In major prospective studies it has been reported that high heart rate at rest predicts the development of coronary heart disease (CHD) or cardiovascular disease (CVD) in men, but the mechanisms producing these relationships are unknown. Since lipoprotein levels contribute strongly to the risk of CHD and CVD, we examined the relationship of resting heart rate to plasma concentrations of high-density (HDL), low-density (LDL), and very low-density (VLDL) lipoproteins, apolipoprotein (apo) A-I and A-II, and serum concentrations of lipoprotein subfractions in 81 men to determine if atherogenic lipoproteins could potentially induce the reported association of heart rate with development of …

Characterization Of Monoclonal Antibodies Against Shiga-Like Toxin From Escherichia Coli, Nancy A. Strockbine, Lilian R. M. Marques, Randall K. Holmes, Alison D. O'Brien
United States Public Health Resources
Three monoclonal antibodies, designated MAb 16E6, MAb 13C4, and MAb 19G8, were produced which recognize Shiga-like toxin (SLT) from Escherichia coli. All three monoclonal antibodies neutralized the cytotoxicity of E. coli SLT and were able to immunoprecipitate intact labeled toxin with Staphylococcus aureus protein A. The three antibodies were of the Gl heavy and kappa light chain classes. MAb 16E6 bound to the B subunit of SLT in Western blots and also neutralized the lethality of the toxin for mice and the enterotoxicity of the toxin in ligated rabbit ileal loops. The ability of MAb 16E6 to neutralize the …

Effects Of Water Quality And Water Quantity On Nutritional Status: Findings From A South Indian Community, James R. Hébert
Faculty Publications
Quantitative assessments of the relative effects on health of various aspects of water supply are virtually absent from the literature. Despite the lack of information, resources are being allocated throughout the developing world, for projects related to water and sanitation. The present study was designed specifically to overcome many of the methodological problems that other researchers have faced. Data were collected concerning the nutritional status of 627 children in three urban communities in South India. Information was also collected on water quality, water quantity, household sanitation, socioeconomic conditions, and housing. A statistical technique is presented that allows for controlling potential …

Faculty Publications
Weight-for-height and height-for-age data were compared for preschool-age Haitian children enrolled in a community health and nutrition intervention programme and children measured in the Haiti national nutrition survey of 1978. Cross-sections of the longitudinal data of the intervention programme corresponding to the season when the national survey was conducted (May to September) were chosen for the three years of available programme data (1969, 1970, 1971). Significantly less stunting was found in children in the 1970 and 1971 intervention group than in the children covered by the national survey. Tests of trend also showed that the height (or length) status of …

Body Composition And Its Outcomes And Management In Multiple Sclerosis: Narrative Review, H W Van Landuyt, J Boelaert, P Piot, L Verbist
Faculty, Staff and Student Publications
Background: There is emerging interest in obesity and its prevalence, outcomes, and management in people with multiple sclerosis (MS). Body mass index (BMI) is the traditional marker of obesity in MS, whereas body composition, inclusive of specific body tissue compartments (e.g., fat, bone, and muscle), is often overlooked despite its relevance.
Objective: This narrative review (a) underscored the use and utility of dual-energy X-ray absorptiometry (DEXA) as an accurate and reliable measure of body composition; (b) thematically analyzed and synthesized the current evidence regarding body composition (using DEXA); and (c) determined gaps to be addressed in future research.
Methods: The …

A study designed to identify the location of a gene for multiple endocrine neoplasia type 2 (MEN-2) is being performed using a single large family segregating for MEN-2A. Here we report our initial results using a set of DNA restriction fragment length polymorphisms spanning a 40 cM segment of the short arm of human chromosome 11. The locus for MEN-2 in this family is excluded from this entire region which includes an oncogene, c-Ha-ras-1, two hormone loci, insulin and parathyroid, and the five hemoglobin genes in the beta hemoglobin gene duster. This is the first exclusion of such a large …

We evaluated the effects of screening for multiple endocrine neoplasia type 2A (MEN-2A) in 12 families. Genealogical studies going back to 1730 show a common ancestry for seven Swedish families and one American family. The total number of patients included 105 individuals, 68 of whom were diagnosed by our screening program. Our screening methods for medullary carcinoma of the thyroid (MTC) had an optimal sensitivity and specificity. The frequency of gene carriers detected in MEN-2A families was 55%. Screening will lead to early diagnosis and early therapy, which in turn, will significantly decrease morbidity, incidence of surgical complications, and mortality …

In 19 patients from nine MEN-2A families, high-resolution C-banded chromosome studies have revealed a visible deletion within sub-band 20p12.2, yet no abnormality was observed in a 10th family. A deletion indistinguishable from that in MEN-2A was observed in five patients from three MEN-2B families but not in two other MEN-2B families. We found no abnormality in the entire karyotype of the four MEN-1 patients studied. These findings suggest that the mutation in most MEN-2 patients is a visible deletion in the short arm of chromosome 20.

Progress in understanding the single gene, cytogenetic, and multifactorial traits that predispose to human cancer suggests possible new directions for research in the multiple endocrine neoplasia (MEN) syndromes. Among the other 200 or so monogenic disorders associated with human neoplasia, advances have come from further delineation of syndromes by various clinical specialists, the recognition of subtypes of syndromes previously thought to be homogeneous, the search for in vitro manifestations of the mutant gene in fibroblasts, and the establishment of cell, tissue and patient registries and of voluntary lay organizations to serve as advocates for the disease. With regard to cytogenetics, …
