Purification Of Maturation-Promoting Factor, An Intracellular Regulator Of Early Mitotic Events, Manfred J. Lohka, Marianne K. Hayes, James L. Maller

Maturation-promoting factor causes germinal vesicle breakdown when injected into Xenopus oocytes and can induce metaphase in a cell-free system. The cell-free assay was used to monitor maturation-promoting factor during its purification from unfertilized Xenopus eggs. Ammonium sulfate precipitation and six chromatographic procedures resulted in a preparation purified >3000-fold that could induce germinal vesicle breakdown within 2 hr when injected into cycloheximide- treated oocytes. Proteins of 45 kDa and 32 kDa were correlated with fractions of highest activity in both assays. These fractions contained a protein kinase activity able to phosphorylate the endogenous 45-kDa protein, as well as histone Hi, phosphatase …

Sulfiting agents, including sodium and potassium bisulfite, sodium and potassium metabisulfite, sodium sulfite, and sulfur dioxide, have enjoyed widespread use as food and drug ingredients. The oral ingestion of these sulfiting agents is now known to trigger asthma in a small subset of the asthmatic population. The best evidence suggests that perhaps 150,000 to 200,000 individuals in the United States may be sulfite sensitive. Although the mechanism of sulfite-induced asthma remains unknown, several possibilities have been considered, including inhalation of sulfur dioxide (SO2) while swallowing, an IgE-mediated reaction, and a deficiency of sulfite oxidase leading to impaired sulfite …

The purpose of the present study was to test the use of visually displayed EMG biofeedback as a means for training hearing impaired adolescents to reduce anterior temporalis or frontalis muscle tension . Five male and four female hearing impaired students between the ages of 13 and 15 were chosen from the California School for the Deaf, Fremont, CA to serve as participants. Each participant was randomly assigned to either an experimental or control condition. Participants in the experimental groups were given five 15 minute EMG biofeedback training sessions. Dimethylpyrimidylornithyl-N-trifluoroethyl-O-TBDMS (DMPO-N-TFE-O-TBDMS) polyaminoalcohol derivatives of arginyl peptides were evaluated for their utility in peptide analysis by capillary gas chromatography-mass spectrometry (GC-MS). These derivatives were hydrolytically stable and sufficiently volatile for capillary GC-MS analysis of low nanomole amounts of peptides as large as RPPGF. Complete amino acid sequence information was generated under electron impact ionization conditions, including several ions diagnostic for arginine residues, with molecular weight information imparted by the [M-15]+ and [M-57]+ pair of ions characteristic of TBDMS derivatives. Analysis of the sequential intermediate derivatives by FAB-MS facilitated optimization of problematic derivatization steps. As little as 20 picomoles of peptide …

There are inconsistent guidelines for, and consequently, inconsistent practice in the screening of hospitalized adults for fever. Using the predictive knowledge of circadian thermal rhythms, the purposes of this study were to identify (1) the hour of the day in which most fevers occurred and (2) the necessary frequency of measurement to detect fever. The sample consisted of 35 subjects, ages 22-62. Temperatures were measured at 6 p.m., 10 p.m., 6 a.m., 10 a.m., 2 p.m., and 6 p.m. for 24 hours. This study used an experimental design to investigate whether the timing of a child's admission physical assessment, either before or after an interview with the child's parent(s), had an effect on the upset and cooperation behaviors exhibited by the child during the physical exam.; Thirty-two children, age 12 to 36 months, who were scheduled for elective outpatient surgery were randomly assigned to control and experimental groups.; The hypothesis of the study was that children who were examined after the interview would exhibit less upset behaviors and be more cooperative with the examining nurse. The purpose of this research project was to evaluate the role of the community pharmacist in screening, identifying, and referring ambulatory patients with high total blood cholesterol (TBC) in a community pharmacy. Fifty seven patients, out of 241 initially screened individuals, met the study inclusion criteria and were accepted into this study. Of these 57 patients, 51 patients completed the six month study period. The normal population group consisted of 164 participants with TBC < 200 mgjdL at the initial cholesterol testing (visit 1). The drop out group represented six patients who failed to continue attending the two follow up tests (visit 2 and 3). For screening purposes, a non-fasting whole blood sample was used to measure TBC using the Boehringer Mannheim Reflotron analyzer.

Polyamine And Acetylpolyamine Levels During Phenylhydrazine-Induced Erythropoiesis In Mouse Spleen, Othman A. Alshabanah

The phenylhydrazine-induced erythropoietic mouse spleen is used as a model system to demonstrate the relationship between tissue growth and polyamine metabolism. Phenylhydrazine produced significant changes in spleen weights, hematocrits and reticulocyte counts in Swiss-Webster mice. The average spleen weight went up from a control of 155 mg to 875 mg at 96 hours after phenylhydrazine administration, while a 49% reduction in the value of hematocrit was observed at 72 hours. Reticulocyte counts in peripheral blood went from 0.8 to 58% at 168 hours after treatment with phenylhydrazine. Phenylhydrazine at a dose of 40 mg/kg produced significant increases in the levels …

We studied the efficiency of a standard-kit preparation using 1 mg 111In-labeled 96.5 monoclonal antibody in combination with 19 mg of unlabeled antibody in the diagnostic imaging of 27 patients with documented metastatic melanoma. Twenty-three of 26 patients (88%) demonstrated immunoscintigraphic localization of tumor. Of 104 metastatic sites previously documented by conventional studies, 62 (60%) were identified by immunoscintigraphy. A total of 77 sites demonstrated localization of radiolabeled antibody. Fifty-four (70%) corresponded to known sites of disease; eight sites (10%) were "discovered" by immunoscintigraphy and subsequently confirmed by conventional studies; 15 imaged sites (20%) could not be confirmed by conventional …

Since the beginnings of the Medical College of Virginia (MCV), the School of Pharmacy at Virginia Commonwealth University has been a leader in pharmacy education. This book traces the history of the School of Pharmacy up to the time of its publication in 1988. Glucocorticoid-Responsive Hepatic Cytochromes P-450 In The Rat: Multiplicity And Differential Regulation, Keith Alan Hostetler

The multiplicity of glucocorticoid-responsive hepatic cytochromes P-450 in the rat was investigated by developing a series of monoclonal antibody probes directed against purified steroid-inducible isozymes from the rat (P-450p) and man (HLp). Two antibodies, termed 1G8 and 13-7-10, showed unique specificity for purified cytochromes P-450 upon immunoblot analyses. These antibodies were further characterized in terms of their reactivity toward proteins in liver microsomes from untreated and xenobiotic-treated rats. Three P-450p-related proteins were identified using 1G8 and 13-7-10 to monitor the expression of P-450p family subtypes. In untreated rats, members of this family exhibited sex-specific expression. Nine brains taken from chicks two weeks of age were used for the development of this atlas. Each chick was first anesthetized with an intravenous (IV) injection of Chloropent2 (1.8 ml/kg). Chicks were then perfused via the heart with 90 ml physiological saline followed by 90 ml Heidenhain's3 solution. Each head was then positioned in a stereotaxic instrument as described in the previous section and three stainless steel (SS) insect pins (#2) were implanted in each brain at known coordinates. An Identified Histaminergic Neuron Can Modulate The Outputs Of Buccal-Cerebral Interneurons In Aplysia Via Presynaptic Inhibition, Hillel J. Chiel

We have identified 2 buccal-cerebral interneurons (BCIs), B17 and B18, that appear to be involved in the coordination of feeding behavior in Aplysia. The BCIs have their cell bodies in the buccal ganglion, but send axons to the cerebral ganglion via the cerebral-buccal connectives. The BCIs appear to make monosynaptic connections with neurons in the cerebral ganglion that modulate extrinsic muscles involved in feeding behavior. B17 and B18 are activated antiphasically during a motor program induced by stimulating the esophageal nerve and appear to 'read out' different phases of the buccal program to different cells in the cerebral ganglion.
