Saat ini, perbanyakan rami menggunakan rizoma sebagai bahan tanamnya, tetapi dalam produksinya membutuhkan waktu yang lama dan sebagai bahan tanam umur simpannya singkat. Kultur jaringan merupakan salah satu teknologi untuk mendapatkan bahan tanam yang seragam dan sehat dalam waktu yang singkat. Peningkatan kualitas kecambah dapat dibantu dengan seed priming, seperti perendaman dalam larutan pupuk organik cair. Penelitian bertujuan untuk mengetahui: pengaruh berbagai bahan dan konsentrasi seed priming terhadap perkecambahan biji aren (Arenga pinnata (Wurmb.) Kerugian akibat serangan penyakit busuk buah kakao (BBK) mencapai 40-100%, produktivitas hanya 669,9 kg/ha/tahun. Upaya pengendalian dapat dilakukan melalui penggunaan cendawan antagonis Trichoderma dan peningkatan dosis pupuk Kalium. Penelitian bertujuan menguji efektivitas Trichoderma viride dan pupuk K dalam mengendalikan penyakit BBK. Penelitian dilakukan pada Januari sampai Desember 2019 di kebun kakao rakyat (Klon CSA 6) berumur 10 tahun dengan tanaman pelindung Gliricidia sp. Integrating crop and livestock is being considered to improve soil health by carbon sequestration. A two-year study (2019-2021) at CPBES in Newton, MS was aimed to evaluate soil microbial diversity in the warm, humid regions, specifically southeastern USA. Amplicons targeting bacterial 16S rRNA genes and fungal ITS2 regions were sequenced. Taxonomic assignment and microbial diversity characterization were performed using QIIME2®. Soil fungal diversity showed significant differences (alpha diversity, p = 0.031 in yr. 2020 and beta diversity, p = 0.037 in yr. 2021). As fossil fuel supplies decrease and concerns of climate change increase, the search for alternative sources of fuel has pushed biomass crops to the forefront of discussion. Saccharum spontaneum readily hybridizes with commercial sugarcane and lends cold tolerance and greater yields to the hybrid progeny, called energycane. Twenty genotypes were tested against an energycane variety (Ho 02-113) as a control. Two locations were tested: the HH Leveck Animal Research Center (planted in 2019), and the Bearden Dairy Research Center (planted in 2020). With new 2,4-Dichlorophenoxyacetic acid (2,4-D) tolerant crops, increases in off-target movement events are expected. New formulations may mitigate these events, but standard lab techniques are ineffective in identifying these 2,4-D formulations. Using Fourier-transform infrared spectroscopy and machine learning algorithms, research was conducted to classify 2,4-D formulations in treated herbicide-tolerant soybeans and cotton and observe the influence of leaf treatment status and collection timing on classification accuracy. Pooled Classification models using k-nearest neighbor classified 2,4-D formulations with over 65% accuracy in cotton and soybean. Tissue collected 14 DAT and 21 DAT for cotton and soybean respectively produced higher accuracies than the …

In 2019, the Mississippi River watershed had a record-long flooding, which was comparable with the 1927 Great Mississippi River Flood. This study leveraged this flooded condition to assess the flood tolerance of eastern cottonwood (Populus deltoides) and black willow (Salix nigra) planted as short rotation woody crops (SRWCs) on seasonally flooded marginal land of the Lower Mississippi Alluvial Valley (LMAV) in 2018. The survival/mortality prediction models developed by using hydrologic and environmental variables suggested that only high flood depth affected the survival of black willow. Soil carbon is the largest sink and source of the global carbon cycle and is disturbed by several natural, anthropogenic, and environmental factors. The global increase of atmospheric CO2 affects soil carbon cycling through varied biogeochemical processes. The first chapter is a compilation of current information on potential factors triggering soil acidification and weathering mechanisms under elevated CO2 and their consequences on soil inorganic carbon (SIC) pool and quality. Potassium (K) deficiency of soybean [Glycine max (Merr.) L.] is common throughout the Mid-Southern region of the United States. Visual symptoms of K deficiency may be absent or subtle during early and mid-reproductive growth stages. Correcting K deficiency in-season is problematic due to limited research. Information regarding soybean yield response to in-season fertilizer K application time is limited to growth stages prior to the onset of R1. However, the use, motivations, and barriers to adopting organic practices in the commercial turfgrass and landscape industry are undocumented. A survey with Louisiana Turfgrass Association (LTA) members to gauge their perceptions on current and future use of organic products in Louisiana was performed; and a field trial was conducted to evaluate organic fertilizers during turfgrass establishment. Floodplain forests support a high diversity of tree species adapted to regenerate under fluctuating water and light availability. Regeneration regulates species composition, and shade and flood tolerance influence the likelihood of regeneration. Regeneration failure of shade-intolerant and flood-tolerant tree species commonly occurs in southcentral and southeastern floodplain forests of the United States, also known as bottomland hardwoods (BLHs). In many BLHs reduced flooding has resulted in a dryer floodplain. These changes in flooding are linked to recent shifts in species composition. Cover crops improve soil health and reduce the risk of soil erosion. However, their impact on the carbon dioxide equivalence (CO2e) is unknown. Therefore, objective of this two-year study was to quantify the effect of cover crop-induced differences in soil moisture, temperature, organic C, and microorganisms on CO2e and to develop machine learning algorithms that predict daily N2O-N and CO2-C emissions. The prediction models tested were multiple linear regression (MLR), partial least square regression (PLSR), support vector machine (SVM), random forest (RF), and artificial neural network (ANN). Nematodes are the most abundant and diverse animals on the planet but lack representation in biodiversity research. This presents a problem for studying nematode diversity, particularly when molecular tools (i.e., barcoding and metabarcoding) rely on well-populated and curated reference databases, which are absent for nematodes. To improve molecular identification and the assessment of nematode diversity, we created and curated an 18S rRNA database specific to nematodes (18S-NemaBase) using sequences sourced from the most recent publicly available 18S rRNA SILVA v138 database. Several weeds serve as alternative soybean cyst nematode (SCN) hosts. Still, the relative reproductive capacity of SCN HG types (Heterodera glycines type) on weed hosts relative to soybean is not well understood. This study examined the reproduction of three South Dakota endemic SCN populations—PSCN-1 (HG 0), PSCN-2 (HG 2.5.7), and PSCN-3 (HG 7)—on purple deadnettle, field pennycress, and henbit. The Relative Female Index (RFI) was calculated to compare SCN reproduction relative to the susceptible soybean check. Weed hosts, HG types, and their interactions influenced SCN reproduction. Soybean plants are vulnerable to a wide range of diseases, and nematodes are among the worst pests of soybean. This dissertation explores three aspects of conservation agriculture (CA) in the Sub-Saharan African region (SSA). The first article examines the demand side of CA and explores whether urban maize (Zea mays L.) consumers in the Democratic Republic of the Congo (DRC) would be willing to pay a premium for CA-produced maize flour. The second article estimates the effects CA provides to adopters and their farms in smallholder farming systems in the DRC, focusing on changes in soil properties and cowpea (Vigna unguiculata) yields. Young peach trees are often rainfed in the southeastern U.S. and periods of dry weather can cause tree water deficit that can be detrimental to orchard productivity. There is a lack of understanding of the influence of water regimes and rootstocks on the nutritional status and gas exchange of the peach tree. Most peach growers in Southeastern United States do not irrigate their young peach trees because they do not produce fruit until the third leaf. Protein prenylation, a common lipid post-translational modification, is required for growth and development in eukaryotes. Rab geranylgeranylation involves the addition of one or two 20-carbon geranylgeranyl moieties to Rab-GTPase target proteins, which regulate intracellular vesicle trafficking. The reaction is carried out by heterodimeric Rab geranylgeranyltransferase (Rab-GGT), which is composed of two associated α- and β-subunits, with the assistance of an additional protein called Rab escort protein (REP). Phytophthora root and crown rot (PRCR) is currently the most important disease on lavender (Lavandula spp.) in the United States. The disease was first described on English lavender (L. angustifolia) in a Maryland nursery in 1991, with Phytophthora nicotianae as the causal agent. Since that time, the disease has been reported on multiple continents and as caused by several species of Phytophthora. This study examined the distribution and pathogenicity of Phytophthora species on lavender in the United States, requested lavender grower feedback regarding their production and concerns, and examined efficacy of selected management options. Key message: We investigate the genetic basis of panicle architecture in switchgrass in two mapping populations across a latitudinal gradient, and find many stable, repeatable genetic effects and limited genetic interactions with the environment. Abstract: Grass species exhibit large diversity in panicle architecture influenced by genes, the environment, and their interaction. The genetic study of panicle architecture in perennial grasses is limited. High demand from the brewing industry has encouraged production outside of the traditional Pacific Northwest, the primary production region. Producers in the Midwest are attempting to grow cultivars adapted to the Pacific Northwest, but environmental differences have caused low yields and changes in secondary metabolite content. Arbuscular mycorrhizal fungi (AMF) remain a vital obligate symbiont of nearly all plants. It is well established that the symbiosis between AMF and host plant improves plant nutrient acquisition, alleviates abiotic and biotic environmental stressors, defends against plant pathogens, and contributes to overall plant fitness and productivity through modification of the soil habitat. Modifications include increased soil aggregation and stability, carbon sequestration through provision of fungal wall precursors to soil organic matter (SOM) formation, and enhanced nutrient cycling in the mycorrhizosphere. Quizalofop-resistant rice technology allows for over-the-top applications of quizalofop, an acetyl-coenzyme A carboxylase-inhibiting herbicide. However, quizalofop caused significant injury to quizalofop-resistant rice in some Arkansas fields during the first year of commercialization. Experiments evaluated the effect of early-season soil moisture and nitrogen availability; pre-exposure to low rates of glyphosate and imazethapyr; planting date; and environmental conditions including, soil moisture content, air temperature, and light intensity on quizalofop-resistant rice tolerance to quizalofop applications. Blueberries’ popularity is in part supported by the health benefits associated with their consumption. Research has strongly focused on their polyphenol and anthocyanin concentration, one of the highest amongst commonly consumed foods. Polyphenols have demonstrated antioxidant effects in vitro, and consuming polyphenol-rich foods is associated with protective effects towards non-communicable diseases, but their direct bioactivity in the human body remains to be fully understood. It is estimated that about 90 to 95% of the initial polyphenol intake reaches the colon without being absorbed. Sustainable intensification of rice farming is crucial to meeting human food needs while reducing environmental impacts. Rice produces 8% of all anthropogenic CH4, which is a potent greenhouse gas. CH4 emissions can potentially be reduced by cultivation practices that minimize the number of days the fields are saturated, such as dry-seeding instead of water-seeding and irrigation using the alternate wetting and drying (AWD) technique instead of delayed, continuous flooding (DF). Within the last four decades, phylogenetic comparative methods have become the defacto method of analysis for comparative biologists. The availability of high-quality comparative datasets has been matched by an explosion of possible phylogenetic models. In large part, the efforts to increase the realism of phylogenetic comparative methods has been successful as evidenced by their widespread use.
