Characterization Of Monoclonal Antibodies Against Shiga-Like Toxin From Escherichia Coli, Nancy A. Strockbine, Lilian R. M. Marques, Randall K. Holmes, Alison D. O'Brien

Three monoclonal antibodies, designated MAb 16E6, MAb 13C4, and MAb 19G8, were produced which recognize Shiga-like toxin (SLT) from Escherichia coli. All three monoclonal antibodies neutralized the cytotoxicity of E. coli SLT and were able to immunoprecipitate intact labeled toxin with Staphylococcus aureus protein A. The three antibodies were of the Gl heavy and kappa light chain classes. MAb 16E6 bound to the B subunit of SLT in Western blots and also neutralized the lethality of the toxin for mice and the enterotoxicity of the toxin in ligated rabbit ileal loops. The central issues raised here are whether the two frozen embryos have a legal right to 1) live and be implanted in a surrogate mother, and, when and if they are born, 2) assert inheritance rights in the Rios' estate. Equally important is the question of the extent to which research into the new reproductive technologies should be allowed or restricted.

Protein Concentration Elevations In Mouse Lungs Following Sudden Transient Cephalad (+Gz) Acceleration, Charles J. Gutierrez
Retrospective Theses and Dissertations
Laboratory and feral lineages of mice were subjected to cephalad (+GZ) accelerations, for 1.8 seconds, aboard a solid fuel rocket. Spectrophotometric analysis of bronchoalveolar lavage retrieved post launch revealed significant (p < .001) elevations of protein in the lungs of experimental mice. Sudden transient imposition of a mean +GZ acceleration of 6.22 ± .47 (SD) G, at lift-off, may have induced hypervolemia of basilar pulmonary microvasculature with concomitant migration of fluid and protein from intravascular to juxta-alveolar perivascular compartments. Exudates may have entered bronchiolar airways subsequently gravitating toward alveoli.

The Role Of The Insulin Receptor In Skeletal Muscle Insulin Resistance: Evaluation Of Two Animal Models, Charles F. Burant
MUSC Theses and Dissertations
The role of the insulin receptor in muscle insulin resistance was studied. Denervation of skeletal muscle resulted in the rapid appearance of a decrease in basal and insulin stimulated glucose transport, glycogen synthesis and a defect in the activation of glycogen synthase which was independent of levels of glycogen in the muscle. The effect was seen in both predominantly red and predominantly white muscle. There was no change in the denervated muscles' sensitivity to epinephrine-mediated decrease in glycogen synthesis or glycogen content, nor was there a major change in the insulin binding capacity of denervated muscles. The primary objective of the research was to investigate the pharmacodynamics of digoxin in dogs. Initially an assay specific for digoxin in the presence of its major metabolites, viz., digoxigenin, digoxigenin mono-digitoxoside, digoxigenin bis-digitoxoside and dihydrodigoxin was developed using HPLC-RIA. Methodology for non-invasive measurement of left ventricular ejection time (LVET) and other systolic time intervals (STI) in beagle dogs were developed. This involved surgery for exteriorization of the carotid artery in the dogs and subsequent measurements of LVET and STI after recovery. Isolates of the most common 0 serogroups of enteropathogenic Escherichia coli (EPEC) associated with infant diarrhea (designated class I) adhere to Hep-2 cells; the genes for this adhesin, termed EPEC adherence factor (EAF), are located on plasmids 50-70 MDa in size. Volunteers ingested 1010 organisms of an 0127:H6 Hep-2-adhesive class I strain (E2348/69) or its plasmid-minus, nonadhesive derivative. Diarrhea occurred in nine of 10 volunteers who ingested the parent strain (mean, 1,178 ml) but in only two of nine who took the plasmid-minus variant (mean, 433 ml; P < .006). All volunteers ill from strain E2348/69 mounted serum IgA and IgG responses to a 94-kDa plasmid-associated outer membrane protein of E2348/69; this protein was found in other class I EPEC but not in enterotoxigenic or meningitic strains. The 50-70-MDa EAF plasmid seems neces- sary for full expression of pathogenicity in EPEC that exhibit Hep-2 adhesiveness. EPEC isolates of certain other, less common, 0 serogroups (044, 086, and 0114) are rarely Hep-2 adhesive. These EPEC, designated class II, possess distinct 50-70 MDa plasmids lacking EAF genes. A number of life-threatening diseases, such as kidney failure, diabetes, and certain kinds of coronary heart disease, can be cured by organ transplantation. However, despite decades of research, graft rejection remains a very real threat to the organ transplant recipient. In attempting to develop methods that interfere with the graft rejection process, scientists have uncovered a remarkably complex system of cellular interactions that allows the total destruction of a transplanted organ while leaving the recipient's own organs untouched. This ability to distinguish self from non-self is achieved through intercellular communication involving cell-to-cell contact and the release of a number of …

Partial Purification And Characterization Of F₄₂₀-Dependent Nadp Reductase From Methanobrevibacter Smithii Strain De1, Scott D. Sheridan

The F420-dependent NADP reductase of Methanobrevibacter smithii has been partially purified employing a combination of affinity chromatography with Blue Sepharose (Cl-6B) and molecular sieve chromatography with Sephacryl S-200, The enzyme, which requires reduced F420 as an electron donor, has been purified over 145 fold with a recovery of 6%. A molecular weight of 120,00 for the native enzyme was determined by Sephacryl S-200 chromatography. A subunit molecular weight of 28,200 was determined by SDS-PAGE, indicating that the native enzyme is a tetramer. The optimal temperature for enzymatic activity was found to be 45°C, with a pH optimum …
