress and shear rate. Coral reefs are one of the world’s most diverse yet heavily impacted marine ecosystems. As a result of many direct and indirect stressors, coral reefs have experienced major degradation over the last several decades. Declines in coral reefs in the Caribbean have been particularly acute and generally associated with the loss of key herbivores and an increase in algae. Herbivorous fishes such as parrotfishes can positively impact coral reefs by removing algae that compete with corals for light and space.
