Biological phenotypes of tri-segmented arboviruses display characteristics that map to mutation/s in the S, M or L segments of the genome. Plaque variants have been characterized for other viruses displaying varied phenotypes including attenuation in growth and/or pathogenesis. In order to characterize variants of Bunyamwera and Ngari viruses, we isolated individual plaque size variants; small plaque (SP) and large plaque (LP) and determined in vitro growth properties and in vivo pathogenesis in suckling mice. We performed gene sequencing to identify mutations that may be responsible for the observed phenotype. The LP generally replicated faster than the SP and the difference …

Mathematical Modeling And Simulation Of Multialleic Migration-Selection Models, Chad N. Vidden
Journal of Undergraduate Research at Minnesota State University, Mankato
Population ecology is concerned with the growth and decay of specific populations. This field has a variety of applications ranging from evolution and survival at the environmental level to the spread of infectious disease at the cellular and molecular levels. Many ecological circumstances require the use of mathematical methods and reasoning in order to acquire better knowledge of the issue at hand. This study considered and analyzed multiple different mathematical models of population dynamics along with their purposes. This foundation was then applied in order to explore the migration of populations from one isolated region to another along with the …
Simple, Label-Free And Non-Instrumented Analyte Quantitation By Flow Distance Measurement In Microfluidic Devices, Debolina Chatterjee
Simple, Label-Free And Non-Instrumented Analyte Quantitation By Flow Distance Measurement In Microfluidic Devices, Debolina Chatterjee
Theses and Dissertations
Rapid determination of the concentrations of molecules related to diseases can provide timely information for treatment options. However, most biomarker quantitation methods require costly and complex equipment. On the other hand, point-of-care systems have less complex instrumentation needs than laboratory-based equipment, but often provide less information; for example, biomarker presence or absence instead of concentration. A complete analysis setup addressing key limitations of both laboratory-based and portable systems is highly desirable. I developed microfluidic devices with visual inspection readout of a target’s concentration from microliter volumes of solution flowed into a microchannel. Microchannels are formed within polydimethylsiloxane (PDMS), and the …

We used the State Inpatient Databases from the United States Agency for Healthcare Research and Quality to provide state-specific age-adjusted blastomycosis-associated hospitalization incidence throughout the entire United States. Among the 46 states studied, states within the Mississippi and Ohio River valleys had the highest age-adjusted hospitalization incidence. Specifically, Wisconsin had the highest age-adjusted hospitalization incidence (2.9 hospitalizations per 100,000 person-years). Trends were studied in the five highest hospitalization incidence states. From 2000 to 2011, blastomycosis-associated hospitalizations increased significantly in Illinois and Kentucky with an average annual increase of 4.4% and 8.4%, respectively. Trends varied significantly by state. Overall, 64% of …

Coexistence Of Multi-Allelic Polymorphism With Migration And Selection, Andrew Flick
Journal of Undergraduate Research at Minnesota State University, Mankato
Population ecology is concerned with the growth patterns of populations. This field has many applications, ranging from survival at the environmental level, to the spread of infectious diseases at the cellular level. Mathematical modeling and computer simulation can be powerful tools in researching this area. I will be investigating the spatial patterns in populations (or gene frequencies) due to migration and selection. My research conditions are for the maintenance of polymorphism under a variety of migration schemes in discrete-space and continuous-time mathematical models. The results will be applicable from the ecological level to the molecular level. Some species are better …

Is There Progress Toward Sustainability?: Despite The Inherent Human Resistance To Change., Jens Pohl
Collaborative Agent Design (CAD) Research Center
The theme of this paper is to briefly survey the status of current efforts to maintain our natural environment and then discuss prospects for achieving future sustainability. The author finds that while there has been general recognition of the need for environmental sensitivity and conservation of natural resources, progress toward achieving sustainability goals has been slow. A principal reason for this lack of action is found in the reactive nature of the human species. Situated by biological design in our environment we typically respond to environmental changes only after they have occurred. Accordingly, while a number of revolutionary proposals have …

Dartmouth Scholarship
Background: Readmissions after hospital discharge are a common occurrence and are costly for both hospitals and patients. Previous attempts to create universal risk prediction models for readmission have not met with success. In this study we leveraged a comprehensive electronic health record to create readmission-risk models that were institution- and patient- specific in an attempt to improve our ability to predict readmission. Methods: This is a retrospective cohort study performed at a large midwestern tertiary care medical center. All patients with a primary discharge diagnosis of congestive heart failure, acute myocardial infarction or pneumonia over a two-year time period were …
Using Smart Grid Technologies To Protect Human Health In Water Distribution Systems, Graham Symmonds
Using Smart Grid Technologies To Protect Human Health In Water Distribution Systems, Graham Symmonds
International Conference on Hydroinformatics
Since the introduction of the Safe Drinking Water Act, there has been marked reduction in the number of water-borne disease outbreaks attributed to water treatment systems. Over the same period, however, the percentage of disease outbreaks attributable to defects in the distribution system has increased exponentially. Interestingly, as a result of the continuous aging infrastructure employed by our water utilities, the number of water-borne disease cases has increased in the last decade. The smart grid for water can be used to significantly improve both the utility's understanding of water quality in the distribution system, but can dramatically increase the response …
Antimicrobial Nanoparticles: A Green And Novel Approach For Enhancing Bactericidal Efficacy Of Commercial Antibiotics, Monic Shah
Masters Theses & Specialist Projects
On the verge of entering the post-antibiotic era, numerous efforts are in place to regain the waning charm of antibiotics which are proving ineffective against most “Superbugs”. Engineered nanomaterials, especially gold nanoparticles (GNPs) capped with antibacterial agents, are proving to be an effective and novel strategy against multidrug resistant (MDR) bacteria. In this study, we report a one-step synthesis of antibioticcapped GNPs (25 ± 5 nm) utilizing the combined reducing and capping ability of a cephalosporin antibiotic, ceftazidime. No signs of aggregation or leaching of ceftazidime from GNP surface was observed upon its storage. Antibacterial testing showed dosedependent broad spectrum …

Gpu-Accelerated Influenza Simulations For Operational Modeling, Peter Holvenstot
Gpu-Accelerated Influenza Simulations For Operational Modeling, Peter Holvenstot
Masters Theses
Simulations of influenza spread are useful for decision-making during public-health emergencies. Policy-makers use models to predict disease spread and estimate the effects of various intervention strategies. Effective modeling of targeted intervention strategies requires accurate modeling of individual-level behavior and transmission. However, this greatly increases the computational costs of these agent-based models. In addition, if the models are used as an outbreak progresses, some operational decisions must occur rapidly in order to contain the spread of the disease.
Graphics Processing Units (GPUs) are a type of specialized processor used to drive graphical displays. Many recent devices also allow users to write …

School of Mathematical & Statistical Sciences Faculty Publications
The goal of the current analysis is to examine relationships between smoking status and anxiety and depression among adults of Mexican heritage to inform the development of culturally relevant smoking cessations efforts. Mexican heritage residents (N=1,791) of the city of Brownsville, TX, aged 18 years or older, enrolled in the Cameron County Hispanic Cohort, were selected through two stage cluster sampling of randomly selected census tracts from the first and third quartile of SES using Census 2000. Among current smokers, anxiety and depression scores were highest among women who had not completed high school (p<0.05). Former smoking women, but not men, with at least a high school education and former smoking women born in the United States reported higher levels of anxiety and depression than never smoking women. Negative affective states may represent a greater barrier to smoking cessation among women than men.
Comparison Of Bacterial Inactivation Efficiency And Disinfection Byproduct Formation From Different Disinfection Processes, Binbin Li
All Theses
Formation of disinfection byproducts (DBPs) during Escherichia coli K12 and Pseudomonas aeruginosa PAO1 inactivation under four disinfection processes, including chlorination, chloramination, photo-Fenton reaction, and TiO2/UV photocatalytic inactivation were examined. Factors of pH (5 or 8) and different disinfectant dosages were also investigated in order to balance the risks of DBP formation and bacterial cell viability in the four treatment processes. Chlorination had the fastest bacterial inactivation whereas chloramination, photo-Fenton, and TiO2/UV requiring longer contact times to achieve the same inactivation efficiency. In spite of the lag phase in the treatment processes, both photo-Fenton and TiO2/UV achieved 100% bacteria reduction after …

Folates are tripartite molecules comprising pterin, para-aminobenzoate (PABA), and glutamate moieties, which are essential cofactors involved in DNA and amino acid synthesis. The obligately intracellular Chlamydia species have lost several biosynthetic pathways for essential nutrients which they can obtain from their host but have retained the capacity to synthesize folate. In most bacteria, synthesis of the pterin moiety of folate requires the FolEQBK enzymes, while synthesis of the PABA moiety is carried out by the PabABC enzymes. Bioinformatic analyses reveal that while members of Chlamydia are missing the genes for FolE (GTP cyclohydrolase) and FolQ, which catalyze the initial …
Investigation Into The Control Of Melittin Secondary Structure And Antimicrobial Activity, Zachary B. Molinets
Investigation Into The Control Of Melittin Secondary Structure And Antimicrobial Activity, Zachary B. Molinets
Open Access Theses
Antimicrobial resistance has been an exponentially growing problem since the discovery of antibiotics. Antibiotics have been misused for many years and this misuse has grown into a real problem for the medical community. While there are countless safeguards to prevent infection by a resistant strain of bacteria, there are still many plagued by it and must be treated with sometimes dangerous antibiotics. Melittin, along with many other peptides, contain potent antimicrobial properties, but are also toxic toward enthrocytes. The control of the secondary structure of peptides provides the key to adjusting their activity.

Use Of System Dynamics Modeling To Explicate The Theory-Of-Change Of A Social Marketing Innovation, Brian J. Biroscak
USF Tampa Graduate Theses and Dissertations
Community coalitions are an important part of the public health milieu and thus subject to many of the same external pressures as public health organizations--including changes in required strategic orientation. Many funding agencies have shifted their funding agenda from program development to policy change. Thus, the Florida Prevention Research Center created the Community-Based Prevention Marketing for Policy Development framework to teach community coalitions how to apply social marketing to policy change. The dissertation research reported here was designed to explicate the framework's theory-of-change. The research question was: "What are the linkages and connections between CBPM inputs, activities, immediate outcomes, intermediate …

A Likelihood Approach To Estimate The Number Of Co-Infections, Kristian A. Schneider, Ananias A. Escalante
Harold W. Manter Laboratory of Parasitology: Library Materials
Article abstract:
The number of co-infections of a pathogen (multiplicity of infection or MOI) is a relevant parameter in epidemiology as it relates to transmission intensity. Notably, such quantities can be built into a metric in the context of disease control and prevention. Having applications to malaria in mind, we develop here a maximum-likelihood (ML) framework to estimate the quantities of interest at low computational and no additional costs to study designs or data collection. We show how the ML estimate for the quantities of interest and corresponding confidence-regions are obtained from multiple genetic loci. Assuming specifically that infections are …

Nondestructive acoustics methods can be used to monitor in situ biofilm growth in porous media. In practice, however, acoustic methods remain underutilized due to the lack of models that can translate acoustic data into rock properties in the context of biofilm. In this paper we present mechanistic models of biofilm growth in porous media. The models are used to quantitatively interpret arrival times and amplitudes recorded in the 29 day long Davis et al. (2010) physical scale biostimulation experiment in terms of biofilm morphologies and saturation. The model pivots on addressing the sediment elastic behavior using the lower Hashin-Shtrikman bounds …

Compartmental modeling has been used to model infectious diseases for roughly 100 years. Since 2009, several papers have modeled zombie outbreak using this method with various results. This paper will develop a unique model for the spread of the The Walking Dead zombie virus throughout the contiguous United States. Frequency dependent and density dependent transmission will be discussed, and density dependent transmission will be shown to be the appropriate choice for this model. Constant parameters, such as birth rate, bite rate, death rate, and turning rate will be determined using real-world and fictional data. After developing a basic model, modifications …
Application Of Nuclear Magnetic Resonance Based Metabolomics To Study The Central Metabolism Of Staphylococci, Bo Zhang
Department of Chemistry: Dissertations, Theses, and Student Research
Metabolomics studies the collection of small molecules (metabolites) involved in enzymatically catalyzed reactions, cell signaling and cellular structure. Perturbations in metabolite concentrations have been used to reflect the activity of corresponding enzymes or proteins. Nuclear magnetic resonance (NMR) spectroscopy is a well-known approach for the structure determination of biological macromolecules. Alternatively, NMR has recently been established as a valuable tool of metabolomics, in which NMR spectral signals correlate small molecules with cellular activities. This has been accomplished through the chemometric analysis of high-throughput one dimensional 1H spectra (metabolic fingerprinting) and quantitative metabolite identification based on two dimensional 1H, …
