Background: Parkinson's disease (PD) is the second most prevalent neurodegenerative disorder. Biomarkers that can help monitor the progression of PD or response to disease-modifying agents will be invaluable in making appropriate therapeutic decisions. Further, biomarkers that could be used to distinguish PD from other related disorders with PD-like symptoms will be useful for accurate diagnosis and treatment. C-Abl tyrosine kinase is activated in PD resulting in increased phosphorylation of the tyrosine residue at position 39 (Y39) of α-synuclein (α-syn) (pY39 α-syn), which contributes to the death of dopaminergic neurons. Because pY39 α-syn may be pathogenic, monitoring pY39 α-syn could allow …

The Ability Of Thaumoctopus Mimicus To Be Operantly Conditioned To A Sound Stimulus, Stephanie Wittman

Honors Projects

This paper focuses on the ability of the mimic octopus Thaumoctopus mimicus to be operantly conditioned to an auditory stimulus. The octopus is known to be the most advanced of the invertebrates and has learning abilities that are comparable to vertebrates in spite of their differences in brain structure. These animals have been shown to react to visual and tactile stimuli and can be operantly conditioned to perform behaviors to obtain a food reward. The goal of this experiment is to determine whether the octopus can be operantly conditioned to swim into a box on the side of its tank …

Arachidonic Acid Inhibits Inflammatory Responses By Binding To Myeloid Differentiation Factor-2 (Md2) And Preventing Md2/Toll-Like Receptor 4 Signaling Activation, Yali Zhang, Hongjin Chen, Wenxin Zhang, Yan Cai, Peiren Shan, Di Wu, Bing Zhang, Hui Liu

Paediatrics Publications

Arachidonic acid (AA) plays a fundamental role in the function of all cells. Metabolites of AA contribute to inflammation as well as for resolving inflammation. Although AA-derived metabolites exhibit well-substantiated bioactivity, it is not known whether AA regulates inflammatory responses independent of its metabolites. With the recent discovery that saturated fatty acids activate toll-like receptor-4 (TLR4), we tested the hypothesis that AA directly regulates inflammatory responses through modulating the activity of TLR4. In cultured cardiomyocytes and macrophages, we found that AA prevents saturated fatty acid-induced TLR4 complex formation with accessory proteins and the induction of proinflammatory cytokines. We discovered that …

Glaucoma is a leading cause of blindness, affecting up to 70 million people worldwide. High intraocular pressure (IOP) is a major risk factor for glaucoma. It is well established that inefficient aqueous humor (AqH) outflow resulting from structural or functional alterations in ocular drainage tissues causes high IOP, but the genes and pathways involved are poorly understood. We previously demonstrated that mutations in the gene encoding the serine protease PRSS56 induces ocular angle closure and high IOP in mice and identified reduced ocular axial length as a potential contributing factor. Here, we show that Prss56−/− mice also exhibit an abnormal …

Increasing incidences of multidrug-resistant (MDR) pathogens causing endophthalmitis threaten our ability to treat this condition, and the modulation of inflammatory responses by MDR bacteria is not known. In this study, using human microglia and retinal pigment epithelial (RPE) cells, we compare the inflammatory responses of sensitive (S-PA) and multidrug-resistant (MDR-PA) clinical isolates of Pseudomonas aeruginosa. Infected cells were subjected to qPCR analysis, enzyme-linked immunosorbent assay (ELISA), and immunostaining to assess the expression of inflammatory mediators. Both microglia and RPE cells, challenged with S-PA and MDR-PA, induced a time-dependent expression of inflammatory cytokines. Significant differences were observed in expression levels of …

Digitalization and digital health are transforming research practices, while economic growth is increasingly driven by the information commons. In the case of biological sciences, information commons, such as public biobanks and free/libre open source software (FLOSS), are of paramount importance for both research and the bioeconomy. In a time of digitalization, however, information commons are vulnerable to violations, such as the free-rider problem, that render the commons unsustainable. Consequently, it has been argued that the enclosure of the informational common resources is the only means to effectively exploit them. Given the social and economic importance of the information commons, the …

Outdoor acoustic event detection is an exciting research field but challenged by the need for complex algorithms and deep learning techniques, typically requiring many computational, memory, and energy resources. These challenges discourage IoT implementations, where an efficient use of resources is required. However, current embedded technologies and microcontrollers have increased their capabilities without penalizing energy efficiency. This paper addresses the application of sound event detection at the very edge, by optimizing deep learning techniques on resource-constrained embedded platforms for the IoT. The contribution is two-fold: firstly, a two-stage student-teacher approach is presented to make state-of-the-art neural networks for sound event …

Widespread Army Ant Aversion Among East African Jumping Spiders (Salticidae), Ximena J. Nelson, Samuel Aguilar-Arguello, Robert R. Jackson

All Peer-Reviewed Publications

Jumping spiders (Salticidae) typically prey on a variety of arthropods of similar size to themselves, but rarely on ants. Using 28 salticid species from East Africa, we first investigated vision-based aversion to ants by recording latency to enter a transparent sealed chamber flanked by chambers containing living army ants (Dorylus sp.) or tsetse flies (Glossina pallidipes) of comparable size. For all species, entry latency was significantly longer when the stimuli were ants. In another experiment, we used dead ants and tsetse flies mounted in a life-like posture as stimuli; except for Goleba puella, a species with unusual retinal ultrastructure, we …

Cerebrovascular Function In Adults With Depression, Rauchelle Richey May 2020

Cerebrovascular Function In Adults With Depression, Rauchelle Richey

Kinesiology Theses - Archive

Cerebral vasodilatory responsiveness is blunted in older adults (~45-70 yrs) with depressive disorders and thought to contribute to the link between depressive symptomology and increased risk for neurocognitive (e.g., dementia) and cerebral vascular (e.g., stroke) diseases. In young adults with major depressive disorder (MDD), peripheral vascular endothelial dysfunction is evident; however, to date, limited investigations have examined cerebral vasodilatory function in young healthy adults with MDD. We tested the hypothesis that cerebral vasodilatory responsiveness to a hypercapnic stimulus would be blunted in healthy young adults with MDD compared to healthy non-depressed adults (HA). Fourteen HA (22 ± 3yrs) and 14 …
