Inactivation Of Brain Hexokinase By An Adenosine 5'-Triphosphate Analog, Subbarao Bondada, Umakant W. Kenkare

An analog of ATP, 6-mercapto-9+nribofuranosylpurine 5’-triphosphate (SH-TP), functions as a phosphoryl donor for the reaction catalyzed by hexokinase. Incubation of the enzyme with millimolar concentrations of this reagent led to its rapid inactivation. However, the corresponding monophosphate derivative of this reagent was much less effective. A plot of the initial rate of inactivation versus the concentration of SH-TP exhibited saturation, suggesting the formation of a reversible complex between the enzyme and SH-TP prior to the inactivation reaction. The dissociation constant of this enzyme-inhibitor complex was the same as the K, of this reagent with respect to hexokinase in the phosphoryl …

Duck plague is a hemorrhagic, necrotic disease found exclusively in the order Anseriformes, family Anatidae (ducks, geese, swans). The etiologic agent has been reported to be a herpes-like virus. The Holland strain of duck plague was found to induce cytopathic effects in cell culture (perinuclear vacuolation followed by lysis). A simple reproducible plaque assay for quantitating the virus was developed. Under conditions of the plaque assay, one-hit kinetics were observed. The host range of duck plague virus in cell culture was found to include the order Galliformes, family Phasianidae (pheasants, quails and partridges). Reaction Of Brain Hexokinase With Tetranitromethane: Oxidation Of Essential Thiol Groups, Subbarao Bondada, Umakant W. Kenkare

Microbiology, Immunology, and Molecular Genetics Faculty Publications

Treatment of bovine brain mitochondrial hexokinase with a fivefold molar excess of tetranitromethane (TNM) at pH 8.0 results in a complete loss of activity of the enzyme. Spectral measurements and amino acid analysis showed that only cysteinyl residues were modified by TNM under these conditions. The products of the reaction of TNM with the enzyme were cysteic acid and disulfides as found by amino acid analysis of the carboxymethylated hexokinase. Failure of 2-mercaptoethanol to reverse the effect of TNM suggested that the thiol group that is oxidized to cysteic acid is the one essential for enzyme activity. Substrates glucose and …

Agatha Rollins’ 1977 master’s project, Electronic Cell Pattern Recognition Manual, explores the feasibility of using the Technicon Hemalog "D" Differential Analyzer as a diagnostic tool for hematologic diseases. The analyzer identifies white blood cell types through electronic signals based on their scatter and absorbance patterns in the peroxidase channel. Rollins hypothesized that similar diseases would yield consistent, recognizable electronic patterns, enabling identification without manual microscopy.

The project involved analyzing patients with various conditions—including chronic lymphocytic leukemia, myeloperoxidase deficiency, eosinophilia, and several leukemias—by comparing manual differential counts with Hemalog “D” results. It is difficult to pick up a magazine, medical or otherwise, that does not include an article about the Chinese method of diagnosis and treatment known as acupuncture. This ancient technique, about which there is so much current interest, is reputed to have originated nearly 30 centuries ago when a Chinese warrior emperor made the observation that certain arrow wounds caused pain to disappear in remote parts of the body.
