Eukaryotic cells are highly heterogeneous. These cells are arranged into different compartments, carrying out separate functions and facilitating biological processes. Proteins are the effector biomolecules targeted to subcellular locations that help fulfill specific tasks in living organisms. Spatial proteomics can help unravel molecularly how protein abundance and localization are altered in cells, which is not feasible in traditional bulk-scale proteomics. To achieve this, our lab has developed a miniaturized sample processing platform called nanoPOTS, reduced separation columns' inner diameter to increase ionization efficiency and concentrate analytes for mass spectrometers and optimized data acquisition modes for increasing proteome coverage in spatial …

This study explored the effect of contextual diversity on speech motor learning in an individual with apraxia of speech (AOS) using virtual reality (VR). One 62-year-old participant with severe AOS participated in the study where he practiced monosyllabic CVC words in both consistent and diverse VR environments. Kinematic, acoustic, and perceptual data were collected and evaluated for evidence of learning. Kinematic data were collected using electromagnetic articulography where sensors were glued to the participant's upper lip, lower lip, tongue blade, and tongue tip and were tracked with high precision. The refugee and newcomer population faces many challenges as they arrive in their host country. Many individuals experience varying degrees of trauma in their country of origin. Trauma can lead to poor mental health outcomes and poor adjustment in host countries. Refugee children in particular may experience post-traumatic stress disorder, depression, and anxiety as a result of traumatic experiences. Children can be resilient when given opportunities to create relationships and gain confidence. These opportunities can be fostered through various avenues, one of which is the school environment. While child abuse, maltreatment, and neglect are clear examples of the most maladaptive types of parent-child interactions (Borchet et al., 2022), there is considerable evidence for the damaging nature of parentification in terms of youth outcomes and parental/familial dysfunction. The research literature contains numerous studies related to parentification, but there is little research overtly demonstrating the systemic effects of parentification (Peris et al., 2008). It is imperative that researchers and clinicians learn more about the mechanisms involved in the parentification process. This dissertation investigates the impact of honeybee introductions on native bee populations, evaluates the efficiency of traditional bee sampling and monitoring methods, and explores the use of drone technology for assessing floral resources on federal lands in the United States. Federal lands, managed for multiple uses, present a valuable opportunity for the summer pasturing of honeybees. These lands produce vast floral resources and are largely free from pesticides and other pollutants that could weaken honeybee populations. The potential use of this resource is significant given the economic and agricultural concerns presented by declining pollinator trends and Colony Collapse Disorder (CCD). …

Congenital Heart Disease (CHD) is the most common birth defect, affecting approximately 1% of live births. This dissertation explores the genetic mechanisms underlying CHD, focusing on the T-box transcription factors Tbx2a and Tbx2b in zebrafish. Utilizing advanced bioinformatics techniques, we reconstructed the ancestral sequences of these genes to understand their evolutionary trajectory and functional divergence. Our analysis highlights the evolutionary conservation of the DNA-binding domain in tbx2a and tbx2b, suggesting its crucial role in maintaining correct gene expression patterns during development. We observed unique and shared amino acid substitutions in these proteins, indicating potential adaptive changes and conserved functions. Comparative …

Liquid microdroplet resonators provide an excellent tool for optical studies due to their innate smoothness and high quality factors, but they can be difficult to control. By using 3D printed mounts to support the droplets, we can obtain precise control over the droplet geometries and positions. We here present our work with oil, water, and ice microdroplets, as well as tools required to enable their study. We first present methods for creating 3D printed mounts for oil microdroplet resonators. The mounts enable precise positioning of the droplets relative to a tapered optical fiber. German abortion law is restrictive for a Western European nation of strong economic standing and with relatively liberal social policy. Additionally, Germany is one of few Western European nations without an embryopathy indication, which allows for exceptional abortions in cases of fetal defects. The law (as it stands in 2024), has been immeasurably shaped by two ideologies, eugenics and feminism, and by modern Germany’s consideration of both the ideologies themselves, and of how they influenced the reproductive policies of the twentieth century. The Domain Name System (DNS) root servers have provided a useful look into the DNS and internet ecosystems for decades. We present a categorization of clients querying DNS root servers. Using two clustering algorithms on DNS traffic sampled in 2020, we can predict the structure and volume of queries originating from different types of clients. Previous research has used unsupervised techniques to better understand DNS traffic patterns, but none have, to our knowledge, considered clients beyond those driven by queries from end users. This study examines current trends of university counseling center (UCC) utilization among Reserve Officer Training Corps (ROTC) students and student-military Veterans, compared to their nonmilitary-minded peers at a large, private, western university. Given likely differences in the lived experiences between these three populations, research is needed to provide insight on the treatment needs of military-minded students. We examine counseling center data for ROTC (n = 88), Veteran (n = 93), and nonmilitary-minded (n = 16,268) students who presented for counseling services from 2014-2021. This thesis addresses the underrepresentation of indigenous Māori people in leadership positions and pursues the need to develop a Māori leadership framework aimed at empowering the next generation of indigenous people. Several frameworks have attempted to exhibit Māori leadership dynamics, each offering unique insights into the cultural and relational aspects of Māori leadership. However, existing frameworks often exhibit limitations in fully encapsulating the breadth of Māori leadership values and practices, necessitating the development of a comprehensive Māori leadership model independent of Western frameworks. Given that the manifestation of schizophrenia varies by sex, it is hypothesized that the interaction between diagnosis and sex significantly influences brain structure and is believed to be a crucial factor in understanding the neurobiology of schizophrenia. Recently, the United States has seen a notable increase in controversy and legislation regarding "divisive" concepts and issues in education. These divisive issues include Critical Race Theory, Social Emotional Learning, book censorship, and other race and gender topics. Legislation and media coverage regarding these issues contributes to a larger trend of challenges to teacher professionalism. Purpose: The purpose of this project was to identify the rates of and reasons for nonadherence in medical follow-up care among patients who utilize the Volunteer Care Clinic (VCC).
Background: The VCC is a free outpatient care center for acute medical conditions. The VCC serves uninsured, low socioeconomic residents of Utah County, and many of the patients who come to the clinic primarily speak Spanish. The director of the VCC expressed concern that patients who were assigned medical follow-up care to complete after their visit were not completing these assignments. Problem behavior has been increasing in classrooms and has significantly contributed to teacher burnout. Students in special education settings have been found to demonstrate problem behaviors more than their general education peers. Schools have a responsibility to implement evidence-based behavioral support to assist students with their behavioral needs, decrease problem behavior, and increase desired behavior in the classroom. There is a relationship between the level of implementation fidelity and the effectiveness of an intervention. Heart rate variability (HRV) and personality are related to both physical and mental health outcomes. Previous research has found a relationship between neuroticism and distress while the findings on the relationship between neuroticism and HRV are mixed. Studying the facets of neuroticism may provide a more nuanced view of the effects of personality on HRV and distress. Previous research has suggested that the vulnerability facet of neuroticism appears to be uniquely related to the cardiovascular response to stress. This dissertation has been written in a three-article journal-ready format and centers on competencies for blended teaching in higher education. I use the PRISMA process and thematic coding to analyze 34 articles from 2002 to 2022, highlighting strategic combination, learner-centered approaches, and additional support. I also use the Google Search API and content analysis to suggest that blended, digital, and flexible may be post-secondary institutions' most frequently used terminology to describe contexts incorporating online and in-person components.
