Changes In Gabaa̳ Receptor Expression And Alterations In Ca⁺⁺/Calmodulin-Dependent Protein Kinase Ii Activity In A Hippocampal Neuronal Culture Model Of Epilepsy, Robert Eagan Blair
Theses and Dissertations
Expression levels of GABAA receptor subunits and activity of the Ca++ /calmodulin-dependent protein kinase II (CaM kinase II) enzyme system were evaluated in an in vitro hippocampal neuronal culture model of epilepsy. Treatment of hippocampal neuronal cultures with Mg++-free media for 3 hours results in the induction of an enduring "epileptic" state as evidenced by the expression of spontaneous recurrent seizure (SRS) discharge. The induction of the SRS activity was shown to be a N-methyl-D-aspartate (NMDA) receptor, Ca++-dependent mechanism (Sombati and DeLorenzo, J Neurophys., 73 (4), 1995). Significant and long-lasting decreases in mRNA expression for the GABAA α2 and α5 …

Exclusion Mapping Of Polycystic Kidney Disease: A Third Locus, Eoin Thompson
Exclusion Mapping Of Polycystic Kidney Disease: A Third Locus, Eoin Thompson
Theses: Doctorates and Masters
The aim of this research was to perform exclusion on a rare form of hereditary autosomal dominant polycystic kidney disease (ADPKD). To-date, two genes for ADPKD have been identified: PKDI which has been localized to the short arm of chromosome 16 and PKD2 which has been localized to the long arm of chromosome 4. However, a small number of families have been reported that have not shown linkage to either of these two loci, thus suggesting the existence of at least one additional locus (PKD3). Two families that are affected with ADPKD and do not show linkage to PKDI or …
Validation Of The Barkemeyer-Callon-Jones Malingering Detection Scale: The Ability Of A Scale Differentiate Simulating Malingers From Controls And Prior Litigants From Those With No Litigation Experience Within A Sample Of Men Who Have All Suffered Chronic Low Back Pain, David Curnow
Theses: Doctorates and Masters
Chronic low back pain costs the community, and several authors have suggested that individuals often attempt to exaggerate chronic low back pain. Currently no reliable and valid scale for assessing malingering in chronic pain populations exists, and there is a large difference in opinion on the ability of experts using clinical judgment to detect malingering. The current study seeks to provide a validation for the BarkemeyerCallon-Jones Malingering Detection Scale (MDS) which has purported to be able to identify individuals attempting to malinger neurological conditions and pain. A simulation design was used, as in previous research, because it is difficult to …
Women's Perceptions Of Birth Centre Care: A Qualitative Approach, Karen Coyle
Women's Perceptions Of Birth Centre Care: A Qualitative Approach, Karen Coyle
Theses: Doctorates and Masters
The purpose of this exploratory study was to describe women's perceptions of the care they received in a birth centre, compared to their previous experiences in a hospital. Australian statistics indicate that five percent of childbearing women now choose to receive care in a birth centre setting. Clinical outcomes of birth centre care are now well documented, but there is limited empirical data about women's experiences of this model of care. Seventeen women, who had recently given birth in a birth centre, and had previously experienced care in a hospital setting, were interviewed about their care experiences. Using content analysis, …

A Descriptive Study Of Non-Accredited Athletic Training Programs, Donald Lee Bishop
A Descriptive Study Of Non-Accredited Athletic Training Programs, Donald Lee Bishop
Dissertations and Theses @ UNI
The purpose of the study was to describe the current status of non-accredited athletic training programs at four-year colleges and universities in relation to receiving future accreditation by the Commission on Accreditation of Allied Health Education Programs (CAAHEP).
Since it was first organized, the National Athletic Trainers' Association, Inc. (NATA) has continually sought to elevate the standards of its members. There were no certification or education requirements for athletic trainers until 1970. These requirements, once very broad and open-ended, have since been refined and delimited to ensure the highest quality for athletic trainers. The NATA has once again raised their …
Eating-Disorder Patterns In The Minnesota Multiphasic Personaity Inventory, Karen Baer-Barkley
Eating-Disorder Patterns In The Minnesota Multiphasic Personaity Inventory, Karen Baer-Barkley
Dissertations
Problem. Early intervention is crucial for effective treatment of eating disorders. While there have recently been many assessment tools developed specifically for eating disorders, a screening tool is still needed. This study sought to identify, in an eating-disordered sample, the existence of a v-shaped pattern on scales 4-5-6 of the MMPI, a commonly used general measure of psychopathology.
Method. The sample consisted of 356 females who were being treated for eating disorders through the HOPE program between 1989 and 1996. Past research on the MMPI has produced equivocal results on identifying significant patterns on the clinical scales. Since prior research …
Predicting The Severity Of Radiation Skin Reactions In Women With Breast Cancer, Davina Porock
Predicting The Severity Of Radiation Skin Reactions In Women With Breast Cancer, Davina Porock
Theses: Doctorates and Masters
Skin reactions are unavoidable side effects of radiotherapy for breast cancer that may limit the amount of treatment a patient is able to receive. As well, the discomfort associated with the treatment may negatively affect the patient's quality of life and their willingness to complete a course of treatment that typically extends over seven weeks. Prior literature suggests that variations in patients' tissue reactions to radiation may be related to Individual patient characteristics. Before health care providers can intervene to prevent or minimise skin reactions, a clinical model that helps predict which patients will experience more skin reactions is needed. …

A Systematic Evaluation Of Methods To Separate X- And Y- Bearing Sperm, Jacquelyn Irving
Theses: Doctorates and Masters
This project was initiated to determine if was possible to enrich either X- or Y- bearing sperm, and hence to preselect the sex of a child. Two of the possible reasons why couple might want to select the sex of a child are firstly because of a family history of an X-linked recessive genetic disorder, which usually only affect sons, and secondly families who have had several children of one sex. For this study, men with three or more children of the same sex were recruited following the publication of an article in The West Australian newspaper. The percentage of …

Alterations In Calcium Homeostasis And The Insulin-Like Growth Factor Signaling Pathway Induced By Carcinogenic Polycyclic And Halogenated Aromatic Hydrocarbons In Human Mammary Epithelial Cells, Stacey L. Tannheimer
Pharmaceutical Sciences ETDs
Breast cancer is a major health concern for women, with only a small percent of the risk factors currently identified. It has been estimated that environmental factors may contribute to up to 80% of breast cancer cases. Many environmental carcinogens, such as polycyclic and halogenated aromatic hydrocarbons (PAHs and HAHs), are proven mammary carcinogens in animal models. Therefore, these studies were conducted to elucidate potential roles of PAHs and HAHs in alterations in known signaling pathways in human mammary epithelial cells (HMEC). Carcinogenic PAHs have previously been shown to produce sustained alterations in the calcium (Ca2') homeostasis of lymphocytes Therefore, …

Mutations In Yeast Proliferating Cell Nuclear Antigen Define Distinct Sites For Interaction With Dna Polymerase Δ And Dna Polymerase Ε, Joel C. Eissenberg, L. Rao Ayyagari, Xavier V. Gomes, Peter M.J. Burgers
Faculty Scholarship
The importance of the interdomain connector loop and of the carboxy-terminal domain of Saccharomyces cerevisiae proliferating cell nuclear antigen (PCNA) for functional interaction with DNA polymerases delta (Poldelta) and epsilon (Pol epsilon) was investigated by site-directed mutagenesis. Two alleles, pol30-79 (IL126,128AA) in the interdomain connector loop and pol30-90 (PK252,253AA) near the carboxy terminus, caused growth defects and elevated sensitivity to DNA-damaging agents. These two mutants also had elevated rates of spontaneous mutations. The mutator phenotype of pol30-90 was due to partially defective mismatch repair in the mutant. In vitro, the mutant PCNAs showed defects in DNA synthesis. Interestingly, the pol30-79 …
Maternal Age: Influence On Length Of Gestation And Birth Weight, Charlotte Bratcher
Maternal Age: Influence On Length Of Gestation And Birth Weight, Charlotte Bratcher
Masters Theses & Specialist Projects
The findings indicated no significant differences among the age groups in relation to infant birth weight nor in relation to the risk factors of smoking, maternal weight gain, trimester of first entry into prenatal care, or marital status They did indicate a significant difference [F(2,85) = 4.1364, p < .05] between the oldest and the youngest group (CD = 3.37, a = .05) in relation to gestational length. These findings are significant for healthcare providers in rural areas, since much of the previous research had been done in highly urbanized or inner city settings.

Characteristics And Resource Utilization Of Patients Of A Proprietary Home Health Agency In Rural South Central Kentucky, Linda Sewell
Masters Theses & Specialist Projects
The home health industry's introduction to managed care raises the prospect of reduced access to health care and poor outcomes for a vulnerable segment of our population—the rural elderly. Before effective intervention strategies can be accurately evaluated, a clearer picture of the sociodemographic features and home care service consumption is needed for this understudied group. The study was intended to provide a basis for future research into the evaluation of alternative methods of delivering effective care in terms of outcome and decreased cost for this population. A retrospective descriptive analysis was made of the patient record for the first six …

Direct Demonstration Of Retroviral Recombination In A Rhesus Monkey, Dawn P. Wooley, Randall A. Smith, Susan Czajak, Ronald C. Desrosiers
Neuroscience, Cell Biology & Physiology Faculty Publications
Recombination may be an important mechanism for increasing variation in retroviral populations. Retroviral recombination has been demonstrated in tissue culture systems by artificially creating doubly infected cells. Evidence for retroviral recombination in vivo is indirect and is based principally on the identification of apparently mosaic human immunodeficiency virus type 1 genomes from phylogenetic analyses of viral sequences. We infected a rhesus monkey with two different molecularly cloned strains of simian immunodeficiency virus. One strain of virus had a deletion in vpx and vpr, and the other strain had a deletion in nef. Each strain on its own induced low virus …
