Total Syntheses Of Hosieines A-C, Jiayang Zhang, Xu Yan, Qing-Bao Zhang, Fang Wang, Bin Yang, Yang Yang

Children’s Nutrition Research Center Staff Publications

The collective total syntheses of (±)-hosieines A-C with a cage-like tetracyclic framework have been realized, which includes the first syntheses of hosieines B-C. The key strategy of the synthesis employs a one-pot domino reaction that involves Cu-catalyzed [3+2] cycloaddition, 1,6-enone formation, and 1,6-aza-Michael addition forming the 5/6/6-aza-tricyclic skeleton. Other salient synthetic tactics comprise a challenging double bond migration and a 1,4-aza-Michael addition reaction to afford the tetracyclic framework.

Metformin is a widely prescribed anti-diabetic medicine that also reduces body weight. There is ongoing debate about the mechanisms that mediate metformin’s effects on energy balance. Here, we show that metformin is a powerful pharmacological inducer of the anorexigenic metabolite N-lactoyl-phenylalanine (Lac-Phe) in cells, in mice and two independent human cohorts. Metformin drives Lac-Phe biosynthesis through the inhibition of complex I, increased glycolytic flux and intracellular lactate mass action. Intestinal epithelial CNDP2+ cells, not macrophages, are the principal in vivo source of basal and metformin-inducible Lac-Phe. Genetic ablation of Lac-Phe biosynthesis in male mice renders animals resistant to the …

Brown adipose tissue (BAT)-mediated thermogenesis plays an important role in the regulation of metabolism, and its morphology and function can be greatly impacted by environmental stimuli in mice and humans. Currently, murine interscapular BAT (iBAT), which is located between two scapulae in the upper dorsal flank of mice, is the main BAT depot used by research laboratories to study BAT function. Recently, a few previously unknown BAT depots were identified in mice, including one analogous to human supraclavicular brown adipose tissue. Unlike iBAT, murine supraclavicular brown adipose tissue (scBAT) is situated in the intermediate layer of the neck and thus …

Nkx6.1 And Stx1a As Binding Partners In Pancreatic Beta Cells, Jakob T. Lenker, Jeff S. Tessem, Connor C. Littlefield

Library/Life Sciences Undergraduate Poster Competition 2024

Diabetes is characterized by a loss in beta cell function within the pancreas and the subsequent inability to produce sufficient insulin to regulate blood glucose. While current diabetes treatments focus on delivering pharmaceutical insulin to diabetic individuals, such treatments are temporary solutions and do not address the root of the issue. Instead, our research focuses on potential mechanisms for inducing greater insulin secretion within the pancreas of the individual. NK6 Homeobox 1 (Nkx6.1) is a major transcription factor in beta cells and its overexpression in beta cells is associated with higher insulin secretion. Previous research indicates that Syntaxin 1A (Stx1A) …

Journal of Extension

About 20% of the U.S. population identifies as Latine. To engage these audiences, many Extension nutrition education programs have translated curricula and other resources into Spanish. Culturally responsive programs need to move beyond translation and address critical issues and needs specific to Latine audiences. We conducted focus groups with 118 foreign-born Latine adults across one state. Participants were interested in nutrition education programs involving the entire family including children offered by bilingual educators. They also wanted programs that focused on healthy meals that included foods from their culture of origin with common US foods.

Consumption of probiotics and/or yogurt could be a solution for restoring the balance of the gut microbiota. This study examined associations of regular intake of probiotic supplements or yogurt with the gut microbiota among a diverse population of older adults (N=1,861; 60–72 years). Fecal microbial composition was obtained from 16S rRNA gene sequencing (V1-V3 region). General Linear Models were used to estimate the associations of probiotic supplement or yogurt intake with microbiome measures adjusting for covariates. Compared to non-yogurt consumers (N=1,023), regular yogurt consumers (≥once/week, N=818) had greater Streptococcus (β=0.29, P=0.0003) and lower Odoribacter (β=−0.33, P<0.0001) abundance. The directions of the above associations were consistent across the five ethnic groups but stronger among Japanese Americans (Streptococcus: β=0.56, P=0.0009; Odoribacter: …

Effects Of Black Raspberry, Blueberry, And Blackberry Volatile Extracts On Intestinal Barrier Dysfunction And Inflammation In A Caco-2/Raw264.7 Cells Co-Culture Model, Inah Gu, Cindi Brownmiller, Luke Howard, Sun-Ok Lee

Food Science Faculty Publications and Presentations

Berry volatiles are responsible for the aroma of berries. However, their impacts on intestinal inflammation are unclear. Thus, this study investigated the effects of black raspberry, blueberry, and blackberry volatile extracts (BVEs) on intestinal barrier dysfunction and inflammation using a Caco-2/RAW264.7 cells co-culture model. BVE treatments significantly enhanced integrity and decreased permeability in LPS-damaged epithelial monolayers with increased tight junctions. Blueberry and blackberry volatile extract treatments significantly inhibited the LPS-induced overproduction of NO, IL-6, and TNF-α (p < 0.05). BVEs also suppressed the LPS-induced phosphorylation of p65, IKKα, and IκBα. These findings revealed that volatile extracts from black raspberries, blueberries, and blackberries improve intestinal barrier dysfunction by enhancing intestinal epithelial monolayer integrity and exhibit intestinal anti-inflammatory effects by suppressing pro-inflammatory mediator and cytokines through modulating NF-κB signaling pathway. It suggests that the three berry volatile extracts have the potential as functional food components with gastrointestinal protective effects with anti-inflammatory properties.

BACKGROUND: Prostate cancer (PCa) continues to be one of the leading causes of cancer deaths in men. While androgen deprivation therapy is initially effective, castration-resistant PCa (CRPC) often recurs and has limited treatment options. Our previous study identified glutamine metabolism to be critical for CRPC growth. The glutamine antagonist 6-diazo-5-oxo-l-norleucine (DON) blocks both carbon and nitrogen pathways but has dose-limiting toxicity. The prodrug DRP-104 is expected to be preferentially converted to DON in tumor cells to inhibit glutamine utilization with minimal toxicity. However, CRPC cells' susceptibility to DRP-104 remains unclear.

METHODS: Human PCa cell lines (LNCaP, LAPC4, C4-2/MDVR, PC-3, 22RV1, …
