Purification And Characterization Of Two Oxidoreductases Involved In Bile Acid Modification By The Intestinal Anaerobe Eubacterium Sp. Vpi 12708, Clifton Franklund

Two enzymes, a 7α-hydroxysteroid dehydrogenase (7α-HSDH) and an NADH-dependent flavin oxidoreductase (NADH:FOR), have been purified to apparent electrophoretic homogeneity from the intestinal anaerobe Eubacterium sp. VPI 12708. Using a protocol consisting of four chromatographic separations, the 7α-HSDH was purified by a factor of over 1200-fold with more than a 30% final recovery. Subunit molecular mass was estimated to be 32 Kdal by SDS-PAGE, while native molecular mass estimates from gel filtration were 124 Kdal. The purified 7α-HSDH was able to utilize a variety of bile acids containing an unhindered 7α-hydroxy moiety as substrates, existing either as free acids or glycine …

Reports from the Aga Khan University indicate that 58% of 400 school children studied had undesirably high serum cholesterol levels. The present study was undertaken to determine whether the high cholesterol levels are present at birth and to determine the relationship between cord blood, maternal blood cholesterol and maternal diet. Cord blood from 58 neonates and fasting venous blood form 45 mothers were analyzed for total serum cholesterol. Mothers were interviewed regarding their usual diet during pregnancy. Mean cord blood cholesterol was 56.90 mg/dl (range 26 to 123 mg/dI). Mean maternal blood cholesterol was 232.4mg/dl (range 141-382 mg/dl). Mean maternal …

The purpose of this study was to examine the interrelationships of the person-environment variables of age, sex, length of illness, pain, social support, and physical function as they affected psychological adaptation in dying. An adaptation paradigm of constructs from nursing, illness, and dying provided the theoretical framework which was used to formulate and test a causal model. This study, in addition, had a qualitative component that identified, from the participants' spontaneous responses, reactions to and perceptions of the dying process. A sample of 97 adults was recruited from two metropolitan home hospice programs, with testing occurring in the home. Regression …

The purpose of this study was to describe and explain the impact of demographic variables (age, educational level, time since diagnosis) and subsequent psychosocial variables (perceived stress, ways of coping, professional support, and social support) on adherence and, ultimately, on health status (blood pressure, psychological symptoms, and subjective health) in patients with essential hypertension whose health care was managed by a nurse. Because nurses manage the therapeutic regimens of these patients, it is necessary to identify those factors that influence adherence and thus the health of hypertensive individuals. The purpose of this study was to compare bioelectrical impedance with hydrostatic weighing in lean subjects with varying amounts of fat-free mass and activity levels. The percentage of body fat of each subject was measured by anthropometry; bioelectrical impedance, using two different equations; and hydrostatic weighing (HW) methods. The two equations used for bioelectrical impedance were versions 1.51 (BIA2) and the weight management program (BIA3) from RJL Systems, Inc. HW was used as the criterion measure. The three groups were body builders, non-athletes, and athletes, with 10 subjects in each group. The mean value for age was 20.4 ± 1.8 …

This study utilized an explanatory correlational design to examine the relationship which exists between the predictor variables of adolescent's perception of family adaptation, cohesion, and communication, parental use of substances, adolescent age and gender and the criterion variables of adolescent behavioral intention and self-reported use of alcohol, tobacco, and illicit drugs. The theoretical perspectives of the study, derived from developmental theory and the Circumplex Model of Marital and Family Systems viewed the adolescent stage as a period in which the entire family is challenged to balance levels of adaptation and cohesion in order to facilitate individuation and autonomy of adolescent …

The purpose of this study was to explore the phenomenon of comforting possessions in order to develop descriptive theory based on the perceptions, beliefs, values, symbolic meanings, and feelings of young children regarding comforting possessions. Using ethnographic methods, children with comforting possessions were observed extensively in a preschool setting (n = 10). Two of these children were later interviewed with a parent present. In addition, 17 other children were interviewed in the home or preschool setting with a parent present for a total study sample of 27 children. The targeting of proven anticancer drugs specifically to cancer cells would provide a unique opportunity to restrict neoplasms without damaging the cancer patient. The present research utilizes the phenomenon of illicit transport, i.e. the coupling of normally impermeant metabolites to permeant metabolites, in targeting the drug melphalan to mouse Ehrlich ascites tumor cells. The dipeptide beta-alanyl-melphalan was synthesized and tested in vitro for toxicity towards mouse Ehrlich ascites tumor cells, mouse liver cells, and mouse 3T3 embryonic cells. The parent compound, melphalan, was used as a control treatment. In addition, both melphalan and beta-alanyl-melphalan were utilized in in vivo chemotherapeutic …

Evidence of an association between use of illicit substances and aggressive behavior is pervasive. But the precise causal mechanisms by which aggression is influenced by intoxicants are still not well understood. Research on intoxication and aggression often has overlooked the nonviolent behavior of most substance users, controlled use of substances, and the evidence from other cultures of a weak or nonexistent relation between substance use and aggression. There is only limited evidence that ingestion of substances is a direct, pharmacological cause of aggression. The temporal order of substance use and aggression does not indicate a causal role for intoxicants. Research …

Vision To reduce suffering and death from cancer using information collected by the Louisiana Tumor Registry. History Cancer registration in Louisiana began in 1947 at the Charity Hospital Tumor Registry in New Orleans. In 1974, as part of its Surveillance, Epidemiology and End Results (SEER) Program, the National Cancer Institute provided funds for a population-based cancer incidence and survival registry covering Jefferson, Orleans, and St. Bernard parishes. The purpose of the study was to investigate the effect of continuous passive motion (CPM) after arthroscopy. A CPM machine was used one hour following knee surgery. Statistical analysis was performed to determine differences in knee range of motion (ROM), strength and circumference, at time intervals preoperatively, one, seven, and twenty-eight days following surgery. The analysis of variance revealed no significant difference between the CPM and control groups for any parameter. A significant interaction effect between treatment and time was demonstrated one day after surgery for extension, joint line circumference, and midthigh circumference; and 28 days after surgery for the …

Simian virus 40 (SV40) deletion mutants dlA2459 and dlA2475 express T antigens that lack the normal carboxy terminus. These mutants are called host range/helper function (hr/hf) mutants because they form plaques at 37 degrees C on BSC-1 and Vero monkey kidney cell lines but not on CV-1p monkey kidney cells. Wild-type SV40 can provide a helper function to permit growth of human adenoviruses in monkey kidney cells; the hr/hf mutants cannot. Progeny yields of hr/hf mutants are also cold sensitive in all cell lines tested. Patterns of viral macromolecular synthesis in three cell lines (Vero, BSC-1, and CV-1) at three …

The purpose of this study was to describe the relationship between perceived touch and self-concept in elders. Johnson's Behavioral Model provided the theoretical framework for this descriptive correlational study. Thirty subjects, who were 60 years or older, and lived in either a nursing home, a retirement home, or an independent living center participated in this study. Data were collected using the Touch Questionnaire and the Tennessee Self-Concept Scale and were analyzed using the Pearson Product Moment Correlation. Experiments were performed in normal and Parkinsonian subjects who were asked to maintain a constant finger position using time-delayed visual feedback. The finger position showed complex dynamics, with characteristic differences between normal and Parkinosonian subjects. Here we discuss some of the theoretical issues that must be resolved in order to understand the mechanisms that underlie the generation of these complex signals.

The location of the translational termination codon for the transmembrane protein (TMP) varies in three infectious molecular clones of simian immunodeficiency virus from macaques (SIVmac). The SIVmac251 and SIVmac142 infectious clones have premature stop signals that differ in location by one codon; transfection of these DNAs into human HUT-78 cells yielded virus with a truncated TMP (28 to 30 kilodaltons [kDa]). The SIVmac239 infectious clone does not have a premature stop codon in its TMP-coding region. Transfection of HUT-78 cells with this clone initially yielded virus with a full-length TMP (41 kDa). …

Linker insertion mutants affecting the simian virus 40 (SV40) large tumor (T) antigen were constructed by inserting a 12-base-pair oligonucleotide linker into restriction endonuclease cleavage sites located within the early region of SV40. One mutant, with the insertion at amino acid 5, was viable in CV-1p and BSC-1 cells, indicating that sequences very close to the amino terminus of large T could be altered without affecting the lytic infection cycle of SV40. A method for the qualitative assessment of oxidative drug metabolism deficiencies, preferably for the qualitative assessment of dextromethorphan metabolism, in a subject comprises analyzing a sample fluid from the subject, for example urine, which contains metabolites of a probe drug using high performance thin layer chromatography.
