DNA contains all the information a cell needs to grow, divide, differentiate, and survive. It also dictates how and when a cell should die. DNA damage (particularly double strand breaks i.e. DSBs) may lead to cell death and/or disease. To circumvent DNA breaks, cells use various DNA repair mechanisms including non-homologous end joining (NHEJ). My ORCA research project focused on identifying novel genes involved in NHEJ. I did this by performing a forward genetic screen (FGS) that tested every known gene in the genome for involvement in NHEJ.

Bisphenol chemicals are commonly used in the manufacturing of polycarbonate plastics, polyvinyl chloride plastics, resins, and thermal printing applications. Humans are inadvertently exposed to bisphenols through contact with consumer products and/or medical devices. Recent reports have shown a link between bisphenol-a (BPA) exposure and adverse cardiovascular outcomes; although these studies have been limited to adult subjects and models. Since cardiac physiology differs significantly between the developing and adult heart, we aimed to assess the impact of BPA exposure on cardiac function, using a neonatal cardiomyocyte model. Voltage-gated proton channels, HV1, were first reported in Helix aspersa snail neurons. These H+ channels open very rapidly, two to three orders of magnitude faster than mammalian HV1. Here we identify an HV1 gene in the snail Helisoma trivolvis and verify protein level expression by Western blotting of H. trivolvis brain lysate. Expressed in mammalian cells, HtHV1 currents in most respects resemble those described in other snails, including rapid activation, 476 times faster than hHV1 (human) at pHo 7, between 50 and 90 mV. We recently identified a voltage-gated proton channel gene in the snail Helisoma trivolvis, HtHV1, and determined its electrophysiological properties. Consistent with early studies of proton currents in snail neurons, HtHV1 opens rapidly, but it unexpectedly exhibits uniquely defective sensitivity to intracellular pH (pHi). The H+ conductance (gH)-V relationship in the voltage-gated proton channel (HV1) from other species shifts 40 mV when either pHi or pHo (extracellular pH) is changed by 1 unit. This property, called ΔpH-dependent gating, is crucial to the functions of HV1 in many species and in numerous human tissues. The HtHV1 channel exhibits normal pHo dependence but …

Autophagy is a widely conserved catabolic process that is necessary for maintaining cellular homeostasis under normal physiological conditions and driving the cell to switch back to this status quo under times of starvation, hypoxia, and oxidative stress. The potential similarities and differences between basal autophagy and stimulus-induced autophagy are still largely unknown. Both act by clearing aberrant or unnecessary cytoplasmic material, such as misfolded proteins, supernumerary and defective organelles. The relationship between reactive oxygen species (ROS) and autophagy is complex. Cellular ROS is predominantly derived from mitochondria. In plants, starch is a major carbon and energy storage compound. Starch is made as a product of photosynthesis while plants are in light and is degraded at night. Our lab is interested in the cellular mechanism of starch degradation in plants and for our studies we use Arabidopsis thaliana as a model. β-amylases are primarily responsible for the hydrolysis of starch in plants and a total of nine β-amylases genes are encoded in Arabidopsis thaliana. Purpose: To test the effect of using an electrically braked ergometer on the validity and reliability of the YMCA submaximal cycle test.

Methods: 22 male and 13 female subjects ages 19 to 31 completed one maximal treadmill test and four submaximal cycle tests to measure and estimate VO2max, respectively. The maximal tests involved recording heart rate and VO2 during graded exercise until volitional fatigue; an actual max was verified when two out of the following criteria: respiratory Exchange Ratio > 1.1, VO2 plateau (< 150 ml/min increase in VO2 during final stage), and achievement of 90% age-predicted HR max (or completed …

In the kidney, endocytic recycling regulates the abundance of channels and transporters in the membrane of the tubular epithelium, and thereby controls the kidney’s ability to regulate water homeostasis. In recent years, a family of proteins called Eps15 homology domain-containing (EHD) proteins has emerged as important regulators of the endocytic recycling pathway. Mammals express four paralogs of EHD proteins, EHD1-4, that are expressed in different tissues. Although EHD4 is expressed in the kidney, the specific physiological role of EHD4 in the kidney remains unknown. Because of its cardio-protective benefits, diets that are low in Na+ and high in K+ are often warranted in conjunction with use of diuretics such as furosemide for treating hypertension. It is critical to understand how such diets influence the diuretic actions on renal K+ handling in order to choose the best drug for patients.

Changes to the embryonic environment that occur within the first few days of development can have significant effects on subsequent fetal development. The following studies investigate the effects of progesterone-deprivation during the early preimplantation period in the mare. The first study described embryo morphology (diameter, stage, and quality grade) collected from mares with induced aluteal cycles. Populations with obesity are more likely to fall and exhibit balance instability. The reason for this is likely multifactorial, but there is some evidence that sensory function is impaired during obesity. We tested the hypothesis that muscle proprioceptor function is compromised in a mouse model of diet induced obesity. An in vitro muscle-nerve preparation was used to record muscle spindle afferent responses to physiological stretch and sinusoidal vibration. Hepatocellular carcinoma (HCC) is the most common form of liver cancer that affects ~14 million people in the world. Like all cancers, HCC is a disease that arises from unstinted cellular growth initiated by genetic alterations, metabolic changes, and dysregulation in key cellular pathways. Of interest is the relationship between metabolism and cell proliferation/degradation for therapeutic targeting. Recently a number of studies have reported various performance enhancing effects of dietary nitrate (NO3-) via beetroot juice (BRJ) supplementation. Objective: The purpose of this study was to observe the weight management habits of division III collegiate wrestlers during the week leading up to a competition. Methods: Subjects reported to the lab on Monday morning after an overnight fast. Subjects were measured for height and weight and were asked to provide urine and blood samples for analysis of hydration and creatine kinase activity, respectively. Objective: The purpose of this study was to determine if caffeine ingestion decreased time to contact (TTC), and increased peak hand speed (HS), peak bat barrel speed (BS), and power (P) during a softball bat swing by fatigued and non-fatigued female athletes from an NCAA Division III varsity softball team. A novel running wearable called the Stryd Summit footpod attaches to a runner’s right or left shoe and measures running power output. The developers of the product purport that the footpod’s power and form power measures may correlate with metabolic data gathered in a lab. Purpose: The purpose of this study was to investigate whether a short-term, unilateral, lower-body resistance training program would significantly improve static and dynamic balance in experienced college-aged resistance-trained participants when compared to a control group’s regular, bilateral lower-body resistance training program. Previous research on lower body positive pressure (LBPP) treadmills have focused primarily on muscle activation (EMG), spatiotemporal variables, and metabolic differences from running on a standard treadmill. The current study aimed to establish the necessary accommodation time needed for runners to display reliable metabolic and neuromuscular responses on the LBPP treadmill. Fifteen habitually trained runners (11 males, 4 females, average± SD VO2Max=62.81 ± 5.43mL.kg-1.min-1) ran a total of 3, 15-minute trials on the AlterG® treadmill during a single testing session. von Willebrand Disease (vWD) is a bleeding disorder caused by a deficiency in von Willebrand Factor (vWF), a large glycoprotein that assists in coagulation. Specifically, large vWF multimers in the blood stream are key components in starting the coagulation cascade. vWF is cleaved by the metalloprotease ADAMTS13, regulating the multimers size, which hinders vWF’s ability to function properly. The three main types of vWD —Type I, II, and III— are not well defined, and as a result are all similarly treated with plasma-derived vWF replacement therapy. And yet, researchers do not fully understand how this bacterium replicates and divides. A chronically elevated vasopressin level has been associated with an impaired glucose regulation system in humans. A recent study in patients with diabetes suggested that low water intake is associated with impaired glucose regulation. The aim of this study was to examine the effect of hypertonic saline infusion on glucose regulation in healthy females. Thirty healthy, non-diabetic females performed a 2-h infusion followed by a 4-h oral glucose tolerance test (OGTT). Purpose: The purpose of this study was to assess relationships among actual motor competence, perceived motor competence, and health-related fitness in a college-aged population. Methods: A total of 76 participants from SUNY Cortland enrolled in an undergraduate Kinesiology course completed an informed consent form. Total data were obtained on 71 participants (male = 53; female = 18). Perceived motor competence (PMC) was assessed via the Physical Self Perception Profile questionnaire, which participants completed one week prior to remaining assessments. Poikilothermic organisms experience trade-offs by differential physiological demands generated by temperature extremes. Many such organisms exhibit acclimatory effects, adjusting their metabolism and physiology to recently experienced temperatures. One such acclimatory effect is metabolic compensation, the deceleration of biological rates below Arrhenius expectations. Daphnia magna is eurythermal, and if acclimated to mildly stressful temperatures first, survives longer in lethal temperatures. This study examined the effect of ambient temperature (5°C-37°C) and acclimation history (lifetime at 10°C or 25°C) on the oxygen consumption rates of 8 genotypes of Daphnia with high or low acute temperature tolerance. Excessive alcohol consumption is a leading cause of death and disability globally and can lead to diseases such as alcoholic skeletal myopathy. Skeletal muscle is the largest organ in the human body and functions to regulate whole-body energy homeostasis. Additionally, skeletal muscle can function as an endocrine organ via secretion of myokines. Two myokines, myonectin and irisin, present a wide range of effects upon metabolism, inflammation, and tissue survival- signaling. We hypothesized that chronic alcohol consumption will result in reduced circulating myonectin and irisin levels. Mice were fed an ethanol-containing or control diet for 10 days or 6 weeks. Tissues …
