Isolation And Selective Culture Of Quail Primordial Germ Cells And The Development Of A Cross Culture System To Harbor Development Of Manipulated Embryos, C. M. Kimmell, A. Saribalis, J. Echeverria, L. Shuey, K. A. Shaw, K. L. Shaw, D. G. Peterson

Animal Science

Primordial germ cells (PGCs) are the embryonic precursors of adult egg and sperm. Isolation, propagation, genetic manipulation and reimplantation of modified PGCs that retain their commitment to the germline have been recently described for the successful generation of transgenic chickens [1]. Here, we report the adaptation and application of these techniques to Japanese quail for the purpose of future applications involving the introduction of a phytase transgene as previously described [2]. Quail PGCs are harvested by aspiration of whole embryonic blood from embryos that have been incubated for 48– 52 h. Blood is aspirated using a mouth pipette fitted with …

Biomechanics Of The Rabbit Knee And Ankle: Muscle, Ligament, And Joint Contact Force Predictions, Dustin M. Grover, Andrew A. Chen, Scott J. Hazelwood Jan 2007

Biomechanics Of The Rabbit Knee And Ankle: Muscle, Ligament, And Joint Contact Force Predictions, Dustin M. Grover, Andrew A. Chen, Scott J. Hazelwood

Biomedical Engineering

Mathematical models of small animals that predict in vivo forces acting on the lower extremities are critical for studies of musculoskeletal biomechanics and diseases. Rabbits are advantageous in this regard because they remodel their cortical bone similar to humans. Here, we enhance a recent mathematical model of the rabbit knee joint to include the loading behavior of individual muscles, ligaments, and joint contact at the knee and ankle during the stance phase of hopping. Geometric data from the hindlimbs of three adult New Zealand white rabbits, combined with previously reported intersegmental forces and moments, were used as inputs to the …

Scapegoating In Group Psychotherapy, J. Kelly Moreno Jan 2007

Scapegoating In Group Psychotherapy, J. Kelly Moreno

Psychology and Child Development

The purpose of this paper is to describe and illuminate the phenomenon of scapegoating in group psychotherapy. Specifically, the role of projective identification - on both individual and group-wide bases - in the evolution of the deviant is delineated. Individual, interpersonal, and whole-group interventions are presented along with the technique of functional subgrouping, a relatively new and particularly potent group intervention. Several case vignettes are detailed for illustration.

The Long And Winding Road: Influences Of Intracellular Metabolite Diffusion On Cellular Organization And Metabolism In Skeletal Muscle, Stephen T. Kinsey, Kristin M. Hardy, Bruce R. Locke Jan 2007

The Long And Winding Road: Influences Of Intracellular Metabolite Diffusion On Cellular Organization And Metabolism In Skeletal Muscle, Stephen T. Kinsey, Kristin M. Hardy, Bruce R. Locke

Biological Sciences

A fundamental principle of physiology is that cells are small in order to minimize diffusion distances for O2 and intracellular metabolites. In skeletal muscle, it has long been recognized that aerobic fibers that are used for steady state locomotion tend to be smaller than anaerobic fibers that are used for burst movements. This tendency reflects the interaction between diffusion distances and aerobic ATP turnover rates, since maximal intracellular diffusion distances are ultimately limited by fiber size. The effect of diffusion distance on O2 flux in muscle has been the subject of quantitative analyses for a century, but the …

Reading Differently, Rereading For Difference In Versions Of Rimbaud's "Mouvement", Brian G. Kennelly Jan 2007

Reading Differently, Rereading For Difference In Versions Of Rimbaud's "Mouvement", Brian G. Kennelly

World Languages and Cultures

How to engage students to reread Arthur Rimbaud’s 1886 poem “Mouvement” differently? What can they learn about the poem and in the process about themselves as readers of literature through its various versions and English language translations? Might rereading for difference hold pedagogical promise? How might a comparative study of the poem’s various versions in French and in English translation favor active reading and help students embrace the poem as differently meaningful because of its modernity?

History And Behavioral Ecology During The Middle-Late Transition On The Central California Coast: Findings From The Coon Creek Site (Ca-Slo-9), San Luis Obispo County, Brian F. Codding, Terry L. Jones Jan 2007

History And Behavioral Ecology During The Middle-Late Transition On The Central California Coast: Findings From The Coon Creek Site (Ca-Slo-9), San Luis Obispo County, Brian F. Codding, Terry L. Jones

Social Sciences

As the focus of intense debate concerning the possible effects of environmental variability on Native populations, the Middle-Late Transition (MLT) is an exceptionally important period in California prehistory. Recent salvage excavations at the Coon Creek Site (CA-SLO-9) on the San Luis Obispo County coast revealed a single, highly discrete component dating cal A.D.900–1300 which is largely synchronous with most definitions of the MLT. With a recovery volume of 23.2 m3, this is the first component to yield artifact and faunal assemblages substantive enough to establish the culture historical markers for this period in this region, and to define …

Proximate Developmental Mediators Of Sexual Dimorphism In Size: Case Studies From Squamate Reptiles, Henry B. John‐Alder, Robert M. Cox, Emily N. Taylor Jan 2007

Proximate Developmental Mediators Of Sexual Dimorphism In Size: Case Studies From Squamate Reptiles, Henry B. John‐Alder, Robert M. Cox, Emily N. Taylor

Biological Sciences

Sexual dimorphism in size (sexual size dimorphism; SSD) is nearly ubiquitous, but the relative importance of genetic versus environmental control of SSD is not known for most species. We investigated proximate determinants of SSD in several species of squamate reptiles, including three species of Sceloporus lizards and the diamond‐backed rattlesnake (Crotalus atrox). In natural populations of these species, SSD is caused by sexual differences in age‐specific growth. Males and females, however, may often share similar potentials for growth: growth is strongly responsive to the availability of food, and sexual differences in growth can be greatly suppressed or completely …

Flavor Partition And Fat Reduction In Cheese By Supercritical Fluid Extraction: Processing Variables, Jessica L. Yee, Hany Khalil, Rafael Jiménez-Flores Jan 2007

Flavor Partition And Fat Reduction In Cheese By Supercritical Fluid Extraction: Processing Variables, Jessica L. Yee, Hany Khalil, Rafael Jiménez-Flores

Dairy Science

Developing low-fat cheese with flavor to match that of full-fat cheese has been a challenge in the dairy industry. The objective of this investigation was to develop lower fat Cheddar and Parmesan grated cheese using supercritical fluid extraction (SFE) and characterize its flavor profile comparative to a full-fat product. Specifically, enabling flavor compound partition between the matrices of cheese and extracted lipids. Carbon dioxide (CO2) was the supercritical fluid for fat extraction. Extraction took place in a 500 mL SFE vessel using 100 g of grated cheese. Fractional factorial design was utilized to investigate two levels of treatment …

Indo-Fijian Children’S Bmi In The Context Of Urbanization, Embodied Capital, And Food Choice Trade-Offs, Dawn B. Neill Jan 2007

Indo-Fijian Children’S Bmi In The Context Of Urbanization, Embodied Capital, And Food Choice Trade-Offs, Dawn B. Neill

Social Sciences

Health research has shown that overweight and obesity in children and adults are becoming significant public health problems in the developing world. Evidence suggests that this phenomenon is more marked in urban than rural areas and may be associated with modernization. However, the underlying reasons for this nutrition transition remain unclear. Dietary shifts, often in conjunction with income and time constraints in urban environments, may entail a greater reliance on more convenient sugar and fat-dense food. Also, the necessity of labor-intensive agricultural work to meet rural subsistence needs is supplanted in urban environments by sedentary work. This paper extends the …

Compartmentalization Of The Broad-Range Phospholipase C Activity To The Spreading Vacuole Is Critical For Listeria Monocytogenes Virulence, P.S. Marie Yeung, Yoojin Na, Amanda J. Kreuder, Helene Marquis Jan 2007

Compartmentalization Of The Broad-Range Phospholipase C Activity To The Spreading Vacuole Is Critical For Listeria Monocytogenes Virulence, P.S. Marie Yeung, Yoojin Na, Amanda J. Kreuder, Helene Marquis

Biological Sciences

Listeria monocytogenes is a bacterial pathogen that multipliesin the cytosol of host cells and spreads directly from cellto cell by using an actin-based mechanism of motility. The broad-rangephospholipase C (PC-PLC) of L. monocytogenes contributes tobacterial escape from vacuoles formed upon cell-to-cell spread.PC-PLC is made as an inactive proenzyme whose activation requirescleavage of an N-terminal propeptide. During infection, PC-PLCis activated specifically in acidified vacuoles. To assess theimportance of compartmentalizing PC-PLC activity during infection,we created a mutant that makes constitutively active PC-PLC(the plcBpro mutant). Results from intracellular growth andcell-to-cell spread assays showed that the plcBpro mutant wassensitive to gentamicin, suggesting …
