A radiation hybrid (RH)-derived physical map of 25 markers on the feline X chromosome (including 19 Type I coding loci and 6 Type II microsatellite markers) was compared to homologous marker order on the human and mouse X chromosome maps. Complete conservation of synteny and marker order was observed between feline and human X chromosomes, whereas the same markers identified a minimum of seven rearranged syntenic segments between mouse and cat/human X chromosome marker order. Within the blocks, the feline, human, and mouse marker order was strongly conserved. Similarly, Y chromosome locus order was remarkably conserved between cat and human …

Long term control of high-grade brain tumors is rarely achieved with current therapeutic regimens. This study sought to determine if pretreatment with plasmid DNA expressing Tumor Necrosis Factor-α (TNF-α) could enhance the effect of ionizing radiation in an aggressive, rapidly growing glioma tumor (C6) model. Plasmid DNA is an attractive vector system for delivery of foreign genes into eukaryotic cells due to its advantages including low immunogenicity in vivo, lesser toxicity and relatively low cost. We have constructed a plasmid-based mammalian expression vector (pGL1-TNF) to secrete human TNF-α from cancer cells. Plasmid DNA was successfully transfected into C6 cells …

With the advancement of medical procedures, heart transplantation has become a viable alternative for infants born with congenital or acquired heart disease. Although these children are thought to experience much improvement in their overall physical functioning post-transplantation, the long-term psychological functioning of these children and their families is currently unknown. This study examines the long-term family functioning and psychosocial development of infant heart transplant recipients in comparison to children with congenital heart disease (CHD) and non-clinical control children. Development of the eye is controlled by a network of genes, often conserved, that regulate the timing and location of cellular differentiation. One approach to understanding this network of genes and their interaction is to focus on mutations, spontaneous or induced, that predictably disrupt the proper function of such networks, and by examining the effect of such disruption on the function of other genes.

Teens in the United States have the highest pregnancy, abortion, and sexually transmitted disease rates among the developed countries. The social and financial costs of teen births are staggering. Research shows that high self-esteem is significantly related to resistance to group pressures, willingness to express unpopular opinions, family adjustment, creativity, and effective communication between parents and youth. Spinal cord injury (SCI) patients confined to a wheelchair for long periods are prone to pressure sores occurring at ischial tuberosity. This study used Expert-Seat pressure evaluation pad to record the pressure of the ischial tuberosities. Twenty-nine spinal cord injury subjects and twenty normal subjects participated in this study. The pressure distributed over ischial tuberosities during sitting was measured five times, including one measured without cushion. Four wheelchair cushions with different types of materials were evaluated in two population groups: spinal cord injured and normal control. We utilized the mouse mammary tumor virus (MMTV) long terminal repeat (LTR) in vivo to understand how the interaction of the glucocorticoid receptor (GR) with a nucleosome-assembled promoter allows access of factors required for the transition from a repressed promoter to a derepressed, transcriptionally competent promoter. A mutation (C644G) in the ligand binding domain (LBD) of the mouse GR has provided information regarding the steps required in the derepression/activation process and in the functional significance of the two major transcriptional activation domains, AF1 and AF2. Perceptions Of Couple Functioning Among Female Survivors Of Child Sexual Abuse, David Dilillo, Patricia J. Long

A sample of 51 college women retrospectively reporting a history of childhood sexual abuse and 91 women failing to report such a history was examined in order to investigate the relationship between victimization history and survivors' self-reports of functioning in adult intimate relationships. Specifically, relationship satisfaction, communication, and trust were examined in heterosexual relationships of at least six months' duration. As hypothesized, even when demographic differences between groups were controlled, survivors reported significantly less relationship satisfaction, poorer communication, and lower levels of trust in their partners than did women with no history of sexual abuse. Qualitative transcriptional regulatory sequences functional in plants, plant tissue and in plant cells for inducible gene expression and quantitative transcriptional regulatory sequences for increasing the transcriptional expression of downstream genetic information in plants, plant tissue and plant cells are disclosed. Also disclosed are methods and recombinant DNA molecules for improving the disease resistance of transgenic plants, especially wherein an inducible promoter controls the expression of a protein capable of evoking the hypersensitive response in a plant.

Why Are Girls Less Physically Aggressive Than Boys? Personality And Parenting Mediators Of Physical Aggression, Gustavo Carlo, Marcela Raffaelli, Deborah J. Laible, Kathryn A. Meyer

The primary goal of the present analysis was to determine whether the commonly observed gender difference in physical aggression could be accounted for by gender differences in selected personality and social contextual factors. Eighty-nine adolescents (M age = 16.0; 52% female; 53% European-Americans, 38% Latinos) completed self-report measures, including sympathy (empathic concern and perspective taking) and parental involvement (support and monitoring). Mediation analyses revealed that relatively high levels of both empathic concern and parental monitoring accounted for relatively low levels of physical aggression. In addition, sympathy (for males) and parental involvement (males and females) were negatively related to physical aggression. …

This study investigated whether parent-teen disagreement in reports of sexual discussions is due to methodological factors. Forty-four mothers and their 12- to 15-yearold daughters (n = 18) and sons (n = 26) completed parallel questionnaires including measures of communication about 18 different sexual topics during the teen’s entire life and during the past year. Analyses examined whether congruence rates were infl uenced by the use of global as compared to specifi c items, assessment of conversations during the teen’s entire life as compared to the past year, and use of forced-choice as opposed to continuous-response categories. An ultrasonic imaging system includes a receive beam former that generates analog receive signals and a scan converter. A receive signal processing path interconnects the receive beamformer and the scan converter, and this processing path includes both an A/D converter characterized by a selectable sampling rate and at least one filter characterized by at least one filter parameter. The filter parameter is selected as a function of the sampling rate to provide enhanced image quality.

The present invention relates to monoclonal antibody 1A7. This is an anti-idiotype produced by immunizing with an antibody specific for ganglioside GD2, and identifying a hybridoma secreting antibody with immunogenic potential in a multi-step screening process. Also disclosed are polynucleotide and polypeptide derivatives based on 1A7, including single chain variable region molecules and fusion proteins, and various pharmaceutical compositions. When administered to an individual, the 1A7 antibody overcomes immune tolerance and induces an immune response against GD2, which comprises a combination of anti-GD2 antibody and GD2-specific T cells. This study examined the tracking of selected measures of physical activity, inactivity, and fitness in a cohort of rural youth. Students (N=181, 54.7% female, 63.5% African American) completed test batteries during their fifth- (age=10.7±0.7 years), sixth-, and seventh-grade years. The Previous Day Physical Activity Recall (PDPAR) was used to assess 30-min blocks of vigorous physical activity (VPA), moderate-to-vigorous physical activity (MVPA), TV watching and other sedentary activities, and estimated energy expenditure (EE). Fitness measures included the PWC 170 cycle ergometer test, strength tests, triceps skinfold thickness, and BMI. Intraclass correlation coefficients (ICCs) for VPA, MVPA, and after-school EE …

Association of a targeting ligand such as transferrin, or an endosome disrupting peptide such as GALA, with cationic liposome-DNA complexes ('lipoplexes') results in a significant enhancement of transfection of several cell types. Although these strategies can overcome some of the barriers to gene delivery by lipoplexes, the mechanisms by which they actually enhance transfection is not known. In studies designed to establish the targeting specificity of transferrin, we found that apo-transferrin enhances transfection to the same extent as transferrin, indicating that internalization of the lipoplexes is mostly independent of transferrin receptors. Consistency And Development Of Prosocial Dispositions: A Longitudinal Study, Nancy Eisenberg, Ivanna K. Guthrie, Bridget C. Murphy, Stephanie A. Shepard, Amanda Cumberland, Gustavo Carlo

The issue of whether there is consistency in prosocial dispositions was examined with a longitudinal data set extending from ages 4 to 5 years into early adulthood (N = 32). Spontaneous prosocial behaviors observed in the preschool classroom predicted actual prosocial behavior, other- and self-reported prosocial behavior, self-reported sympathy, and perspective taking in childhood to early adulthood. Prosocial behaviors that were not expected to refl ect an other-orientation (i.e., low cost helping and compliant prosocial behavior) generally did not predict later prosocial behavior or sympathy. Sympathy appeared to partially mediate the relation of early spontaneous sharing to later prosocial dispositions. …

Monolayer cultures of lapine articular chondrocytes were transduced with first-generation adenoviral vectors carrying lacZ or transforming growth factor β1 genes under the transcriptional control of the human cytomegalovirus early promoter. High concentrations of transforming growth factor β1 were produced by chondrocytes following transfer of the transforming growth factor β1 gene but not the lacZ gene. Transduced chondrocytes responded to the elevated endogenous production of transforming growth factor β1 by increasing their synthesis of proteoglycan, collagen, and noncollagenous proteins in a dose-dependent fashion. The increases in collagen synthesis were not accompanied by alterations in the collagen phenotype; …

As follow-up to the NHPF's February 1999 session entitled "Biological Terrorism: Is the Health Care Community Prepared?" the Forum organized a site visit on October 4 and 5, 1999, to Baltimore and Fort Detrick, Maryland. The site visit provided federal congressional and agency staff with the opportunity to learn first-hand how one local area is preparing for the possibility of a bioterrorist incident. Bovine Herpesvirus 1 Can Infect Cd4+ T Lymphocytes And Induce Programmed Cell Death During Acute Infection Of Cattle, M. T. C. Winkler, Alan R. Doster, Clinton J. Jones

Acute infection of cattle with bovine herpesvirus 1 (BHV-1) represses cell-mediated immunity, which can lead to secondary bacterial infections. Since BHV-1 can induce apoptosis of cultured lymphocytes, we hypothesized that these virus-host interactions occur in cattle. To test this hypothesis, we analyzed lymph nodes and peripheral blood mononuclear cells (PBMC) after calves were infected with BHV-1. In situ terminal deoxynucleotidyltransferase- mediated dUTP nick end-labeling (TUNEL) staining of lymphoid tissues (pharyngeal tonsil, cervical, retropharyngeal, and inguinal) was used to detect apoptotic cells. Calves infected with BHV-1 for 7 days revealed increased apoptotic cells near the corticomedullary junction in lymphoid follicles and …
