Exploring Elementary Students’ Agricultural And Scientific Knowledge Using Evidence Centered Design, Molly R. Brandt May 2016

Exploring Elementary Students’ Agricultural And Scientific Knowledge Using Evidence Centered Design, Molly R. Brandt

School of Natural Resources: Dissertations, Theses, and Student Research

The public is more disconnected from agriculture than ever. Americans are two to four generations removed from the farm, and a majority of Americans, even in rural agricultural states have no direct link to agriculture. As a result, the public lacks the knowledge and appreciation of the food, fuel, and fiber it demands. The National Center for Agricultural Literacy (NCAL) uses National Agricultural Learning Objectives (NALOs) to measure student’s agricultural knowledge. The purpose of this project is to develop assessments aligned with the NALOs and to use results from the assessments to further understand students’ agricultural literacy. This study focuses …

Early Growth And Development Impairments In Patients With Ganglioside Gm3 Synthase Deficiency, H. Wang, A. Wang, Dan Wang, A. Bright, V. Sency, Aimin Zhou, B. Xin

Chemistry Faculty Publications

Ganglioside GM3 synthase is a key enzyme involved in the biosynthesis of gangliosides. GM3 synthase deficiency (GSD) causes a complete absence of GM3 and all downstream biosynthetic derivatives. The individuals affected by this disorder manifest severe irritability, intractable seizures and profound intellectual disability. However, we have found that most newborns seem symptom-free for a period of time after birth. In order to further understand the onset of the disease, we investigated the early growth and development of patients with this condition through this study. We compared 37 affected individuals with their normal siblings and revealed that all children with GSD …

Impacts To Phytoplankton After The Establishment Of Quagga Mussels In Lake Mead, Nevada, Warren Bruce Turkett May 2016

Impacts To Phytoplankton After The Establishment Of Quagga Mussels In Lake Mead, Nevada, Warren Bruce Turkett

UNLV Theses, Dissertations, Professional Papers, and Capstones

Quagga mussels (Dreissena rostriformis bugensis) are an invasive species that were discovered in Lake Mead in January of 2007, and rapidly spread throughout the lake. Quagga mussels are about the size of a fingernail, but are known to clog pipes, damage infrastructure and alter ecosystems. In large numbers, mussels can have a dramatic impact on an ecosystem by reducing the phytoplankton and potentially increasing toxin producing cyanobacteria. This research analyzed data collected from three separate basins in Lake Mead to determine if water quality characteristics that are commonly impacted by invasive mussels have changed. Transparency, chlorophyll-a, phytoplankton and zooplankton samples …

Vision Based Extraction Of Nutrition Information From Skewed Nutrition Labels, Tanwir Zaman

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Vision-based extraction of nutritional information from nutrition labels (NLs) available on most product packages is critical to proactive nutrition management, because it improves the user’s ability to engage in continuous nutritional data collection and analysis. However, even users who are health conscious find it difficult to keep track of their nutrition intake due to lack of time, motivation, or training. In order to make nutrition management more proactive we present a Proactive NUTrition Management System (PNUTS), which aims to make nutrition management more user friendly and proactive using computer vision techniques running on smartphones which are ubiquitous and powerful computers …

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

The first three projects involve the synthesis and the mode of action study of 1,4-naphthoquinone based anticancer agents. These anticancer agents are highly potent against a wide range of cancer cell lines. These compounds showed ~8-fold selectivity towards the lung cancer cell line than the normal cell line. We also studied the mode of action and observed that generation of reactive oxygen species is the primary mode of action.

The final project involves the synthesis of multi-cationic antimicrobial agents. These compounds are active against both bacteria and fungi. These compounds are very easy to synthesize and can be scaled up …

Hydra Effects In Stable Communities And Their Implications For System Dynamics, Michael H. Cortez, Peter A. Abrams May 2016

Hydra Effects In Stable Communities And Their Implications For System Dynamics, Michael H. Cortez, Peter A. Abrams

Mathematics and Statistics Faculty Publications

A hydra effect occurs when the mean density of a species increases in response to greater mortality. We show that, in a stable multispecies system, a species exhibits a hydra effect only if maintaining that species at its equilibrium density destabilizes the system. The stability of the original system is due to the responses of the hydra-effect species to changes in the other species’ densities. If that dynamical feedback is removed by fixing the density of the hydra-effect species, large changes in the community make-up (including the possibility of species extinction) can occur. This general result has several implications: (1) …

Preparation Of Novel Steroidal Conjugates As Potential Diagnostic And Therapeutic Agents With An Emphasis On Quinoid And Halogenated Moieties, Nuha Obaid Juma Alsoom

Theses

Steroids bonded to pharmaceutically active molecules are interesting candidates as therapeutic agents. In this work, a number of potentially bioactive residues, which include the quinoid moiety, have been synthesized as a simple steroid quinone hybrid, where the steroid component will be estradiol derived or the steroid cholesterol derived. For this purpose, two novel reactions have been explored namely, the modification of the APPEL reaction to connect residues by using PPh3/BrCCl3 for amidation and esterification of carboxylic acids. Another one is the hydrogenation reaction of alkenes by using NaBH4, AcOH, Pd/C and toluene or benzene in the presence of benzyl groups …

Effects Of Creatine, Leucine, And Ethanol On Glial Cell Line-Derived Neurotrophic Factor In C2c12 Skeletal Muscle Cells, Daniel Waters

Honors Theses

Glial Cell Line-Derived Neurotrophic Factor (GDNF) is a protein expressed throughout the mammalian nervous system that serves several protective functions. Originally discovered in the brain, where it supports the survival of dopaminergic neurons, GDNF has since been shown to be expressed in skeletal muscle and is vital for the maintenance of motor neurons. The relationship between muscle hypertrophy, and GDNF is not well understood. The purpose of this study was to examine the relationship between hypertrophy and GDNF by inducing hypertrophy in C2C12 skeletal muscle cells using leucine and creatine, inhibiting hypertrophy using ethanol and measuring GDNF content. As measured …

Improved Microplate Fluorometric Soil Enzyme Assay For Β-Glucosidase Detection, Emily C. Hoehn Apr 2016

Improved Microplate Fluorometric Soil Enzyme Assay For Β-Glucosidase Detection, Emily C. Hoehn

School of Natural Resources: Dissertations, Theses, and Student Research

Soil microbes produce extracellular enzymes responsible for degrading complex organic compounds to release energy and nutrients. Measurement of soil enzymes can be considered an indicator of soil health and microbial community composition because of its sensitivity to agricultural and management practices. Fluorescence enzyme assays tend to be more sensitive than spectrophotometric (ie. colorimetric) assays and a 96-well plate has the potential capacity for high-throughput use. Development of a newly modified enzyme assay using fluorometric (4-methylumbelliferone), automated pipetting system and sonication, as well as a reduction in replicates allows for a higher sample throughput rate suitable for service laboratory use. The …

Does Predation Influence The Seasonal And Diel Timing Of Moose Calving In Central Ontario, Canada?, Brent R. Patterson, Kenneth J. Mills, Kevin R. Middel, John F. Benson, Martyn E. Obbard

School of Natural Resources: Faculty Publications

Birth synchrony is well documented among ungulates and is hypothesised to maximize neonate survival, either by minimizing the risk of predation through predator swamping or by synchronising birthing with increased seasonal food availability. We used encapsulated vaginal implant transmitters to locate and capture neonatal moose calves and document the seasonal and diel timing of parturition in two adjacent study areas with different predation pressure in central Ontario, Canada. We tested the hypothesis that predation promotes earlier and more synchronous birth of moose calves. Across both areas, proportionately more births occurred during the afternoon and fewer than expected occurred overnight. Mean …

Evaluation Of Pulmonary Function Among Workers Engaged In The Manufacture Of Hydraulic Fracking Ceramic Proppant, Humairat H. Rahman

USF Tampa Graduate Theses and Dissertations

Workers involved in hydraulic fracking processes are exposed to various types of chemicals and dusts in their workplaces, such as proppants, which hold open the fissures created in the fracking process. Recently, ceramic proppants have been developed that may be less hazardous to workers than traditional proppants. Pulmonary function testing of workers producing ceramic proppant was used to assess the potential inhalation hazards of ceramic proppant. Male workers (n = 100) from a producer of ceramic proppant were evaluated with pulmonary function test data collected and evaluated using The American Thoracic Society (ATS) acceptability criteria. A comparison group was selected …

Biology, Chemistry, and Environmental Sciences Faculty Articles and Research

1.Global climate models suggest that many ecosystems will experience reduced precipitation over the next century and the consequences for invasive plant performance are largely unknown. Annual invasive species may be able to quickly evolve traits associated with drought escape or tolerance through rapid genetic changes.

2.We investigated the influence of five years of water and nitrogen manipulations on trait values in a southern California grassland system. Seeds from two annual grass species (Avena barbata, Bromus madritensis) were collected from experimental plots and grown in a common environment over two generations. We measured 14 physiological, morphological, phenological, and …

Modelling Latent Variables For Bayesian Networks, Charles Cain Apr 2016

Modelling Latent Variables For Bayesian Networks, Charles Cain

Undergraduate Research Symposium 2016

Bayesian Networks are networks of interconnected variables used to explain causal relationships with conditional probability. Latent variables or hidden variables are variables that cannot be directly measured, like depression or physical activity. They can be used inside of a Bayesian Network. This research looks at latent variables as a weighted sum of observed variables. We use these modeled latent variables as continuous variables in a Bayesian Network. As an example, we look at a Bayesian Network of the causation of diabetes using data from the National Health and Nutrition Examination Survey (NHANES) that is publicly available from the CDC and …

Comparison Of Growth Rates In Two Captive Bred Species Of Atelopus (Anura; Bufonidae), At El Valle Amphibian Conservation Center, Cecile Avery Apr 2016

Comparison Of Growth Rates In Two Captive Bred Species Of Atelopus (Anura; Bufonidae), At El Valle Amphibian Conservation Center, Cecile Avery

Independent Study Project (ISP) Collection

Since their evolution, amphibians have managed to survive four mass extinctions. But today’s amphibians are now facing severe decline due to a plethora of causes including habitat destruction, climate change, pathogens, and pollution. Of all the possible causes of decline and extinction of amphibian populations, one of the most startling has been the effect of Batrachochytrium dendrobatidis (Bd), or chytrid fungus. It was decided that rapid action was needed to preserve the amphibian populations in the area, since it was clear that current in situ conservation methods were ineffective against Bd. EVACC, the El Valle Amphibian Conservation Center, was created …

The Effect Of Diet On The Bovine Rumen Microbial Community Structure And Composition And Its Effects On Methane Production In Growing And Finishing Cattle, Allison L. Knoell Apr 2016

The Effect Of Diet On The Bovine Rumen Microbial Community Structure And Composition And Its Effects On Methane Production In Growing And Finishing Cattle, Allison L. Knoell

Department of Animal Science: Dissertations, Theses, and Student Research

Methane is a potent greenhouse gas (GHG) that contributes to global warming. A natural by-product of ruminant fermentation is the production and eructation of methane. Methane is produced by a small unique group of microorganism’s called methanogens that belong to the domain Archaea. Enteric methane represents 2-12% energy loss in ruminants. It is well established that diet affects the microbial community structure and composition. Fermentative products of the mixed microbial population (bacteria, fungi, and protozoa) become the substrates for methanogens. These substrates influence which microorganisms will thrive. However, the effect of diet on the microbial community while simultaneously calculating …

Binding Of Oxaliplatin And Its Analogs With Dna Nucleotides At Variable Ph And Concentration Levels, Rippa Sehgal Apr 2016

Binding Of Oxaliplatin And Its Analogs With Dna Nucleotides At Variable Ph And Concentration Levels, Rippa Sehgal

Masters Theses & Specialist Projects

Oxaliplatin is one of the three FDA-approved platinum anticancer drugs and considered a third generation drug, discovered after the first generation drug cisplatin and second generation drug carboplatin. It is known to react with proteins and DNA nucleotides in the body. Reaction with DNA occurs primarily at guanosine residues and secondarily at adenine residues for oxaliplatin and other platinum drugs. We have previously studied oxaliplatin and an analog with additional steric hindrance in the amine ligand and found that the analog had different reactivity with methionine. Now, we have prepared oxaliplatin and its three analogs Pt(Me2dach)(ox), Pt(en)(ox) and Pt(Me4en)(ox) and …

Folate-Peg-Histidine For Use In Detection, Capture, And Analysis Of Fr+ Cells, Sean Dwyer Apr 2016

Folate-Peg-Histidine For Use In Detection, Capture, And Analysis Of Fr+ Cells, Sean Dwyer

All Capstone Projects

Folate receptor alpha is a membrane-bound protein displaying high affinity for folic acid. This receptor serves as a receptor-mediated transport system of folic acid into cancer and cells associated with inflammation (e.g., macrophages). Folate is a basic component of cell metabolism in both the synthesis of DNA and proteins, which is hypothesized to be necessary for maintaining adequate processes during increased metabolic demand. Interestingly, most normal cells in the body have little or no high affinity folate receptor alpha. Based on these attributes, folate based drug delivery, imaging systems, and diagnostic systems are in several stages of development worldwide. To …

Mechanistic Binding Insights For 1-Deoxy-D-Xylulose-5-Phosphatesynthase, The Enzyme Catalyzing The First Reaction Of Isoprenoid Biosynthesis In The Malaria-Causing Protists, Plasmodium Falciparum And Plasmodium Vivax, Matthew R. Battistini, Christopher Shoji, Sumit Handa, Leonid Breydo, David J. Merkler

Chemistry Faculty Publications

We have successfully truncated and recombinantly-expressed 1-deoxy-D-xylulose-5-phosphate synthase (DXS) from both Plasmodium vivax and Plasmodium falciparum. We elucidated the order of substrate binding for both of these ThDP-dependent enzymes using steady-state kinetic analyses, dead-end inhibition, and intrinsic tryptophan fluorescence titrations. Both enzymes adhere to a random sequential mechanism with respect to binding of both substrates: pyruvate and D-glyceraldehyde-3-phosphate. These findings are in contrast to other ThDP-dependent enzymes, which exhibit classical ordered and/or ping-pong kinetic mechanisms. A better understanding of the kinetic mechanism for these two Plasmodial enzymes could aid in the development of novel DXS-specific inhibitors that might prove useful …

Purification And Preparation Of Intrinsically Disordered Proteins For Nmr Spectroscopy, Kyle Chamberlain Apr 2016

Purification And Preparation Of Intrinsically Disordered Proteins For Nmr Spectroscopy, Kyle Chamberlain

Open Access Theses

NMR spectroscopy has recently become a promising field for protein characterization and dynamic studies. As the technology and pulse sequences improve for tracking proteins, a greater demand for developing effective purification protocols to produce NMR grade protein samples have arisen. This thesis explores two proteins: histone H4 tail and Mucin 1; Two very different proteins that require different methods of expression and purification to achieve a high enough yield for NMR analysis. H4 is a water soluble protein that weighs ~2.7 kDa, and has no extinction coefficient. Since the protein is too small for many methods of expression and purification, …
