This study explores the impact of using a diesel, biodiesel, and toluene additive fuel blend in a multi-cylinder direct injection diesel engine, focusing on both performance and emissions characteristics. Biodiesel, made from renewable sources like vegetable oils, is often added to diesel to reduce reliance on fossil fuels and improve combustion due to its higher oxygen content. In this work, a 45% biodiesel blend (B45) led to significant reductions in particulate matter (PM) emissions by up to 40% compared to conventional diesel. Additionally, carbon monoxide (CO) and hydrocarbon (HC) emissions decreased by up to 30% and 25%, respectively, due to …

The field of radiation oncology seeks novel techniques to increase the therapeutic ratio to treat malignancies while minimizing damage and toxicities in healthy tissues. While it was observed that delivering therapeutic doses at high dose rate could elicit lower biological effects over a half-century ago, the field of ultra-high dose rate (UHDR) radiation therapy (colloquially known as FLASH RT) has seen a resurgence in recent years due to an alignment of accelerator capability, dosimetric advances, and improved biotechnological techniques. Obesity is caused by an energy imbalance that increases chronic low-grade inflammation, including macrophage cell infiltration. Adipose tissue macrophages are polarized into pro-inflammatory macrophage type 1 (M1), secrete large amounts of pro-inflammatory cytokines, and activate transcription factors. Yoghurt with probiotics is popular at all ages for its health benefits and must be protected by microencapsulation. The CRISPR (clustered regularly interspaced short palindromic repeats)/Cas (CRISPR-associated protein) programmable nuclease system continues to evolve, with in vivo therapeutic gene editing increasingly applied in clinical settings. However, off-target effects remain a significant challenge, hindering its broader clinical application. To enhance the development of gene-editing therapies and the accuracy of prediction algorithms, we developed CRISPRoffT (https://ccsm.uth.edu/CRISPRoffT/). Users can access a comprehensive repository of off-target regions predicted and validated by a diverse range of technologies across various cell lines, Cas enzyme variants, engineered sgRNAs (single guide RNAs) and CRISPR editing systems. In nuclear medicine, theranostics utilizes radionuclide pairs for diagnosis and therapy. Radioscandium isotopes (43Sc, 44Sc, and 47Sc) offer a promising alternative to the 68Ga/177Lu theranostic pair due to their identical radiolabeling conditions, complexes and in vivo pharmacokinetics. 43Sc (t1/2=3.89h, β+=88%) and 44Sc (t1/2 =3.97h. β+=94.3%) are suitable for Positron Emission Tomography (PET) imaging whereas 47Sc (t1/2=3.3d. Eβ-ave=162keV) is suitable for targeted radiotherapy and Single Photon Emission Computed Tomography (SPECT). These isotopes require production routes that lead to high radionuclidic purity and radiochemistry development for incorporation into radiopharmaceuticals. Also, this project addresses the growing challenges posed from Generative AI. In particular, issues surrounding Deepfake Phishing and Polymorphic Malware are discussed. The spatiotemporal variation of vegetation cover in the mining areas of YSR Kadapa district, Andhra Pradesh, using Remote Sensing and GIS techniques. By focusing on the years 2014, 2018, and 2023, the analysis provides insights into changes observed in water bodies, bare soil, sparse vegetation, moderate vegetation, and dense vegetation. The results reveal dynamic trends in land cover categories, highlighting the environmental impact of mining activities in the region. A significant decline in water bodies is observed, with the area reducing from 3.48 km2 in 2014 to 1.91km2 in 2023. This decrease raises concerns about the potential degradation …

Eastern Diamondback Rattlesnakes (Crotalus adamanteus; EDBs) are a declining species endemic to the Southeastern Coastal Plain. Reproduction is costly for females, which are ovoviviparous, slow to mature, reproduce infrequently, and are often emaciated after parturition. Yet, we have a limited understanding of the trade-offs associated with their reproductive biology. Since the advent of the modern automobile, manufacturers have provided means of tracking various critical data points associated with automobile operation, with the most prominent and standardized method being the instrument cluster. These data points include, but are not limited to, automobile speed, engine speed, fuel level, oil temperature, radiator (water) temperature, and battery charge. While this data is updated in real-time as the automobile is running, traditional instrument clusters cannot be modified or adjusted to the automobile driver’s needs, unless extensive after-market modifications are made. The Australian plague locust (Chortoicetes terminifera) is an agricultural and ecological pest that causes tens of millions of dollars in crop damage each year. In this thesis, we develop mathematical models of hopper bands, dense formations of juvenile locusts that move across a vegetated field and destroy plants in their path. We develop agent-based and PDE models of hopper bands moving through vegetation to examine how recently discovered behavior where locusts slow down to avoid collisions with other locusts may influence the shape, speed, and destructiveness of hopper bands. Harmful Algal Blooms (HABs) threaten aquatic ecosystems, local economies, and water quality. While extensively studied in lakes (lentic systems), their impact on flowing waters (lotic systems) remains underexplored. This study examined algal drivers in both ecosystems within the same watershed, focusing on Beech Fork Lake (lentic) and Twelve Pole Creek (lotic). Key environmental factors, including pH, turbidity, dissolved oxygen, and nutrient levels, were measured using in situ sensors and laboratory analysis. Water samples were analyzed for nitrogen, phosphorus, and algal biomass for comparison to environmental conditions. Phytoplankton was preserved for identification in future research. The oil sands in Alberta, Canada are being destroyed by oil extraction because remediation methods have proven largely ineffective. One toxic byproduct of oil extraction is a family of cyclical surfactant organic acids known as naphthenic acids (NAs). They are toxic to aquatic life, including fish, frogs, and aquatic birds. It is suspected that NAs bind to humic acid, a major organic component of soil, in dissolved organic matter in natural water, making them difficult to remove by conventional remediation methods. Individual driving behavior is a pivotal element that shapes the overall traffic dynamics in a city. In this work, we study and analyze the complex web of relationships between individual driving behaviors and their impact on the overall traffic dynamics of a smart city with two primary objectives: first, understanding the spatial interaction between individual vehicles and their impact on each other, and second, finding anomalous driving behaviors, which lead to congestion and traffic incidents. Amphibian populations have been declining globally since the 1950s. Many species that have experienced declines are viewed as 'environmental indicators' that reveal habitat degradation. Declines can be attributed to several factors, including chemicals used in agriculture, such as Roundup and Atrazine. Agrochemicals contaminate water bodies through runoff or leaching and cause alterations to aquatic ecosystems. This study evaluated the effects of herbicide usage on two wild anuran populations using wood frogs (Boreorana sylvatica) and Cope’s gray tree frogs (Dryophytes chrysoscelis) as a model. Although North America contains the largest diversity of freshwater mussels in the world, only ~24% of species in the United States and Canada have stable populations. The oil sand regions in Alberta, Canada have proven to be costly to the environment despite its economic benefits. The toxins produced during oil processing are sent to tailing ponds to be recycled, though often leaking into nearby waterways and soil. Naphthenic acids, NAs, are one of the most common toxins produced and have shown to be toxic to aquatic life, animals, and humans. This thesis aims to elucidate the molecular mechanisms by which nucleotide binding dictates the intrinsic dynamics and to understand the functional conformational changes of Human Dynamin-1 during the GTP hydrolysis cycle through the use of Elastic Network Models (ENM). A comprehensive understanding of this process is crucial given dynamin's role in clathrin-mediated endocytosis (CME). The research utilized both experimentally derived crystal structures and AlphaFold-predicted structures of dynamin, specifically examining the GTPase and Bundle Signaling Element (BSE) domains across nucleotide-free, GTP-bound, GDP-bound, and transition states. Kentucky’s renewable energy landscape beats with a distinct rhythm shaped by ever-changing variations in sunlight, wind, rainfall, and the ever-modernizing grid that connects them. This paper analyzes minute-by-minute performance data from seven renewable and storage assets owned and operated by the PPL Corporation in Kentucky, including hydroelectric, solar, wind, and lithium-ion battery systems. Using a full year of synchronized, high-resolution data from multiple sites in the Commonwealth, the study examines daily and seasonal capacity factor trends, explores correlations among generation types, and evaluates their alignment with utility load profiles. When making decisions, it is important to take into account high-impact low-probability events. For such events, traditional probability-based approach -- which considers the product of the probability p that this event happens and the probability P that a randomly selected building will be destroyed -- often underestimates risks. Available data has lead to an empirical table that provides a more adequate risk estimate. Most of the entries in this table correspond to the fuzzy-like formula min(p,P). This paper explains this empirical result. Specifically, it explains both the effectiveness of the min formula -- and also explains deviations from this formula.

Human development continues to alter natural landscapes, often with significant consequences for wildlife populations. Among the most sensitive taxa to such disturbances are bats, which serve vital ecological roles as insect predators, pollinators, and seed dispersers. Because bats depend on specific roosting and foraging habitats, even minor environmental changes can strongly influence their activity and diversity. A new road is proposed through an area of Warner Parks, Nashville, Tennessee which is otherwise untouched. This may affect the biodiversity and the populations of bats as a result.
