Complete Genome Sequence Of Moraxella Bovis Strain Epp-63 (300), An Etiologic Agent Of Infectious Bovine Keratoconjunctivitis, John Dustin Loy, Aaron M. Dickey, Michael L. Clawson
School of Veterinary and Biomedical Sciences: Faculty Publications
We report here the complete closed genome sequence of Moraxella bovis strain Epp-63 (300) (Epp63). This strain was isolated from an infectious bovine keratoconjunctivitis (IBK) case in 1963. Since then, Epp63 has been used extensively for IBK research. Consequently, the genome sequence of Epp63 should help elucidate IBK host-pathogen interactions.
Comparative Analysis Of Low Molecular Weight Fraction Of Conditioned Media Derived From Starvation-Dormant And Anaerobically-Dormant Mycobacterium Avium Subsp. Paratuberculosis, Marina Buciuc
All Graduate Theses, Dissertations, and Other Capstone Projects
Johne's disease (JD) caused by Mycobacterium avium subsp. paratuberculosis (MAP) is a contagious chronic enteritis affecting all ruminants and causing economic loss of at least $250 million annually in the US dairy industry. One of the major limitations in the diagnosis of JD is the low sensitivity of fecal culture in animals with no clinical signs of the disease, but who are actively shedding MAP. Persistence of MAP in the host with no apparent signs of infection is attributed to its ability to enter a dormant state wherein the bacterium is viable, but is not able to replicate until conditions …
Rhodococcus Equi In The Foal – Improving Diagnostic And Prevention Measures, Fernanda Bicudo Cesar
Rhodococcus Equi In The Foal – Improving Diagnostic And Prevention Measures, Fernanda Bicudo Cesar
Theses and Dissertations--Veterinary Science
Although Rhodococcus equi (R. equi), previously known as Corynebacterium equi, was first isolated from pneumonic foals almost a century ago, it remains the most common cause of subacute or chronic granulomatous bronchopneumonia in foals. While the majority of foals exposed to R. equi develop a protective immune response (regressors), others exhibit a unique susceptibility to infection (progressors). The determinants for either outcome are not completely understood. Therefore, current diagnostic and preventive measures are suboptimal and require betterment. In light of this current need, we hypothesized that immunoglobulin G subisotype T [IgG(T)] against the virulence-associated protein A (VapA) …
Cellular And Molecular Basis Of Equine Arteritis Virus Persistent Infection In The Stallion Reproductive Tract: Characterization Of Local Host-Pathogen Interactions Mediating Long-Term Viral Persistence, Mariano Carossino
Theses and Dissertations--Veterinary Science
Equine arteritis virus (EAV) has a global impact on the equine industry being the causative agent of equine viral arteritis (EVA), a reproductive, respiratory, and systemic disease of equids. A distinctive feature of EAV infection is that it establishes long-term persistent infection in the reproductive tract of stallions and is continuously shed in the semen (carrier state). Recent studies showed that long-term persistence is associated with a specific allele of the CXCL16 gene (CXCL16S). However, the cellular and molecular mechanisms underlying the establishment and maintenance of persistent infection are yet to be determined. The studies were undertaken herein …

Prevalence Of Cytauxzoon Felis (Protista: Apicomplexa) In Feral Cats In Russellville Arkansas, Cynthia H. Jacobs
Journal of the Arkansas Academy of Science
Abstract
Cytauxzoon felis (C. felis) is a protozoan hemoparasite of domestic and wild felids. Transmitted by ixodid ticks, the sylvatic reservoir for this organism in North America is the bobcat (Lynx rufus) in which the infection is apparently self-limiting. In domestic cats (Felis catus), C. felis causes a highly fatal disease with a distribution that covers much of the central, southcentral and southeastern U.S. and parallels that of the primary vector, the lone star tick (Amblyomma americanum). Interestingly, there appears to be an increased survival rate in domestic cats in the geographic …

Turkish Journal of Veterinary & Animal Sciences
This is the first report of the diagnosis and treatment of Apatemon gracilis infection in domestic geese in mainland China confirmed at the molecular level. The mortality of geese reached 10% (150/1500). Necropsy examinations confirmed hemorrhagic inflammation of the small intestine and blood and catarrhal mucus in the enteric cavity. A large number of flukes were found in the first third of the small intestine, with one end buried in the intestinal mucosa, forming lesions at the absorption sites. The ITS-1 sequence of the parasites was amplified by polymerase chain reaction and sequence analysis confirmed that the isolated worms were …

Anticoagulant Rodenticide Residues In Game Animals In California, Stella C. Mcmillin, Robert H. Poppenga, Shannon C. Chandler, Deana L. Clifford
United States Department of Agriculture Wildlife Services: Staff Publications
Anticoagulant rodenticides (ARs) are used to control rodents around homes, buildings, and in agriculture. They have been found widely in predatory and scavenging wildlife as a result of secondary exposure and less commonly in herbivores and omnivores from primary exposure. While predators and scavengers have been monitored for AR exposure, very little information is available about AR residues in edible muscle tissue of game animals. Game animals may be exposed to ARs through direct consumption of bait, ingestion of contaminated food or vegetation, or consumption of contaminated prey items. Carcasses of three species of game animals (black bear, wild pigs, …

Rhodococcus equi is one of the most important causes of disease in foals. Infection is typically characterized by pyogranulomatous pneumonia although extrapulmonary infections occur occasionally. Uveitis and polysynovitis have been reported in foals naturally infected with R. equi and are thought to be the result of an immune-mediated process. However, the pathogenesis of these conditions is poorly understood. The objectives of this study were to document the occurrence of uveitis and polysynovitis after experimental infection with R. equi and to determine if these disorders are the direct result of infection at these sites. Foals between 3 and 4 weeks of …

Hydrated lime manure treatment was evaluated to determine porcine epidemic diarrhea virus (PEDV) susceptibility to alkaline stabilization. At pH 10, PEDV decreased (quantitative polymerase chain reaction) and lost infectivity (swine bioassay). Although ammonium decreased above pH 9 (up to 25%), alkaline stabilization managed to control potential infection from manure sources.

Infectious bovine keratoconjunctivitis (IBK) is an economically significant disease caused by Moraxella bovis. Moraxella bovoculi, although not reported to cause IBK, has been isolated from the eyes of cattle diagnosed with IBK. Identification of M. bovis and M. bovoculi can be performed using biochemical or DNA-based approaches, both of which may be time consuming and inconsistent between laboratories. We conducted a comparative evaluation of M. bovoculi and M. bovis identification using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) with a database provided by Bruker Daltonics (termed the BDAL database), the BDAL database supplemented with spectra generated in …

School of Veterinary and Biomedical Sciences: Faculty Publications
Intrinsically photosensitive retinal ganglion cells (ipRGCs) innervate the hypothalamic suprachiasmatic nucleus (SCN), a circadian oscillator that functions as a biological clock. ipRGCs use vesicular glutamate transporter 2 (vGlut2) to package glutamate into synaptic vesicles and light-evoked resetting of the SCN circadian clock is widely attributed to ipRGC glutamatergic neurotransmission. Pituitary adenylate cyclase-activating polypeptide (PACAP) is also packaged into vesicles in ipRGCs and PACAP may be coreleased with glutamate in the SCN. vGlut2 has been conditionally deleted in ipRGCs in mice [conditional knock-outs (cKOs)] and their aberrant photoentrainment and residual attenuated light responses have been ascribed to ipRGC PACAP release. However, …

Bovine respiratory disease complex (BRDC) is one of the most significant diseases of cattle. Bacterial pathogens involved in BRDC include Mannheimia haemolytica, Mycoplasma bovis, Histophilus somni, and Pasteurella multocida. We developed and evaluated a multiplexed real-time hydrolysis probe (rtPCR) assay using block-based Peltier and rotary-based thermocycling on lung tissue, nasal swabs, and deep nasopharyngeal swabs. The rtPCR results were compared to culture or a gel-based M. bovis PCR using statistical analysis to determine optimum quantification cycle (Cq) cutoffs to maximize agreement. The limits of detection were 1.2–12 CFU/reaction for each pathogen. M. haemolytica was the most prevalent organism detected …

The success of Staphylococcus aureus as a pathogen is due in part to its ability to adapt to changing environmental conditions using signal transduction pathways, such as metaboliteresponsive regulators and two-component systems. S. aureus has a two-component system encoded by the gene pair sav0224 (hptS) and sav0223 (hptR) that regulate the hexose phosphate transport (uhpT) system in response to extracellular glucose-6-phosphate. Glycolytic intermediates such as glucose-6-phosphate are important carbon sources that also modulate the activity of the global metabolite-responsive transcriptional regulator CcpA. Because uhpT has a putative CcpA binding site in its promoter and …

Skin cancer is among the most common cancer types accompanied by rapidly increasing incidence rates, thus making the development of more efficient therapeutic approaches a necessity. Recent studies have revealed the potential role of decosahexaenoic acid ester of phloridzin (PZDHA) in suppressing proliferation of liver, breast, and blood cancer cell lines. In the present study, we investigated the cytotoxic potential of PZDHA in an in vitro model of skin cancer consisting of melanoma (A375), epidermoid carcinoma (A431), and non-tumorigenic (HaCaT) cell lines. Decosahexaenoic acid ester of phloridzin led to increased cytotoxicity in all cell lines as revealed by cell viability …

Moraxella bovoculi is frequently cultured from the ocular secretions and conjunctiva of cattle with Infectious Bovine Keratoconjunctivitis (IBK). Previous work has shown that single nucleotide polymorphism (SNP) diversity in this species is quite high with 81,284 SNPs identified in eight genomes representing two distinct genotypes isolated from IBK affected eyes (genotype 1) and the nasopharynx of cattle without clinical IBK signs (genotype 2), respectively. The goals of this study were to identify SNPs from a collection of geographically diverse and epidemiologically unlinked M. bovoculi strains from the eyes of IBK positive cattle (n = 183) and another from the eyes …

Background: The amino acid taurine (2-Aminoethanesulfonic acid) modulates inhibitory neurotransmitter receptors. This study aimed to determine if the dual action of taurine on GABAC- ρ1R relates to its structure. To address this, we tested the ability of the structurally related compounds homotaurine, hypotaurine, and isethionic acid to modulate GABAC- ρ1R.
Results: In Xenopus laevis oocytes, hypotaurine and homotaurine partially activate heterologously expressed GABAC- ρ1R, showing an increment in its deactivation time with no changes in channel permeability, whereas isethionic acid showed no effect. Competitive assays suggest that hypotaurine and homotaurine compete for the GABA-binding site. In addition, their effects were …

Background The global planetary health community increasingly recognises the need to prepare students to investigate and address connections between environmental change and human health. As we strive to support education on planetary health themes for students of all ages, understanding students’ concepts of linkages between the health of people and animals, and their shared environments might advance educational approaches. Children living in villages bordering Ruaha National Park in Iringa Region, Tanzania, have direct experience of these connections as they share a water-stressed but biodiverse environment with domestic animals and wildlife. Livelihoods in these villages depend predominantly on crop and livestock …

Potassium tellurite ( K2TeO3) is an effective selective agent for O157:H7 Shiga toxin-producing Escherichia coli (STEC), whereas tellurite resistance in non-O157 STEC is variable with information on O45 minimal. High-level K2TeO3 resistance in STEC is attributable to the ter gene cluster with terD an indicator of the cluster’s presence. Polymerase chain reactions for terD and K2TeO3 minimum inhibitory concentration (MIC) determinations in broth cultures were conducted on 70 STEC and 40 non-STEC control organisms. Sixty-six STEC strains (94.3%) were terD+ compared to 28 control organisms (70.0%; P < 0.001). The prevalence of terD in …

Sarcoplasmic/endoplasmic reticulum Ca2+ adenosine triphosphatase (SERCA)2a, a critical regulator of calcium homeostasis, is known to be decreased in heart failure. Patients with myocarditis or dilated cardiomyopathy develop autoantibodies to SERCA2a suggesting that they may have pathogenetic significance. In this report, we describe epitope mapping analysis of SERCA2a in A/J mice that leads us to make five observations: 1) SERCA2a contains multiple T cell epitopes that induce varying degrees of myocarditis. One epitope, SERCA2a 971–990, induces widespread atrial inflammation without affecting noncardiac tissues; the cardiac abnormalities could be noninvasively captured by echocardiography, electrocardiography, and magnetic resonance microscopy imaging. 2) SERCA2a …

Wildlife-aircraft collisions (wildlife strikes) pose a serious safety risk to aircraft. Wildlife strikes can be evaluated at different levels, include efforts to examine these problems at the national, regional, or state level, or for an individual airport. Similarly, wildlife strikes involving individual wildlife species or guilds can be examined at varying scales. Although wildlife strike analyses at the national, regional, or species/guild level are valuable, airport-specific analyses are essential for the effective implementation and evaluation of integrated wildlife damage management programs as these actions are conducted at the airport level. The species that present hazards to safe aircraft operations varies …

Flavobacterium columnare is Gram negative bacteria which caused columnaris disease in various kinds of freshwater fishes. This study is the first report of F. columnare infection in freshwater culturing Asian sea bass in Thailand. Phenotypic characterization, antimicrobial susceptibility test and quinolone resistant-associated genes of F. columnare were performed. A total 15 of F. columnare were isolated from diseased fishes and phylogenetic analysis revealed that these isolates were belonged to genetic group 2 and 4. From disk diffusion test results, 5 and 6 isolates were resistant to oxolinic (OA) and nalidixic acid (NA) respectively. The highest MIC values to OA and …

Surveillance Of Influenza A Viruses In A Live Bird Market, Yangon, Myanmar During 2017-2018, Khin Thurain
Chulalongkorn University Theses and Dissertations (Chula ETD)
Live bird markets play a significant role in highly pathogenic avian influenza (HPAI) virus spread and transmission. In Myanmar, the first HPAI-H5N1 outbreak was reported in 2006. Since 2014, routine influenza A virus surveillance in LBMs have been conducted in Myanmar by the Livestock Breeding and Veterinary Department in cooperation with FAO. However, there is limited information on virus characterization and evolution of influenza viruses in Myanmar. The objectives of this thesis were to detect influenza virus subtypes circulating in different poultry species from Mingalar-Taung-Nyunt live bird market (MTN-LBM) and to determine genetic characteristics and genetic diversity of influenza A …
Genetic Characteristics Of Avian Influenza Viruses Isolated From Ducks In Java Island, Indonesia Year 2015-2017, Lestari -
Chulalongkorn University Theses and Dissertations (Chula ETD)
Avian influenza becomes public health concern over years due to the serious impacts in both animal and human health. This study aimed to identify and characterize avian influenza viruses isolated from ducks in Java Indonesia during 2015-2017. Total 50 samples of previously identified as avian influenza viruses were acquired from the virus/culture collections of Disease Investigation Center (DIC) Wates Yogyakarta, Indonesia. The samples were recovered from either the avian influenza surveillance or avian influenza outbreaks in ducks in 3 provinces, East Java (n=26), Central Java (n=16), and Yogyakarta (n=8). Then the samples were processed for influenza A virus screening by …

Genome Characterization Of Hahella Chejuensis Strain Hn01, A Pathogen Of Red Egg Disease In Tilapia (Oreochromis Sp.), Putu Cri Devischa Gallantiswara
Chulalongkorn University Theses and Dissertations (Chula ETD)
Red eggs disease is an emerging disease occurred in Tilapia hatcheries which caused by a Gram-negative bacterium called Hahella chejuensis HN01. The disease infects to all the stage of fish eggs and reduces the hatchability. However, there are limited study about the bacterial pathogenicity. In this study, we analyzed the phylogenetic relationship and characterized the virulence traits of Hahella chejuensis by using genomic approach. The bacterial genome was sequenced using Illumina Miseq platform. The phylogenetic tree based on several housekeeping genes confirmed that the bacterium was belong to Genus Hahella and species chejuensis. the Average Nucleotide Identity calculation with the …
