Patients with recurrent/metastatic (r/m) head and neck squamous cell carcinoma (HNSCC) have limited treatment options and a dismal prognosis, especially when their cancer is resistant to standard treatments like anti-programmed cell death protein 1 and platinum-based therapies. Petosemtamab - a human, common light chain, bispecific antibody with enhanced antibody-dependent cellular cytotoxicity targeting epidermal growth factor receptor (EGFR) and leucine-rich repeat-containing G-protein coupled receptor 5 (LGR5) - demonstrated antitumor activity in r/m HNSCC. In many tumor types, including HNSCC, EGFR is an oncogenic driver, while LGR5 is upregulated. LGR5 can potentiate the wingless-type integration site (WNT)/β-catenin signaling pathway in response to …

Single-cell multiomic techniques have sparked immense interest in developing a comprehensive multi-modal map of diverse neuronal cell types and their brain-wide projections. However, investigating the complex wiring diagram, spatial organization, transcriptional, and epigenetic landscapes of brain-wide projection neurons is hampered by the lack of efficient and easily adoptable tools. Here we introduce Projection-TAGs, a retrograde AAV platform that allows multiplex tagging of projection neurons using RNA barcodes. By using Projection-TAGs, we performed multiplex projection tracing of the cortex and high-throughput single-cell profiling of the transcriptional and epigenetic landscapes of the cortical projection neurons in female mice. BACKGROUND: EpCAM (epithelial cell adhesion molecule) is a key regulator of epithelial cell-cell adhesion, signal transduction, tissue regeneration, and serves as a stem cell marker. It is frequently overexpressed in epithelial cancers and is linked to tumor progression, survival, and metastasis. However, the functional impact of EpCAM mutations in cancer remains poorly understood.
METHODS: To investigate the role of EpCAM mutations, we performed a comprehensive analysis of cancer cohorts from multiple genomic datasets, identifying novel somatic EpCAM mutations across diverse epithelial cancers. Barth syndrome (BTHS) is a rare X-linked recessive disorder characterized by mutations in the TAFAZZIN gene, leading to mitochondrial dysfunction, cardioskeletal myopathy, neutropenia, exercise intolerance, and growth delay. While cardiac transplantation can improve heart function in BTHS patients, the metabolic effects of this procedure remain poorly understood. BACKGROUND: Development of readily available Rapid COVID-19 Antigen tests essential for promptly identifying SARS-CoV2 infection and preventing its spread.
METHODS: This study evaluated the clinical performance of the Healgen Rapid COVID-19 antigen test as a point-of-care diagnostic tool with 806 evaluable subjects who were within 6 days post-symptom onset. The results from the Healgen test were compared to the results from Emergency Use Authorization (EUA) approved SARS-CoV-2 RT-PCR tests.
RESULTS: Out of the 806 evaluable subjects, 140 tested positive and 640 tested negative for SARS-CoV-2 with the Healgen COVID-19 test, showing good agreement with the EUA RT-PCR results. There were …

The basis and impact of functional asymmetries in the brain, particularly language lateralization, are not fully understood, and the relationship between functional and structural asymmetries remains largely untested. This study investigated the degree to which asymmetries in hemispheric language laterality are concordant with asymmetries in gray matter (GM) structure and whether the hemispheric organization of memory is influenced by functional language asymmetries. Although much IPE research focuses on competence as an outcome, little research examines learner confidence in competence, which is likely to translate didactic knowledge into application of clinical skills. Background: Pulmonary emboli (PE) have a significant incident rate of 112.3 per 100,000 adults. When age-weighted, the population over 75 has an incident rate of 1 out of 100. 90% of deaths from acute cases occur within two hours of the onset of symptoms. In pregnancy-related deaths, 20% are attributed to PE, and pregnancy increases the risk of the development of PE by 400 – 600%. Significant risk factors for PE include COPD, obesity, recent trauma or surgery, a history of deep vein thrombosis (DVT), a history of hormonal therapy, smoking, and pregnancy. Immune checkpoint inhibitors, such as nivolumab, have become integral in treating metastatic malignancies by enhancing immune responses against tumors. However, they are associated with immune-related adverse events (irAEs), including cutaneous reactions. We report a case of persistent lichenoid dermatitis during therapy and eruptive keratoacanthomas (KAs) following the cessation of nivolumab.

A 77-year-old Caucasian man presented with a subcutaneous melanoma on the left lateral chest, treated with excision followed by adjuvant nivolumab. Migraine affects about 1 billion people worldwide and is associated with a significant negative impact on personal and occupational functioning. Migraine accompanied by aura is seen in about 15% of migraineurs. Visual aura is the most common type of aura. Much less common though are auras that involve higher mental function, such as confusional state. The confusional state may manifest with a wide variety of cortical dysfunction, such as speech impairment, increased alertness, agitation, and amnesia, often lasting longer than a typical migraine aura.

Sarcoidosis is a systemic immune disease with an unknown trigger, identified histologically by non-caseating granulomas. Neurosarcoidosis is a subset in which granulomatous inflammation infiltrates the central or peripheral nervous system. Sarcoid myelopathy affects a very small fraction of sarcoidosis patients. The present study provides the characterization of the phytochemical content and the antibacterial activity of ethanolic extracts from the leaves (LM) and stems (SM) of Micromeria barbata (M.barbata) against Pseudomonas aeruginosa . Important functional groups were determined by analyzing the FTIR spectra of LM and SM. The phytochemical profiles were analyzed by GC-MS, and these characterized the chemicals according to retention periods and peak regions. Despite preventative efforts, malarial pregnancies still cause birth complications, therefore emphasizing the necessity to study these adverse outcomes in malarial pregnancy. Malaria in pregnancy can lead to placental malaria (PM), which is the presence and accumulation of Plasmodium-infected red blood cells in the placental intervillous space (IVS). The growing incidence of overweight and obesity in young adults has been closely associated with unhealthy lifestyle habits, particularly the high intake of sugar-sweetened beverages (SSBs) and low levels of physical activity. College students are especially at risk due to lifestyle changes that occur during their university experience. BACKGROUND: Hospitalized older adults with cognitive impairment (CI) have an increased risk for adverse outcomes. Acute Care for Elders (ACE) Units were developed to reduce functional decline in hospitalized older adults. INTERVENTION: We developed a quality improvement (QI) initiative to disseminate ACE care processes to non-ACE units, converting them into "Virtual ACE" units. The aim of this evaluation is to examine the impact of this intervention on the mobilization of patients with CI. Stress is a physiological state of altered neuroendocrine signaling and heightened indices of behavioral vigilance and arousal in response to threatening stimuli. This project aimed to investigate the adaptations to stress that occur in the noradrenergic locus coeruleus (LC) and how these changes affect stress coping behavior. We began by investigating how adolescent restraint and predator odor stress affects coping when presented with a secondary noxious stimulus in the defensive shock probe burying task one week following stressor exposure, finding that stressed male rats demonstrate a more active coping response and stressed female rats demonstrate a more passive coping response, …

Abstract
Systemic Sclerosis (SSc) is a rare autoimmune disease characterized by multiple-organ involvement and notably by fibrosis. Current therapies for SSc focus on symptom managements and treating specific organ related complications. However, heterogeneous nature of the disease further complicates treatment responses and hinders the development of new therapies. Therefore, there is a critical unmet need for the development of patient-specific therapeutics.
The objectives of this thesis were: (1) to evaluate the therapeutic potential of Bardoxolone Methyl (CDDO-Me) in both SSc fibroblasts and SSc macrophages. To achieve these goals, I utilized both wet-lab analysis tools and computational analysis.

It is anticipated that between 60 and 80 percent of healthy, full-term newborns would exhibit idiopathic neonatal jaundice. Mode of delivery has recently been associated with idiopathic neonatal jaundice. As seen at Melati Husada Women and Children Hospital in Malang, the increasing number of caesarean section was followed by increased frequency of neonatal hyperbilirubinemia. The widespread use of Visual Display Terminals (VDTs) is associated with visual and musculoskeletal complaints, particularly Asthenopia, which may reduce productivity. A 32-year-old female financial worker presented with eye discomfort, including sore eyes, blurred vision, dizziness, and twitching on her left temple, particularly during extended computer use. The symptoms were relieved with rest. Most VDTs now use Light Emitting Diodes, which emit more blue light than LCDs and may cause eye fatigue and disrupt sleep. This research is a narrative literature review, in which 3,610 articles were initially identified and then filtered based on inclusion-exclusion criteria, resulting in 8 articles.
