Oxidative Stress In Health And Disease, V. Prakash Reddy Nov 2023

Oxidative Stress In Health And Disease, V. Prakash Reddy

Chemistry Faculty Research & Creative Works

Oxidative stress, resulting from the excessive intracellular accumulation of reactive oxygen species (ROS), reactive nitrogen species (RNS), and other free radical species, contributes to the onset and progression of various diseases, including diabetes, obesity, diabetic nephropathy, diabetic neuropathy, and neurological diseases, such as Alzheimer's disease (AD), amyotrophic lateral sclerosis (ALS), and Parkinson's disease (PD). Oxidative stress is also implicated in cardiovascular disease and cancer. Exacerbated oxidative stress leads to the accelerated formation of advanced glycation end products (AGEs), a complex mixture of crosslinked proteins and protein modifications. Relatively high levels of AGEs are generated in diabetes, obesity, AD, and other …

Which Activation Function Works Best For Training Artificial Pancreas: Empirical Fact And Its Theoretical Explanation, Lehel Dénes-Fazakas, Lásló Szilágyi, György Eigner, Olga Kosheleva, Martine Ceberio, Vladik Kreinovich

Departmental Technical Reports (CS)

One of the most effective ways to help patients at the dangerous levels of diabetes is an artificial pancreas, a device that constantly monitors the patient's blood sugar level and injects insulin based on this level. Patient's reaction to insulin is highly individualized, so the artificial pancreas needs to be trained on each patient. It turns out that the best training results are attained when instead of the usual ReLU neurons, we use their minor modification known as Exponential Linear Units (ELU). In this paper, we provide a theoretical explanation for the empirically observed effectiveness of ELUs.

Therefore, this study analyzed information, motivation, and behavioral skills associated with diabetes self-management. Data were collected in the Special Capital Region of Jakarta, with 277 diabetic patients selected using a questionnaire by a systematic random sampling method. The analyzed variables were information (with indicator variables of information on physical activity, nutritional intake, drug consumption, and blood sugar monitoring); sociodemographic (age, sex, occupation, education level, and duration …

Investigating The Interactions Of The Iron-Sulfur Mitochondrial Protein, Mitoneet With Its Binding Partners, Ebenezer Osei Newton Jan 2023

Investigating The Interactions Of The Iron-Sulfur Mitochondrial Protein, Mitoneet With Its Binding Partners, Ebenezer Osei Newton

Graduate Theses, Dissertations, and Problem Reports

MitoNEET belongs to the CDGSH Iron-Sulfur Domain (CISD)-gene family of proteins and is a [2Fe-2S] cluster-containing protein found on the outer membrane of mitochondria. The specific functions of mitoNEET/CISD1 remain to be fully elucidated, but the protein is involved in regulating mitochondrial bioenergetics in several metabolic diseases. Unfortunately, drug discovery efforts targeting mitoNEET to improve metabolic disorders are hampered by the lack of ligand-binding assays for this mitochondrial protein. We have developed a protocol amenable for high-throughput screening (HTS) assay, by modifying an ATP fluorescence polarization method to facilitate drug discovery targeting mitoNEET. Based on our observation that adenosine triphosphate …

Research outputs 2022 to 2026

Higher intakes of cruciferous and allium vegetables are associated with a lower risk of cardiometabolic-related outcomes in observational studies. Whilst acknowledging the many healthy compounds within these vegetables, animal studies indicate that some of these beneficial effects may be partially mediated by S-methyl cysteine sulfoxide (SMCSO), a sulfur-rich, non-protein, amino acid found almost exclusively within cruciferous and alliums. This scoping review explores evidence for SMCSO, its potential roles in human health and possible mechanistic action. After systematically searching several databases (EMBASE, MEDLINE, SCOPUS, CINAHL Plus Full Text, Agricultural Science), we identified 21 original research articles meeting our inclusion criteria. These …

Predicting Insulin Pump Therapy Settings, Riccardo L. Ferraro, David Grijalva, Alex Trahan

SMU Data Science Review

Millions of people live with diabetes worldwide [7]. To mitigate some of the many symptoms associated with diabetes, an estimated 350,000 people in the United States rely on insulin pumps [17]. For many of these people, how effectively their insulin pump performs is the difference between sleeping through the night and a life threatening emergency treatment at a hospital. Three programmed insulin pump therapy settings governing effective insulin pump function are: Basal Rate (BR), Insulin Sensitivity Factor (ISF), and Carbohydrate Ratio (ICR). For many people using insulin pumps, these therapy settings are often not correct, given their physiological needs. While …

Advanced Glycation End Products In Health And Disease, V. Prakash Reddy, Puspa Aryal, Emmanuel K. Darkwah

Chemistry Faculty Research & Creative Works

Advanced glycation end products (AGEs), formed through the nonenzymatic reaction of reducing sugars with the side-chain amino groups of lysine or arginine of proteins, followed by further glycoxidation reactions under oxidative stress conditions, are involved in the onset and exacerbation of a variety of diseases, including diabetes, atherosclerosis, and Alzheimer's disease (AD) as well as in the secondary stages of traumatic brain injury (TBI). AGEs, in the form of intra- and interprotein crosslinks, deactivate various enzymes, exacerbating disease progression. The interactions of AGEs with the receptors for the AGEs (RAGE) also result in further downstream inflammatory cascade events. The overexpression …

Diabetes is a chronic disease that effects 10 percent of the world’s population and causes more than 1.5 million deaths a year and billions of dollars in associated health care cost. It can lead to very serious complications such as renal failure, liver cirrhosis, heart attack, and vision loss. The most common type of diabetes is type 2 diabetes. Type 2 diabetes arises when blood glucose levels remain chronically high due to insulin resistance. The reason for this elevation is due to the failure of insulin to allow tissues to uptake glucose causing problems in subsequent metabolic pathways. Over the …

Why We Need A Ruminant Revolution: Combating Malnutrition And Metabolic Illnesses To Enable Sustainable Development, Peter J. Ballerstedt, David B. Hannaway, T. D. Noakes

IGC Proceedings (1997-2023)

Animal source foods (ASF) are essential for proper human development and function. Livestock in general, and ruminants in particular, are essential components of our sustainable global food systems. Of significant worldwide impact, diets with higher-than-recommended levels of ASF can correct the symptoms of metabolic illnesses, offering hope in arresting the current worldwide epidemic of diabetes and other metabolic diseases. Most dietary policy and recommendations are based on the ill-founded belief that plant-based, high-carbohydrate diets are “healthy.” High-quality scientific evidence does not support the belief that vegetarian diets are healthier than omnivorous or animal-based diets. A Therapeutic Carbohydrate Reduction (TCR) lifestyle …

Dissertations (1934 -)

Diabetic Retinopathy (DR) is one major complication of diabetes and is the leading cause of blindness worldwide. Progression of DR and complete vision loss can be prevented by keeping diabetes in control and by early diagnosis through annual eye screenings. However, cost, healthcare disparities, cultural limitations, lack of motivation, etc., are the main barriers against regular screening, especially for a few ethnically and racially minority communities. On the other hand, to well-manage and control diabetes, the diabetic population needs to be physically active and keep their weight healthy. From the perspective of Behavioral Science, Some self-management techniques based on motivational …

Building An Ins-1 Cdna Library For A Genome-Wide Crispr-Cas9 Screen, Idongesit Ekpo

Undergraduate Honors Theses

By the year 2040, an estimated 642 million people are expected to have diabetes globally. Diabetes results from an elevation of metabolic stressors, such as glucotoxicity, lipotoxicity, oxidative stress and apoptosis. In type 2 diabetes, these stressful conditions contribute to the malfunction and loss of functional insulin-producing β-cells. Current treatment methods for diabetes include insulin therapy, islet transplant and anti-diabetes medication. These treatments are not curative and ignore other factors that contribute to the pathogenesis of diabetes beyond insulin resistance and islet β-cell failure. Previous research on β-cells has focused on ways to replace functional β-cell mass, trigger β-cell proliferation, …
