Understanding the role of various chemical messengers in altering behaviors and physiological processes is a common goal for scientists across multiple disciplines. The main focus of this dissertation is on characterizing the action of an important neurotransmitter, acetylcholine (ACh), modulating larval Drosophila melanogaster neural circuits and heart. In this dissertation, I provide important insights into the mechanisms by which ACh influences the formation and performance of select neural circuits, while also revealing significant details regarding its role in additional physiological functions, including cardiac pace making. Ectothermic animals are susceptible to temperature changes such as cold shock with seasons. To survive through a cold shock, ectotherms have developed unique strategies. My interest is focusing on the physiological function of during cold shock and prolonged cold exposure in the fruit fly (Drosophila melanogaster) and crayfish (Procambarus clarkii). The variability in the number of basal axillary shoots (ground suckers) among all tobacco (Nicotiana tabacum L.) varieties, has increased since the hydroponic transplant production system became the standard. The larger root ball of hydroponically produced transplants compared to traditionally produced transplants potentially generates a difference in the ratio of auxin (inhibits axillary shoot formation) to cytokinin (promotes lateral branching), that induces basal axillary shoot development. Starting in 2014, studies were conducted to investigate whether the addition of synthetic auxins or cytokinins to hydroponic transplant production could prevent ground sucker formation. The Arctic has warmed by at least 3°C over the past 50 years and this rapid warming is expected to continue. Climate warming is driving the proliferation of shrubs across the tundra biome with implications for energy balance, climate, hydrology, nutrient cycling, and biodiversity. Changes in tundra plant water use attributable to shrub expansion are predicted to increase evapotranspirative water loss which may amplify local warming and reduce run-off. However, little is known about the extent to which shrubs will enhance evapotranspirative water loss in these systems. Direct measures of shrub water use are needed to accurately predict …

Bone mineral density reflects an athlete’s cumulative history of energy availability, physical activity, and menstrual status, as well as nutritional and environmental factors. Although sports with high-impact loading are associated with higher bone mineral density than low-impact or non-impact sports, confounding variables are differences in the athletes’ body size and sport-specific training. The primary objective of this study was to evaluate the synergistic effects of repeated sprint training in hypoxia (RSH) and beta-alanine supplementation on performance in recreationally active men. Participants were randomly assigned to one of the following groups: hypoxia + beta-alanine (HB, n = 10), hypoxia + placebo (HP, n = 9), normoxia + beta-alanine (NB, n = 11) and normoxia + placebo (NP, n = 8). ischemia/reperfusion injury that evokes cardioprotection through endogenous neural

and/or humoral signaling. A recent study has reported that protective signaling is

mediated by exosomes through the circulation; however this concept is supported by

limited and inconsistent evidence. Despite overwhelming success in preclinical studies,

This Dissertation is the first to estimate the variables D and S of the nonlinear theory of acute cell injury in hippocampal regions CA1 and CA3 using a rat model of global brain ischemia and reperfusion (I/R). D was estimated by quantifying protein aggregates using ubiquitin Western blotting. S was estimated by quantifying changes in polysomal mRNAs as measured by microarray chips. The miR-17- 92 cluster is necessary for cell proliferation and development of the cardiovascular system. Deletion of this cluster leads to death in neonatal mice. The role of this cluster still needs to be defined following ischemia and reperfusion. Methods and Results: Adult male mice were injected with Tamoxifen- was to induce inducible cardiac-specific miR-17- 92-deficient (miR-17- 92-def: MCM:TG:miR-17- 92 flox/flox ) and wild type (WT: MCM:NTG:miR-17-92 flox/flox ) mice were subjected to 30 minutes of myocardial ischemia via left anterior descending coronary artery ligation followed by reperfusion for 24 hours. The development and survival of the malaria parasite Plasmodium within its mosquito vector is not very well understood. When the parasite is taken up by an Anopheles mosquito, it is exposed to digestive enzymes, reactive oxygen species, increasing pH, decreasing temperature, and a diverse microbial flora. Our lab is studying the adaptive mechanisms underlying parasite survival in this hostile environment, one of which is movement out of the midgut. This study aimed to investigate the protective effects of trehalose on osmotic tolerance and the quality of equine sperm before and after freezing and thawing. Equine ejaculated semen was collected from six stallions (3 ejaculates per stallion; >50% motility and >70% normal morphology). Transient receptor potential vanilloid 2 channel (TRPV2) is a Ca2+-permeable ion channel that is highly expressed in leukocytes but is also present in skeletal and cardiac muscle and endocrine cells. The TRPV2 function is implicated in a number of physiological processes, including bacterial phagocytosis, pro-inflammatory cytokine production, cardiac hypertrophy, and cancer development. TRPV2 knockout mice exhibit a high incidence of perinatal mortality, arguing that the channel plays essential roles in physiology. Rationale: Tissue Factor (TF) is a transmembrane glycoprotein that canonically functions as the initiator of the coagulation cascade. Increased levels of TF have been associated with inflammatory airway diseases. Since lipopolysaccharide (LPS) is known to elicit and inflammatory response in airway epithelium, we hypothesized that airway epithelial cells release TF when exposed to LPS. Since TF aids in local wound healing, we also hypothesized that inhibition of TF would decrease NHBE growth. Impaired walking performance is a key determinant of morbidity among older adults. A distinctive characteristic of impaired walking performance among older adults is a greater metabolic cost compared to young adults. Specifically, healthy older adults have been shown to have a 15-20% greater metabolic cost of walking compared to young adults. However, a recent study suggests that older adults who routinely run for exercise have a lower metabolic cost of walking compared to older adults who walk for exercise. Supplementation with beetroot juice (BR) has been shown to reduce blood pressure (BP) at rest and improve several performance parameters during submaximal and maximal exercise. BR effects on BP during submaximal exercise have not been investigated. Furthermore, the effects of BR on VO2max are inconclusive. The purpose of this study was to investigate the effects of BR on VO2max and BP during submaximal exercise. 20 healthy, recreationally trained volunteers (age 21.8±2.35 years, weight 75.10±10.62 kg, height 177.4±6.39 cm) participated in this study, which had a double-blind placebo controlled randomized crossover design. Purpose: Our project was designed to determine the effect of time of day on multiple mechanisms influencing breathing stability and respiratory plasticity. We investigated if the number and duration of breathing events coupled to upper airway collapsibility, as well as the carbon dioxide reserve, chemoreflex sensitivity and arousal threshold during non-rapid eye movement (NREM) sleep were affected by the time of day. In addition, we examined if mild intermittent hypoxia (IH) initiates long-term facilitation of upper airway muscle activity leading to a reduction in the therapeutic continuous positive airway pressure required to eliminate breathing events.

Control of calcium binding to and dissociation from cardiac troponin C (TnC) is essential to healthy cardiac muscle contraction/relaxation. There are numerous aberrant post-translational modifications and mutations within a plethora of contractile, and even non-contractile, proteins that appear to imbalance this delicate relationship. The Southwestern US is predicted to become hotter and drier, as global climate change forces increasing temperatures and variability in timing and size of precipitation inputs. Drought stress has become more frequent in recent decades, and resulted in massive forest mortality in piñon-juniper woodlands. During recent severe droughts (2000-2003, 2009-2012), piñon pine (Pinus edulis Englem.) suffered disproportionately high mortality compared to co-occurring one-seed juniper (Juniperus monosperma [Engelm.] Sarg.). A large-scale precipitation manipulation experiment was established in a piñon-juniper woodland in central New Mexico to test hypotheses regarding tree survival and mortality with respect to altered water regimes. A new class of N-linked protein glycosylation – arginine rhamnosylation – has recently been discovered as a critical modification for the function of bacterial elongation factor P (EF-P). Herein, we describe the synthesis of suitably protected α- and β-rhamnosylated arginine amino acid “cassettes” that can be directly installed into rhamnosylated peptides. Preparation of a proteolytic fragment of Pseudomonas aeruginosa EF-P bearing both α- and β-rhamnosylated arginine enabled the unequivocal determination of the native glycosidic linkage to be α through 2D NMR and nano-UHPLC-tandem mass spectrometry studies.

Polycystic ovarian syndrome (PCOS) is a common endocrine disorder in women of reproductive age. The pathophysiology of PCOS has conventionally thought to originate from androgen excess. However, recent evidence suggests that androgen excess is a downstream consequence to inflammatory dysregulation and subsequent metabolic abnormalities. Inflammatory mRNA gene expression of TNFa and IL-1ß in mononuclear cells isolated from women diagnosed with PCOS was explored using qPCR. Additionally, the correlations between body mass index (BMI) and fasting glucose on mRNA expression of TNFa and IL-1ß were explored. mRNA expression of both TNFa and IL-1ß were found to be significantly higher in PCOS …
