Key Success Factors In Using The Lifting Method To Construct A Steel Bridge Pylon, Ming-Chao Su, Hui-Ping Tserng

Constructing the inclined steel pylon of cable-stayed bridges is challenging because of the pylon’s height, spatial limitations, and safety concerns. To address these challenges, this study developed a modular lifting method that can enhance the efficiency and precision of pylon construction. The proposed method involves using rotation jigs to temporarily assemble the pylon, horizontal positioning on temporary supports, and synchronized lifting through computer-controlled hydraulic systems. Field implementation on a large-span bridge project revealed that compared with the traditional method, the proposed method resulted in higher welding quality, lower risks associated with great-height operations, and a shorter on-site assembly time. The coastline stands as a critical domain encompassing industry and the environment. The escalating global warming, leading to elevated sea levels and intensified wave-current interactions, has given rise to substantial coastal erosion. Understanding the factors that control ocean productivity is crucial in fisheries management. Taiwan Bank, located between the Taiwan Strait and the South China Sea, is a highly productive ecosystem due to upwelling events. The total zooplankton community (holozooplankton and merozooplankton), diversity, seasonal changes, and environmental drivers remain unexplored despite its productivity. This study evaluates the seasonal and vertical environmental dynamics in the Taiwan Bank and their impacts on the zooplankton community structure. This study focuses on assessing and improving the predictive performance of sea surface temperature (SST) in the Gulf of Thailand under unique environmental challenges. Data from the Hybrid Coordinate Ocean Model (HYCOM) were utilized, covering the period from July 1994 to June 2024 at depths ranging from 0 to 30 meters. After cleaning the data using the Interquartile Range (IQR) method, the processed dataset was tested using four models: Recurrent Neural Network (RNN) architectures, including Long Short-Term Memory (LSTM) and Gated Recurrent Unit (GRU), as well as Convolutional Neural Networks (CNNs). Inflammatory cytokines such as interleukin-6 (IL-6) and interleukin-12 (IL-12) are central regulators of immune signaling and key biomarkers of disease, yet existing assays for their detection remain slow, invasive, and lack multiplexing ability. This dissertation advances the development of single-walled carbon nanotube (SWCNT) optical nanosensors capable of real-time, multiplexed, and molecularly specific cytokine detection through innovative use of single-stranded DNA (ssDNA) interfaces.

First, an IL-6-specific DNA aptamer was employed as both a dispersing agent and recognition probe for SWCNT fluorescence sensing. They can manipulate light in unprecedented ways by altering amplitude, phase, and polarization at subwavelength scales. The ability of these two-dimensional elements to perform multiple optical functions within compact optical systems—while offering high functionality, small form factors, and easy integration into optoelectronic devices—has sparked significant interest across various fields and industries. We previously found that the levels of soluble amyloid precursor protein α (sAPPα) are partly regulated by acetylcholinesterase (AChE) in human non-small cell lung cancer cell lines A549 (p53 wild-type) and H1299 (p53-null). In this study, we hypothesize that sAPPα is partly regulated by leptin, a hormone classically recognized for its anorexigenic role in appetite regulation— yet recently shown to play a role in cancer cell signaling. It was observed that cells treated with leptin exhibited greater concentrations of sAPPα and lower concentrations of Aβ40/42 in the media compared to those of cells left untreated. Although the term “stakeholder” has been widely used in science communication, recent expansion in the discourse around the negative connotations of the term necessitate that scientists and practitioners cease using the term. We discuss why this shift is necessary, how it affects our own praxis, and suggest resources for working through more respectful and just language. Looking to the science communication literature can help recalibrate terminology in positive ways.

Byte-pair encoding (BPE) is pivotal for processing text into chunksize tokens, particularly in Large Language Model (LLM). From a topic modeling perspective, as these chunksize tokens might be mere parts of valid words, evaluating and interpreting these tokens for coherence is challenging. Most, if not all, of coherence evaluation measures are incompatible as they benchmark using valid words. We propose to interpret the recovery of valid words from these tokens as a ranking problem and present a model-agnostic and training-free recovery approach from the topic-token distribution onto a selected vocabulary space, following which we could apply existing evaluation measures. Recent research into solving vehicle routing problems (VRPs) has gained significant traction, particularly through the application of deep (reinforcement) learning for end-to-end solution construction. However, many current construction-based neural solvers predominantly utilize Transformer architectures, which can face scalability challenges and struggle to produce diverse solutions. To address these limitations, we introduce a novel framework beyond Transformer-based approaches, i.e., Adversarial Generative Flow Networks (AGFN). This framework integrates the generative flow network (GFlowNet)-a probabilistic model inherently adept at generating diverse solutions (routes)-with a complementary model for discriminating (or evaluating) the solutions. MicroRNAs (miRNA) are a class of small non-coding RNAs that play a key role in gene silencing. miRNAs form an RNA-induced silencing complex (RISC) with the argonaute 2 (AGO2) protein in order to control gene expression through silencing messenger RNAs (mRNA). The ability for a miRNA-mRNA complex to fit into AGO2 and form the RISC is based on the location and characteristics of a central bulge, but the mechanism and interactions that lead to a functional RISC remain unknown. Conventional computed tomography produces a 2D slice or 3D image from multiple conventional images to avoid overlapping structures and increase visualization of the internal features of the object. A set up, macro controls and processing for computed tomography was created for the Center for X-ray Optics for the first time, and demonstrated in this work. Adding differential phase contrast to conventional x-ray images highlights edges. Two more channels of information, dark field and integrated phase, improve the contrast and increase the signal-to-noise ratios of the image. CRISPR-Cas12a is a rapidly evolving technology for improved gene editing in cells, as well as in vivo. The system, consisting of Cas12a nuclease and approximately 40-nt long guide RNA (gRNA), can efficiently recognize and cut genomic double-stranded DNA (dsDNA). Chemical modifications to the gRNA can potentially improve its dsDNA binding and enhance Cas12a-mediated nuclease activity. In the presence of underlying cyclical patterns, the integrity of time series analysis depends critically on the ability to detect, model, and impute structured missing data without compromising the temporal structure. Azobenzene is a photoswitchable molecule known for its high conversion yield and repeatability. The inclusion of an azobenzene moiety with a known inhibitor of your target of interest allows for greater study of function as well as potential therapies. Three derivatives were designed from azobenzene and known inhibitor of EAATs, TBOA, to synthesize photoswitchable inhibitors of EAATs. Further investigation into the function of EAATs and development of a specific inhibitor for EAAT3, an important glutamate transporter in the brain required for proper function, can then be conducted. Functional Data Analysis (FDA) is a statistical approach used to analyze data that vary across a domain, such as curves or functions. This dissertation investigates Bayesian Functional Data Analysis (BFDA) through three applications. First, we explore the use of BFDA in outcome-dependent follow-up (ODFL) studies. After conducting simulation studies, we apply our model to cardiotoxicity and kidney function data. In this work, we discovered a novel organometallic gold(III) macrocycle, Au-Mac1, that demonstrates anticancer potency in a panel of triple-negative breast cancer cells (TNBC), and based on this complex, a biotinylated-Au-Mac1 probe was designed for target identification via chemoproteomics, which uncovered the engagement of HMOX2 of the heme-energy metabolism pathway. Using orthogonal chemical biology and molecular biology approaches, including immunoblotting, flow cytometry, and cellular thermal shift assays, it was confirmed that Au-Mac1 engages HMOX2 in cells. Downstream effects of Au-Mac1 on the depletion of mitochondrial membrane proteins and bioenergetics point to the potential role of HMOX2 in cancer. Importantly, Au-Mac1 …

Metallo-phosphines are ubiquitous in organometallic chemistry with widespread applications as catalysts in various chemical transformations, precursors for organic electronics, and chemotherapeutic agents or chemical probes. Here, we provide a comprehensive review of the exploration of the current biological applications of Au complexes bearing phosphine donor ligands. The goal is to deepen our understanding of the synthetic utility and reactivity of Au-phosphine complexes to provide insights that could lead to the design of new molecules and enhance the cross-application or repurposing of these complexes.

Background: Pediatric cancer incidence has been increasing in the United States, despite improvement in pediatric cancer survival. This steady increase in incidence trends is not completely understood but maybe associated with social and environmental factors. In this study we aimed to assess the cumulative incidence, temporal trends, and mortality rates in pediatric cancer. Additionally, we examined sub-group variability in both incidence and mortality rates. Methods: Data from Surveillance, Epidemiology, and End Results (SEER) −18 from 1973–2014 were used for the purpose of analysis in this study. Age-adjusted incidence rates were used to assess temporal trends in cancer among children aged < 1–19 years. Univariable and multivariable binomial regression models were used to examine the association between race and cancer mortality while adjusting for potential confounders. Results: There were 92,594 cancer diagnoses during this period. White children comprised 74,758, (80.7%), black children 10,030, (10.8%), and other races 6648, (7.2%). Overall the age-adjusted cumulative incidence was slightly higher among white children (16.4%) than black children (12.4%) and other (13.0%). Children aged 15–19 years and those in metropolitan regions were more likely to be diagnosed with pediatric cancer. Relative to females, males were 16% more likely to die from the disease [adjusted Risk Ratio (aRR): 1.16, 95% Confidence Interval (CI): 1.09–1.22]. Additionally, compared to white children, black children had higher mortality rates [(aRR): 1.37, 99% CI: 1.23–1.52]. Conclusions: There is an increasing trend in pediatric cancer incidence; while white children have the highest incidence, black children and males indicated a survival disadvantage, indicative of racial and sex variability in overall pediatric cancer in the United States.

This thesis examines whether cyber warfare now poses a more immediate threat to U.S. national security than weapons of mass destruction (WMD). Cyber operations have become a dominant instrument of contemporary conflict, with malicious actors operating in a persistent “grey zone” that blurs traditional boundaries between war and peace. These operations target military and civilian entities, both directly and often as collateral damage due to the uncontrollable nature of cyber threats. WMDs, despite their catastrophic destructive potential, remain largely constrained by established deterrence frameworks.

This thesis presents a hierarchical motion planning framework for SoftRafts, a modular and deformable aquatic robot capable of performing locomotion and manipulation tasks on water surfaces. SoftRafts consist of soft and rigid components that enable structural reconfiguration, offering adaptability in unstructured aquatic environments.

To address the complexity of planning in high-dimensional, deformable systems, the proposed method uses a bounding-shape abstraction, specifically, enclosing circles and rectangular bounding boxes to simplify motion planning. These enclosures abstract the robot's overall shape, reducing the high-dimensional planning problem into a lower-dimensional problem. The Belt Supergroup of the northwestern United States is one of the most extensive Mesoproterozoic sedimentary successions in North America. Although certain stratigraphic units have been well studied, correlations across the entire outcrop region remain unresolved. This project uses magnetostratigraphy and lithostratigraphy to investigate a potential lateral equivalency between the 1.4 billion-year-old (Ga) Appekunny Formation (Fm.) in Glacier National Park, MT and the 1.4 Ga Greyson Fm. in the Helena Embayment, MT. Both formations consist predominantly of shallow-water mudstones and contain the only North American occurrences of Horodyskia moniliformis, one of the oldest known macrofossils.
